Eco-Friendly Amylase Production and Immobilization on Macadamia-Based Carbon Using Aspergillus niger

This study demonstrates the valorization of macadamia nutshells,a lignocellulosic agricultural waste,as both a carbon source for amylase production and a support matrix for enzyme immobilization.Under optimized solid-state fermentation conditions,Aspergillus niger ICP2 synthesized amylase with a peak activity of 0.312 U/mL after 72 hours.A four-step purification process of the crude enzyme extract resulted in a 188.54-fold increase in specific activity,albeit with a final recovery yield of 0.0031%.In parallel,nutshells were carbonized at 600℃,700℃,and 800℃,then chemically activated with ZnCl_(2).The carbon derived at 700℃ exhibited superior physicochemical characteristics,including enhanced porosity and increased availability of functional groups,which enabled effective enzyme adsorption,improved catalytic performance,and enhanced reusability.Immobilized amylase on this support retained approximately 30% of its initial activity after five hydrolysis cycles,demonstrating moderate operational reusability and potential for repeated use in bioprocesses.In contrast,carbon materials from 600℃ and 800℃ showed lower stability and enzyme performance.These findings highlight the critical role of carbonization conditions in designing effective immobilization matrices and underscore the potential of macadamia nutshells as a renewable and sustainable resource for biocatalyst development.This“biowaste-to-biocatalyst”strategy exemplifies a circular bioeconomy model with implications for green chemistry,industrial biocatalysis,and environmental sustainability. 展开更多

Vitamin C,a potent antioxidant with broad therapeutic applications,is limited by rapid degradation under environmental stressors,which compromises its stability and bioactivity.This study addresses these limitations by formulating a double nano-emulsion(W/O/W)system incorporating macadamia oil and tea tree oil,using homogenization and phase inversion temperature(PIT)techniques.Comprehensive physicochemical charac-terization,including droplet size,polydispersity index(PDI),zeta potential,turbidity,Fourier transform infrared spectroscopy(FTIR),and SEM,was conducted alongside stability assessments under varying pH,temperature,and storage conditions.The optimized nano-emulsions exhibited nanoscale droplet sizes(10-40 nm),low PDI values(indicating high uniformity),and robust stability.Interestingly,the formulation with 2%W/O loading,with a particle size of 11.57 nm and a PDI of 0.04,demonstrated an antioxidant capacity of 4622.62μg ascorbic acid equivalents(AA)/g,which was significantly higher(p<0.05)compared to both natural oils(macadamia oil:20.91μg AA/g,tea tree oil:16.86μg AA/g)and a 10%Vitamin C aqueous solution(592.94μg AA/g).FTIR analysis confirmed the molecular integrity of Vitamin C and its successful encapsulation with macadamia and tea tree oils,while SEM images revealed uniformly spherical and well-dispersed droplets.Moreover,the formulation retained its structural integrity and antioxidant functionality under diverse pH and thermal conditions.These findings underscore the potential of double nano-emulsion systems to overcome the stability challenges of Vitamin C,offering a promising approach to enhance its bioavailability and therapeutic performance in phar-maceutical and cosmetic applications. 展开更多

Oleic acid (18:1) was the main monounsaturated fatty acid (MUFA) (51.3%), followed by palmitoleic acid (16:1, 22.6%). The content of polyunsaturated fatty acids (PUFAs), C18:2 and C18:3 represented 5.4%. Thus, MUFAs and PUFAs together constituted more than 80% of the total fatty acids present. Trans-vaccenic acid was also present (3%). As regards to other phytochemical compounds, tocopherols and tocotrienols were not found in the sample, but the presence of squalene was detected.The antioxidant activity (44.2%) of the extract was produced by the phytochemicals present. 展开更多

On the 15th of every month in 2011, 1 leaf in the second round from top-branch in 4 directions of the 20 trees was col ected. The content of 9 mineral elements of N, P, K, Ca, Mg, Zn, Cu, Fe and Mn were deter-mined. [Result] The results showed that the N, P, K content in leaves reached a peak in April, then N content decreased slowly, P content stayed stable, and K con-tent increased slightly; Ca, Fe content decreased in April to different degrees and then increased slowly. Mg content was consistent al year round, meanwhile Mn content decreased slightly. Cu and Zn content reached a peak in April, and then presented a rise-fal trend. In Kenya, the crop is predominantly grown by smallholder farmers mainly for export markets. Macadamia trees also enhance agro-forestry conservation, in the East African farmlands, by providing habitat and floral resources to beneficial arthropods such as insect pollinators. Allogamy of macadamia flowers is largely dependent on insects that pollinate and consequently influence the nut set, retention and yield. However, there is limited information on macadamia insect flower visitors in Kenya. This article assessed the diversity of insects that forage flowers of macadamia crop, and further evaluated their temporal distribution, in Murang’a county, central Kenya. Insect flower visitors were sampled weekly using a butterfly net for twelve consecutive months, from January 2021 to December 2021. Sixty-one insect species were recorded foraging macadamia flowers in Murang’a county, central Kenya. There was a statistical difference in the Simpson diversity index among weather seasons, dry, long rain, cold and short rain (p Apis mellifera), a key pollinator species, being the most abundant flower visitor. This study provides strong baseline information, to scientists and farmers, on probable macadamia pollinator species, in central Kenya. . 展开更多

