胸部创伤患者血清Sestrin 2、OSM、TLR9的表达及其与细菌感染的相关性

目的探讨胸部创伤患者血清Sestrin 2、抑瘤素M(OSM)、Toll样受体9(TLR9)的表达水平及其与细菌感染的相关性。方法选取2022年7月至2024年7月河北医科大学第一医院收治的148例胸部创伤(以胸部创伤为主)患者作为研究对象(创伤组)。根据创... 目的探讨胸部创伤患者血清Sestrin 2、抑瘤素M(OSM)、Toll样受体9(TLR9)的表达水平及其与细菌感染的相关性。方法选取2022年7月至2024年7月河北医科大学第一医院收治的148例胸部创伤(以胸部创伤为主)患者作为研究对象(创伤组)。根据创伤程度将患者分为轻伤组(52例)、重伤组(47例)、危重组(49例),根据细菌感染并发情况分为感染组(54例)、未感染组(94例)。另纳入同期于该院体检的100例健康志愿者作为对照组。采用酶联免疫吸附法检测血清Sestrin 2、OSM、TLR9水平。应用多因素Logistic回归分析胸部创伤患者并发细菌感染的影响因素。应用ROC曲线分析血清Sestrin 2、OSM、TLR9水平对胸部创伤患者并发细菌感染的预测价值。结果与对照组相比,创伤组血清Sestrin 2、OSM、TLR9水平均升高(P<0.05)。与轻伤组相比,重伤组、危重组血清Sestrin 2、OSM、TLR9水平升高;与重伤组相比,危重组血清Sestrin 2、OSM、TLR9水平升高(P<0.05)。与未感染组相比,感染组机械通气时间、创伤严重程度(ISS)评分及白细胞、Sestrin 2、OSM、TLR9水平均升高(P<0.05)。ISS评分、Sestrin 2、OSM、TLR9为影响胸部创伤患者并发细菌感染的危险因素(P<0.05)。血清Sestrin 2、OSM、TLR9单独及联合预测胸部创伤患者并发细菌感染的AUC为0.847、0.819、0.856、0.951,三者联合的预测价值优于Sestrin 2(Z=3.562)、OSM(Z=4.069)、TLR9(Z=3.815)单独预测(P<0.001)。结论胸部创伤患者血清Sestrin 2、OSM、TLR9水平升高,与创伤严重程度、细菌感染有关,且三者联合检测在胸部创伤患者并发细菌感染预测中具有一定价值。 展开更多

利妥昔单抗治疗M型磷脂酶A2受体相关特发性膜性肾病的疗效及预后分析

目的分析M型磷脂酶A2受体相关特发性膜性肾病运用利妥昔单抗治疗的效果及预后。方法39例M型磷脂酶A2受体相关特发性膜性肾病患者,均运用利妥昔单抗治疗。分析患者的临床疗效以及不良反应发生情况,比较不同时间点实验室指标(24 h尿蛋白... 目的分析M型磷脂酶A2受体相关特发性膜性肾病运用利妥昔单抗治疗的效果及预后。方法39例M型磷脂酶A2受体相关特发性膜性肾病患者,均运用利妥昔单抗治疗。分析患者的临床疗效以及不良反应发生情况,比较不同时间点实验室指标(24 h尿蛋白定量、血肌酐、白蛋白、甘油三酯、免疫球蛋白G)。结果经过12个月随诊,39例患者完全缓解22例、占比56.41%,部分缓解15例、占比38.46%,未缓解2例、占比5.13%;总有效率为94.87%。经过12个月随诊,39例患者出现恶心1例,占比2.56%;呕吐1例,占比2.56%;肝功能异常0例;感染1例,占比2.56%;血糖升高0例。不良反应总发生率为7.69%。治疗后1、3、6、12个月的24 h尿蛋白定量、血肌酐、甘油三酯均较治疗前降低,白蛋白较治疗前升高,治疗后12个月的免疫球蛋白G水平较治疗前升高,有差异(P<0.05)。结论对M型磷脂酶A2受体相关特发性膜性肾病患者运用利妥昔单抗治疗的效果显著,可有效改善实验室相关指标,值得临床推荐。 展开更多

