Dissipation behavior, residue distribution and dietary risk assessment of chlorfenapyr and clothianidin in leek using RRLC-QqQ-MS/MS technique 被引量:8

Commercial suspension emulsion(SE) of(8% chlorfenapyr + 20% clothianidin), as a pre-registered product in China, was firstly investigated under Chinese open-field conditions. A MWCNTs-based QuEChERS method for simultaneous determination of chlorfenapyr and clothianidin in leek was established and validated through rapid resolution liquid chromatography triples quadrupole tandem mass spectrometry(RRLC-QqQ-MS/MS). Based on this method, the dissipation behaviors, residue distributions and dietary risk probability of these fungicides in leek from two representative locations in China in 2016, which were previously treated with these insecticides at the doses of 420–630 g a.i./ha twice applications, were further investigated for food safety. Dissipation behavior of chlorfenapyr and clothianidin in leek followed first-order kinetics with the half-lives of 2.9–7.2 days, and the highest residues(HR) of these insecticides in leek were below 0.3811 mg/kg and 0.2989 mg/kg, respectively, at the pre-harvest interval(PHI, 7 days). For dietary risk assessments, the risk quotients(RQs) of chlorfenapyr and clothianidin in leek were evaluated by comparing the value of national estimated daily intake(NEDI) with acceptable daily intake(ADI), based on the trials data under good agricultural practices(GAP) conditions. The results indicated that RQs of chlorfenapyr and clothianidin in leek were21.5% and 0.29%, respectively, which exhibited an acceptably low health risk to leek consumption. The current study could not only guide reasonable usage of the formulation, but also facilitate the setting of maximum residue limits(MRLs) of chlorfenapyr and clothianidin in leek for Chinese 展开更多

To optimize the leek peeling performance, a new nozzle has been developed in which the nozzle has a design Mach number of 1.68, an inner diameter of 2.0 mm at the throat, and an inner diameter of 2.3 mm at the exit. Experiments have been conducted over a range of nozzle pressure ratios from 3.0 to 6.0. Flow field issued from the new nozzle is quantitatively visualized by the rainbow schlieren deflectometry and compared with that from a conventional nozzle. Density fields in the free jets are reconstructed by the Abel inversion method for the schlieren images with the horizontal rainbow filter. The density values at the exit of the conventional nozzle obtained by the rainbow schlieren are compared with the analytical results by the flow model proposed in the past. In addition, Pitot probe surveys along the jet centerline were made to obtain the impact pressure distributions. The Mach number and velocity distributions along the jet centerline are obtained from a combination of the density and Pitot pressure data to clarify the fundamental flow structure of leek peeler nozzle jets. 展开更多

[Methods] The species of thrips on leek were identified in Sichuan Province, and the damage symptoms, occurrence regularity and control methods of thrips in leek field were studied. [Result] The morphological characteristic of thrips on leek was Frankliniella occidentalis(Pergande). ITS molecular identification results showed that the amplified fragment was 520 bp, which shared the similarity of 99.81% with the base se-quence of F. occidentalis. F. occidentalis concentrated at the height of 80 cm from the ground; in sunny days, the occurrence frequency of F. occidentalis was relatively high during 15:00 and 17:00; rainwater had obvious lethal effect on F. occidentalis, and the lethal rate was 29.30%. Field trial showed that 20% dinotefuran SG and 40% acetamiprid GR had good control effect against F. occidentalis. The control effect of 20% dinotefuran SG 1 500 times dilution was the best, up to 92.56%. [Conclusion] The results will provide effective technical support for large-scale control of thrips in leek field. 展开更多

