Genotypic and allelic profiles were identified using polymerase chain reaction based in real time and amplification-refractory mutation system.Results: We observed a protective association of IL-10(-819 C/T) C allele(P = 0.028,OR = 0.56, CI = 0.34–0.91) against DHF, while the C/T(P = 0.047, OR = 2.10,CI = 1.01–4.38) and T/T(P = 0.008, OR = 3.82, CI = 1.38–10.59) genotypes were associated with DHF and DF, respectively. The dominant model TNFA-308 GA + AA(P = 0.043, OR = 0.45, CI = 0.20–1.00) genotypes were found to have protective effect against dengue infection. A protective association among the IFNG(+874 A/T) A/T genotype against DF(P = 0.02, OR = 0.46, CI = 0.24–0.89) and DHF(P = 0.034,OR = 0.43, CI = 0.19–0.95) was observed. When the studied single-nucleotide polymorphism was analyzed in combination, the combination GTA(P = 0.022, OR = 2.95,CI = 1.18–7.41) was statistically significantly associated with susceptibility to DF and the combination GCT(P = 0.035, OR = 0.28, CI = 0.08–0.90) with protection against the development of DHF.Conclusions: This research identifies the association of the IFNG(+874 A/T), TNFA(-308 G/A), IL-10(-819 C/T) genotypes as a factor for protection, susceptibility and severity to dengue.展开更多

Its underlying mechanisms are complex, involving multiple inflammatory and immune signaling pathways. This study integrates bioinformatics and experimental validation to identify key genes closely associated with burn pain and to explore their potential clinical value as therapeutic targets. Gene expression profiles (GSE19743 as the training set and GSE37069 as the validation set) were retrieved from the GEO database, and pain-related genes were screened via the GeneCaRNA database. Subsequent analyses included differential analysis, GO and KEGG enrichment analyses, and PPI network construction and visualization. Three machine learning algorithms—LASSO regression, SVM, and RF—were employed to identify key genes, following which a diagnostic model was established and evaluated using ROC curves and DCA. RT-qPCR validated candidate gene expression in peripheral blood samples. A total of 117 differentially expressed burn pain-related genes were identified, primarily enriched in inflammatory signaling pathways such as PI3K-Akt and MAPK. IFNG, IL10, and TLR4 were consistently identified as key feature genes across all three machine-learning methods. The diagnostic model based on these genes demonstrated excellent performance in the GSE37069 validation set, achieving an AUC of 0.959. RT-qPCR validation indicated that IL10 was significantly upregulated in burn patients, IFNG expression was decreased, while TLR4 expression showed no significant difference, partially in line with bioinformatics predictions. These findings suggest that IFNG, IL10, and TLR4 may be involved in burn pain occurrence and maintenance by modulating immune and inflammatory responses, thus showing promise as diagnostic biomarkers and therapeutic targets. Further research into their signaling pathway mechanisms and clinical intervention value is warranted.展开更多
Background:Now that the epidemic of new coronavirus pneumonia(corona virus disease 2019)is spreading all over the world,Jinhuaqinggan granules in the Chinese treatment plan has been proved to be an effective Chinese p...Background:Now that the epidemic of new coronavirus pneumonia(corona virus disease 2019)is spreading all over the world,Jinhuaqinggan granules in the Chinese treatment plan has been proved to be an effective Chinese patent medicine for the treatment of corona virus disease 2019.Methods:This study aims to clarify the possible therapeutic mechanism governing the efficacy of Jinhuaqinggan granules in the treatment of corona virus disease 2019,through using network pharmacology and molecular docking.During the analysis,227 active components were obtained and screened by using the ADME method.Furthermore,282 Jinhuaqinggan granule targets and 56 common targets with corona virus disease 2019 were gathered from various databases.Then the protein-protein interaction network of Jinhuaqinggan granules and corona virus disease 2019 targets were constructed and 6 core targets were selected through network topology analysis.In addition,A total of 262 biological function annotation entries(P<0.01)and 101 pathways(P<0.01)were obtained by gene ontology functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes pathway analysis.Results:Molecular docking showed that quercetin,luteolin,kaempferol,wogonin and naringin had an affinity for SARS-CoV-23CL hydrolase and angiotensin-converting enzyme 2.Conclusion:corona virus disease 2019 can be prevented by the primary targets of Jinhuaqinggan granules.The most important bioactive components in Jinhuaqinggan granules-quercetin,naringenin,luteolin and wogonin-can play antiviral effect,anti-inflammatory storm,regulate immunity by regulating signal transducers and activators of transcription 1,interleukin 4,interferon-γ,heme oxygenase 1 and acting on the lipopolysaccharide response,toll-like receptor signaling pathway,mitogen-activated protein kinase signaling pathway,etc.展开更多
