[ Result ] The extract 1 had fairly strong in vitro bacteriostasis activities and than the extract2 and extract3. The MIC and MBC was 3.91 mg/ml and 31.25 mg/ml on S. aureus and E. coil [ Conclusion] The result showed that the different extract from Ilex Kudingcha C. J. Tseng had varying amount of different bacteriostatic activities,the Ilex Kudingcha C. J. Tseng was developed new and safe bacteriostatic reagent to provide reference.展开更多

The random amplified polymorphic DNA marker (RAPD) was applied to detect the genetic relationships and diversity among 21 germplasm materials of Kudingcha species in Oleaceae, which involved 8 species, i.e., Ligustr...The random amplified polymorphic DNA marker (RAPD) was applied to detect the genetic relationships and diversity among 21 germplasm materials of Kudingcha species in Oleaceae, which involved 8 species, i.e., Ligustrum robustum (Roxb.) Blume, L. henryi Hemsl., L. japonicum Thunb, L. japonicum Thunb. var. pubscens Koidz, L. lucidum Ait., L. pedunculare Rehd, Osmanthus masumuranus Hayata, and L. delavayanm Hariot. 20 RAPD primers selected were applied for the amplification on the 21 germplasm materials mentioned above. 427 bands were obtained, and the percentage of polymorphic bands (PPB) was 97.7%. The genetic similarity coefficients (GS) ranged from 0.1522 to 0.8322 with an average of 0.5466. There was a significant genetic difference among germplasm materials of Kudingcha species in Oleaceae, and UPGMA cluster based on the GS of RAPD could distinguish all test germplasm materials clearly and indicated the relationship of the 8 species mentioned above, all of which indicated that RAPD markers could be used for the studies of genetic diversity and relationship and classification of germplasm resources of Kudingcha species in Oleaceae. Analysis results of RAPD showed that L. japonicum Thunb. var. pubscens Koidz has closer genetic relationship with L. pedunculare Rehd and further genetic relationship with L. japonicum Thunb. among all tested species. The authors suggest that further research is needed to study whether L. japonicum Thunb. var. pubscens Koidz should be classified into a variata of L. japonicum Thunb, or should be considered as an independent species. The analysis results supported that L. pururascens Y. C. Yang should be combined into L. robustum (Roxb.) Blume.展开更多
This research the preventive effect of Kudingcha polyphenols(Kp)on colitis based on animal experiments.Experimental mice divided into four groups,including normal group,model group,low-concentration Kp(LKP)group and h...This research the preventive effect of Kudingcha polyphenols(Kp)on colitis based on animal experiments.Experimental mice divided into four groups,including normal group,model group,low-concentration Kp(LKP)group and high-concentration Kp(HKP)group,and they all smeared and given 0.15 mL 1%oxazolone solution by lavage to induce BALB/c mice colitis.DAI,colon weight/length ratio,serum levels of cytokines,related antioxidant activities of colon tissues such,and the mRNA expressions.The experimental results show that KP can significantly(p<0.05)reduce DAI in mice with colitis and inhibit shortening of colon caused by colitis,increase colon weight/length ratio.It can also significantly reduce(p<0.05)the activities of MPO,NO,MDA in mice colon tissues with colitis,increase the activity of GSH,and significantly increase(p<0.05)the level of IL-2 and decrease the level of IL-10 in mice serum with colitis.The results of qPCR assay show that Kp can significantly increase(p<0.05)the expression of Cu/Zn-SOD,Mn-SOD,GSH-Px,CAT,c-Kit,SCF and decrease the expression of IL-8,CXCR2 in mice colon tissues with colitis.Kp has a good preventive effect on colitis,and high concentration of PT has better preventive effect.展开更多
Ethanol supernatant of Ilex kudingcha methanol extract(ESIKME)was identified to be rich in phenolic com-pounds and inhibited murine squamous cell carcinoma(SCC)-7 growth through slightly restoring adaptive im-munity a...Ethanol supernatant of Ilex kudingcha methanol extract(ESIKME)was identified to be rich in phenolic com-pounds and inhibited murine squamous cell carcinoma(SCC)-7 growth through slightly restoring adaptive im-munity and Th1-inclination immune balance in the SCC7-bearing mice.To further evaluate the inhibitory effects and mechanisms of ESIKME against SCC7-induced inflammation status,the effects of ESIKME on inflammatory stress and related cachexia induced by SCC7 cells were investigated in vitro and in vivo.Chemical components of ESIKME were measured using an advanced LC-MS/MS analysis.SCC7 cells were treated with mouse splenocyte(SCM)and macrophage conditioned media(MCM)cultured without/with ESIKME in vitro.SCC7-bearing mice were orally administered with ESIKME for 26 days in vivo.Cachexia-related biomarkers in the SCC7-bearing mice were analyzed.Most of the detected compounds in ESIKME were polymeric or oxidized phenolic compounds.Either SCM or MCM without ESIKME significantly inhibited SCC7 cell growth.The high-dose ESIKME-admin-istered group(400 mg/kg b.w./day)had significantly higher body weights,feed efficiency,and energy efficiency than the dietary control(DC)group,suggesting non-adverse toxic effects for ESIKME supplementation in the SCC7-bearing mice.The DC group had a higher TNF-αand TNF-α/IL-10 secretion ratio but lower anti-inflammatory IL-10 cytokine secretions by macrophages than the vehicle control group.ESIKME administra-tion dose-dependently and significantly reversed these adverse statuses.Medium dose ESIKME(100 mg/kg b.w./day)decreased tumor sizes but increased gastrocnemius weights and foreleg muscle grip strength.Our results suggest that ESIKME alleviated inflammatory stress and related cachexia in vitro and in vivo.展开更多