Background: The pharmacological properties of Allium family have been reported to include antibacterial, antioxidant, anticancer, anti-aging and antilipidemic properties. This investigation was conducted to evaluate the antibacterial properties of ethanolic and aqueous extracts of Allium sativum and Allium porrum on both Gram-positive and Gram-negative organisms from chronic wound infections. Methods: Ten (10) isolates were obtained from infected open wounds from patients at Chukwuemeka Odumegwu Ojukwu University teaching Hospital Amakwu and Nnamdi Azikiwe University Teaching Hospital Nnewi, Anambra state, Nigeria. Gram reaction and other biochemical tests namely: Indole, Citrate, Catalase, coagulase and oxidase tests were done for identification of the isolates. The isolates are K. pneumoniae (three), S. aureus (two), Enterococcus (two), P. aeruginosa (two) and E. coli (one). The antibacterial properties of ethanolic and aqueous extracts of Allium sativum and Allium porrum were determined using the agar well diffusion method while Gentamicin (10 mcg) and Tetracycline (30 mcg) were used as positive controls. Result: The results of the study showed that the ethanolic extract of Allium sativum (500 mg/ml and 250 mg/ml) respectively had an inhibitory effect on all the bacteria isolates under study, except a strain of P. aeruginosa which was also resistant to Allium porrum and the conventional antibiotics (Gentamicin and Tetracycline) used as positive controls. The ethonolic extract of Allium porrum (500 mg/ml and 250 mg/ml) respectively inhibited all the bacteria under investigation except one strain of P. aeruginosa and E. coli. Worthy of note is that the extracts of both plants inhibited two (2) strains of K. pneumoniae and one (1) strain of S. aureus that were resistant to Gentamicin and Tetracycline. The combination of these two plant extracts did not produce any synergistic effects. Conclusion: The present study provides evidence that extracts of Allium sativum and Allium porrum possess antibacteria properties and could serve as alternatives to conventional antibiotics in the treatment of bacterial infections from wounds. 展开更多

Antibacterial Effects of Water Extracts of Various Broadleaf Wild Leeks (Allium ampeloprasum)

There are numerous reports regarding the antimicrobial effects of herbal extracts on bacteria and fungi. We have prepared water extracts of Wild Leek (A. ampeloprasum var. porrum), Kurrat (A. ampeloprasum var. kurrat) and Elephant Garlic (A. ampeloprasum var. ampeloprasum) by rotary vacuum distillation. As a result, the extraction was carried out in 40 ℃ thus omitting any risk of loss of anti-bacterial activity due to high temperatures reported in prior similar studies [ 1]. The five bacteria Bacillus subtilis, Pseudomonas aeruginosa, Escherichia coli, Enterococcus faecalis, and Staphylococcus aureus were tested against the extracts. While some extent of inhibition was observed when the bacteria were cultured in presence of the leek and kurrat extracts, elephant garlic was determined to possess an extreme inhibitory quality against all the aforementioned bacteria, completely inhibiting the growth of the bacteria after 24 hrs of culture in exposure to the extract. We propose that elephant garlic extract could be an effective alternative to chemical antibiotic treatments and disinfectants in hospital and other settings in which infections are common. 展开更多

针对目前韭菜收割机在收割韭菜时会出现收割不完整、收割效率低和无法有效收割倒伏韭菜等现象,为提高韭菜的收割效率及完整度,结合韭菜风力敏感特性与种植模式,设计一款气力扶禾装置。在传统锥形筒扶禾装置的基础上设置出风口,以传统直... 针对目前韭菜收割机在收割韭菜时会出现收割不完整、收割效率低和无法有效收割倒伏韭菜等现象,为提高韭菜的收割效率及完整度,结合韭菜风力敏感特性与种植模式,设计一款气力扶禾装置。在传统锥形筒扶禾装置的基础上设置出风口,以传统直接接触的物理扶禾为主,气力间接接触为辅,完成扶禾作业。运用Standard k-epsilon湍流模型,采用有限元软件分析影响出风口速度的3个主要因素:出风口角度、直径与总数量,寻求最佳参数组合。以韭菜倒伏率、茎叶损失率为评价指标,进行田间收割试验。结果表明,当出风口角度为30°、出风口直径为8 mm、出风口总数量为8个时,扶禾效果最好;与传统扶禾装置相比,新型气力扶禾装置的韭菜倒伏率降低3.071%,茎叶损失率降低1.691%,总体满足韭菜的收割要求。研究结果为风力扶禾装置的开发提供理论和试验依据。 展开更多

