To solve the problems of microbial contamination and no rooting during the tissue culture of Huperzia serrata, wild seedlings of Huperzia serrata were cultured by indoor hydroponics or soil cultivation methods, then the stem tips were used in tissue culture. The above operation significantly reduced microbial contamination during tissue culture process. Different types and concentrations of hormones were added into the basic medium MS to screen the optimal formula for induced plantlet growth and rooting. It was found that 1/2MSCO was the best medium on which stem, leaves and roots of tissue culture plantlets of Huperzia serrata grew quickly. At the same time, sand planting was a convenient and effective approach for gemmae propagation of Huperzia serrata, the survival rate of seedlings germinated from gemmae was nearly 100%. 展开更多

[Objectives]This paper aims to study the function of the ARF7 gene family of Huperzia serrata.[Methods]Based on the full-length transcriptome sequencing results of H.serrata,the unigenes of 17 H.serrata ARF7 candidate genes were analyzed using biogenetics technology.[Results]The molecular weight of the ARF7 proteins ranges from 11.28 to 608.54 kDa.HsARF7-4,HsARF7-5,HsARF7-8,HsARF7-9,HsARF7-10,HsARF7-13,HsARF7-16 and HsARF7-17 are basic proteins;all the proteins are unstable;except that HsARF7-6 and HsARF7-15 are hydrophilic,the rest are lipophilic proteins;there are 197 types of cis-acting elements,and 17 of the proteins contain as more as 20 acting elements such as GTGANTG10,CBFHV,GT1CONSENSUS,NODCON2GM,CAATBOX1,MYBCORE,GATABOX,WRKY71OS and RAV1AAT;HsARF7-1,HsARF7-2,HsARF7-4,HsARF7-6,HsARF7-8,HsARF7-10,HsARF7-12,HsARF7-14 and HsARF7-16 all have the 6 types of motif predicted;the proteins have domains such as PABP-1234 super family,RRM_SF super famliy,PRK10263 super family,AUX_IAA super family and MFS super family;the proteins all can cross cell membrane and have no signal peptide,and they are all located in the nucleus.On the level of secondary structure,the proportion ofβ-sheet is the smallest.In HsARF7-7 and HsARF7-9,α-helix has the largest proportion,and in the remaining proteins,random coil has the largest proportion;and tertiary structure prediction found that HsARF7-1,HsARF7-2,HsARF7-4,HsARF7-14 and HsARF7-17 have a tertiary structure of polyadenylate-binding protein,and HsARF7-7 has a tertiary structure of auxin-induced protein iaa4.[Conclusions]The above analysis results can provide a certain theoretical basis for further research on the biological function and regulatory mechanism of the ARF gene of H.serrata in the future,and provide a reference for the study of the growth and development of H.serrata. 展开更多

Background:The male reproductive system consists of the testes,epididymis,accessory gonads,and genital ducts.The physiological integrity of the testicles is essential for successful sperm production,but the testes are susceptible to a variety of exogenous and endogenous toxins,which can lead to structural damage to tissues and sperm abnormalities.Spermatogenesis is always in a homeostatic process and is sensitive to changes in the external environment.Huperzia serrata flavonoids(HSF)have a variety of pharmacological effects,including the treatment of bruises,strains,etc.HSF can improve the reproductive function of male mice,so the purpose of this experiment is to explore whether huperzine flavonoids are involved in and regulate the male reproductive process.Methods:Our experimental animals were ICR mice,and samples were collected after 28 days of HSF treatment,and verified by a series of experimental methods such as immunofluorescence,immunohistochemistry,HE staining,protein hybridization,and screening of related genes.Results:RNA sequencing results showed that HSF up-regulated genes related to sperm development and differentiation in testes and promoted spermatogenesis.HSF is effective in promoting steroid hormone synthesis by modulating STAR,CYP11A1,and CYP17A1.Conclusion:Thus,HSF can regulate testicular gene expression,stabilize the gut microbiome,alter serum metabolites,and regulate steroid homeostasis. 展开更多

Qian ceng Ta,the whole plant of Huperzia serrata,is an important landscape and medicinal herbs and contains abundant bioactive lycopodium alkaloids.Although the structures of more than 100 lycopodium alkaloids in Huperzia serrata have been isolated and identified,the content and distribution of these alkaloids in different tissues are still unclear.In current study,an ultra-performance liquid chromatography-mass spectrometry based comprehensive metabolomics strategy was developed,including the extraction,separation,identification,and statistical analysis.The results showed that different types lycopodium alkaloids could be separated at different time-windows,which was helpful for further metabolite identification.Peak4388 and peak3954 were metabolite biomarkers for the different tissues according to the principle component analysis and partial least squares-discriminant analysis model.A computational tool based in-house database was also built up and used for putative identification.Of the 2354 true peaks after four-step filtration,118 peaks were putatively identified as lycopodium alkaloids by using in-house database,and four of which was identified by authentic standards.Alternatively,another computational software was used to predict the fragmentation pattern,to dereplicate the structure of identified peaks,and identified the peak3585 to N-methylhuperzine A.The integration of both computational tools could be used for more metabolites identification. 展开更多

