Objective:To determine the effects of Hydrangeae Dulcis Folium(EHDF)on physical stress,changes in the whole-body cortisol level and behaviour in zebrafish(Danio rerio).Methods:One hundred and seventy-four fish were randomly divided into 4[adrenocorticotropin hormone(ACTH)challenge test:4 fish per group]or 6 groups(behavioural test:10–12 fish per group,whole-body cortisol:4 fish per group).Net handling stress(NHS)was used to induce physical stress.Fish were treated with vehicle or EHDF(5–20 mg/L)for 6 min before they were exposed to stress.And then,fish were sacrificed for collecting body fluid from whole-body or conducted behavioural tests,including novel tank test and open field test,and were evaluated to observe anxiety-like behaviours and locomotion.In addition,to elucidate the mode of action of the anti-stress effects of EHDF,ACTH(0.2 IU/g,i.p.)challenge test was performed.Results:The increased anxiety-like behaviours in novel tank test and open field test under stress were prevented by treatment with EHDF at 5–20 mg/L(P<0.05).Moreover,compared with the unstressed group,which was not treated with NHS,the whole-body cortisol level was significantly increased by treatment with NHS(P<0.05).Compared with the NHS-treated stressed control group,pre-treatment with EHDF at concentrations of 5–20 mg/L for 6 min significantly prevented the NHS-increased whole-body cortisol level(P<0.05).In addition,ACTH challenge test showed that EHDF completely blocked the effects of ACTH on cortisol secretion(P<0.05).Conclusion:EHDF may be a good antistress candidate and its mechanism of action may be related to its positive effects on cortisol release. 展开更多

花色是植物重要的观赏性状之一,而黄烷酮羟化酶是植物花色素物质积累的关键酶,对植物花色差异具有重要影响。本研究利用PCR技术从盛花期蓝色花绣球品种‘无尽夏’不孕花中克隆获得了Hm F3H基因的全长序列,该基因的ORF序列长度为1 095 bp... 花色是植物重要的观赏性状之一,而黄烷酮羟化酶是植物花色素物质积累的关键酶,对植物花色差异具有重要影响。本研究利用PCR技术从盛花期蓝色花绣球品种‘无尽夏’不孕花中克隆获得了Hm F3H基因的全长序列,该基因的ORF序列长度为1 095 bp,编码364个氨基酸,编码蛋白的理论分子量为41.08 k D,理论等电点为5.48,为亲水性蛋白,该蛋白属于PLNO2515超家族成员,具有2-酮戊二酸双加氧酶结构域。系统进化树分析表明,该蛋白与咖啡树和佛手瓜的亲缘关系最近。实时定量PCR分析表明,Hm F3H基因在绣球花不同组织器官中均有表达,红色绣球品种‘粉黛’盛花期不孕花比蓝色绣球品种‘无尽夏’盛花期不孕花中表达量高,说明Hm F3H基因对绣球花花色差异形成具有一定影响。本研究获得了绣球花Hm F3H基因序列及其表达情况,为探讨绣球花品种花色差异形成机制具有一定指导意义。 展开更多

大叶绣球(Hydrangea macrophylla)是广受欢迎的花灌木,且品种繁多。为筛选园林应用价值较高的品种,本研究选取了株型、株高、枝条硬度、花序数量、显花度、花序直径、萼片颜色、萼片轮数、花期早晚、单花期10个生长及观赏性指标,应用灰... 大叶绣球(Hydrangea macrophylla)是广受欢迎的花灌木,且品种繁多。为筛选园林应用价值较高的品种,本研究选取了株型、株高、枝条硬度、花序数量、显花度、花序直径、萼片颜色、萼片轮数、花期早晚、单花期10个生长及观赏性指标,应用灰色关联度分析法和主成分分析法对25个大叶绣球品种的观赏性状进行了综合评价,并结合聚类分析法划分了各个品种的观赏性等级。分析结果将25个大叶绣球品种分为了4个等级,初步筛选出‘花手鞠’‘汉堡’‘康士坦’‘魔幻贵族’‘奇迹’以及‘雪舞’6个园林应用价值较高的品种。研究结果为大叶绣球的园林应用提供了借鉴。 展开更多

