Coconut(Cocos nucifera L.),a major oil and fruit crop of the Arecaceae family,is extensively cultivated across the Asia—Pacific region.Despite its agricultural importance,genome assembly in coconut remains challenging due to its large genome size and high proportion of repetitive sequences.Allele-specific expression(ASE)plays a key role in regulating plant development and evolution,yet research on ASE in coconut is limited(Shao et al.,2019;Li et al.,2021;Zhang et al.,2021;Hu et al.,2022).Among phenotypic traits,fruit color is especially important as an indicator of maturity,guiding harvest timing and post-harvest processes(Kapoor et al.,2022).While prior studies have explored various coconut traits such as salt tolerance,fiber content,and plant height(Wang et al.,2021;Yang et al.,2021),investigations into ASE and fruit color remain scarce. 展开更多

目的研究中性粒细胞弹性蛋白酶(NE)、干扰素-γ(IFN-γ)、α1-抗胰蛋白酶(α1-AT)及调节性T细胞(Treg)/辅助性T细胞17(Th17)对慢性阻塞性肺疾病急性加重期(AECOPD)患者并发呼吸衰竭(RF)的预测价值。方法选取2019年10月-2022年12月秦皇... 目的研究中性粒细胞弹性蛋白酶(NE)、干扰素-γ(IFN-γ)、α1-抗胰蛋白酶(α1-AT)及调节性T细胞(Treg)/辅助性T细胞17(Th17)对慢性阻塞性肺疾病急性加重期(AECOPD)患者并发呼吸衰竭(RF)的预测价值。方法选取2019年10月-2022年12月秦皇岛市第一医院收治的160例AECOPD患者资料,将其按照是否并发RF分作RF组53例及无RF组107例。比较两组NE、IFN-γ、α1-AT及Treg/Th17以及各项临床资料。采用多因素Logistic回归分析AECOPD患者并发RF影响因素。采用受试者工作特征(ROC)曲线分析上述各项指标预测AECOPD患者并发RF的效能。结果RF组外周血NE、IFN-γ及α1-AT水平均高于无RF组,Treg/Th17低于无RF组,差异有统计学意义(均P<0.05)。RF组FEV1%预计值低于无RF组,差异有统计学意义(P<0.05)。经多因素Logistic回归分析证实:NE、IFN-γ、α1-AT升高均是AECOPD患者并发RF的危险因素,Treg/Th17、FEV1%预计值升高为AECOPD患者并发RF的保护性因素(均P<0.05)。经ROC曲线分析发现,NE、IFN-γ、α1-AT及Treg/Th17联合预测AECOPD患者并发RF的效能优于上述四项指标单独预测,差异有统计学意义(均P<0.05)。结论NE、IFN-γ、α1-AT水平升高及Treg/Th17降低会导致AECOPD患者并发RF的风险增加。 展开更多

