Methods: Forty ejaculates were harvested from four buffalo bulls by means of the artificial vagina. Ejaculated semen samples were diluted with each of the tris citrate-fructose egg yolk glycerol and lecithin-based extender diluents. The semen samples diluted with each of the two extenders were added to pre-warmed dried test tubes containing BHT (prepared in ethanol) to get concentrations at 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0 mM/mL BHT. These ingredients were put at 37 ℃ for 5 min to allow the proper BHT spermatozoal permeation. The diluted semen samples were cooled to 5 ℃ and then frozen to -196 ℃ in 0.25 mL ministraws before dipping in liquid nitrogen pending its evaluation. Sperm motility, viability, morphology, intact acrosome and membrane integrity were tested. Visual motility was tested using a high power ordinary microscope (at 400 × ) with closed circuit television, and sperm concentration was tested using Neubauer haemocytometer and abnormality % using eosin-nigrosin stain. Spermatozoal membrane integrity was tested using the hypo-osmotic swelling test. The sperm with swollen twisting tail was normally intact. Sperm acrosomal integrity % was tested as mentioned by Watson. Results: Addition of BHT improved (P<0.01) progressive motility, viability, morphology and acrosome as well as plasma membrane integrities at 0.5-2.0 mM/mL depending upon types of used extenders and stages of pre-and post-freezing process. Higher levels of 2.5 and 3.0 mM/mL BHT had a deteriorating (P<0.01) result if compared to the control and all extenders assayed. Conclusions: BHT addition at lower concentration can improve pre-frozen and post-thawed buffalo sperm quality.展开更多

Semen samples were extended gradually 1:10 with TCFYG control extender and tris-citric acid-fructose-glycerol (TCFG) extender plus variable concentrations of soybean lecithin (0.5%, 1.0%, 1.5%, 2.0%, 2.5% and 3.0%) to ensure 60 million active spermatozoa/mL of the extended semen. The diluted semen samples were refrigerated slowly (roughly for 2 h) up to 5℃ and equilibrated for 2 h. Semen was filled into 0.25 mL polyvinyl French straws (IMV, France). After equilibration period, the straws were placed horizontally on a rack and frozen in a vapor 4 cm above liquid nitrogen for 10 min and were then dipped stored in liquid nitrogen at -196℃. Results:The respective overall percentages of forward motile spermatozoa, live spermatozoa, morphologically normal spermatozoa, acrosome integrity and hypo-osmotic swelling reactivity observed primarily in fresh semen, after equilibration (pre-freeze stage) and post freezing (post-thaw stage) in TCFYG (control) extended semen declined progressively and statically (P<0.01) during these periods of study. Pre-freezing stage: replacement of egg yolk into TCFG with soybean lecithin at concentrations of 1.0% and 1.5% significantly (P<0.01) ameliorated the maintenance of (motility, viability, acrosome and membrane integrity %), meanwhile it had significantly (P<0.01) reduced the abnormality % of spermatozoa to the lowest value compared to control TCFYG and to some other concentrations in use. Post-thaw stage: the replacement of egg yolk with 1.0% soybean lecithin (SL) showed significantly (P<0.01) higher percentage of sperm progressive motility compared to 1.5% SL and TCFYG control. These values were significantly (P<0.01) higher than 0.5%, 2.0%, 2.5% and 3.0% SL. The post thawing live sperm percentage mean values were significantly (P<0.01) higher in 1.0% SL and 1.5% SL compared to control. These values were significantly (P<0.01) higher than in 0.5%, 2.0%, 2.5% and 3.0% SL. The mean values of post-thaw morphological normal sperm percentage did not differ between 1.0% SL and control groups but significantly (P<0.01) higher than 0.5%, 1.5%, 2.0%, 2.5% and 3.0% SL. The respective percentage mean values of post-thaw sperm with head, mid-piece and tail abnormalities were significantly (P<0.01) lower in 1.0% SL than all other SL concentrations. Concerning the post-thaw percentages of acrosome and sperm membrane integrity, the respective mean values were significantly (P<0.01) higher in 1.0% SL and 1.5% SL as compared to control. Mean values of both parameters in the 0.5% SL were intermediate between 1.0% and 1.5% SL versus control groups. The previously mentioned mean values in acrosome/membrane integrity were significantly (P<0.01) higher than 2.0% SL, 2.5% SL and 3.0% SL.Conclusions: Lecithin-based diluent can be a potent proper alternative extender for preservation of spermatozoa during pre- and post-freezing process. SL 1.5% extenders have supplied an optimal environment and condition for ameliorating the quality of pre-freezing and post-thaw buffalo spermatozoa by means of improved motility, viability, functional acrosome, sperm membrane integrity and morphologically normal spermatozoa.展开更多

