[Objective]The paper was to provide theoretical basis for selection of parental combination and early identification of hybrids.[Method]The soluble protein and peroxidase of LiLum davidii var.unicolor,Lilium Asiatic hybrids and their filial generations were analyzed using polyacrylamide gel electrophoresis technique.[Result]The protein spectrum of filial generation with L.davidii var.unicolor as parent not only appeared the homologous band as parent with darker coloring,but also had new bands compared with parent.Peroxidase zymogram of hybrid F1 mainly displayed incomplete complementary and hybrid type of parent.[Conclusion]Protein spectrum and peroxidase zymogram could be used as the biochemical markers for the identification of hybrids of lily,which could also detect the target traits of plant. 展开更多

'Taisi N0.3' is an early mid-maturing towel gourd F1 hybrid developed by crossing Ps17-6-5-2-10-8 as female parent and Ps39-3-8-4-6-5 as male parent. The period from sowing to first harvest is about 75 days in spring cropping. It grows exuberantly in early stage and better in middle and later stages; leaves are green and palmately divided; the maximum leaf's length and width are about 25.5 and 33.7 cm. It bears fruit at the main and lateral branch. The first female flower sets on the 7^th-8^th node, and the fruit-setting rate is 72.8%. The fruit-setting rate is high. Its fruit is long round stick shape, 47.4 cm in length, and 3.75 cm in width, with smooth and green skin covered by dark green stripes, dense flesh. The fruit tastes good, with high Vc content. It is resistant to downy mildew, moderately resistant to CMV, and highly resistant to blight. The average fruit weight is about 282.3 g with the general yield of 82 500 kg/hm2. 展开更多

Male sterility induced by a chemical hybridization agent (CHA) is an important tool for utilizing crop heterosis. Leaves, especially the flag leaves, as CHA initial recipients play a decisive role in inducing male sterility. To investigate effects of different treatment times of CHA-SQ-1 used, morphological, biochemical and physiological responses of wheat flag leaves were detected in thistudy. CHA induced programmed cell death (PCD) as shown in terminal deoxynucleotidyl transferase-mediated dUTP nick end-labelling (TUNEL) and DNA laddering analysis. In the early phase, CHA-SQ-1 trig- gered organelle changes arid PCD in wheat leaves accompanied by excess production of reactive oxygen species (O2- and H202) and down-regulation of the activities of superoxide dismutase (SOD), catalase (CAT) and guaiacol peroxidase (POD). Meanwhile, leaf cell DNAs showed ladder-like patterns on agarose gel, indicating that CHA-SQ-1 led to the activation of the responsible endonuclease. The oxidative stress assays showed that lipid peroxidation was strongly activated and photosynthesis was obviously inhibited in SQ-l-induced leaves. However, CHA contents in wheat leaves gradually reduced along with the time CHA-SQ-1 applied. Young flags returned to an oxidative/antioxidative balance and ultimately developed into mature green leaves. These results provide explanation of the relations between PCD and anther abortion and practical application of CHA for hybrid breeding. 展开更多

One of the main diseases that adversely impacts the global citrus industry is citrus bacterial canker(CBC),caused by the bacteria Xanthomonas citri subsp.citri(Xcc).Response to CBC is a complex process,with both proteinDNA as well as protein–protein interactions for the regulatory network.To detect such interactions in CBC resistant regulation,a citrus high-throughput screening system with 203 CBC-inducible transcription factors(TFs),were developed.Screening the upstream regulators of target by yeast-one hybrid(Y1H)methods was also performed.A regulatory module of CBC resistance was identified based on this system.One TF(CsDOF5.8)was explored due to its interactions with the 1-kb promoter fragment of CsPrx25,a resistant gene of CBC involved in reactive oxygen species(ROS)homeostasis regulation.Electrophoretic mobility shift assay(EMSA),dual-LUC assays,as well as transient overexpression of CsDOF5.8,further validated the interactions and transcriptional regulation.The CsDOF5.8–CsPrx25 promoter interaction revealed a complex pathway that governs the regulation of CBC resistance via H2O2homeostasis.The high-throughput Y1H/Y2H screening system could be an efficient tool for studying regulatory pathways or network of CBC resistance regulation.In addition,it could highlight the potential of these candidate genes as targets for efforts to breed CBC-resistant citrus varieties. 展开更多