目的:探讨抗感合剂对脓毒症急性肺损伤大鼠肺组织中STING、IRF3、IFN-β和NLRP3表达的影响。方法:40只大鼠随机均分为空白对照组、模型组、地塞米松(1.08 mg/kg灌胃3 d)组和抗感合剂(20 mL/kg灌胃3 d)组,第4天除空白对照组外其他3组气... 目的:探讨抗感合剂对脓毒症急性肺损伤大鼠肺组织中STING、IRF3、IFN-β和NLRP3表达的影响。方法:40只大鼠随机均分为空白对照组、模型组、地塞米松(1.08 mg/kg灌胃3 d)组和抗感合剂(20 mL/kg灌胃3 d)组,第4天除空白对照组外其他3组气管内注射LPS建立脓毒症急性肺损伤大鼠模型。第7天通过HE染色评估肺组织损伤程度;运用荧光免疫双染技术检测肺组织STING、IRF3、IFN-β与NLRP3的相对荧光强度;采用ELISA法测定血清和肺组织匀浆中STING、IRF3、IFN-β、NLRP3和炎症因子的含量;采用qRT-PCR和全自动蛋白表达分析系统检测大鼠肺组织中STING、IRF3、IFN-β、NLRP3和TBK1 mRNA及蛋白表达水平。结果:空白对照组大鼠肺泡整齐且完整,未见明显炎性细胞浸润;模型组肺间质变厚且肺泡水肿,可见中性粒细胞浸润;与模型组比较,抗感合剂组和地塞米松组肺组织病理改变减轻。与空白对照组比较,模型组肺组织STING、IRF3、IFN-β和NLRP3蛋白的荧光强度升高,血清及肺组织匀浆中STING、IRF3、IFN-β、NLRP3和炎症因子IL-6、IL-1β、TNF-α、IFN-α含量升高,肺组织中STING、IRF3、IFN-β、NLRP3和TBK1 mRNA及蛋白表达升高,p-IRF3和TBK1蛋白表达升高;抗感合剂组上述指标均降低(P<0.05)。结论:抗感合剂可能通过抑制STING、IRF3、IFN-β和NLRP3的表达减少炎症因子的释放,减轻脓毒症急性肺损伤大鼠肺损伤。 展开更多

In order to investigate the correlation between the expression of the apoptotic regulatory proteins (Fas, Bcl 2) in peripheral blood lymphocytes (PBLC) and the level of IFN γ and IL 4 in serum of the patients with condyloma acuminata (CA) in the immune pathogenesis of CA, indirect immunofluorescence labeling method of flow cytometer and solid sandwich ELISA method were performed for detecting the expression of Fas, Bcl 2 in PBLC and the level of IFN γ and IL 4 in serum of 60 cases of CA. The results showed the expression level of Fas in PBLC of CA was significantly higher than in the normal control group, but the expression level of Bcl 2 was significantly lower (both P< 0.01). The level of IFN γ in serum of CA was significantly lower than in the normal control group ( P< 0.01), but IL 4 was significantly lower (both P< 0.01). The expression of Fas in PBLC had a negative correlation with the level of IFN γ in serum of patients with CA, but had a positive correlation with the level of IL 4; The expression of Bcl 2 had a positive correlation with the level of IFN γ, but had a negative correlation with the level of IL 4. All the correlation coefficients had significant differerce by t test ( P< 0.01). It was suggested abnormal apoptosis in PBLC, the suppressed secretion of the THl associated cytokines (eg: IFN γ) and the increased secretion of the TH2 asssociated cytokines (eg: IL 4) existed in the patients with CA and might play an important role in the immune pathogenesis of CA. 展开更多

目的:探索结核特异性细胞因子γ干扰素(IFN-γ)、白细胞介素-2(IL-2)联合检测在老年发热患者结核病诊断中的价值。方法:回顾性分析2022年1月—2024年3月于安徽医科大学附属阜阳人民医院感染科就诊的≥60岁因反复发热住院患者127例。所... 目的:探索结核特异性细胞因子γ干扰素(IFN-γ)、白细胞介素-2(IL-2)联合检测在老年发热患者结核病诊断中的价值。方法:回顾性分析2022年1月—2024年3月于安徽医科大学附属阜阳人民医院感染科就诊的≥60岁因反复发热住院患者127例。所有患者在进行相关检查后不排除结核感染,并进行结核特异性细胞因子IFN-γ、IL-2联合检测及结核分枝杆菌DNA(TB-DNA)扩增检测。比较结核病(TB组)与非结核发热(NTB组)患者的IFN-γ、IL-2水平,并分析IFN-γ、IL-2联合检测诊断结核病的价值。结果:127例发热患者中,结核病18例,占14.2%(18/127),其中肺结核14例,肺外结核4例,非结核发热109例,以感染性发热、慢性阻塞性肺疾病急性加重、肺癌为主。TB组IFN-γ、IL-2水平均显著高于NTB组(P<0.05)。以TB-DNA为金标准,IFN-γ、IL-2联合检测的敏感度为83.3%、特异度为84.4%。受试者操作特征(ROC)曲线显示,IFN-γ、IL-2联合诊断结核病的AUC为0.940[95%CI(0.886,0.994)]。结论:结核特异性细胞因子IFN-γ、IL-2联合检测在老年发热患者结核病诊断中具有良好价值。 展开更多