About 67% cropland in China lacks sufficient phosphorus, especially that with red soil. Extensive soil phosphorus reserves exist in the form of organic phosphorus, which is unavailable for root uptake unless hydrolyzed by secretory acid phosphatases. Thus, many microorganisms with the ability to produce phosphatase have been exploited. In this work, the activity of an extracellular acid phosphatase and yeast biomass from Candida mycoderma was measured under different culture conditions, such as pH, temperature, and carbon source. A maximal phosphatase activity of 8.47 × 10^5±0.11× 10^5 U/g was achieved by C. Mycoderma in 36 hr under the optimal conditions. The extracellular acid phosphatase has high activity over a wide pH tolerance range from 2.5 to S.0 (optimum pH 3.5). The effects of different phosphorus compounds on the acid phosphatase production were also studied. The presence of phytin, lecithin or calcium phosphate reduced the phosphatase activity and biomass yield significantly. In addition, the pH of the culture medium was reduced significantly by lecithin. The efficiency of the strain in releasing orthophosphate from organic phosphorus was studied in red soil (used in planting trees) and rice soil (originating as red soil). The available phosphorus content was increased by 230% after inoculating 20 days in rice soil and decreased by 50% after inoculating 10 days in red soil. This work indicates that the yeast strain C. mycoderma has potential application for enhancing phosphorus utilization in plants that grow in rice soil.展开更多

Two sources of soybean lecithin (20% and 95% soybean phosphatidylcholine; PC20 and PC95) and three concentrations (1%, 2% and 3% v/v) of PC20 and PC95 supplemented in Tris-citric acid-fructose (TCF) extender were tested. The TCF extender supplemented with 20% hen egg yolk was used as a control. Fresh semen samples were collected from 3 goats by artificial vagina. Seminal plasma was removed by centrifugation and sperm pellets were pooled together and divided into 7 groups according to types of extender. The diluted semen samples were kept at 4 ℃ (equilibration). The semen qualities including progressive motility, sperm viability, sperm plasma membrane integrity and tail abnormalities were evaluated before dilution and after 4 hrs equilibration. It was found that the progressive motility of equilibrated semen in egg yolk and PC20 extenders were higher than those in PC95 extender (P 〈 0.05). Sperm viability was lower in 1% and 2% PC95 extender compared to other extenders (P 〈 0.05). PC20 extender maintained the sperm membrane integrity and normal tail morphology at low temperature better than egg yolk and PC95 (P 〈 0.05). It can be concluded that 20% soybean phosphatidylcholine supplemented in TCF extender at 1%-3% (v/v) is as effective as hen egg yolk to preserve goat semen during equilibration at 4 ℃ for 4 hrs .展开更多

Besides, the PCP effect on critical micelle concentration (CMC) of SL and the effect of PCP and SL on the ligninolytic activity of A. discolor were evaluated. The PCP solubilization test was performed for 100, 200 and 400 mg·L-1 with SL solution varying concentrations of ranging from 0 to 0.9 g·L-1. The effect of PCP (5 to 20 mg·L-1) on CMC of SL by conductivity was evaluated. The effect of SL (0 to 0.9 g·L-1) on biodegradation of PCP (21.4 mg·L-1) in modified Kirk medium was studied. The results showed that SL addition enhanced PCP solubility in water. The solubilities of 100, 200 and 400 mg·L-1 of PCP were about 4.2, 5.0 and 2.5 times higher than their solubility in pure water when the system was added with 0.9 g·L-1 of SL. We found that the increase in PCP concentration caused a decrease in the CMC value. In the biodegradation assay, in liquid medium supplemented with 0.9 g·L-1 of SL, the 94.4% of PCP was biodegraded after 26 days of incubation. In contrast, in the control assay (without SL), the 76.5% of PCP was removed. The PCP degradation was confirmed by the presence of its metabolites. During biodegradation assay, A. discolor mainly produced manganese peroxidase reaching a maximum of 96.8 U·g-1 (dw) when the culture medium was added with 0.6 g·L-1 of SL. These results show that SL can be applied to increasing the bioavailability and biodegradation of PCP.展开更多