The proprietary Chinese medicine preparation Kaiyu Granule is made of bupleurum, nutgrass ga- lingale rhizome, szechwan Iovage rhizome, turmeric root tuber, white peony alba, cape jasmine fruit fried semen ziziphi jujubae, and prepared liquorice root. It is a common recipe for the clinical treatment of depression in China. In this study, after 21 days of unpredictable stress exposure, Wistar rats exhibited similar behavioral changes to patients with depression. Moreover, G-protein-coupled inwardly rectifying K+ channel 1 mRNA and protein expression were significantly reduced in rat hippocampal CA1 and CA3 regions. However, G-protein-coupled inwardly rectifying K+ channel 1 mRNA, protein expression, and rat behavior were clearly better after administration of 12, 8, or 4 g/kg of Kaiyu Granule when depression model rats underwent stress. 12 g/kg of Kaiyu Granule had the most obvious effects on the increased expression of G-protein-coupled inwardly rectifying K+ channel 1 mRNA and protein in rat hippocampal CA1 and CA3 regions. These results suggested that Kaiyu Granule improved depression by affecting G-protein-coupled inwardly recti- fying K+ channel 1 expression in the rat hippocampus. 展开更多

A submerged internal circulating membrane coagulation reactor (MCR) was used to treat surface water to produce drinking water. Polyaluminum chloride (PAC1) was used as coagulant, and a hydrophilic polyvinylidene fluoride (PVDF) submerged hollow fiber microfiltration membrane was employed. The influences of trans-membrane pressure (TMP), zeta potential (ZP) of the suspended particles in raw water, and KMnO4 dosing on water flux and the removal of turbidity and organic matter were systematically investigated. Continuous bench-scale experiments showed that the permeate quality of the MCR satisfied the requirement for a centralized water supply, according to the Standards for Drinking Water Quality of China (GB B749-2006), as evaluated by turbidity (〈1 NTU) and total organic carbon (TOC) (〈5 mE/L) measurements. Besides water flux, the removal of turbidity, TOC and dissolved organic carbon (DOC) in the raw water also increased with increasing TMP in the range of 0.01-0.05 MPa. High ZP induced by PAC1, such as 5-9 mY, led to an increase in the number of fine and total particles in the MCR, and consequently caused serious membrane fouling and high permeate turbidity. However, the removal of TOC and DOC increased with increasing ZP. A slightly positive ZP, such as 1-2 mV, corresponding to charge neutralization coagulation, was favorable for membrane fouling control. Moreover, dosing with KMnO4 could further improve the removal of turbidity and DOC, thereby mitigating membrane fouling. The results are helpful for the application of the MCR in producing drinking water and also beneficial to the research and application of other coagulation and membrane separation hybrid processes. 展开更多

Objective: Vascular-targeted therapy is gradually becoming more appealing for patients with lung cancer. It is unclear whether vascular endothelial growth factor receptor 2(VEGFR2) and neuropilin-1(NRP-1) can be biomarkers for clinical treatment. We aimed to investigate the expression levels of VEGFR2 and NRP-1 in human non-small cell lung cancer(NSCLC) and their clinical significance by observing patient prognosis. Methods: VEGFR2 and NRP-1 were assessed by immunohistochemistry(IHC) in 40 patients with NSCLC and in 10 patients with benign lesions of lung; kinase insert domain receptor(KDR) and NRP-1 copy number gain(CNG) was assessed by fluorescence in situ hybridization(FISH). The distributions of overall survival(OS) and progression-free survival(PFS) were estimated using the Kaplan-Meier method and compared between groups by log-rank test.Results: Rates of positive immunostaining for VEGFR2 and NRP-1 were 58% and 55%, respectively. KDR and NRP-1 CNG(+) were detected in 32.5% and 30% of tumors, respectively. Levels of both VEGFR2 and NRP-1 in lung tumors were significantly different than in the control tissue(χ2=11.22, P=0.001; χ2=9.82, P=0.001, respectively); similar results were obtained using CNGs(χ2=4.39, P=0.036; χ2=3.95, P=0.046, respectively). Statistically significant correlations were observed with histological grade, clinical TNM stage and the lymph node status(P〈0.05), but not age, gender or pathology type(P〉0.05). VEGFR2 showed a strong correlation with NRP-1(Rs=0.68, P=0.00); similar results were observed with KDR and NRP-1 CNG(Rs=0.32, P=0.04). Significant differences in OS and PFS were observed between the groups with higher VEGFR2 and NRP-1 and those with lower expression(P〈0.05). Conclusions: According to these data, VEGFR2 and NRP-1 are highly expressed in NSCLC. We can conclude that they play a key role in NSCLC occurrence, development and metastasis and are associated with patient prognosis(P〈0.05 for OS and PFS). This information will be beneficial for clinical antiangiogenic treatment in NSCLC. 展开更多