目的 分析白细胞介素-1β(IL-1β)、干扰素-γ(IFN-γ)、诱导型一氧化氮合酶(iNOS)表达水平与膀胱癌术后感染的相关性。方法 收集96例膀胱癌患者临床资料。依照患者术后感染情况分为非感染组(76例)和感染组(20例)。比较两组年龄、体质... 目的 分析白细胞介素-1β(IL-1β)、干扰素-γ(IFN-γ)、诱导型一氧化氮合酶(iNOS)表达水平与膀胱癌术后感染的相关性。方法 收集96例膀胱癌患者临床资料。依照患者术后感染情况分为非感染组(76例)和感染组(20例)。比较两组年龄、体质指数、性别、文化程度、肿瘤分期、吸烟、饮酒、IL-1β、IFN-γ、iNOS表达水平;分析IL-1β、IFN-γ、iNOS表达水平与膀胱癌术后感染的相关性;受试者操作特征曲线(ROC)下面积(AUC)评估IL-1β、IFN-γ、iNOS表达水平对膀胱癌术后感染的预测效能。结果 两组年龄、体质指数、性别、文化程度、肿瘤分期、吸烟、饮酒比较差异无统计学意义(P>0.05)。感染组IL-1β、IFN-γ、iNOS水平均高于非感染组,差异有统计学意义(t=14.278,P=0.000、t=7.633,P=0.000、t=11.749,P=0.000)。IL-1β、IFN-γ、iNOS水平与膀胱癌术后感染呈显著正相关性,P均<0.05。将膀胱癌术后感染=1,非感染=0作为因变量,IL-1β、IFN-γ、iNOS水平作为自变量,进行二元逻辑回归分析,并进行受试者操作特征曲线(ROC)分析,结果显示,IL-1β、IFN-γ、iNOS三者联合预测膀胱癌术后感染的预测效能相比单一指标较高,三者联合的ROC下面积(AUC)最大,为0.967,灵敏度为0.850,特异度为0.908。IL-1β、IFN-γ、iNOS的预测阈值分别为1.785 pg/ml、40.200 pg/ml、143.430 U/ml, IL-1β、IFN-γ、iNOS的AUC分别为0.938、0.889、0.908。结论 IL-1β、IFN-γ、iNOS在膀胱癌术后感染患者中表达水平显著升高,IL-1β、IFN-γ、iNOS与膀胱癌术后感染呈正相关关系,三者联合对于膀胱癌术后感染有良好的预测价值,可作为临床上膀胱癌术后感染的评估因子。 展开更多

目的探讨柴桂苓汤加减在慢性前列腺炎患者中的临床疗效及其对患者尿流动力学和血清白介素-8(interleukin-8,IL-8)、γ-干扰素(gamma-interferon,IFN-γ)水平的影响。方法选择174例慢性前列腺炎患者,随机分对照组和观察组,各87例。对照... 目的探讨柴桂苓汤加减在慢性前列腺炎患者中的临床疗效及其对患者尿流动力学和血清白介素-8(interleukin-8,IL-8)、γ-干扰素(gamma-interferon,IFN-γ)水平的影响。方法选择174例慢性前列腺炎患者,随机分对照组和观察组,各87例。对照组采用常规西医药物治疗方案,观察组在对照组基础上联合柴桂苓汤加减。7d为1个疗程,连续治疗5个疗程。比较两组患者的临床资料、临床疗效、治疗前后尿流动力学指标、治疗前后炎症指标。结果两组患者临床资料、治疗前尿流动力学指标及炎症指标方面的差异并无统计学意义(P>0.05)。与对照组相比,观察组患者的总有效率显著升高(P<0.05)。治疗后,与对照组相比,观察组患者最大尿流率、平均尿流率升高,排尿前等待时间、有效排尿时间缩短(P<0.05)。治疗后,与对照组相比,观察组患者IL-8、IFN-γ水平显著降低(P<0.05)。结论柴桂苓汤加减可提高慢性前列腺炎患者临床疗效,缓解临床症状,改善排尿情况,减轻炎症反应,具有较好的临床应用价值。 展开更多

