Background:Attention deficit hyperactivity disorder(ADHD)is one of most common disorders in child and adolescent psychiatry.Jingling oral liquid,a traditional Chinese patent medicine,is frequently used in clinical pra...Background:Attention deficit hyperactivity disorder(ADHD)is one of most common disorders in child and adolescent psychiatry.Jingling oral liquid,a traditional Chinese patent medicine,is frequently used in clinical practice for treating ADHD,and its therapeutic efficacy is widely recognized.However,its underlying mechanisms are not fully understood.Objective:This study used network medicine and metabolomics to investigate the mechanisms by which Jingling oral liquid alleviates ADHD.Methods:First,dopamine levels in the brain tissue of Wistar-Kyoto and spontaneously hypertensive rats were measured using high-performance liquid chromatography with a fluorescence detector(HPLC-FLD).Second,the Kunming mice were used to analyze the ingredients that reached the brain.The ingredients and targets of Jingling oral liquid for improving ADHD were analyzed through network medicine,and nontargeted metabolomics was used to screen differentially abundant metabolites in brain tissue and analyze possible metabolic pathways.Results:The medium-and high-dose Jingling oral liquid groups showed significantly increased dopamine levels in rat brain tissue.The combined analysis of network medicine and metabolomics revealed that quercetin,berberine,tenuifolin,phellodendrine and gallic acid in Jingling oral liquid regulate 12 key proteins,including APP,BDNF,DRD1-4,and 5-HTR2C.These compounds modulate four metabolites,namely dopamine,γ-aminobutyric acid(γGABA),calcitriol,and N-acylsphingosine,which are involved in four metabolic pathways,butanoate metabolism,sphingolipid metabolism,tyrosine metabolism and steroid biosynthesis,to treat ADHD.展开更多