The selected formulation was a stable oil/water emulsion system. The particles in the emulsion system were distributed evenly, and the particle size ranged from 20 to 260 nm( average size : 67.5 nm ). Soybean lecithin played an important role in the emulsion system due to its abilities of acting as both absorption enhancer for insulin uptake through sublingual mucosa and oily phase for the emulsion system. The laser confocal scanning microscopic(LCSM) study showed that FITC-labelled insulin could penetrate the sublingual mucosa of rabbits, and the phase diagrams of the emulsion system suggested that soybean lecithin could take the place of oily phase to construct a stable emulsion system even if the traditional oil was absent. The applications of soybean lecithin as pharmaceutical biomaterial were extended for the further usage by present studies.展开更多

Methods: The ejaculates from 4 mature male Damascus goats were obtained by using an artificial vagina. Semen samples were pooled and diluted in Tris extender supplemented with soybean lecithin at different concentrations of 1.5%, 3.0%, 6.0% and 10.0% to get better concentration to be used for further experiments, with a final concentration of 240×106 spermatozoa/mL. Semen samples were packed in straws (0.25 mL), frozen by using an automated system and stored in liquid nitrogen at -196 ℃ for 48 h. After thawing (37 ℃/30 s), the samples were evaluated for sperm quality parameters, including sperm motility, membrane integrity and acrosome integrity. Malondialdehyde concentration was estimated as a marker for lipid peroxidation. Based on the previous investigations, only Tris extender supplemented with 3.0% soybean lecithin (based on its positive results) was used versus BullXcell and OptiXcell for sperm ultrastructure evaluation and artificial insemination by using electron microscope and artificial insemination of the synchronized does. Results: There was no significant difference between Tris-soybean lecithin at 3.0% and BullXcell/OptiXcell diluents in post-thaw sperm parameters and fertility following artificial insemination;meanwhile, the other concentrations of soybean lecithin (1.5%, 6.0% and 10%) showed lower sperm parameters following cryopreservation. Conclusions: Using of Tris-soybean lecithin based extender at the level of 3.0% can be an appropriate alternative to either BullXcell or OptiXcell for Damascus goat sperm cryopreservation.展开更多

文摘As a natural oil-soluble emulsifier to replace Polyglycerol Polyricinoleate(PGPR),lecithin's application in double emulsions(DEs)remains constrained due to low encapsulation efficiency and stability, while incorporating Pickering particles within the inner aqueous phase(W_(1))has emerged as an effective approach to enhance DEs stability. However, the interfacial interactions between Pickering particles in W_(1) and lecithin in oil phase and their effects on the physical, delivery and digestion properties of DEs remain to be explored. In this study,a natural Pickering DEs encapsulating blueberry anthocyanins(ANCs)was developed using lecithin and gliadinbased particles, with gliadin colloid particles(GCPs)introduced into W_(1).The effects of GCP concentration in W_(1) on emulsion droplet size,microstructure,W_(1)/O interfacial rheology,in vitro digestion, and ANCs delivery performance were systematically investigated. Results showed that GCPs in W_(1) accelerated the decreasing of W_(1)/O interfacial tension, indicating that, beyond competitive adsorption, combination between the two might also occur at the interface, which could accelerate the interfacial adsorption of lecithin.GCPs significantly improved the strength of the W_(1)/O interface, thus improving ANCs encapsulation efficiency from 49.66%to 70.60%.Moreover,the droplet size of DEs decreased from 38.94μm to 24.09μm as GCPs concentration increased, indicating that some GCPs might transfer to the outer aqueous phase through phase exchange, acting as hydrophilic emulsifiers.GCPs in W_(1) can modulate ANCs in vitro release, enhancing ANCs'bioaccessibility(31.10%)and antioxidant capacity.

文摘Identifying a potential dietary non-pharmacological treatment to prevent cerebrovascular damage in Alzheimer's disease is crucial for alleviating cognitive decline in older adults and enhancing quality of life. This study featured the combined supplementation of soy lecithin(SL)and soy isoflavones(SIF),using in vivo animal models,in vitro vascular ring preparation, and cell studies to investigate the potential advantages and mechanisms of SL combined with SIF on cognitive function and cerebrovascular health from multiple perspectives. The results show that Aβcan significantly induce learning and memory impairment in rats,as well as pathological changes in brain blood vessels, exacerbating damage to cerebral vasodilation function and subsequently reducing cerebral blood flow in the brain. The above-mentioned phenomena induced by Aβcan be significantly improved by the combined intervention of SL and SIF.Further research has revealed that the combined intervention of SL and SIF can reverse the downregulation of the PI3K/PIP3/PDK-1/Akt/eNOS signaling pathway and phosphorylated protein expression induced by Aβin rat brain vascular tissues and bEND.3 cells.Silencing PDK-1 expression in bEND.3 cells showed that the upregulation effect of SL and SIF on Akt and eNOS disappeared. Here we find that prophylactically supplementation with SL in conjunction with SIF appears to effectively activate the PI3K/PIP3/PDK-1/Akt/e NOS pathway within cerebral vascular. This activation improves cerebrovascular vasodilation, offering potential protective effects for both cerebral vascular health and cognitive function.