The use of natural fibers as an additive in polymeric matrices has attracted interest of the automotive industries,for its low cost,mechanical properties,biodegradability and lightness.However,the hydrophilic nature of the fiber makes polymer compatibility difficult.Fiber surface treatments can be used to enhance the fiber/matrix interface.In the present work,polypropylene(PP)composites reinforced with fibers from macadamia nutshell were obtained and characterized.Macadamia nutshell fibers were treated by an alkaline treatment with sodium hydroxide(NaOH 4%)to improve adhesion between fibers and matrix.Fibers were characterized by techniques of Scanning Electron Microscopy(SEM)and X-Ray Diffraction(XRD).The PP was mixed with the macadamia fibers(treated and untreated),in proportions of 5 and 10%(wt/wt)using a thermokinetic mixer.Furthermore tensile,flexural and impact specimens(Pure PP and composites)were prepared by an injection molding process and tested for evaluation of mechanical properties.The results showed that the insertion of treated fibers in the PP matrix increased the stiffness of the composites.However,the composites reinforced with untreated fibers presented higher impact energy absorption when compared to composites reinforced with treated fibers. 展开更多

A greenhouse experiment was conducted at Mahidol University, Thailand from January to May 2009. Macadamia plants were planted in pots with mixed substrate of soil: sand: coconut-coir: rice-husk: charcoal; 3.0: 1.0: 0.5: 1.0, respectively. Micro drip-irrigation system was operated 10:00 to 16:00 h daily with a controller scheduled to pump 12 minutes at 12 intervals. The study showed that application of Enshi tablet (18 tablets/plant/year) in a drip-irrigation system enhances growth of Macadamia compared to the application of OsmocoteTM tablet, Modified-enshi and Enshi solution. Significant variation in chlorophyll (a + b) content was observed in Enshi tablet treatment. Soil chemical analyses indicated that the nutrient content in Enshi tablet appeared low compared to other formulations but nutrient removal was considerably brought down by the system, which gives a balance between nutrient removals by crops and addition through fertilizers. Hence, application of Enshi tablet is suggested for Macadamia in a drip-irrigation system. 展开更多

In the present context, the objective of this study was to synthesize and analyze the content of AA of macadamia protein and the impact of hydrogen ion concentration (pH) on AA composition. The determination of AA mainly by cation-exchange chromatography was also investigated. Reproducible and reliable techniques for quantification and identification of AA usually require derivatization. However, techniques such as AA analyzer are composed of cation-exchange chromatography and other components can sideline the derivatization with significant accuracy. The present analysis revealed a higher concentration of essential amino acids especially acidic AA, Glu and Asp and basic AA, Arg than other AA in macadamia protein. The study constitutes first report of use of bubble chart for evaluation of AA and explaination of AAS. The results may elaborate that the degradation of AA of macadamia protein for extraction of pH 11 is caused by the impact of pH. Moreover, the nutritional values of AA present in macadamia protein could change for the better by adjusting pH of extraction. 展开更多

The relative antioxidant capacity was investigated through some in vitro models such as scavenging activity of DPPH radical, scavenging of ABTS<sup>+</sup> radical and evaluation of the total antioxidant capacity assay. Macadamia protein was characterized by methods of DEAE cellulose cation-exchange chromatography and SDS-PAGE. The aim of this study was to apply the existing techniques that enable examination ofmacadamia kernels to provide a better understanding of physico-chemical properties of kernels during postharvest processing. These techniques, such as X-ray tomography, could be applied for quality monitoring in the macadamia industry. The objectives of this study were to investigate the browning centre symptoms that usually occur in macadamia nuts-in-shell. The applied techniques included confocal microscopy, X-ray tomography and magnetic resonance imaging (MRI). Five different varieties of macadamia nuts (A38, 246, 816, 842 and Daddow) were selected to include distinct characteristics, such as drop pattern and growing location. Analysis of the microstructure of kernels by confocal microscopy showed the distribution of possible brown pigment compounds as well as the distribution of lipids, carbohydrates and proteins inside macadamia cells. Physical properties data, including shell density and seed to volume ratio, were obtained by X-ray tomography. Magnetic resonance diffusion tensor imaging used in this study showed marked differences in microstructure which indicate that different varieties exhibit different microstructures expressed as fraction ofanisotropy and apparent diffusion coefficient that appear to be related to the occurrence of the brown centre. Hence, the findings of this study have potential to improve the existing postharvest techniques used in the macadamia processing industry. They will be of benefit to the industry in terms of improved quality control and cost reduction. 展开更多

