With the increasing accumulation of plastic pollutants in various environments,research on microorganisms(including bacteria,fungi,and algae)with plastic degradation capabilities has gained significant attention.Howev...With the increasing accumulation of plastic pollutants in various environments,research on microorganisms(including bacteria,fungi,and algae)with plastic degradation capabilities has gained significant attention.However,only a limited number of microbial plastic-degrading enzymes have been identified to date.This highlights that the degradation mechanisms employed by many plastic-degrading microorganisms,particularly filamentous fungi,remain insufficiently explored.In this study,we utilized a versatile fungal plasmid(pCT74)to express green fluorescent protein(GFP)in a marine-derived fungus Alternaria alternata strain FB1 with plastic degradation capabilities.Upon evaluating the degradation effect of polyester-type polyurethane(PU)film,we observed that different transformants exhibited three kinds of activities(the same,reduced,or enhanced degradation capability)compared to the FB1 wild-type strain.Further analysis of the plasmid fragment insertion sites in different transformants revealed that pCT74 integrates randomly into the genome of the host fungus.Notably,a direct correlation was found between the plasmid insertion site and the degradation capability of the corresponding transformant.Our findings not only redefine the potential applications of plasmid pCT74 in filamentous fungi but also show a novel research approach to identifying key enzymes involved in plastic degradation by fungi.展开更多
Mycobacterium marinum can cause disease in both human beings and fishes.Infected fish exhibits symptoms commonly referred to as fish tuberculosis.In this research,we screened a 14-Membered Resorcylic Acid Lactones(RAL...Mycobacterium marinum can cause disease in both human beings and fishes.Infected fish exhibits symptoms commonly referred to as fish tuberculosis.In this research,we screened a 14-Membered Resorcylic Acid Lactones(RALs)library constructed in the laboratory for in vitro anti-Mycobacterium marinum activity and found that several compounds,including compounds 3,4,6,10,and 17–26,showed good activity.Notably,the trace natural product cochilimycin B(compound 3)possesses significant long-lasting inhibitory function to Mycobacterium marinum.It also shows synergistic effects with drugs like isoniazid,ethambutol,and bedaquiline(FICI≤0.5).In addition,cochilimycin B showed a significant inhibitory effect on the biofilm produced by Mycobacterium marinum,with an inhibition rate of 65.9%.To solve the bottleneck problem of natural product drug sources,we realized an efficient one-step semi-synthetic method to obtain compound 3 with a yield of 32.1%.In vivo,inflammatory changes in BALB/c mice infected with Mycobacterium marinum were characterized.After 12 h of infection,IL-1βwas 20%–30%lower in the spleen and tail of the3-treated group compared to the control group.In the thymus and tail of the treated group,TNF-αlevels were 30%–60%lower than in the control group.This is the first time that the significant anti-Mycobacterium marinum effect of cochilimycin B from 14-Membered Resorcylic Acid Lactones(14-RALs)was reported,offering a new chemical molecule for anti-Mycobacterium marinum drug research.展开更多
Ferroptosis of chondrocytes is a significant contributor to osteoarthritis(OA),for which there is still a lack of safe and effective therapeutic drugs targeting ferroptosis.Here,we screen for anti-ferroptotic drugs in...Ferroptosis of chondrocytes is a significant contributor to osteoarthritis(OA),for which there is still a lack of safe and effective therapeutic drugs targeting ferroptosis.Here,we screen for anti-ferroptotic drugs in Food and Drug Administration(FDA)-approved drug library via a high-throughput manner in chondrocytes.We identified a group of FDA-approved anti-ferroptotic drugs,among which vitamin K showed the most powerful protective effect.Further study demonstrated that vitamin K effectively inhibited ferroptosis and alleviated the extracellular matrix(ECM)degradation in chondrocytes.Intra-articular injection of vitamin K inhibited ferroptosis and alleviated OA phenotype in destabilization of the medial meniscus(DMM)mouse model.Mechanistically,transcriptome sequencing and knockdown experiments revealed that the anti-ferroptotic effects of vitamin K depended on growth arrest-specific 6(Gas6).Furthermore,exogenous expression of Gas6 was found to inhibit ferroptosis through the AXL receptor tyrosine kinase(AXL)/phosphatidylinositol 3-kinase(PI3K)/AKT serine/threonine kinase(AKT)axis.Together,we demonstrate that vitamin K inhibits ferroptosis and alleviates OA progression via enhancing Gas6 expression and its downstream pathway of AXL/PI3K/AKT axis,indicating vitamin K as well as Gas6 to serve as a potential therapeutic target for OA and other ferroptosis-related diseases.展开更多

