Background In a previously identified locus linked to hypertension on chromosome 15q, we identified three blood pressure candidate genes: insulin-like growth factor 1 receptor gene (IGF1R), myocyte specific enhancer f...Background In a previously identified locus linked to hypertension on chromosome 15q, we identified three blood pressure candidate genes: insulin-like growth factor 1 receptor gene (IGF1R), myocyte specific enhancer factor 2A gene (MEF2A), and paired basic amino acid cleaving enzyme 4 gene (PACE4). In this study, we te sted their associations with hypertension using haplotype analysis.Methods A total of 288 unrelated individuals, including 163 high diastolic blood pressure (DBP) subjects and 125 normal DBP subjects were enrolled in this case-control study. Twenty single nucleotide polymorphisms (SNPs) in the three genes were genotyped using polymerase chain reaction followed by restriction enzyme digestion. Haplotype analysis was accomplished in the following stages: (1) pair-wise linkage disequilibrium test among SNPs on the same gene was performed to explore blocks in which recombination is very unlikely to happen; (2) Estimation-Maximization algorithm was applied to estimate haplotype frequencies in each block; (3) the chi-square test was used to examine the specific haplotype difference, and a permutation test was used to examine the overall haplotype profile difference between cases and controls in each block.Results An estimated haplotype “CCCCG” frequency in the haplotype block on the PACE4 gene was significantly higher in high DBP cases than in controls (P<0.01). The overall estimated haplotype profile in this block was also significantly different between the cases and the controls (P<0 .001). This association indicates. Conclusions This study for the first time demonstrated that PAC E4 gene may play an important role in the regulation of DBP. This association indicates that variations influencing DBP resides in or near this genomic region.展开更多

The Solanum genus is known for its diverse bioactive compounds,yet its diuretic potential remains understudied.This research commenced with an analysis of polyphenol and flavonoid content in Solanum elaeagnifolium lea...The Solanum genus is known for its diverse bioactive compounds,yet its diuretic potential remains understudied.This research commenced with an analysis of polyphenol and flavonoid content in Solanum elaeagnifolium leaf extract(SEFE)using colorimetric techniques,followed by HPLC-DAD to delineate its chemical composition.The aqueous extract revealed prominent constituents:naringin(12.38%),quercetin 3-O-B-D-Glucoside(27.25%),and flavone(15.26%).A 15-day study on normal rats investigated the diuretic potential of SEFE at repeated doses.SEFE significantly increased urine volume and urinary sodium/potassium levels without inducing hypokalaemia,contrasting with furosemide,a standard diuretic that induced hypokalaemia.Conversely,furosemide,a standard diuretic,increased urinary sodium and potassium while inducing hypokalaemia.It was evident that the diuretic effect of S.elaeagnifolium is dose-dependent,with a dosage of 500 mg/kg body weight exerting a more potent diuretic effect compared to furosemide.The diuretic activity of this plant was supported by an in silico study of the diuretic effect.The findings demonstrate how S.elaeagnifolium leaves have a potent diuretic impact on rats.However,more in-depth studies are needed to examine the following aspects:identifying the specific molecules responsible for the diuretic effect,understanding the molecular and cellular mechanisms underlying this activity,and assessing the long-term safety and clinical efficacy of this plant in different contexts.展开更多

The potherb mustard Xuecai(XC)cultivar is a cruciferous vegetable that is popular either fresh or pickled.Due to the deep notches in the edges of leaves in mustard XC,this plant can be said to have multipinnately lobe...The potherb mustard Xuecai(XC)cultivar is a cruciferous vegetable that is popular either fresh or pickled.Due to the deep notches in the edges of leaves in mustard XC,this plant can be said to have multipinnately lobed leaves.The net photosynthesis of lobed leaves is significantly greater than that of simple leaves.However,the molecular mechanism of leaf shape variation has not been determined.Here,we used HiFi and Hi-C data to assemble the XC genome.The genome was 961.72 Mb in size,with a contig N50 value of 6.565 Mb.The XC genome was compared with four previously sequenced mustard genomes,and the genomic collinearity regions,SNPs,and indels were identified.Five BjRCO genes were found on chromosome(Chr.)A10 in potherb mustard XC when the BjRCO gene locus was compared against other sequenced B.juncea genomes.Segmental duplication was found to contribute to the BjRCO gene copy number.The transcript expression of BjRCO genes was greater in multipinnately lobed leaves than in sawtooth-like leaves.Together,these findings indicate that both the greater copy number and the expression level of BjRCO genes regulate leaf shape from simple to complex in B.juncea.Gene editing of the BjRCO gene from XC changed the leaf shape from multipinnately lobed to simple.The high-quality XC genome sequence not only provides new insight into B.juncea leaf-type genomics but also helps in deciphering leaf shape variation.Our study provides insights into the variation and evolution of important traits in Brassica plants through a comparative analysis of the sequenced genomes.展开更多