CCCH型锌指蛋白在植物生长发育及响应逆境胁迫中发挥重要的调控作用。本研究以澳洲坚果JW和桂热1号品种为试验材料,采用RT-PCR技术获得澳洲坚果锌指蛋白基因MiZFP11,分析其编码蛋白的结构特征和亚细胞定位,并利用实时荧光定量PCR技术分析其在不同组织中以及干旱、高温胁迫下的表达水平。RT-PCR克隆结果表明,克隆获得的澳洲坚果锌指蛋白基因MiZFP11(GenBank注册号为MT332641),与CCCH型锌指蛋白基因较高同源。生物信息学分析表明,澳洲坚果MiZFP11蛋白相对保守的C端含有LCCL结构域,属于C3H13亚类锌指蛋白。蛋白基本性质分析表明,MiZFP11蛋白是不稳定的亲水蛋白和非分泌非跨膜蛋白,且以丝氨酸磷酸化修饰为主。蛋白高级结构分析表明,MiZFP11蛋白的二级和三级结构主要由无规则卷曲、α-螺旋和延伸链交错组成。烟草叶片的瞬时表达和共聚焦显微分析显示,MiZFP11蛋白定位于细胞核,与亚细胞定位预测结果一致。实时荧光定量PCR分析表明,澳洲坚果JW和桂热1号品种的MiZFP11基因在叶片中的表达量最高,在根、小花和小果中也有较高的表达量,在茎中的表达量则最低。干旱胁迫1~36h,叶片中的MiZFP11基因在JW品种中表现出“升-降-升-降”的表达模式,在桂热1号品种中表现出“升-降”的表达模式,2个品种整体上均受诱导上调表达。高温胁迫1~36h,叶片中的MiZFP11基因在JW和桂热1号品种中均表现出“升-降”的上调表达模式,且JW品种的表达量明显高于桂热1号品种。因此,MiZFP11基因在不同组织中均有显著差异表达,且受干旱和高温胁迫诱导后显著上调表达,推测MiZFP11基因在澳洲坚果组织生长发育,尤其在响应干旱和高温胁迫中起重要作用。 展开更多

该研究以澳洲坚果青皮多酚为原料,采用生物法绿色合成纳米银(Silver Nanoparticles,AgNPs)研究澳洲坚果青皮多酚质量浓度以及反应时间对AgNPs合成的影响,同时利用傅里叶变换红外光谱仪、纳米粒度及Zeta电位分析仪、透射电子显微镜等现... 该研究以澳洲坚果青皮多酚为原料,采用生物法绿色合成纳米银(Silver Nanoparticles,AgNPs)研究澳洲坚果青皮多酚质量浓度以及反应时间对AgNPs合成的影响,同时利用傅里叶变换红外光谱仪、纳米粒度及Zeta电位分析仪、透射电子显微镜等现代仪器解析其结构特性。结果表明,澳洲坚果青皮多酚纳米银绿色合成条件为澳洲坚果青皮多酚质量浓度2.0 mg/mL、AgNO_(3)溶液浓度0.1 mmol/L、反应时间48 h、多酚溶液与AgNO_(3)溶液体积比1:9;傅里叶变换红外光谱图谱表明,苯环上的-OH参与了AgNPs的合成;纳米粒度及Zeta电位分析结果表明,合成的AgNPs水合粒径介于70~130 nm之间,粒径小,分散性良好,且带负电荷;透射电子显微镜及能谱仪图像显示,合成的AgNPs形貌近似球形,Ag元素周围紧密包裹着C、O元素,Ag元素占比为62.31%。综上,澳洲坚果青皮多酚可作为绿色合成AgNPs的天然还原剂和稳定剂,合成的AgNPs分散性良好,为澳洲坚果青皮的综合利用提供新思路。 展开更多