Conclusions: BHT addition at lower concentration can improve pre-frozen and post-thawed buffalo sperm quality.
文摘In the present study, gel formulations of organogels, hydrogels, and oleo-hydrogel(bigels)were evaluated as transdermal drug delivery systems for diltiazem HCL(DH).Organogels were prepared using soya-bean oil(SO)as a solvent and span 60(Sp 60),cetyl alcohol(CA)or lecithinpluronic(PLO)as organogelators without and with different surfactants(2%w/w)namely span 80(Sp80),tween 20(T20)and tween 80(T80).On the other hand, hydrogels were formulated using Hydroxypropyl-methylcellulose(HPMC)polymer and bigels were prepared by mixing organogels with HPMC hydrogels. The prepared gels were analyzed microscopically, thermally by DTA and for pH,and viscosity. The effect of gelator used, surfactant types and pH of the sink on DH release from cellophane membrane was investigated. In addition, the DH permeability across the rabbit skin was evaluated. Finally, the in vivo performance of various gel formulationswas assessed based on the hypotensive effects of the drug using hypertensive albino male rat models. The microscopical analysis indicated that the solid fibers formed by gelator particles form the backbone of the organogels while bigels appeared as emulsion like. The addition of surfactants showed an increase in organogel viscosity. The thermal analysis of organogels indicated that the drug present in amorphous not in crystalline form. The release studies indicated that DH release from organogels, hydrogels and bigels could be controlled. The included surfactants decreased the DH release and permeation from organogels compared to those without surfactants using either Sp60 or CA.HPMC hydrogel and Bigels showed higher DH release and permeation rates when compared to organogels. The percent DH released in different pH values was in the following descending order:pH5.5>pH1.2>pH6.8>pH7.4.The in vivo antihypertensive activity of DH using different transdermal gels is arranged as following:hydrogels>PLO organogel>bigel>Sp 60 organogel.

文摘Fast and high-throughput determination of drugs is a key trend in clinical medicine.Single particles have increasingly been adopted in a variety of photoanalytical and electroanalytical applications, and microscopic analysis has been a hot topic in recent years, especially for electrochemiluminescence(ECL).This paper describes a simple ECL method based on single gold microbeads to image lecithin.Lecithin reacts to produce hydrogen peroxide under the successive enzymatic reaction of phospholipase D and choline oxidase.ECL was generated by the electrochemical reaction between a luminol analog and hydrogen peroxide, and ECL signals were imaged by a camera.Despite the heterogeneity of single gold microbeads, their luminescence obeyed statistical regularity. The average luminescence of 30 gold microbeads is correlated with the lecithin concentration, and thus,a visualization method for analyzing lecithin was established.Calibration curves were constructed for ECL intensity and lecithin concentration, achieving detection limits of 0.05 m M lecithin. This ECL imaging platform based on single gold microbeads exhibits outstanding advantages, such as high throughput, versatility and low cost, and holds great potential in disease diagnostics, environmental monitoring and food safety.
文摘Mantle pieces of Hyriopisis cumingii was treated by three kinds of solution,These mantle pieces were inserted together with paraffin nucleuses into the connective tissue of three groups of Hyriopisis cumingii. These operated animals were cultured in a pool for a month.Several pearl sacs were put out and immersed by Bouin's solution after every five days.Sections of each animals were made by histological method. Those without treatment were in a control group.Observations of these sections showed that cytochrome c and yolk lecithin have an accelerated roles on pearl sac development and pearl layer secretion. No effects on pearl sac development by polyvinyplyrrolidone(PVP).
文摘Objective:To estimate the result of egg yolk replacement with alternative cryopreservatives such as plant-derived lecithin from soybean on sperm quality parameters pre and post freezing in buffalo bulls. Methods: The control cryopreservation extender was tris-citric acid-fructose-egg yolk-glycerol (TCFYG) diluent. SL 1.5% extenders have supplied an optimal environment and condition for ameliorating the quality of pre-freezing and post-thaw buffalo spermatozoa by means of improved motility, viability, functional acrosome, sperm membrane integrity and morphologically normal spermatozoa.