Laser-Induced Graphene (LIG) is regarded as a promising sensor carrier due to its inherent three-dimensional porous structure. However, as two mutually exclusive properties of the pressure sensor, sensitivity and work...Laser-Induced Graphene (LIG) is regarded as a promising sensor carrier due to its inherent three-dimensional porous structure. However, as two mutually exclusive properties of the pressure sensor, sensitivity and working range are difficult to be further improved by the single porous structure. Inspired by the unique geometry of Oxalis corniculata L. leaves, we here propose a novel method consist of laser pre-etching and inducing steps to fabricate LIG-based electrodes with a two-stage architecture featuring microjigsaw and microporous structures. The following injection of liquid-silicone significantly improves the friction resistance and bending reliability of LIG materials. The interface contact between external microjigsaw structures induces substantial resistance changes, and the internal microporous structure exhibits reversibility during dynamic deformation. Consequently, the jigsaw-like pressure sensor achieves a balanced performance with sensitivities of 3.64, 1.20 and 0.03 kPa^(- 1) in pressure range of 0 -20, 20 - 40 and 40 - 150 kPa, respectively. The bionic LIG-based pressure sensor serves as the core component and further integrated with an all-in-one wireless transmission system capable of monitoring various health parameters such as subtle pulse rates, heartbeat rhythms, sounds, etc., indicating broad prospects in future wearable electronics.展开更多
BACKGROUND Upper respiratory tract infections(URTIs)are one of the most frequent causes of childhood school leave and morbidity.AIM To study the present trend of medications’prescribing pattern utilized in URTIs amon...BACKGROUND Upper respiratory tract infections(URTIs)are one of the most frequent causes of childhood school leave and morbidity.AIM To study the present trend of medications’prescribing pattern utilized in URTIs among the pediatric population attending outpatient clinics in pediatric hospitals.METHODS This analytical observational cross-sectional research was conducted in 200 children aged 1–10 years with URTIs attending the pediatric outpatient clinics in pediatric hospitals,one of which is an educational hospital,from July 2018 to August 2020.RESULTS Most of the prescriptions in our study included antibiotics(116/58%).The most commonly prescribed antibiotic family was ampicillin/sulbactam or amoxici-llin/clavulanic acid(53/26.5%),followed by first-generation cephalosporin(25/12.5%)and third-generation cephalosporin(20/10%).Macrolides and second-generation cephalosporins were prescribed less frequently,in 16(8%)and 2(1%)patients,respectively.Most of our study population(155/77.5%)was satisfied with their prescriptions,whereas the rest of the study population(45/22.5%)was unsatisfied.CONCLUSION Overprescription of antibiotics is a significant issue among clinicians in pediatric outpatient clinics.Stewardship of drugs,particularly antibiotics,is a must to prevent the development of drug resistance.Most cases of URTIs were treated in accordance with the existing national treatment guidelines.展开更多
The aromatic Baeckea frutescens leaves(BFL)known as“Gang-Song-Cha”is a popular drink(Tea)in South China.To explore the healthcare value,a systematic and in-depth study on BFL alleviating rheumatoid arthritis(RA)was ...The aromatic Baeckea frutescens leaves(BFL)known as“Gang-Song-Cha”is a popular drink(Tea)in South China.To explore the healthcare value,a systematic and in-depth study on BFL alleviating rheumatoid arthritis(RA)was carried out by integrating network pharmacology,molecular docking,and experimental verification.The efficacy was first evaluated,and the BFL extract could ameliorate RA by inhibiting articular swelling,lowering immune organ index,improving the ankle pathologies,and regulating serum levels of tumor necrosis factorα(TNF-α),interleukin(IL)-6,and IL-10 in CIA rats.Then the anti-RA mechanism of BFL was investigated by leveraging the high-content and active component 5-hydroxy-7-methoxy-2-isopropylchromone(Chro),and the potential signaling pathway and related target proteins for Chro acting on RA were analyzed by Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment and docking simulation.Finally,the experimental validations were conducted on rheumatoid arthritis fibroblast-like synoviocytes(RAFLS),and compound Chro could inhibit cell invasion and migration,block cell cycle at G2/M phase and induce apoptosis,and reduce the secretion of inflammatory cytokines by suppressing the PI3K-Akt pathway.These results provided pharmacological basis for the folk usage of“Gang-Song-Cha”for intervening RA.展开更多

文摘The aromatic Baeckea frutescens leaves(BFL)known as“Gang-Song-Cha”is a popular drink(Tea)in South China.To explore the healthcare value,a systematic and in-depth study on BFL alleviating rheumatoid arthritis(RA)was carried out by integrating network pharmacology,molecular docking,and experimental verification.The efficacy was first evaluated,and the BFL extract could ameliorate RA by inhibiting articular swelling,lowering immune organ index,improving the ankle pathologies,and regulating serum levels of tumor necrosis factorα(TNF-α),interleukin(IL)-6,and IL-10 in CIA rats.Then the anti-RA mechanism of BFL was investigated by leveraging the high-content and active component 5-hydroxy-7-methoxy-2-isopropylchromone(Chro),and the potential signaling pathway and related target proteins for Chro acting on RA were analyzed by Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment and docking simulation.Finally,the experimental validations were conducted on rheumatoid arthritis fibroblast-like synoviocytes(RAFLS),and compound Chro could inhibit cell invasion and migration,block cell cycle at G2/M phase and induce apoptosis,and reduce the secretion of inflammatory cytokines by suppressing the PI3K-Akt pathway.These results provided pharmacological basis for the folk usage of“Gang-Song-Cha”for intervening RA.