目的探讨血清乳酸清除率(LCR)、瞬时受体电位阳离子通道亚家族M成员7(TRPM7)水平与脓毒症并发急性肾损伤(SA-AKI)患者病情进展的关系。方法选取2021年1月至2024年9月西安大兴医院收治的SA-AKI患者125例(AKI组)和同期收治的单纯脓毒症患... 目的探讨血清乳酸清除率(LCR)、瞬时受体电位阳离子通道亚家族M成员7(TRPM7)水平与脓毒症并发急性肾损伤(SA-AKI)患者病情进展的关系。方法选取2021年1月至2024年9月西安大兴医院收治的SA-AKI患者125例(AKI组)和同期收治的单纯脓毒症患者125例(非AKI组)作为研究对象,将SAAKI患者根据AKI分期分为1期组(34例)、2期组(42例)、3期组(49例),根据入院后28d预后分为死亡组(51例)和存活组(74例)。计算2、4、6hLCR,并检测血清TRPM7水平。采用多因素Logistic回归分析血清6hLCR、TRPM7水平与SA-AKI患者死亡的关系,采用受试者工作特征曲线分析确定血清6hLCR、TRPM7水平对SA-AKI患者死亡的预测效能。结果与非AKI组比较,AKI组血清6hLCR降低(P<0.05),TRPM7水平升高(P<0.05)。1期组、2期组、3期组血清6hLCR依次降低(P<0.05),TRPM7水平依次升高(P<0.05)。125例SA-AKI患者28d死亡率为40.80%(51/125)。死亡组血清6hLCR低于存活组(P<0.05),TRPM7水平高于存活组(P<0.05)。调整年龄、基础疾病和炎症指标后,6hLCR为SA-AKI患者死亡的独立保护因素(P<0.05),TRPM7为SA-AKI患者死亡的独立危险因素(P<0.05)。血清6hLCR、TRPM7水平联合预测SA-AKI患者死亡的曲线下面积为0.884,大于血清6hLCR(0.782)、TRPM7(0.786)水平单独预测(P<0.05)。结论SA-AKI患者血清6hLCR降低,TRPM7水平升高,与病情分度加重及预后不良密切相关,可能成为评估SA-AKI患者病情进展的指标。 展开更多

It was found in acupuncturegroup that the fibrillation-liability of ischemic myocardium was efficiently decreased, the affinity ofAch-M receptors on membranes of ischemic myocardium was markedly increased, and the tolerance ofischemic myocardium to atropine was elevated in the experiment of atropine inducing fibrillation.These results indicate that acupuncture may play a therapeutic role on ischemic arrhythmia throughactivating the activity of muscarine-like receptors of cholinergic nervous system. 展开更多

The cholinergic system plays an important role in the central nervous system of insects and is closely related to the complex behavior of insects. The immunohistochemical technique was performed to detect the expression of like-muscarinic acetylcholine receptor M2 in the brain of three castes of Polyrhachis vicina. A positive expression of like-muscarinic acetylcholine receptor M2 was observed in the mushroom body, central body and antennal lobes of the ant brain; but there is great diversity in their location and intensity among worker, queen and male ants. It is speculated that like-muscafinic acetylcholine receptor M2 plays a critical role in the central nervous system, in terms of projecting visual information and olfactory information into the protocerebrum and integrating many inputs. 展开更多

However, the exact molecular mechanism remains unclear. In this study, muscarinic acetylcholine receptor M1 (CHRM1) expression was measured via immunohistochemical analysis in human prostate cancer tissue array slides. PC-3, LNCaP, and A549 cells were treated with pirenzepine or carbachol, and the cell migration and invasion abilities were evaluated. Western blotting and quantitative real-time PCR were performed to measure GLI family zinc finger 1 (GLI1), patched 1 (PTCH1), and sonic hedgehog (SHH) expression levels. High expression of CHRM1 was found in early-stage human prostate cancer tissues. In addition, the selective CHRM1 antagonist pirenzepine inhibited PC-3, LNCaP, and A549 cell migration and invasion, but the agonist carbachol promoted the migration and invasion of these three cell lines. Muscarinic signaling can be relayed by hedgehog signaling. These data show that CHRM1 is involved in the regulation of prostate cancer migration and invasion through the hed^eho~ si^nalin~ ~athwav. 展开更多

