Niobium carbide MXene quantum dots(Nb2 C MQDs)derived from 2 D Nb2 CTx(MXene)are the rising-star material recently.Herein,a sulfur and nitrogen co-doped Nb2 C MQDs(S,N-MQDs)were synthesized through a hydrothermal method.The obtained Nb2 C MQDs have excellent green fluorescence with a quantum yield(QY)of 17.25%.In addition,they exhibited excitatio n-dependent photoluminescence,antiphotobleaching and dispersion stability.They emit light at 520 nm when excited at 390 nm.The Nb2 C MQDs could be successfully applied to copper ion detection with detection limit of 2μmol/L and Caco-2 cells imaging. 展开更多

OBJECTIVE:To investigate the impact of electro-acupuncture at the Neiguan(PC 6) acupoint on protein and RNA expression of acid-sensing ion channel 2(ASIC2) and ASIC3 in myocardial ischemia rats.METHODS:Fifty male Sprague-Dawley rats were used,weighing(230 ± 50) g.The rats were randomized into a normal group A,model group B,Neiguan(PC 6) group C,Lieque(LU 7) group D,and A-shi points group E.There were 10 rats in each group.Rats were continuously administered 85 mg/kg intravenous isoproterenol daily to establish the model.Successfully modeled rats in groups C,D,and E were given electro-acupuncture treatment.Each group of rats was sacrificed with chloral hydrate(1 mL/100 g) intraperitoneal injection.The left ventricular myocardium was extracted and placed at- 70 ℃ until use.Western blot analysis and real-time PCR were performed to assay protein and RNA expressions of ASIC2 and ASIC3,respectively.Fold changes in RNA expression were quantified with the 2~^(-△△Ct) method.Blood samples were drawn from the aorta abdominalis and tested for creatine kinase-MB(CK-MB) and lactate dehydrogenase(LDH) levels using enzyme-linked immunosorbent assay.RESULTS:Myocardial ischemia rats given electro-acupuncture at the Neiguan(PC 6) acupoint had significantly lower protein and RNA expression of ASIC2 and ASIC3,and CK-MB and LDH levels,compared with model rats(P < 0.01).CONCLUSION:Electro-acupuncture at the Neiguan(PC 6) acupoint can not only decrease the protein and RNA expression of ASIC2 and ASIC3,but also inhibit the opening of ASICs and reduce the cardiomyocyte damage in myocardial ischemia rats. 展开更多

Modern medicine is increasingly interested in advanced sensors to detect and analyze biochemical indicators.Ion sensors based on potentiometric methods are a promising platform for monitoring physiological ions in biological subjects.Current semi-implantable devices are mainly based on single-parameter detection.Miniaturized semi-implantable electrodes for multiparameter sensing have more restrictions on the electrode size due to biocompatibility considerations,but reducing the electrode surface area could potentially limit electrode sensitivity.This study developed a semi-implantable device system comprising a multiplexed microfilament electrode cluster(MMEC)and a printed circuit board for real-time monitoring of intra-tissue K^(+),Ca^(2+),and Na^(+)concentrations.The electrode surface area was less important for the potentiometric sensing mechanism,suggesting the feasibility of using a tiny fiber-like electrode for potentiometric sensing.The MMEC device exhibited a broad linear response(K^(+):2–32 mmol/L;Ca^(2+):0.5–4 mmol/L;Na^(+):10–160 mmol/L),high sensitivity(about 20–45 mV/decade),temporal stability(>2weeks),and good selectivity(>80%)for the above ions.In vitro detection and in vivo subcutaneous and brain experiment results showed that the MMEC system exhibits good multi-ion monitoring performance in several complex environments.This work provides a platform for the continuous real-time monitoring of ion fluctuations in different situations and has implications for developing smart sensors to monitor human health. 展开更多