【目的】花的开放过程伴随着各种花器官,特别是花瓣的协调运动,然而花瓣运动和花朵开放的机制尚不清楚。本研究通过对百合和花韭花朵开放的物理过程和表皮细胞解剖观察,探讨两者花瓣开放运动机理与解剖结构的异同。【方法】以东方百合... 【目的】花的开放过程伴随着各种花器官,特别是花瓣的协调运动,然而花瓣运动和花朵开放的机制尚不清楚。本研究通过对百合和花韭花朵开放的物理过程和表皮细胞解剖观察,探讨两者花瓣开放运动机理与解剖结构的异同。【方法】以东方百合“西伯利亚”和花韭“玫红”为试验材料,结合解剖学和生理学方法,测量不同开放时期的花瓣长度,统计细胞数量,并观察细胞的形态变化,分析影响花朵开放的影响因素。【结果】百合花朵开放过程分为5个时期(S1~S5),S1—S5期花瓣长度逐渐增长且细胞形态发生明显变化。S1—S2期细胞数量明显增长且内表皮细胞数量增长显著大于外表皮,但S2—S5期细胞数量增长不明显。花韭花朵开放过程分为3个过程(T1~T3),T1—T3期花瓣长度和细胞形态均无明显变化,在T1和T3时期,内表皮细胞数量大于外表皮细胞数量,但T2期则相反。【结论】百合内外表皮细胞数量在S2期存在明显差异,促使百合花瓣突然绽放,S3、S4和S53个时期内外表皮细胞数量差异较小,可能是由于细胞的膨胀生长导致花瓣进一步开放,直到花朵枯萎凋谢;花韭在T2时期外表皮细胞数量明显大于内表皮,在T3时期内表皮细胞数量明显大于外表皮,推测花瓣两侧细胞数量不对称增长是导致花韭在T2期花朵开放T3期闭合的原因。 展开更多

基于Box—Behnken设计的韭菜收割机工作参数优化与田间性能验证

针对韭菜机械化收获过程中存在的植株损伤率高、漏割损失大等关键问题,以手扶式韭菜收割机为作业机具,以韭菜为作业对象,基于作物—机械互作机理开展收获参数优化研究。采用Box—Behnken试验设计构建三因素三水平正交试验方案,系统分析... 针对韭菜机械化收获过程中存在的植株损伤率高、漏割损失大等关键问题,以手扶式韭菜收割机为作业机具,以韭菜为作业对象,基于作物—机械互作机理开展收获参数优化研究。采用Box—Behnken试验设计构建三因素三水平正交试验方案,系统分析前进速度(0.40~0.60 m/s)、割刀转速(1 800~2 200 r/min)和输送速度(0.48~0.72 m/s)对损失率与损伤率的影响规律。通过二次回归正交试验建立多因素交互作用的数学模型,并利用响应面分析法优化参数组合。验证试验表明,最佳参数组合:前进速度为0.50 m/s、割刀转速为2 000 r/min、输送速度为0.60 m/s,此时损失率与损伤率分别为4.71%、2.26%,研究揭示关键参数交互作用对收获性能的影响机制,可为韭菜收割装备的智能化控制系统开发提供理论支撑。研究结果可为韭菜收割机械的结构改进与参数优化提供理论依据,提出参数优化方法对叶菜类作物收获机械的农艺适配性提升具有普适性参考价值,有助于推动设施蔬菜生产全程机械化发展。对提升蔬菜收获装备的农艺适应性具有实际意义。 展开更多

为探究不同栽培模式(水培、土培和基质培)对韭菜采后贮藏品质及风味物质的影响,采用水培、土培和基质培3种栽培方式种植‘航研791’韭菜,采后放入20℃、相对湿度80%~85%的冷库中贮藏,测定其贮藏期间生理品质和风味物质的变化。结果表明... 为探究不同栽培模式(水培、土培和基质培)对韭菜采后贮藏品质及风味物质的影响,采用水培、土培和基质培3种栽培方式种植‘航研791’韭菜,采后放入20℃、相对湿度80%~85%的冷库中贮藏,测定其贮藏期间生理品质和风味物质的变化。结果表明:土培和基质培栽培方式能够较好地维持韭菜在贮藏期间的质量损失率和腐烂率,水培韭菜则质量损失率和腐烂率都较高,与水培相比,土培和基质培韭菜能够较好地维持VC和可溶性蛋白含量,具有较高的膳食纤维含量,土培还能促进纤维素、半纤维素、果胶和木质素含量的积累,基质培韭菜的木质素呈下降趋势,水培韭菜在贮藏过程中纤维素含量呈下降趋势,木质素含量则基本不变。韭菜的风味物质主要是醛类、醇类、酯类、醚类、杂环类化合物等,α-松油醇、丁酸异戊酯、水杨酸甲酯和苯乙醛等物质是3种韭菜贮藏前后的主要风味物质。土培韭菜能够在贮藏过程中更好地维持韭菜的品质和风味物质,耐贮性最强,其在贮藏过程中品质劣变以衰老为主,基质培次之,水培韭菜则耐贮性最差,品质劣变以软化腐烂为主。 展开更多