The aim of this study was to establish a model by single injection of Aβ1-40+IA into rat brain basal ganglion and examine the effect of ZDY101, an active component of a Yin tonic from Chinese traditional medicinal herb. The results showed that the amnestic effect of co-injection of Aβ and IA lasted for at least 2 months. At same time, the total M-receptor density in model brain was significantly lower than blank control, indicating the change is profound enough for long term pathological studies or drug screening. It can be clearly seen that the decreased brain M-receptor density caused by Aβ was significantly increased by ZDY101. Such an elevation effect was significantly correlated with dose of ZDY101 when the dose was examined in a certain reasonable range. It can also he clearly seen that such an elevation of M-cholinergic receptor density was significantly correlated with the improvement of memory which indicated that the increase of M-cholinergic receptor density was an important factor in improving the memory of such animal model. 展开更多

Objective: To study the potential role of cellular macrophage colony-stimulating factor (cM-CSF) and cellular macrophage colony-stimulating factor receptor (cM-CSF-R) with breast cancer and hepatoma and search the way for clinical application. Methods: Frozen surgical specimens from 48 breast cancer patients, including 29 cases of histological grade II and 19 eases of grade III, and 16 hepatoma patients were investigated by Avidin Biotin Complex (ABC) immunohistochemical assay with anti-M-CSF monoclonal antibody (Mab) and anti-M-CSF-R Mab. Pathohistological examination was performed as well. Results: cM-CSF and cM-CSF-R were detected in tested specimens. The expression levels of cM-CSF and cM-CSF-R in grade III group were higher than in grade II group and more higher than control group hyperplasia of breast. Hepatoma tissues also showed higher expression level of cM-CSF and cM-CSF-R than normal adult and fetal liver. Conclusion: Breast cancer and hepatoma tissues presented higher expression levels of cM-CSF and cM-CSF-R than control and expression level might be related with tumor’s process. 展开更多

Objective: To investigate the potential role of macrophage colony-stimulating factor (M-CSF) and macrophage colony-stimulating factor receptor (M-CSF-R) on the growth of human hepatoma cells. Methods: Specimens of different origin, including tissues of human hepatocellular carcinoma (HCC), human fetal liver (FL) and normal liver (NL), the hepatoma cell lines, as well as the peripheral blood mononuclear cells (PBMC) from patients with HCC or liver metastatic tumor (LMT), were used to detect the expression levels of M-CSF and M-CSF-R by ABC immunohistochemistry staining and reverse transcription polymerase chain reaction methods the expression levels of M-CSF and M-CSF-R. Influence of monoclonal antibody against M-CSF (B5) or M-CSF-R (RE2) on proliferation ability of hepatoma cell linesin vitro was also studied. Results: The results showed that hepatoma tissues produced elevated levels of both M-CSF and M-CSF-R compared with those of fetal liver (P<0.001). The M-CSF/M-CSF-R expression levels of PBMC from hepatoma patients were higher than those of LMT patients (P<0.01,P<0.05) and the normal people (P<0.001). The hepatoma cell lines showed strong positive for M-CSF and M-CSF-R production. Both B5 and RE2 displayed a dose-dependent inhibitory effect on the growth and proliferation of hepatoma cells. Conclusion: The study indicates a co-expression model for M-CSF-R in hepatoma cells, suggesting an involvement of M-CSF/M-CSF-R in growth signaling of those malignant cells. The M-CSF/M-CSF-R seems to function through an autonomy mechanism in human hepatoma. 展开更多

Whether M3 cholinergic receptor signal transduction pathway is involved in regulation of the activation of NF-κB and the expression of chemokine MOB-1, MCP-1genes in pancreatic acinar cells was investigated. Rat pancreatic acinar cells were isolated, cultured and treated with carbachol, atropine and PDTC in vitro. The MOB-1 and MCP-1 mRNA expression was detected by using RT-PCR. The activation of NF-κB was monitored by using electrophoretic mobility shift assay. The results showed that as compared with control group, M3 cholinergic receptor agonist (10 -3 mol/L, 10 -4 mol/L carbachol) could induce a concentration-dependent and time-dependent increase in the expression of MOB-1, MCP-1 mRNA in pancreatic acinar cells. After treatment with 10 -3 mol/L carbachol for 2 h, the expression of MOB-1, MCP-1 mRNA was strongest. The activity of NF-κB in pancreatic acinar cells was significantly increased (P<0.01) after treated with M3 cholinergic receptor agonist (10 -3 mol/L carbachol) in vitro for 30 min. Either M3 cholinergic receptor antagonist (10 -5 mol/L atropine) or NF-κB inhibitor (10 -2 mol/L PDTC) could obviously inhibit the activation of NF-κB and the chemokine MOB-1, MCP-1 mRNA expression induced by carbachol (P<0.05). This inhibitory effect was significantly increased by atropine plus PDTC (P<0.01). The results of these studies indicated that M3 cholinergic receptor signal transduction pathway was likely involved in regulation of the expression of chemokine MOB-1 and MCP-1genes in pancreatic acinar cells in vitro through the activation of NF-κB. 展开更多