Interferon(IFN) with antiviral and im-munomodulatory activities is one of the most important therapeutic agents for the treatment of chronic hepatitis. The apoptotic effect of IFN is influenced by cell type and the types of IFN, which suppresses proliferation and induces apoptosis in some cell types while inhibiting apoptosis in others. The aim of this study was to explore the effect of IFNα-2a on Fas expression and the apoptosis rate of peripheral blood cytotoxic T cells (CTLs) in patients with hepatitis B. METHODS:Peripheral blood mononuclear cells were isolated from 26 patients with hepatitis B including 16 patients with chronic hepatitis B and 10 patients with chronic severe hepatitis B. Fas expression and apoptosis rate of CTLs were analyzed with flow cytometry before and after IFNα-2a treatment. RESULTS:Before IFNα-2a treatment, Fas expression and apoptosis rate of CTLs from patients with chronic hepatitis B were significantly higher than those from patients with chronic severe hepatitis B and healthy controls respectively. No significant difference was observed between Fas expression and apoptosis rate of CTLs from patients with chronic severe hepatitis B and healthy controls. After IFNα-2a treatment,Fas expression and apoptosis rate of CTLs from different groups were compared with those before IFNα-2a treatment, showing no significant difference despite alternation of different degree. CONCLUSIONS:Activation induced cell death (AICD) exists in peripheral blood CTLs from patients with hepatitis B. No effect of IFNα-2a exerts on Fas expression and apoptosis rate of Fas in patients with hepatitis B. 展开更多

Objective: To study the expression of the inducible nitric oxide synthase (iNOS) gene and the effects of tumor necrosis factor-α(TNF-a) and interferon-γ(IFN-g)on proliferation of the continuous cultured human colon cancer cell line CCL229. Methods: Using the molecular and biochemical techniques and electron microscopy to analyze the expression of iNOS, production of NO and growth characteristics of human colon cancer cells. Results: cytokine treatment can induce expression of the iNOS gene and production of nitric oxide was significantly higher after treatment of CCL229 cells with TNF-αor IFN-γ. Treatment with either cytokine or a combination of both significantly increased levels of Malondialdehyde (MDA) over control. Furthermore, cytokine treatment increased the proliferation inhibition rate as assessed in vitro and decreased the cell proliferation index on flow cytometry. Electron microscopy showed that cells treated with cytokines had fewer pseudopodia or cell processes than control cells and that cytokine treated cells had dilatation of the mitochondria and endoplasmic reticulum and dilated vesicular or tubular cisternae. Conclusion: Our findings indicate that TNF-α and IFN-γ induce the expression of iNOS gene in CCL229 cells, which increases the production of nitric oxide, inhibits proliferation, causes lipid peroxidation, and results in ultrastructural changes. 展开更多