为高值化利用碎澳洲坚果仁制备坚果酱,以烘烤温度、烘烤时间、研磨时间为考察因素,采用模糊数学感官评价法结合正交试验优化坚果酱的制备工艺,并比较8种不同品种原料的坚果酱在外观品质(色泽)、食用涂抹性品质(黏度)和食用营养品质(脂肪... 为高值化利用碎澳洲坚果仁制备坚果酱,以烘烤温度、烘烤时间、研磨时间为考察因素,采用模糊数学感官评价法结合正交试验优化坚果酱的制备工艺,并比较8种不同品种原料的坚果酱在外观品质(色泽)、食用涂抹性品质(黏度)和食用营养品质(脂肪酸)上的差异。结果表明,澳洲坚果酱的最优工艺为烘烤温度130℃、烘烤时间24 min、研磨时间9.5 min,其模糊数学感官评分最高(85.41分)。澳洲坚果酱呈棕黄色,具有高黏度(1813~5753 mPa·s)、高油酸(56.566%~64.178%)、低动脉粥样硬化指数值(0.105~0.148),是一种适宜涂抹的可食化健康食品。8种坚果酱的色泽具有显著性差异(P<0.05),品种791坚果酱的脂肪酸组成更优,其单不饱和脂肪酸、多不饱和脂肪酸的相对含量均最高且动脉粥样硬化指数值最低,分别为84.626%、2.941%、0.105。该研究结果为澳洲坚果酱的制备和品种的选择提供了基础参考。 展开更多

夏威夷果输送过程离散元参数标定

为优化夏威夷果开口机的设计,对夏威夷果在机器中的输送过程进行离散元仿真分析,提取夏威夷果本征参数并搭建试验台验证离散元参数标定的可靠性。通过单轴压缩试验得到夏威夷果的泊松比为0.29,弹性模量为50.1 MPa,密度为1.019 g/cm^(3)... 为优化夏威夷果开口机的设计,对夏威夷果在机器中的输送过程进行离散元仿真分析,提取夏威夷果本征参数并搭建试验台验证离散元参数标定的可靠性。通过单轴压缩试验得到夏威夷果的泊松比为0.29,弹性模量为50.1 MPa,密度为1.019 g/cm^(3)。采用Central Composite Design得出堆积角最佳组合方案和最佳计算方案,标定出夏威夷果间的碰撞恢复系数为0.502,夏威夷果间的滚动摩擦因数为0.03,夏威夷果间的静摩擦因数为0.318,夏威夷果—硬铝合金板的碰撞恢复系数为0.759,夏威夷果—硬铝合金板的静摩擦因数为0.502,夏威夷果—硬铝合金板的滚动摩擦因数为0.00177。对标定后的参数进行圆筒提升与输送试验验证,结果表明,标定后的夏威夷果仿真堆积角与实测堆积角误差小于1%,仿真条件下颗粒分布情况与实际试验的夏威夷果分布情况基本相同,标定结果可靠,可为夏威夷果相关机具的设计优化提供理论依据。 展开更多

澳洲坚果核心种质的初步构建

构建澳洲坚果的核心种质是提升澳洲坚果种质资源利用效率的关键策略。本研究基于208份澳洲坚果种质资源的全基因组重测序数据,利用Core Hunter软件和基因覆盖度指标,构建了一套核心种质。通过遗传多样性、等位基因和主成分分析等方法对... 构建澳洲坚果的核心种质是提升澳洲坚果种质资源利用效率的关键策略。本研究基于208份澳洲坚果种质资源的全基因组重测序数据,利用Core Hunter软件和基因覆盖度指标,构建了一套核心种质。通过遗传多样性、等位基因和主成分分析等方法对构建的核心种质进行综合评估,共鉴定出5470885个SNP标记,并以此为基础进行分析。最终筛选出41份种质作为核心种质,其中美国种质11份(26.83%),澳大利亚种质12份(29.27%),中国种质18份(43.90%)。在18份国内种质中,包含了经实生选种(11份)、杂交育种(5份)和诱变育种(2份)途径获取的种质。核心种质与原始种质的平均观测杂合度(分别为0.193和0.202)、平均期望杂合度(分别为0.311和0.274)、平均Nei遗传多样性(分别为0.315和0.275)、平均香浓维纳指数(分别为0.481和0.434)、平均多态信息含量(分别为0.256和0.228)均显示出较高的一致性。此外,10种基因型在核心种质和原始种质中的比例几乎一致,次等位基因的分布差异很小。另外,核心种质的主成分分布图和原始种质的分布图的趋势相吻合。本研究构建的核心种质能够有效代表原始种质的遗传变异,为澳洲坚果种质资源的高效利用提供了科学依据。 展开更多