A technique for studying in vivo the production rate and turnover rate constant of mouse brain M-receptors was established. A single injection of 25 mg / kg of Benzilylcholine Mustard to living mice resulted in 90 % irreversible block of brain M-receptors. The time course of the receptor density was then monitored by 3H-QNB binding assay and the production rate and turnover rate constant were calculated from the time course curve with a computer program. It was found that in normal mice the turnover rate constant was about 0.035 h-1 (half-life was about 20 h) and the production rate was 30-42 fmol / (h ·mg protein). Parallel experiments revealed a significant slow down of the turnover of brain M-receptors in hypothyroid mice (turnover rate constant was 0.0257±0.0012 h-1 in hypothyroid vs. 0.0356±0.0021 h-1 in normal) while the production rate was not changed significantly. The results suggest that thyroid hormones have a regulatory action on the turnover of brain M-receptors and the elevation of brain M-receptor density together with slow down of the turnover of brain M- receptors is probably one of the important mechanisms relevant to the brain dysfunction in hypothyroidism. 展开更多

OBJECTIVE Abnormal striatal dopaminergic and glutamatergic neurotransmis⁃sion is central to the pathophysiology of schizo⁃phrenia.In this study,we investigated the roles of M4 receptor interplay with D1 signaling in stria⁃tal neurotransmission that affect glutamatergic transmission to control the etiology of neuropsy⁃chiatric disorders.METHODS To study dorsal striatum(DS)region-specific neuronal and behav⁃ioral responses modulated by M4 receptors,we used clustered regularly interspaced short palin⁃dromic repeats-associated protein 9 technology to generate mice lacking M4 in the dorsal stria⁃tum(DS-M4-KD).The M4 positive allosteric modu⁃lator,VU0467154,were used to study the phar⁃macologically profiles with M4 receptor stimula⁃tion in WT mice.Oxotremorine M(Oxo-M),a no subtype-selective muscarinic agonist,was used to show that mAchRs activation,in order to dissect the particular function of M4,in DS-M4-KD mice.Open filed test and forced swim test were used to assess the change of psychiatric-like behav⁃iors.Western blotting and immunohistochemistry were used to detect protein levels of phosphory⁃lation site of dopamine-and cAMP-regulated phosphoprotein of 32 ku(DARPP-32).Whole-cell patch-clamp recording was used to assess M4-mediated cholinergic inhibition of glutamater⁃gic synaptic input transmission.RESULTS West⁃ern blotting and immunohistochemistry assay showed VU0467154(5 mg·kg-1,ip)promoted phosphorylation of DARPP-32 at Thr75,and atten⁃uated D1-dependent phosphorylation of DARPP-32 at Thr34 within the mouse DS.Consistently,the Oxo-M(4μg,icv)also increased DARPP-32 phosphorylation at site Thr75 to reversed phos⁃phorylation at site Thr34 in WT mice,but not in DS-M4-KD mice.In parallel with altered DARPP-32 responses,VU0467154 or Oxo-M evoked a psychological stress response and reversed D1-induced hyperlocomotion in mice in open field test and force swim tests.However,Oxo-M sup⁃pression of D1-depengdeng behavioral respons⁃es was impaired in DS-M4-KD mice.Whole-cell patch recording showed that VU0467154 or Oxo-M mediated endogenous cholinergic inhibition of miniature excitatory postsynaptic currents through M4 receptors,which in turn suppressed D1-depen⁃dent glutamatergic synaptic transmission in the DS.CONCLUSION This study provides evidence for the role of M4 receptors in regulation of dopa⁃mine/DARPP-32 signaling and glutamate respons⁃es in the DS,and therefore modulation of psychi⁃atric behaviors associated with D1 signaling.This results indicate the mechanisms of treatments targeting M4 in psychiatric disorders. 展开更多