The aim of this article is to present a unique interpretation of the holistic worldview,both in theory and in practice.It explores how this approach,along with a planning process fundamentally different from conventio...The aim of this article is to present a unique interpretation of the holistic worldview,both in theory and in practice.It explores how this approach,along with a planning process fundamentally different from conventional methods,was implemented in the design and construction of the Music Center and Library in the city of Tel Aviv.This process forms a coherent representation of a complete worldview:a humanistic,holistic worldview developed and adopted through more than five decades of architectural practice across all scales of design.In this approach,a building is not regarded as a collection of isolated design elements,but as one hierarchical language,in which the building,its interior,and its ornamented details down to the handle of the door is one continuous system.Within this system,the building,its interior spaces,and even the smallest ornamental details,down to the door handles,are conceived as parts of a single,continuous whole.This worldview aligns with contemporary scientific discourse in fields such as cosmology,neurobiology,psychology,complexity theory,and Buddhist philosophy,disciplines with which this body of work is closely associated.展开更多

Objective:To enhance the reading skills of clinical pathology residents,it is essential to establish a well-structured electronic pathology reading library.Methods:In accordance with the Resident Standardization Train...Objective:To enhance the reading skills of clinical pathology residents,it is essential to establish a well-structured electronic pathology reading library.Methods:In accordance with the Resident Standardization Training Content and Standards(2022 Edition),clinical pathology residents are required to master pathological diagnoses across 11 systems:skin,head and neck,mediastinum and respiratory,digestive,urinary and male reproductive,female reproductive and breast,lymphatic and hematopoietic,bone and soft tissue,cardiovascular,central nervous,and endocrine diseases.Senior pathologists specializing in each subspecialty selected classic pathological slides,which were systematically scanned and compiled into an electronic pathology library.Results:A questionnaire survey was conducted to gather feedback on the electronic pathology reading library.Residents generally found it to be convenient,efficient,and conducive to learning.Conclusion:Training in clinical pathology diagnosis is a core component of standardized resident training.The electronic pathology reading library has been well-received and recognized by resident doctors.However,further efforts are needed to explore diverse teaching methods that align with modern educational approaches,ultimately contributing to the development of highly skilled resident doctors.展开更多
The construction of a borehole correction library for the multi-component array induction tool in deviated boreholes involves extensive 3D forward modeling and typically requires significant computational resources.To...The construction of a borehole correction library for the multi-component array induction tool in deviated boreholes involves extensive 3D forward modeling and typically requires significant computational resources.To address this challenge,this paper proposes an efficient algorithm for the library construction based on the 3D finite volume method(FVM)and contraction high-order Born approximation(CHBA).First,the electromagnetic(EM)field solution region is divided into two symmetric subregions based on the symmetry of the correction library model and the EM field.Numerical solution on a single subregion,combined with the symmetry boundary extension technique,enhances the efficiency of the 3D numerical simulation.Second,three reference mud conductivities are selected based on the mud conductivity range,and the CHBA is applied to calculate the EM responses at all mud conductivity nodes rapidly.Third,the number of forward simulation operations is further reduced by exploiting the principle that models with different frequencies and different formation conductivities have equivalent EM responses.Numerical experiments demonstrate the correctness and feasibility of the proposed algorithm.Compared to conventional 3D modeling,the proposed algorithm achieves approximately a 20-fold speedup in library construction,effectively reducing computational resources and time consumption.展开更多

The textual expos...This article introduces several of the more significant service delivery innovations and their resulting accomplishments instigated by Dongguan Library(thereafter abbreviated as DGL) in recent years. The textual exposition of this paper is based on a case study of DGL by this author about its user-centered vision, mission, immediate objectives and the exhibited service performance within a contextual environment of collegial support from its professional peers. After a five-year period of intensive efforts on such focused professional development and practice, DGL has completed its information service mapping and information delivery for the entire municipality of Dongguan on a 7-day and twentyfour-hour(7/24) basis. This singular feast of accomplishment seems to suggest the moral that any significant development of scale of a municipal library has to be in keeping pace closely with the progress of the society at large in general and with the changing information demands of its local clientele in particular.展开更多

F...Digital libraries are complex systems and this brings difficulties for their evaluation. This paper proposes a hierarchical model to solve this problem, and puts the entangled matters into a clear-layered structure. Firstly, digital libraries(DLs thereafter)are classified into 5 groups in ascending gradations, i.e. mini DLs, small DLs, medium DLs,large DLs, and huge DLs by their scope of operation. Then, according to the characteristics of DLs at different operational scope and level of sophistication, they are further grouped into unitary DLs, union DLs and hybrid DLs accordingly. Based on this simulated structure,a hierarchical model for digital library evaluation is introduced, which evaluates DLs differentiatingly within a hierarchical scheme by using varying criteria based on their specific level of operational complexity such as at the micro-level, medium-level, and/or at the macro-level. Based on our careful examination and analysis of the current literature about DL evaluation system, an experiment is conducted by using the DL evaluation model along with its criteria for unitary DLs at micro-level. The main contents resulting from this evaluation experimentation and also those evaluation indicators and relevant issues of major concerns for DLs at medium-level and macro-level are also to be presented at some length.展开更多