绣球(Hydrangea macrophylla)切花花大色艳、寓意美好,越来越受市场欢迎,但绣球切花花序硕大,对短暂失水反应强烈,极易因此丧失观赏价值。本研究以绣球切花为材料,研究了失水胁迫及复水对绣球切花寿命、花径、弯颈程度、鲜质量、可溶性... 绣球(Hydrangea macrophylla)切花花大色艳、寓意美好,越来越受市场欢迎,但绣球切花花序硕大,对短暂失水反应强烈,极易因此丧失观赏价值。本研究以绣球切花为材料,研究了失水胁迫及复水对绣球切花寿命、花径、弯颈程度、鲜质量、可溶性糖、呼吸强度、电导率及脯氨酸含量变化的影响。结果表明,失水主要引起绣球切花萎蔫,导致弯颈和花径变小,降低观赏价值。但不长于12 h的失水胁迫各时间点复水后瓶插寿命差异不显著。尽管稍长时间的失水对细胞膜有一定程度损害,但失水引起脯氨酸积累以抵御水分亏缺,短期失水可以经复水接近原始状态。绣球切花不长于12 h失水胁迫经复水后对其观赏品质及生理的影响不大,因此在采后流通中,如果条件限制只能干藏,建议时间不超过12 h,并应在此期间及时采用深水法复水以保证切花品质。 展开更多

Slow growth rate restricts the development and growth of seedlings due to nutrients deficiency or nutrient imbalance.Exponential fertilization can enhance the internal nutrient reserves in seedlings at the nursery-stage and strengthen their resistance to adverse conditions.In this study,nitrogen requirements for producing Hydrangea macrophylla‘Hanatemari’that robust seedlings,nutrient dynamics,biomass and growth,was examined utilizing exponential fertilization.The potted seedlings were fertilized with urea under exponential regime at rates of 0.5,1.5 and 2.0 g nitrogen/plant(EF1,EF2,and EF3),respectively.In addition,an unfertilized group treated with equal volume of deionized water was used as control.The results showed that seedlings under 1.5 g N/plant(EF2)had the highest plant growth index and total biomass.The nutrient concentrations of different organs varied in different fertilization treatments.Based on the results of current study,it is concluded that 1.5 g N/plant(EF2)is suitable exponential fertilization treatment for the culture of hydrangea seedlings.Our treatments results showed that 2.0 g N/plant is not suitable for seedling culturing,because of serious nutrient toxicity.These findings will help to improve seedling quality and strengthen the production of H.macrophylla for plantation. 展开更多

Hydrangea bretschneideri Dipp is a highly popular ornamental plant for garden decoration.Genetic engineering technology has been successfully used in many plant species,but it is limited in Hydrangea.Here we established an efficient regeneration system by using stem segments as explants for the first time.In our study,the plant growth regulators(PGRs)were evaluated at the different regeneration processes,including axillary shoots regeneration and root induction.We found that the optimal concentration for axillary buds’induction was 2.0 mgL^(-1)6-BA and 0.5 mgL^(-1)1 IAA,its highest induction rate was 70%.Moreover,the highest axillary shoots proliferation coefficient was 10.7 on the Murashige and Skoog(MS)medium with 2.0 mgL^(-1)6-benzyladenine(BA),0.2 mgL^(-1)indole-3-butyric acid(IBA),and 1.0 mgL^(-1)gibberellin A3(GA3).The highest frequency of root induction was 80.0±0.06%by culturing the elongated shoots in 1/2 MS medium containing 0.1 mgL^(-1)IBA.In summary,our study will provide an effective technology for large-scale propagation and important pathway for promoting the popularization and application of Hydrangea bretschneideri Dipp. 展开更多

Objective:To examine the inhibitory effect of Hydrangea serrata extract against hepatocellular carcinoma HepG2 cells and its underlying mechanisms.Methods:The effects of Hydrangea serrata extract on growth inhibition of tumor cells and spheroids were assessed using MTT and 3D culture assays.Quantitative real-time PCR and Western blot analyses were employed to investigate the changes in mRNA and protein expression levels of molecules related to cell cycle and apoptosis.Results:Hydrangea serrata extract effectively inhibited the growth of both tumor cells and spheroids.The extract also significantly upregulated p27 mRNA expression and downregulated CDK2 mRNA expression,leading to cell cycle arrest.Moreover,increased BAX/Bcl-2 ratio as well as caspase-9 and-3 were observed after treatment with Hydrangea serrata extract,indicating the induction of tumor cell apoptosis.Conclusions:Hydrangea serrata extract has the potential to alleviate tumors by effectively modulating cell-cycle-related gene expressions and inducing apoptosis,thereby inhibiting tumor growth. 展开更多