BACKGROUND: Organophosphorus insecticides may promote the accumulation of acetylcholine at synapses and the neuromuscular junction by inhibiting acetylcholinesterase activity to cause disturbance of neural signal conduction and induce a toxic reaction. Organophosphorus insecticides may act on M2 muscarinic acetylcholine receptors, whose combination with G proteins is regulated by phosphorylation of G protein-coupled receptor kinase 2. OBJECTIVE: To investigate the effects of organophosphorus insecticides on the phosphorylation of G protein-coupled receptor kinase 2-mediated M2 muscarinic acetylcholine receptors and to reveal other possible actions of organophosphorus insecticides. DESIGN, TIME AND SETTING: An observational study, which was performed in the Central Laboratory of Shenyang Medical College, and Department of Physiological Sciences, College of Veterinary Medicine, Oklahoma State University from June 2002 to December 2004. MATERIALS: Paraoxon, parathion, chlorpyrifos, and chlorpyrifos oxon were provided by Chem Service Company, USA, [γ -p^32] ATP and [^35S]GTP γ S by New England Nuclear Life Science Products, and recombinant β 2-adrenergic receptor membrane protein by Sigma Company, USA. METHODS: The M2 muscarinic acetylcholine receptor was extracted and purified from pig brain using affinity chromatography. Subsequently, the purified M2 muscarinic acetylcholine receptor, G protein-coupled receptor kinase 2, and [γ -p^32] ATP were incubated with different concentrations of paraoxon and chlorpyrifos oxon together. The mixture then underwent polyacrylamide gel electrophoresis, and the gel film was dried and radioactively autographed to detect phosphorylation of the M2 muscarinic acetylcholine receptor. Finally, the radio-labeled phosphorylated M2 receptor protein band was excised for counting with an isotope liquid scintillation counter. MAIN OUTCOME MEASURES: Effects of chlorpyrifos oxon, paraoxon, chlorpyrifos, and parathion in different concentrations on the phosphorylation of the M2 muscarinic acetylcholine receptor; effects of chlorpyrifos oxon on the phosphorylation of the β -adrenergic receptor. RESULTS: Chlorpyrifos oxon could completely inhibit the phosphorylation of the M2 muscarinic acetylcholine receptor, and its IC50 was 70 μ mol/L. Chlorpyrifos could also inhibit the phosphorylation of the M2 muscarinic acetylcholine receptor. However, paraoxon and parathion could not inhibit the phosphorylation of the M2 muscarinic acetylcholine receptor. Chlorpyrifos oxon in different concentrations could also not inhibit the phosphorylation of the β 2-adrenergic receptor catalyzed by G protein-coupled receptor kinase 2. CONCLUSION: Different kinds of organophosphorus insecticides have different effects on the phosphorylation of the G protein-coupled receptor kinase 2-mediated M2 muscarinic acetylcholine receptor. Organophosphorus insecticides possibly have different toxic effects. 展开更多

The role of Src family kinases in mediating M-CSF receptor signaling and monocytic cell survival

Previously, we reported that M-CSF induced monocyte survival through the activation of Akt, p38MAPK and Erk1/2 kinases. Here, we found that Src family kinases were upstream of these kinases and played a central role in regulating M-CSF-induced monocyte survival. We observed that M-CSF promoted c-Src activation in monocytes and MDMs in a time-dependent manner. Src inhibitors reduced M-CSF-mediated phosphorylation of the M-CSF receptor (M-CSFR), Akt, Erk1/2, and p38 MAPK. We also observed that Src directly phosphorylated the M-CSFR. Notably, the inhibitors blocked phosphorylation of specific tyrosine residues within the M-CSFR. We further demonstrated that the Src inhibitor, PP2, attenuated M-CSF-induced NF-κB activation and M-CSF-induced monocyte survival. These findings indicated that Src family kinases mediate monocyte survival through the regulation of receptor phosphorylation and modulation of downstream signaling events. Thus, we predict that targeting Src family kinases may have therapeutic implication in inflammatory diseases. 展开更多

