The Psychiatric Genomics Consortium(PGC)has recently identified 10 potential functional coding variants for schizophrenia.However,how these coding variants confer schizophrenia risk remains largely unknown.Here,we investigate the associations between eight potential functional coding variants identified by PGC and schizophrenia in a large Han Chinese sample(n=4022 cases and 9270 controls).Among the eight tested single nucelotide polymorphisms(SNPs),rs3617(a missense variant,p.K315 Q in the ITIH3 gene)showed genome-wide significant association with schizophrenia in the Han Chinese population(P=8.36×10-16),with the same risk allele as in PGC.Interestingly,rs3617 is located in a genomic region that is highly evolutionarily conserved,and its schizophrenia risk allele(C allele)was associated with lower ITIH3 mRNA and protein expression.Intriguingly,mouse neural stem cells stably overexpressing ITIH3 with different alleles of rs3617 exhibited significant differences in proliferation,migration,and differentiation,suggesting the impact of rs3617 on neurodevelopment.Subsequent transcriptome analysis found that the differentially expressed genes in neural stem cells stably overexpressing different alleles of rs3617 were significantly enriched in schizophrenia-related pathways,including cell adhesion,synapse assembly,MAPK and PI3 K-AKT pathways.Our study provides convergent lines of evidence suggesting that rs3617 in ITIH3 likely affects protein function and neurodevelopment and thereby confers risk of schizophrenia. 展开更多

血清蛋白质谱筛选及验证类风湿关节炎患者肌肉量减少的生物标志物

目的:探索类风湿关节炎(rheumatoid arthritis,RA)患者肌肉量减少的生物标志物。方法:本研究为横断面研究,采用非标记液相色谱-串联质谱技术检测基线病情活动且2年随访期间保持肌肉量正常或肌肉量减少的RA患者血清蛋白组学特征,并通过... 目的:探索类风湿关节炎(rheumatoid arthritis,RA)患者肌肉量减少的生物标志物。方法:本研究为横断面研究,采用非标记液相色谱-串联质谱技术检测基线病情活动且2年随访期间保持肌肉量正常或肌肉量减少的RA患者血清蛋白组学特征,并通过生物信息学分析两组患者血清中的差异蛋白;进一步纳入102例RA患者(肌肉量减少组和肌肉量正常组各51例)及招募志愿者51例作为健康对照组(healthy controls,HC)进行差异蛋白的ELISA验证,并采用Logistic回归分析RA患者基线肌肉量减少的相关因素。结果:10例肌肉量减少组与9例肌肉量正常组RA患者通过基线血清蛋白组学分析筛选出38个差异蛋白,其中间-α-胰蛋白酶抑制蛋白重链3(inter-alpha-trypsin inhibitor heavy chain H3,ITIH3)表达差异显著(log 2FC=2.09),并在所有血清样本中稳定表达。ELISA验证结果显示102例RA患者血清ITIH3水平显著高于HC组[(119.4±79.7)mg/L vs.(42.3±16.6)mg/L,P<0.001],且肌肉量减少组RA患者血清ITIH3显著高于肌肉量正常组[(148.1±94.7)mg/L vs.(90.8±46.6)mg/L,P<0.001]。多因素Logistic回归分析结果显示,校正混杂因素后,血清ITIH3水平仍是RA患者肌肉量减少的独立危险因素(OR=1.024,95%CI:1.013~1.038,P<0.001)。结论:血清ITIH3水平是RA患者肌肉量减少的独立危险因素,提示ITIH3可能是RA肌肉量减少的潜在生物标志物。 展开更多

近年来,我国集约化牛场卵巢静止性乏情率较高,但临床奶牛卵巢静止的发生尚未有完整的预警体系,为临床预测奶牛卵巢静止提供预警指标,选取产后60~90d卵巢静止组和对照组奶牛血液样本,在蛋白组学前期研究的基础上筛选出检测试验奶牛血液... 近年来,我国集约化牛场卵巢静止性乏情率较高,但临床奶牛卵巢静止的发生尚未有完整的预警体系,为临床预测奶牛卵巢静止提供预警指标,选取产后60~90d卵巢静止组和对照组奶牛血液样本,在蛋白组学前期研究的基础上筛选出检测试验奶牛血液中与卵巢静止相关指标。3种能量指标:谷草转氨酶(AST)、葡糖糖(Glu)、非游离性脂肪酸(NEFA);4种生殖激素:雌二醇(E2)、孕酮(P4)、促卵泡生长激素(FSH)、促黄体素(LH);4种关键差异表达蛋白:胰岛素样生长因子结合蛋白(IGFBP-2)、α-2-HS糖蛋白(AHSG)、载脂蛋白A4(ApoA4)、视黄醇结合蛋白4(RBP-4);6种关键酶活性:果糖二磷酸醛缩酶(ALDOB)、L-乳酸脱氢酶(LDHB)、inter-α-胰蛋白酶抑制剂重链H3(ITIH3)、谷胱甘肽过氧化物酶(GPX3)、血清衍生的透明质酸相关蛋白(SPAM1)、丙酮酸激酶(PKM2)。结果显示,经过相关性分析、二元逻辑回归建模以及ROC分析,建立了ApoA4和ITIH3单一指标的预警技术,其预警值分别为ApoA4>28.825μg/L,ITIH3>195.07ng/L。建立了基于ApoA4+ITIH3和ApoA4+ITIH3+E2潜在生物标志物的多指标预警体系,前者预警值为ApoA4>19.55μg/L,ITIH3>191.14ng/L;后者预警值为ApoA4>47.56μg/L,ITIH4>187.80ng/L,E2<69.63ng/L。 展开更多

Early Warning System of Risk in Dairy Cows with Inactive Ovaries

This test provides early warning indica... The incidence of Inactive ovaries of dairy cows in China is relatively high. There is no complete early warning system for the occurrence of ovarian quiescence in clinical cows. This test provides early warning indicators for clinical prediction of ovary cessation in dairy cows. This experiment selected blood samples of dairy cows from 60 to 90 days postpartum in the inactive ovaries group and control group. Differential proteins were selected on the basis of proteomics, three energy indexes: AST, Glu, NEFA. Four reproductive hormones: E2, P4, FSH, LH, and four differentially expressed proteins: IGFBP-2, AHSG, APO-A4, and RBP-4. Key enzyme activities: ALDOB, LDHB, ITIH3, GPX3, SPAM1, PKM2. The ELISA test kit was used to detect the content and activity of the above markers in the test bovine serum. Through correlation analysis, binary logistic regression modeling and ROC analysis, a single indicator early warning technique for APOA4 and ITIH3 was established. The early warning values were APOA4 > 28.825 μg/L and ITIH3 > 195.07 ng/L. A multi-index early warning system based on potential biomarkers of APOA4 + ITIH3 and APOA4 + ITIH3 + E2 was established. The former had an early warning value of: APOA4 > 19.55 μg/I;ITIH3 > 191.14 ng/L;the latter has an early warning value: APOA4 > 47.56 μg/L, ITIH4 > 187.80 ng/L, E2 < 69.63 ng/L. 展开更多