文摘Orthophosphate is an essential but limiting macronutrient for plant growth. This work indicates that the yeast strain C. mycoderma has potential application for enhancing phosphorus utilization in plants that grow in rice soil.
文摘The molecular structure of phospholipids can be changed enzymatically to obtain different tailor-made phospholipids.Incorporation ofω-3 fatty acids into phospholipids structure increased their oxidative stability, suggesting more health beneficial phospholipids. This study aimed to optimize eicosapentaenoic acid(EPA)incorporation into phospholipids structure by acidolysis reaction using free lipase(EC 3.1.1.3)from Rhizomucor miehei.Deoiled soy lecithin from anjasmoro variety was used as phospholipids source,whileω-3 fatty acid enriched oil was used as acyl source.Oil enriched withω-3 fatty acids was obtained from low temperature solvent crystallization of lemuru(Sardinella longiceps)by-product.Response surface methodology(RSM)was used in this study to determine the relationship between the three factors(enzyme concentration, reaction time and substrate ratio)and their effects on EPA incorporation into soy lecithin structure. The results showed that the relation between EPA content with three factors(reaction time, enzyme concentration and substrate ratio)was quadratic. The significant factors were substrate ratio and reaction time.Optimum conditions at a ratio of 3.77:1 betweenω-3 fatty acids enriched oil and soy lecithin,30%lipase concentration, and 24.08 h reaction time, gave 22.81%of EPA content of structured phospholipids.
文摘The objective of the present study was to determine the proper sources and concentrations of soybean lecithin (phosphatidylcholine, PC) to be used as substitute for hen egg yolk in extender for preserving goat semen. It can be concluded that 20% soybean phosphatidylcholine supplemented in TCF extender at 1%-3% (v/v) is as effective as hen egg yolk to preserve goat semen during equilibration at 4 ℃ for 4 hrs .
文摘The effect of soya lecithin (SL) on solubilization and biodegradation of pentachlorophenol (PCP) by Anthracophyllum discolor was evaluated. These results show that SL can be applied to increasing the bioavailability and biodegradation of PCP.

文摘An insulin-loaded emulsion system (IES) was developed as a hypoglycaemic drug for transmucosal delivery. The applications of soybean lecithin as pharmaceutical biomaterial were extended for the further usage by present studies.
文摘Objective:To test the effects of extenders and packaging methods on morphological and functional characteristics of frozen thawed Ossimi ram semen. Methods:Ram semen was pooled, diluted in 3 different extenders:Tris-egg yolk(TEY),Tris-soybean lecithin(TSBL),and Tris-butylatedhydroxytoluene(TBHT),equilibrated at 5℃for 4 h,and packaged in straws or pellets for freezing.Semen was evaluated for sperm progressive motility, viability, abnormality, and membrane integrity after dilution, equilibration and thawing. The percentages of viable, early apoptotic, apoptotic, and necrotic spermatozoa as well as comet assay parameters were determined in post-thawed semen.Total antioxidants capacity, malondialdehyde and lactic dehydrogenase were assayed in thawed seminal plasma. Results:After equilibration, only sperm membrane integrity was significantly higher(P<0.05)in TEY and TSBL than in TBHT.After thawing,TEY or TSBL in straws significantly improved sperm progressive motility and vitality(P<0.05).In thawed seminal plasma,TBHT or TSBL in straws and TBHT in pellets significantly reduced malondialdehyde, and TBHT in pellets significantly increased lactic dehydrogenase(P<0.05).TEY in straws increased viable sperm, while significantly decreased early apoptotic and apoptotic sperm(P<0.05).DNA damage was significantly decreased(P<0.05)in straws with TEY and TSBL,and tail moment decreased in straws with all extenders(P<0.05).Conclusions:Despite the disadvantages of dilution of cryopreserved semen with egg yolk, ram semen cryopreserved with TEY gives the best physical, morphological and functional characteristics in straws compared with pellets, followed by semen diluted with TSBL.However, semen diluted with TBHT or TSBL,regardless of packaging method, showed the highest impact on antioxidant status of cyopreserved ram semen.
文摘Objective: To study the effect of adding different concentrations of soybean lecithin in Tris based extender and to compare the results of Tris soybean lecithin extender with two commercial diluents (egg yolk-based: BullXcell and plant-based: OptiXcell) for Damascus goat sperm cryopreservation. Conclusions: Using of Tris-soybean lecithin based extender at the level of 3.0% can be an appropriate alternative to either BullXcell or OptiXcell for Damascus goat sperm cryopreservation.