Heading date is a crucial agronomic trait.However,rice usually delays heading due to the photoperiod,temperature,hormones or age.The present research was conducted to analyze the mechanism cotrlling heading date in F,hybrid rice.We constructed two test-crossing populations using two introgression lines(ILs),P20 and P21 coming from SH527/FH838 as the male parent,respectively,and male sterile line Jin23A as the female parent.Meanwhile,the F,hybrids of H20,obtained by mating P20 with Jin23A and having no heading,and H21,from the crossing between P21 and Jin23A having normal heading,were both observed under long days.Here,we analyzed the photoperiodic response of F,hybrids by transcriptome and metabolome profiling.The greater differences displayed in the transcriptome and the metabolome were caused by photoperiod(exogenous)instead of genes(endogenous).The coping mechanism resulted from long days(LD)in H20,leading to differences in the circadian rhythm and glutathione metabolism relative to other samples.The circadian oscillator and GSH/GSSG cycle typically regulate ROS homeostasis,and both of them are responsible for modulating ROS in H20 under LD condition.Both circadian rhythm genes and the reported genes related to heading date function via the DHD1/OsMFT1-Ehd1-RFT1-OsMADS14/OsMADS18 pathway and the glutathione metabolism pathway by regulating oxidative reduction processes.Both pathways are involved in the heading process and they interacted through the oxidative reduction process which was induced by photoperiod regulation,and all of them collectively modulated the heading process.The results of this study will be helpful for unraveling the mechanism of F,hybrid responses to unheading under LD condition. 展开更多

Through the use of broccolini innovation resources, a group of unique ex- cellent broccolini and male sterile lines, as well as inbred lines were bred. In the research, 4 hybrid strains were bred by the use of male sterile lines which had vigorous growth, high resistance, good quality, good commodity and high yield, in- cluding Beauty No.l, Beauty No.2, Beauty No.3, Beauty No.4. The results showed that Beauty No.2 behaved the best comprehensive characters of high yield, high commercial value and good taste. The yield of Beauty No.2 was 28 185 kg/hm2 and 25% higher than the control Qianxiu, the sulforaphane content was as high as 48 mg/kg and 77.8%, 272.1% higher than the control of Qianxiu and Manuolv broc- coli, respectively. The comprehensive character of Beauty No.1 ranked the second, the yield was the highest (29 265 kg/hm2) and 30% than the control Qianxiu. 展开更多

Proteomic analysis was carried out on the Crab(upper-shore)and Wave(lower-shore)ecotypes of Littorina saxatilis from a hybrid zone at Silleiro Cape,Spain.Proteome profiles of individual snails were obtained.Protein expression in F_(1)hybrid snails bred in the laboratory and snails with intermediate shell phenotypes collected from the mid-shore were compared with Crab and Wave ecotypes using analytical approaches used to study dominance.Multivariate analysis over many protein spots showed that the F_(1)snails are distinct from both ecotypes but closer to the Wave ecotype.The intermediate snails are highly variable,some closer to the Crab and others to the Wave ecotype.Considered on a protein by protein basis,some proteins are significantly closer in expression to the Crab and others to the Wave ecotype for both F_(1)and intermediate snails.Furthermore,a significant majority of proteins were closer in expression to the Wave ecotype for the F_(1),consistent with the multivariate analysis.No such significant majority toward either the Crab or Wave ecotype was observed for the intermediate snails.The closer similarity of F_(1)and Wave ecotype expression patterns could be the result of similar selective pressures in the similar mid-shore and low-shore environments.For a significantly larger number of proteins,intermediate snails were closer in expression to the ecotype having the lower expression,for both Crab and Wave ecotypes.This is somewhat unexpected as lower expression might be expected to be an indication of impairment of function and lower fitness.Proteomic analysis could be important for the identification of candidate proteins useful for gaining improved understanding of adaptation and barriers to gene flow in hybrid zones. 展开更多

