Hypertrophic Scar Formation and Wound Healing Modulation Fatty Acids as Modulators of Severe Scars

There are several treatm... Scar tissue usually generates severe discomfort in the short and long term. Common symptoms include anesthetics sequelae, pruritus, joint malfunction, new wounds on the scar surface, and pain. There are several treatments for scars, like compression, topical or intralesional steroid infiltration, 5-fluorouracil, dermabrasion, and surgeries with new scar tissue. For adult patients, it is easier to choose the treatment. However, compression is commonly applied in children to prevent treatments that have adverse effects. This study reports the outcomes of 15 patients submitted to abdominoplasty, traumatic wounds and post-burn scar treatments, which showed significant changes after the continuous use of an ointment composed of petrolatum, cod liver oil, BHT, Chamomilla recutita (chamomile) oil, Helianthus annuus (sunflower) oil, and Prunus amygdalus dulcis (sweet almond) oil. As components of the stratum corneum, unsaturated fatty acids influence the cutaneous structural and immune status and permeability. They also interfere with the maturation and differentiation of the stratum corneum and inhibit the production of proinflammatory eicosanoids, reactive species (ROS and RNS), and cytokines, thereby influencing the inflammatory response and possibly wound healing. This article aims to share our experience with the regular use of an ointment in adult and pediatric patients for three months. The increase in proinflammatory cytokine production at wound sites, resulting in a noninvasive, therapeutical, and effective cutaneous wound healing and scarring modulation, may provide a physiopathological explanation for the fast improvement of scars. 展开更多

Treatments of atherogenesis,one of the most common cardiovascular diseases(CVD),are continuously being made thanks to innovation and an increasingly in-depth knowledge of percutaneous transluminal coronary angioplasty(PTCA),the most revolutionary medical procedure used for vascular restoration.Combined with an expanding balloon,vascular stents used at stricture sites enable the long-time restoration of vascular permeability.However,complication after stenting,in-stent restenosis(ISR),hinders the advancement of vascular stents and are associated with high medical costs for patients for decades years.Thus,the development of a high biocompatibility stent with improved safety and efficiency is urgently needed.This review provides an overview of current advances and potential technologies for the modification of stents for better treatment and prevention of ISR.In particular,the mechanisms of in-stent restenosis are investigated and summarized with the aim to comprehensively understanding the pathogenesis of stent complications.Then,according to different therapeutic functions,the current stent modification strategies are reviewed,including polymeric drug eluting stents,biological friendly stents,prohealing stents,and gene stents.Finally,the review provides an outlook of the challenges in the design of stents with optimal properties.Therefore,this review is a valuable and practical guideline for the development of cardiovascular stents. 展开更多

Conductive polyphenol microneedles coupled with electroacupuncture to accelerate wound healing and alleviate depressive-like behaviors in diabetes

Inflammation and depression are serious complications of diabetes that interact to form a feedback loop and may hinder diabetic wound healing.They share a common pathophysiological basis of abnormal interactions between diabetic wounds and the brain.Here,we propose a strategy combining electroacupuncture(EA)stimulation of the Dazhui acupoint(GV14)with polyphenol-mediated conductive hydrogel microneedles to promote diabetic wound healing and alleviate depression through local wound-brain interactions.The conductive microneedles comprised methacrylated gelatin,dopamine(DA),DA-modified poly(3,4-ethylenedioxythiophene),and Lycium barbarum polysaccharide.EA at GV14 activated the vagus-adrenal axis to inhibit systemic inflammation while DA coupled electrical signals for long-term inhibition of local wound inflammation.EA at GV14 was also found to elevate 5-hydroxytryptamine levels in rats with diabetic wounds,consequently mitigating depressive-like be-haviors.Additionally,the polyphenol-mediated conductive hydrogel microneedles,and coupled with EA stim-ulation promoted healing of wound tissue and peripheral nerves.This strategy regulated both local and systemic inflammation while alleviating depressive-like behaviors in diabetic rats,providing a new clinical perspective for the treatment of diabetes-related and emotional disorders. 展开更多

Synergistic nanozyme-macrophage hybrid system for targeted therapy of acute lung injury

Due to its complex pathogenesis involving dysregulated reactive oxygen species(ROS)storms,cytokine hyperactivation and immune cell infiltration,acute lung injury(ALI)presents critical clinical challenges,and conventional therapies often fail to address the dual requirements of precise inflammatory targeting and microenvironment regulation.Herein,we report an intelligent biomimetic nanosystem(CeGM)featuring(i)synergistic catalytic circuits,where hollow CeO_(2)nanoenzymes are combined with glycyrrhizic acid(GA)through density functional theory(DFT)-optimized binding configurations,achieving charge redistribution-induced dual-site catalytic activation relative to conventional CeO_(2)alone;(ii)pathology-responsive delivery,where macrophage membrane camouflage enables lipopolysaccharide(LPS)recognition and inflammatory chemotaxis,yielding 7.8-fold higher alveolar accumulation than non-coated counterparts in LPS-induced ALI models;(iii)multidimensional immunomodulation,where NOD-like receptor family pyrin domain containing 3(NLRP3)inflammasome activation(56.20%interleukin-1β(IL-1β)reduction)and M1 macrophage polarization(2.4-fold M2/M1 ratio increase)are concurrently suppressed through GA-mediated nuclear factor kappa B(NF-κB)pathway inhibition and CeO_(2)-mediated redox homeostasis restoration.This dual-functional nanoplatform demonstrates favourable therapeutic outcomes in mitigating pulmonary edema by clearing ROS(47.20%reduction in ROS levels),neutrophil infiltration(58.16%myeloperoxidase(MPO)activity decrease)and cytokine storms(tumor necrosis factor-alpha(TNF-α)reduction of 45.90%),offering a paradigm-shifting strategy for precision nanomedicine in acute inflammatory disorders. 展开更多