In peripheral artery disease patients,the blood supply directed to the lower limbs is reduced.This results in severe limb ischemia and thereby enhances pain sensitivity in lower limbs.The painful perception is induced and exaggerate during walking,and is relieved by rest.This symptom is termed by intermittent claudication.The limb ischemia also amplifies autonomic responses during exercise.In the process of pain and autonomic responses originating exercising muscle,a number of receptors in afferent nerves sense ischemic changes and send signals to the central nervous system leading to autonomic responses.This review integrates recent study results in terms of perspectives including how nerve growth factor affects muscle sensory nerve receptors in peripheral artery disease and thereby alters responses of sympathetic nerve activity and blood pressure to active muscle.For the sensory nerve receptors,we emphasize the role played by transient receptor potential vanilloid type 1,purinergic P2X purinoceptor 3 and acid sensing ion channel subtype 3 in amplified sympathetic nerve activity responses in peripheral artery disease. 展开更多

Metal ions play crucial roles in biological processes,and their dysregulation can lead to various diseases.Understanding the distribution of these metal ions provides deeper insight into their roles in both health and disease.DNAzymes offer a general approach for detecting nearly any metal ion within cells and in vivo with high sensitivity and selectivity,including different oxidation states of the same metal ion.This Review summarizes recent developments in signal transduction strategies for DNAzyme-based sensing and imaging of metal ions in living cells and in vivo.We examine various signal transduction strategies to convert metal ion binding by DNAzymes into measurable signals.These strategies include fluorescence imaging using small molecules,nanostructures and nanoparticles,motors and machines,and other fluorescent materials as well as bioluminescence imaging.We then provide recent examples of applying these DNAzyme-based fluorescence or bioluminescence imaging methods to understand metal ion dynamics and their roles in diseases in living cells and in vivo.Finally,we discuss future directions for advancing intracellular and in vivo metal ion imaging using DNAzymes. 展开更多

Herein,three novel tetraphenylethylene hydrazone chemosensors TC12,SC16,and TC16 are prepared for the selective detection of F−.Two NH and one C=N units are incorporated into the sensors for better colorimetric responses,whereas the tetraphenyl unit is in charge of the aggregation-induced emission effect.Among them,compounds SC16 and TC16 form stable gels with some organic solvents.All the tetrahydrofuran/H2O solutions of the three compounds exhibit aggregation-induced emission effect,whereby the fluorescence emission increases by varying degrees with the volume of poor solvent water.Moreover,good aggregation-induced emission effects are observed in the self-assembly of SC16 and TC16.As a sample chemosensor,TC12 in tetrahydrofuran responds to F−selectively with high sensitivity,with the colorimetric and fluorometric detection limits of 8.25×10^(−7) mol·L^(-1) and 2.69×10^(−7) mol·L^(-1),respectively.The reversible gel-sol-gel phase transition and color changes indicate that both SC16-dimethyl sulfoxide and TC16-ethyl acetate gels specifically respond to F-with good sensitivity.The detection results are well supported by ultraviolet-visible spectroscopy,fluorescent spectroscopy,and 1H nuclear magnetic resonance.More importantly,the driving forces of gelation are visually clarified through the single crystal X-ray analysis of compound TOMe. 展开更多

Plants have developed strategies to cope with sulfate deficiency, and other nutrient ion limitations. However, the regulation of these adaptive responses and the coordinating signals that underlie them are still poorly characterized. O-acetylserine (OAS) is a marker metabolite of sulfate starvation and has been speculated to have a signaling function. OAS is synthesized by the enzyme serine acetyltransferase (SERAT), which is encoded by five distinct genes in Arabidopsis. We investigated quadruple knockout mutants of SERAT that retained only one functional isoform. These mutants displayed symptoms of sulfate starvation. Furthermore, some of them displayed phenotypes typical of prolonged sulfate starvation, in particular, developmental programs associated with senescence or stress responses. Thus, we compared metabolite and transcriptome data from these mutants with N-, P-, K-, and S-depleted plants. This revealed many similarities with general nutrient-depletion-induced senescence (NuDIS), in- dicating the recruitment of existing regulatory programs for nutrient-starvation responses. Several candidate genes that could be involved in these processes were identified, including transcription factors and other regulatory proteins, as well as the functional categories of their target genes. These results outline components of the regulatory network controlling plant development under sulfate stress, forming a basis for further investigations to elucidate the complete network. In turn, this will advance our broader understanding of plant responses to a range of other nutrient stresses. 展开更多