目的 分析2型糖尿病(T2DM)合并代谢相关脂肪性肝病(MAFLD)患者血清干扰素(IFN)-α和IFN-β水平变化及其临床意义。方法 2021年9月~2024年8月我院收治的T2DM患者87例和T2DM合并MAFLD患者63例。采用ELISA法检测血清IFN-α和IFN-β水平。... 目的 分析2型糖尿病(T2DM)合并代谢相关脂肪性肝病(MAFLD)患者血清干扰素(IFN)-α和IFN-β水平变化及其临床意义。方法 2021年9月~2024年8月我院收治的T2DM患者87例和T2DM合并MAFLD患者63例。采用ELISA法检测血清IFN-α和IFN-β水平。应用二元多因素Logistic回归分析T2DM合并MAFLD发生的危险因素,应用受试者工作特征曲线(ROC)分析预测效能。结果 T2DM合并MAFLD组BMI、合并高血压占比、血清TC、TG、LDL-C、UA、IFN-α和IFN-β水平分别为(26.9±2.6)kg/m^(2)、74.6%、(6.8±1.4)mmol/L、(4.3±1.7)mmol/L、(3.7±0.6)μmol/L、(477.1±31.2)μmol/L、(27.9±8.5)pg/mL和(260.1±59.2)pg/mL,均显著高于T2DM组【分别为(24.9±2.2)kg/m^(2)、24.1%、(5.6±1.1)mmol/L、(2.2±0.6)mmol/L、(3.2±0.8)mmol/L、(329.7±30.1)μmol/L、(21.9±4.1)pg/mL和(205.7±51.8)pg/mL,P<0.05】;采用多因素Logistic回归分析结果显示BMI、血清UA、IFN-α和IFN-β水平均为T2DM合并MAFLD的危险因素(P<0.05);ROC曲线分析显示,血清IFN-α和IFN=β水平联合预测T2DM合并MAFLD的AUC为0.832,其灵敏度为73.0%,特异度为87.4%,显著优于两指标单独预测(P<0.05)。结论 监测T2DM患者血清IFN-α和IFN-β水平变化可能有助于早期发现MAFLD的存在,指导临床及时进行合适的干预。 展开更多

目的明确锌指蛋白Krüppel样转录因子9(KLF9)在Ⅰ型单纯疱疹病毒(HSV-1)诱导巨噬细胞Ⅰ型干扰素表达中的作用及机制。方法通过琼脂糖凝胶电泳、实时荧光定量PCR及Western blot法检测KLF9在Klf9^(-/-)(gKO)小鼠骨髓来源的巨噬细胞(BM... 目的明确锌指蛋白Krüppel样转录因子9(KLF9)在Ⅰ型单纯疱疹病毒(HSV-1)诱导巨噬细胞Ⅰ型干扰素表达中的作用及机制。方法通过琼脂糖凝胶电泳、实时荧光定量PCR及Western blot法检测KLF9在Klf9^(-/-)(gKO)小鼠骨髓来源的巨噬细胞(BMDM)中的敲除效果;RNA-seq筛选gKO小鼠及对照小鼠Klf9^(+/+)(WT)BMDM中HSV-1刺激后Klf9下游关键差异基因;实时荧光定量PCR验证HSV-1刺激后干扰素β1(Ifnb1)及下游干扰素刺激基因(ISG)如干扰素诱导蛋白与四肽重复1(Ifit1)、干扰素刺激的核酸外切酶基因20(Isg20)、胆固醇25-羟化酶(Ch25h)及2′-5′-寡聚腺苷酸合成酶1(Oasl1)的表达情况;ELISA检测HSV-1刺激后gKO及WT小鼠BMDM上清中β干扰素(IFN-β)的含量;Western blot法检测HSV-1刺激后gKO及WT小鼠BMDM中磷酸化干扰素调节因子3(p-IRF3)、干扰素调节因子3(IRF3),p-IRF7、IRF7,磷酸化的核因子κB p65(p-NF-κB p65)、核因子κB p65(NF-κB p65)的表达;CUT-Tag及ChIP-qPCR技术检测KLF9在Ifnb1上的结合。结果KLF9在gKO小鼠的BMDM中表达明显减少;RNA-seq结果显示BMDM中Klf9缺失导致Ⅰ型干扰素信号通路受损;实时荧光定量PCR结果表明Klf9敲除后BMDM中除了Isg20以外,Ifit1、Ch25h及Oasl1表达均明显降低;ELISA结果显示Klf9敲除导致BMDM分泌的IFN-β明显减少。在机制研究方面,Western blot法检测结果显示Klf9的缺失不影响p-IRF3、p-IRF7及p-NF-κB p65的表达;CUT-Tag及ChIP-qPCR结果显示KLF9能直接结合至Ifnb1的启动子区。结论KLF9对巨噬细胞抵抗HSV-1感染至关重要。 展开更多