Idiopathic pulmonary fibrosis is an untreatable lethal lung disease, which is related to the aberrant proliferation of fibroblasts. M<sub>3</sub> muscarinic acetylcholine receptor (M<sub>3</sub>-mAChR) activation exerts proliferative effect on various kinds of cells. However, whether M<sub>3</sub>-mAChR inhibition has a protective effect on pulmonary fibrosis remains unexplored. A rat model of pulmonary fibrosis was established by intratracheal instillation of bleomycin. Darifenacin was used to block M<sub>3</sub>-mAChR. Histological changes were observed using Masson’s Trichrome and hematoxylin and eosin (HE) staining. Hydroxyproline was measured by Hydroxyproline detection kit. Transforming growth factor β1 (TGF-β1) and tumor necrosis factor-α (TNF-α) were measured by enzyme-linked immunosorbent assay (ELISA). In vitro, pulmonary fibroblasts were isolated from lungs of neonatal rat. After treatment, the cell viability, Hydroxyproline level was measured by MTT and Hydroxyproline detection kit respectively. The expression level of extracellular signal-regulated kinase (ERK), nuclear factor kappa-B (N-NF-κB), and microRNA-21 (miR-21) was detected by western blot or quantitative real-time PCR (qRT-PCR). Darifenacin relieved the fibrotic effects provoked by bleomycin. The expression level of hydroxyproline, TGF-β1 and TNF-α level was all downregulated after darifenacin treatment. In lung fibroblasts, darifenacin decreased cell viability and hydroxyproline level induced by bleomycin. Besides, phosphorylation-ERK and nuclear N-NF-κB protein level was downregulated, as well as miR-21 level. M<sub>3</sub>-mAChR antagonist darifenacin attenuates bleomycin-induced pulmonary fibrosis in rats, which may relate to the ERK/NF-κB/miRNA-21 signaling pathway. 展开更多

Objectives To explore the relationship between serum autoantibodies against myocardial β1-adrenergic, M2-cholinergic receptors and chronic Keshan disease (CKD). Methods The second extracellular loops of β1 and M2 receptors on human cardiomyocytes were used as the antigens. Enzyme linked immunosorbent assay (ELISA) was applied to determine serum autoantibodies against myocardial β1 and ME receptors in 32 CKD patients. 31 healthy subjects from endemic area were selected as the control. Results Positive rate of autoantibodies against myocardial β1 adrenergic (51.3%, 17/32) and M2 cholinergic (56.3% , 18/32) receptors were significantly higher than those in the control (9.7%, 3/ 31; 12.9%, 4/31) (both P〈 0.01). Both positive rate and titers of above autoantibodies in NYHA Ⅱ - Ⅲ CKD patients were significantly higher than those in NYHA Ⅳ, demonstrating an apparently positive correlation between serum antibodies against myocardial β1 and M2 receptors (r=0.95). Conclusions Autoantibodies against myocardial β1 and M2 receptors were found in sera of CKD patients; distribution of positive rate and titers of the autoantibodies in CKD patients in various NYHA are significantly different. classes of cardiac function 展开更多

Methods Using polymerase chain reaction (PCR) and DNA sequencing techniques, the exons of AMH and AMHRII were analyzed in 27 OHSS patients (OHSS group) and 22 non-OHSS patients (control group) who were applied controlled ovarian hyper- stimulation (COH). Single nucleotide polymorphisms (SNPs) were also analyzed. Results SNPs G〉 T at position 146 of AMH exon 1 and G〉 A at position 134 of AMH exon 2 showed significant differences between the OHSS group and control group (P〈0.05). SNP G〉 T at position 303 of AMH exon 1 showed no significant difference between the OHSS group and control group (P〉0.05). No SNP was detected from the AMHR H exons 1 to 11 in either groups. Conclusion Genetic polymorphisms in the AMH gene may be a cause of ovarian hypersensitivity to exogenous hormone stimulation and the development of OHSS. 展开更多

The pharmacologic characteristics of ^(99)Tc^m-N-ethyl-N_2S_2-memantine,an NMDA receptor imaging agent,was investigated.It was prepared by a one-step reaction from N-ethyl-N_2S_2-memantine.The affinity and specificity were determined by radio-ligand receptor binding assay(RRA).Biodistribution in vivo in mice was performed.The results showed that ^(99)Tc^m-N-ethyl-N_2S_2-memantine bound to a single site on NMDA receptor with a K_d of 584.32 nmol/L and a B_(max)of 267.05 nmol/mg.A competitive analysis showed that such specific binding could be inhibited by specific inhibitors of NMDA receptor,such as ketamine and(+)-MK-801.The biodistribution exhibited rapid uptake and favorable retention in mice brains.The major radioactivity was metabolized by the hepatic system.A two-compartment model of C=4.49e^(-0.083t)+ 1.42e^(-0.0016t)was established,and the half life was 8.35 min in blood.In conclusion,the new radio-ligand ^(99)Tc^m-N-ethyl-N_2S_2-Memantine has a moderate affinity and specific binding to NMDA receptor,and can easily cross the blood-brain barrier(BBB).Therefore,it may be a potential NMDA receptor imaging agent. 展开更多