该研究采用Illumina miseq高通量测序技术对红酸汤和韭菜红酸汤微生物菌群多样性及群落结构进行分析。结果表明,韭菜红酸汤样品的细菌菌群相对丰度高于红酸汤样品,多样性低于红酸汤样品;真菌菌群则相反。两种红酸汤样品的微生物群落存... 该研究采用Illumina miseq高通量测序技术对红酸汤和韭菜红酸汤微生物菌群多样性及群落结构进行分析。结果表明,韭菜红酸汤样品的细菌菌群相对丰度高于红酸汤样品,多样性低于红酸汤样品;真菌菌群则相反。两种红酸汤样品的微生物群落存在一定的差异,差异主要体现在相对丰度上。从红酸汤样品中共注释到8个细菌门、4个真菌门,从韭菜红酸汤样品中共注释到18个细菌门、3个真菌门,优势细菌门(相对丰度>1%)均为厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)、蓝菌门(Cyanobacteria),优势真菌门均为子囊菌门(Ascomycota)、担子菌门(Basidiomycota)。从红酸汤样品中共注释到43个细菌属、25个真菌属,从韭菜红酸汤样品中共注释到103个细菌属、18个真菌属,优势细菌属(相对丰度>1%)均为乳酸杆菌属(Lactobacillus)、迟缓乳杆菌属(Lentilactobacillus)、片球菌属(Pediococcus)、小坂菌属(Kosakonia)、乳酪杆菌属(Lacticaseibacillus)、Lactiplantibacillus;优势真菌属均为酵母属(Saccharomyces)、糙缘腺革菌属(Amphinema)、毕赤酵母属(Pichia)、Ascomycota_gen_Incertae_sedis、Tetracladium、哈萨克斯坦酵母属(Kazachstania)。 展开更多

One of the most important physicochemical properties of meat is its tenderness.The impact of Leek (Allium ampeloprasum ) extract and ultrasound on meat quality and molecular weight distribution of Longissimus lumborum muscles was studied.Samples have been treated with ultrasound (100 and 300 W during 10,20,and 30 min) and Leek-derived exogenous proteases (16.9 and 33.8U/g) simultaneously and separately.The combined effect of enzyme and ultrasound in 10 and 20 min increased the pH value.It was also increased the proteolytic activity of meat.Regarding meat quality,the filtering residues,cooking loss,shear force,and hardness were reduced;water-binding capacity,emulsion capacity,and emulsion stability were improved.The electrophoretic pattern of myofibrillar proteins showed the muscle fibers were severely degraded in the combined treatments.It can be concluded that this process could be used as an alternative to the chemical tenderizing agents in the meat industry. 展开更多

本文建立了一种基于QuEChERS-气相色谱串联三重四极杆质谱的检测方法,用于分析韭菜中五氯硝基苯、毒死蜱、联苯菊酯等5种农药残留。以阴性韭菜为基质,有效排除了基质干扰,消除了外在因素对检测结果的影响。样品经1.0%甲酸酸化的乙腈溶... 本文建立了一种基于QuEChERS-气相色谱串联三重四极杆质谱的检测方法,用于分析韭菜中五氯硝基苯、毒死蜱、联苯菊酯等5种农药残留。以阴性韭菜为基质,有效排除了基质干扰,消除了外在因素对检测结果的影响。样品经1.0%甲酸酸化的乙腈溶液提取,并采用QuEChERS法进行净化处理,气质联用仪检测。结果表明,5种农药在5~200 ng·mL^(-1)均表现出良好的线性关系,相关系数R~2均大于0.999 0。加标回收率在82.84%~104.11%,相对标准偏差为3.2%~8.5%。该方法准确、可靠,可满足日常检测要求,适用于韭菜中农药残留检测。 展开更多