目的:系统探讨干扰素⁃γ(interferon⁃gamma,IFN⁃γ)在克罗恩病(Crohn’s disease,CD)肠纤维化中的调控作用及机制。方法:提取分离CD患者肠成纤维细胞,体外予以不同的细胞因子[20 ng/mL干扰素⁃γ(interferon⁃gamma,IFN⁃γ)、50 ng/mL白介... 目的:系统探讨干扰素⁃γ(interferon⁃gamma,IFN⁃γ)在克罗恩病(Crohn’s disease,CD)肠纤维化中的调控作用及机制。方法:提取分离CD患者肠成纤维细胞,体外予以不同的细胞因子[20 ng/mL干扰素⁃γ(interferon⁃gamma,IFN⁃γ)、50 ng/mL白介素(interleukin,IL)⁃17A、10 ng/mL IL⁃1β、100 ng/mL IL⁃33、200 ng/mL IL⁃36α、20 ng/mL肿瘤坏死因子(tumor necrosis factor,TNF)⁃α]处理48 h,RT⁃PCR检测人肠成纤维细胞中双调蛋白(amphiregulin,Areg)基因表达水平。20 ng/mL IFN⁃γ处理人肠成纤维细胞48 h后,进行转录组测序(RNA⁃seq),结合STRING数据库构建蛋白质互作(protein⁃protein interaction,PPI)网络。免疫荧光检测α平滑肌肌动蛋白(α⁃smooth muscle actin,α⁃SMA)、抗原Kiel 67(antigen Kiel 67,Ki67)等肌成纤维细胞标志物,评估IFN⁃γ对人肠道成纤维细胞活化、增殖的影响。结果:Areg显著促进肠成纤维细胞活化、增殖及胶原合成;IFN⁃γ可显著抑制肠成纤维细胞Areg表达,并下调α⁃SMA、COL1A1和COL6A1等纤维化相关基因,同时IFN⁃γ抑制成纤维细胞增殖与活化能力。RNA⁃seq分析发现IFN⁃γ调控的差异基因显著富集于细胞外基质(extracellular matrix,ECM)重构通路,蛋白互作(protein⁃protein interaction,PPI)网络鉴定出肉瘤原癌基因(sarcoma proto⁃oncogene,SRC)为核心节点,提示其可能介导IFN⁃γ的抗纤维化作用。结论:Areg是促CD肠纤维化关键介质,IFN⁃γ通过转录调控抑制Areg表达,并发现SRC可能是IFN⁃γ下游的关键效应分子。IFN⁃γ可能通过抑制SRC/Areg信号轴来发挥其抗纤维化作用。这些发现为开发靶向IFN⁃γ/SRC/Areg通路的抗纤维化策略提供了理论依据。 展开更多

Methods Human DSC was isolated and cultured in vitro, and the expression of IL-10R1 and IL-10R2 gene was analyzed after cells had been treated with TH2-type cytokines IL-10 and TH1-type cytokines IFN-γ within 60 rain with semiquantitative reverse transcriptase-PCR, then the influence of IL-10 and IFN-γ on expression of IL-10R protein was examined by first trimester DSC using flow cytometry. In addition, the vitality of DSC was detected by MTT. Results IL-10R1 mRNA levels of DSC treated with IL-10 (10 ng/ml) reached the peak level within 15 rain, and were significantly lower at 30 rain, then were not detected at 45 min. The expression of IL-10R1 were induced to moderate level by IFN-γ(10 ng/ml) within 30 rain, and reduced to undetected levels at 60 min. There was no significant difference of IL-10R2 expression (P〉0.05) between treated and not with the abovementioned cytokines. The IL-10R protein expression and vitality of DSC were significantly enhanced by IL-10 (10 ng/ml) and IFN-γ (10 ng/ml) which treated DSC 48 h (P〈0.05). Coneclusion IL-10 and IFN-γ may play an important role of biologic function in early pregnancy by influencing IL-10R expression of DSC. 展开更多