文摘Objective:To enhance the reading skills of clinical pathology residents,it is essential to establish a well-structured electronic pathology reading library.Methods:In accordance with the Resident Standardization Training Content and Standards(2022 Edition),clinical pathology residents are required to master pathological diagnoses across 11 systems:skin,head and neck,mediastinum and respiratory,digestive,urinary and male reproductive,female reproductive and breast,lymphatic and hematopoietic,bone and soft tissue,cardiovascular,central nervous,and endocrine diseases.Senior pathologists specializing in each subspecialty selected classic pathological slides,which were systematically scanned and compiled into an electronic pathology library.Results:A questionnaire survey was conducted to gather feedback on the electronic pathology reading library.Residents generally found it to be convenient,efficient,and conducive to learning.Conclusion:Training in clinical pathology diagnosis is a core component of standardized resident training.The electronic pathology reading library has been well-received and recognized by resident doctors.However,further efforts are needed to explore diverse teaching methods that align with modern educational approaches,ultimately contributing to the development of highly skilled resident doctors.

文摘The construction of a borehole correction library for the multi-component array induction tool in deviated boreholes involves extensive 3D forward modeling and typically requires significant computational resources.To address this challenge,this paper proposes an efficient algorithm for the library construction based on the 3D finite volume method(FVM)and contraction high-order Born approximation(CHBA).First,the electromagnetic(EM)field solution region is divided into two symmetric subregions based on the symmetry of the correction library model and the EM field.Numerical solution on a single subregion,combined with the symmetry boundary extension technique,enhances the efficiency of the 3D numerical simulation.Second,three reference mud conductivities are selected based on the mud conductivity range,and the CHBA is applied to calculate the EM responses at all mud conductivity nodes rapidly.Third,the number of forward simulation operations is further reduced by exploiting the principle that models with different frequencies and different formation conductivities have equivalent EM responses.Numerical experiments demonstrate the correctness and feasibility of the proposed algorithm.Compared to conventional 3D modeling,the proposed algorithm achieves approximately a 20-fold speedup in library construction,effectively reducing computational resources and time consumption.

文摘As an important method of practical teaching,case-based teaching has become increasingly prominent in international Chinese language education.However,the development of case-based teaching for Chinese characters remains insufficient,with limited research outcomes,making it difficult to effectively support the teaching and research of Chinese character instruction.The establishment of a case library for international Chinese character teaching can provide a wealth of teaching cases,meeting the developmental needs of international Chinese language education in the new era.
文摘This article introduces several of the more significant service delivery innovations and their resulting accomplishments instigated by Dongguan Library(thereafter abbreviated as DGL) in recent years. The textual exposition of this paper is based on a case study of DGL by this author about its user-centered vision, mission, immediate objectives and the exhibited service performance within a contextual environment of collegial support from its professional peers. After a five-year period of intensive efforts on such focused professional development and practice, DGL has completed its information service mapping and information delivery for the entire municipality of Dongguan on a 7-day and twentyfour-hour(7/24) basis. This singular feast of accomplishment seems to suggest the moral that any significant development of scale of a municipal library has to be in keeping pace closely with the progress of the society at large in general and with the changing information demands of its local clientele in particular.
文摘Digital libraries are complex systems and this brings difficulties for their evaluation. This paper proposes a hierarchical model to solve this problem, and puts the entangled matters into a clear-layered structure. Firstly, digital libraries(DLs thereafter)are classified into 5 groups in ascending gradations, i.e. mini DLs, small DLs, medium DLs,large DLs, and huge DLs by their scope of operation. Then, according to the characteristics of DLs at different operational scope and level of sophistication, they are further grouped into unitary DLs, union DLs and hybrid DLs accordingly. Based on this simulated structure,a hierarchical model for digital library evaluation is introduced, which evaluates DLs differentiatingly within a hierarchical scheme by using varying criteria based on their specific level of operational complexity such as at the micro-level, medium-level, and/or at the macro-level. Based on our careful examination and analysis of the current literature about DL evaluation system, an experiment is conducted by using the DL evaluation model along with its criteria for unitary DLs at micro-level. The main contents resulting from this evaluation experimentation and also those evaluation indicators and relevant issues of major concerns for DLs at medium-level and macro-level are also to be presented at some length.

文摘As an important part of National Digital Library(NDL),the National Digital Library Project(NDLP),which was launched in 2005,has attracted wide attention of the society in general.This paper introduces the general situation of the project from the following aspects,namely,the characteristics,objectives and contents.It also expounds on the basic ideas and the overall structure of the project and gives an introduction to the design of its basic platform,its application platform,its business management system and its standardization control system.In this paper,the digital resources construction and service delivery of the National Library of China(NLC)are introduced to cover both its present operation as well as those in conjunction with the opening of its new library building during its second-phase of library development.Finally,the most important and difficult point in the construction of the National Digital Library Project is analyzed from the following four perspectives:1)The long-term preservation of digital resources;2)the construction of knowledge organization system;3)the organic integration of the digital library and the traditional library;and 4)the risk of project management and system integration.