Five of each buffalos were bred by freely grazing and slaughtered at 35. 8-month old age to investigate the fatty acid composition and amino acid content in beef of Dehong buffalo,MD buffalo and ND buffalo. The results showed that,saturated fatty acid content of Dehong buffalo( 44. 33%) was significantly higher than that of ND buffalo( 41. 39%) and MD buffalo( 41. 48%)( P 【 0. 05),and the latter two were not significantly different( P 】 0. 05). The content of PUFA in Dehong buffalo( 53. 90%) was significantly lower than ND( 56. 83%) and MD( 56. 83%)( P 【0. 05),however,the n- 6∶ n- 3( 4. 25) of Dehong buffalo was significantly higher than ND( 3. 14) and MD( 3. 52)( P 【 0. 05). Hybrid rice Fanyou 7206(FY7206), derived from the cross between a sterile line Fanyuan A and a restorer line Fuhui 7206, was bred by the Rice Research Institute, Fujian Academy of Agricultural Sciences, China. FY7206 was characterized by moderate blast resistance, cold tolerance, as well as wide adaptability, and high yields. The blast resistance results indicated that the frequencies of blast races in race B, race C and the total resistance frequency for FY7206 were 95.5%, 100.0% and 97.2%, respectively. The disease resistance results showed that the leaf blast grade for FY7206 was level 1 and panicle blast was level 5. The indoor spray results indicated that FY7206 was resistant to 11 isolates of Magnorpathe oryzae. The blast resistance of FY7206 might be derived from the high expression of blast resistance gene Pid3. The results for simulated cold resistance in an artificial climate chamber indicated that the cold tolerance for FY7206 was moderate at the booting and flowering stages. The cold tolerance results also indicated that FY7206 could be tolerant to temperatures as low as 10 °C at the seedling stage. The q RT-PCR results showed that the expression of cold tolerance gene Ctb1 in FY7206 was relatively high. These results suggested that FY7206 is a hybrid indica rice variety with good comprehensive characteristics, including blast resistance and cold tolerance. 展开更多

We explore the decay properties of the isovector and isoscalar 1^(-+)light hybrids,π_(1)andη_(1),in N_(f)=2 lattice QCD at a pion mass m_(π)≈417 MeV.The Mc Neile and Michael method is adopted to extract the effective couplings for individual decay modes,which are used to estimate the partial decay widths ofπ_(1)(1600)andη_(1)(1855)by assuming SU(3)symmetry.The partial decay widths ofπ_(1)(1600)are predicted to be(Γ_(b_(1)π),Γ_(f1(1285)π),Γ_(ρπ),Γ_(K*K))=(325±75,O(10),52±7,8.6±1.3)Me V,and the total width is estimated to be 396±90 Me V,considering only statistical errors.Ifη_(1)(1855)and the 4.4σsignal observed by BESIII(labeled asη_(1)(2200))are taken as the two mass eigenstates of the isoscalar 1^(-+)light hybrids in SU(3),then the dominant decay channel(s)ofη_(1)(1855)(η_(1)(2200))is K_(1)(1270)K(K_(1)(1270)K and K_(1)(1400)K) through the 1^(+(-))0^(-(+))mode.The vector-vector decay modes are also significant for the twoη_(1)states.Using the mixing angleα≈22.7°obtained from lattice QCD for the twoη_(1)states,the total widths are estimated to beΓ_(η1(1855))=282(85)MeV andΓ_(η1(2200))=455(143)MeV.The former is compatible with the experimental width ofη_(1)(1855).Although many systematic uncertainties are not well controlled,these results are qualitatively informative for the experimental search for light hybrids. 展开更多