After digestion and linearization, the recombinant vector was cloned into P. pastor/s GS115, high copy transformant and its methanol utilization type were screened, and its expression conditions were optimized. The results showed that ChlFN-α successfully expressed in P. pastor/s, the expression amount of recombinant protein was the largest after induced expression in 0.5 % methanol containing 2% acid hydrolyzed casein at 28 ℃ for 72 h, and its antiviral activity was the highest. 展开更多

The cell growth. rates were measured by crystal violet assay, and the expressionof PML was detected by immunofluomcent staining with PML Polyclonal antibody. Our study indicated that the growth rates or PC-3 and DU145 were significantly inhibited by IFN-α, whereas nosuch effect can be found on LNCaP cell line. The immunofluorescent staining showed that both thestaining intensity and the number or the PML nuclear boles were significantly increased in PC-3 andDU145 cells, however, no significant changes in the PML nuclear body were found in the IFs-αtreated LNCaP cells. Our study demeastrated that the antiproliferative,effct of IFN in prostate cancer cells may be directly related to the induced expression of PML in response to its treatment. Ourfinding supports the hypothesis that PML may mediate some antiproliferative erfects of IFN. 展开更多

Methods: The effects of treatment with rh-TRAIL alone and/or chemotherapetic drugs on MG-63 cell growth inhibition and apoptosis were measured using the MTT assay, FACS analysis ofAunexin V labeled cells, and the mRNA changes of DR5 and YY1 were detected by RT-PCR. Results: Rh-TRAIL protein inhibited the growth of MG-63 cells, and this inhibition was increased by adriamycin and IFN-γ. Adriamycin and IFN-γ significantly facilitated the induction of the expression of DR5 and reduced the expression of YY1. Conclusion: The apoptosis-inducing effect of rh-TRAIL in MG-63 cells was enhanced by chemotherapeutic drugs. 展开更多

AIM: To study the association between host immunity and hepatitis B virus (HBV) recurrence after liver transplantation. METHODS: Peripheral blood mononuclear cells (PBMC) were isolated from 40 patients with hepatitis B and underwent orthotopic liver transplantation (OLT) before and 2, 4, 8 wk after surgery. After being cultured in vitro for 72 h, the levels of INF-γ and TNF-α in culture supematants were detected with ELISA. At the same time, the quantities of HBV DNA in serum and PBMCs were measured by real time PCR. RESULTS: The levels of INF-γ and TNF-α in PBMC culture supernatants decreased before and 2, 4 wk after surgery in turns (INF-γ 155.52±72.32 ng/L vs 14.76±9.88 ng/L vs 13.22±10.35 ng/L, F= 6.946, P= 0.027〈0.05; TNF-α 80.839±46.75 ng/L vs18.59±17.29 ng/L vs9.758±7.96 ng/L, F= 22.61, P = 0.0001〈0.05). The levels of INF-γ and TNF-α were higher in groups with phytohemagglutinin (PHA) than in those without PHA before surgery. However, the difference disappeared following OLT. Furthermore, INF-γ and TNF-α could not be detected in most patients at wk 4 and none at wk 8 after OLT. The HBV detection rate and virus load in PBMC before and 2, 4 wk after surgery were fluctuated (HBV detected rate: 51.4%, 13.3%, 50% respectively; HBV DNA: 3.55±0.674 log(10) copies/mL vs 3.00±0.329 log(10) copies/mL vs 4.608±1.344 log(10) copies/mL, F= 7.582, P= 0.002〈0.05). HBV DNA in serum was 4.48±1.463 log(10) copies/mL before surgery and 〈 10^3 copies/mL after OLT except for one with 5.72×10^6 copies/mL 4 wk after OLT who was diagnosed as HBV recurrence. The levels of INF-γ and TNF-α were lower in patients with a high HBV load than in those with a low HBV load (HBV DNA detected/undetected in PBMCs: IFN-γ 138.08±72.44 ng/L vs 164.24±72.07 ng/L, t = 1.065, P= 0.297〉0.05, TNF-α 80.75±47.30 ng/L vs 74.10±49.70 ng/L, t= 0.407, P= 0.686〉 0.05; HBV DNA positive/negative: IFN-γ 136.77±70.04 ng/L vs 175.27±71.50 ng/L, t= 1.702, P= 0.097〉0.05; TNF-α 75.37±43.02 ng/L vs 81.53±52.46 ng/L, t = 0.402, P = 0.690〉0.05). CONCLUSION: The yielding of INF-γ and TNF-α from PBMCs is inhibited significantly by immunosuppressive agents following OLT with HBV load increased, indicating that the impaired immunity of host is associated with HBV recurrence after OLT. 展开更多

Objective:To investigate the effects of interferon-a (IFN-a) and IFN-a combined with interleukin-6 (IL-6) on growth and expression of bcr-abl, bcl-2 and c--myc genes in the mononuclear cells (MNCs) from bone marrow (BM) of patients with chronic myelogenous leukemia (CML). Methods: MNCs were collected from BM of the patients with CML in chronic phase by centrifugation in lymphocyte separation medium and cultured in liquid with IFN-a (200U/ml) or IFN-a (200U/ml) plus IL-6 (100 ng/ml). The growing cells were counted every day. The expression levels of b-actin, bcr-abl, bcl-2 and c-myc genes in the MNCs incubated for 24 h were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and relatively quantitative analysis of the amplified fragments by optical density scanning for the bands on gel. Results: The cell growth was markedly suppressed by IFN-a but the degree of cell-growth inhibition was slightly decreased by IL-6 on the basis of IFN-a effect. The expression of bcr-abl chimeric gene was intensely inhibited by IFN-a or IFN-a plus IL-6. The expression of bcl-2 gene was suppressed by either IFN-a or IFN-a plus IL-6, whereas that of c-myc gene was also inhibited by IFN-a but strongly elevated by IL-6 on the basis of IFN-a action. Conclusions: Both IFN-a and IFN-a plus IL-6 can inhibit the expression of anti-apoptosis gene such as bcr-abl and bcl-2 and regulate the expression of the cs.hn.cn gene related to cell proliferation and differentiation such as c-myc. Either IFN-a or IFN-a combined with IL-6 will serve as a trustful strategy of clinical treatment for CML. 展开更多

The level of FcγR on peripheral blood mononuclear cells (PBMC) was determined by ABC-ELIsA in 43 patients with primary hepatocellular carcinoma (PHC) and in 8 patients with space-occupying benign lesion (BL) in liver, twelve patients who received carcinomaectomy were followed up to detect their FcγR.Nine PBMC samples from PHC were treated with rIFNγ-The results showed that: 1. the level of FcγR in PHC patients was significantly lower than that in BL and In normal control; 2. the expression of FcγR was enhanced in postoperated PHC patients; and 3. the production of IFNγ in PHC patients was defective. The level of FcγR on PBMC was increased obviously in PHC after PBMC being treated with rIFNγ but was still lower than that in control. These results suggested; (1) FcyR detection is helpful tor distinguishing malignant liver lesion from benign; (2) the inhibited expression of FcγR in PHC could be related to some factors secreted by cancer tissues and also to low level of IFNγ; and (3) IFNγstimulates the expression of FcyR in vitro as one of the anti-tumor mechanism forIFNγ. 展开更多