【背景】从植物中分离得到的内生细菌能产生多种多样的活性物质,能够促进宿主植物的生长发育,而目前对于珍稀野生药用植物蛇足石杉内生细菌分离及其促生功能的研究鲜见报道。【目的】从蛇足石杉中分离内生细菌并进行鉴定,从中筛选出具... 【背景】从植物中分离得到的内生细菌能产生多种多样的活性物质,能够促进宿主植物的生长发育,而目前对于珍稀野生药用植物蛇足石杉内生细菌分离及其促生功能的研究鲜见报道。【目的】从蛇足石杉中分离内生细菌并进行鉴定,从中筛选出具有多种促生功能的菌株,为蛇足石杉内生菌资源的开发利用提供参考。【方法】通过使用不同培养基对蛇足石杉不同组织部位的内生细菌进行分离,再根据菌落形态特征及16S rRNA基因序列测序比对进行菌种鉴定及系统发育分析,最后对菌株的固氮、溶磷、产铁载体、产吲哚乙酸(indole-3-acetic acid,IAA)这4种促生功能进行检测。【结果】从蛇足石杉根、茎、叶3种组织中共分离得到168株内生细菌,在分类学上归属于3门4纲5目6科16属,其中芽孢杆菌属(Bacillus)占55.95%,类芽孢杆菌属(Paenibacillus)占19.05%,为优势菌属;不同组织分离出的内生细菌数量与种类均有明显差异,表现为根部>茎部>叶部;从分离的内生细菌中筛选出95株具有固氮能力,76株具有溶磷能力,23株具有产铁载体能力,81株具有产IAA能力。【结论】从蛇足石杉分离的内生细菌具有丰富的物种多样性和促生功能,分布具有明显的组织特异性,并获得多株具有良好促生功能的菌株,为进一步开发菌肥资源提供了新的菌株材料,同时为进一步开发利用蛇足石杉内生菌资源奠定了基础。 展开更多

YUCCA基因编码黄素单加氧酶,在植物生长发育过程中起重要调控作用。为探究YUCCA基因对蛇足石杉生长发育的作用,对蛇足石杉YUCCA基因家族进行生物信息学及组织特异性表达分析。结果表明,在蛇足石杉全长转录组中共鉴定出18个HsYUCCA基因家族成员,氨基酸长度为150~544 aa,蛋白质相对分子量为16105.53~59058.24 Da,等电点在4.36~9.63,其中,有9个为碱性蛋白,有4个为稳定蛋白,有15个为亲水蛋白,有7个具有跨膜结构,分布在细胞核、细胞膜、细胞质、线粒体、叶绿体以及过氧化物酶体中。HsYUCCA4在孢子、茎、芽胞中的表达量最高,HsYUCCA6在叶中的表达量最高。HsYUCCA启动子区拥有激素响应、光响应、胁迫响应等35种顺式作用元件,并具有15种保守基序。HsYUCCA1、HsYUCCA2和HsYUCCA4,HsYUCCA5和HsYUCCA6,HsYUCCA7和HsYUCCA13具有相似的理化性质、亚细胞定位、二级结构、三级结构及保守基序,系统发育表现出一定的同源性,进化过程趋于一致。以上研究结果为解析蛇足石杉YUCCA基因在生长发育和繁殖过程中的作用机理提供了理论依据。 展开更多

Trev.)铜胺氧化酶(copper-containing amine oxidase, CAO)是石杉碱甲(huperzine A, HupA)生物合成途径中的关键酶之一。研究蛇足石杉CAO家族不同成员间的相互作用,筛选出高活性的蛇足石杉CAO或CAO组合,有望为后续通过生物工程方法实现HupA的合成、加快新药创制进度提供理论依据。本研究基于蛇足石杉转录组数据中得到的一个CAO-like基因片段,利用cDNA末端快速扩增(rapid amplification of c DNA ends, RACE)技术克隆到其cDNA全长,并将其命名为Hs CAO3。Hs CAO3的cDNA全长为2 736 bp,CDS为2 202 bp,共编码733个氨基酸。氨基酸序列同源性对比和进化树分析结果表明Hs CAO3与蛇足石杉中的另一个CAO (HsCAO1)具有较高同源性;蛋白三级结构模拟结果暗示Hs CAO3可能以同源二聚体的形式存在并发挥生物学功能。在此基础上进一步构建了pGBKT7-Hs CAO3重组载体,并进行了转录自激活检测,结果表明Hs CAO3无自激活活性。本研究为后续探究不同Hs CAOs间的相互作用奠定了基础。 展开更多

通过对我国蛇足石杉主要分布区35个野外样点和总样方量71300m2的调查,结合国内12个标本馆470余号蛇足石杉馆藏标本及现有文献的研究表明:蛇足石杉的主要分布区位于我国中亚热带和南亚热带所在的地理区域,幅员面积260×104km2,实际... 通过对我国蛇足石杉主要分布区35个野外样点和总样方量71300m2的调查,结合国内12个标本馆470余号蛇足石杉馆藏标本及现有文献的研究表明:蛇足石杉的主要分布区位于我国中亚热带和南亚热带所在的地理区域,幅员面积260×104km2,实际分布生境总面积约39×104km2;性喜阴湿,所适应的生态环境为中亚热带常绿阔叶林和南亚热带季风常绿阔叶林及其次生生态系统;野外生物量的调查得到了蛇足石杉在不同的地理区域的生物量分布格局,并由此结合专家系统对直接取样结果和间接推导数据的权重分析及环境参数的评估,初步确定我国蛇足石杉天然生物量为4656.6t;并以此为基础,在进一步考虑人的采集能力的前提下,判定我国蛇足石杉的理论可采量为1604t;同时指出,在目前收购价格条件下(30~40元/kg),我国蛇足石杉商业可采量大约不会超过200t。由于蛇足石杉资源分散,而且许多相对集中的产地均位于自然保护区和风景区范围内,因此,不可大规模开采,解决资源短缺的问题还有赖于寻求其他更加科学合理的途径。另外,论文为小型植物的生物量和可采量评估提供了一些新思路和方法。 展开更多

目的:研究药用植物蛇足石杉内生真菌的分布及其多样性,并筛选具有乙酰胆碱酯酶抑制活性的内生真菌。方法:采用平板分离法分离蛇足石杉内生真菌,结合形态学和ITS-rDNA序列分析鉴定内生真菌;并采用DTNB显色法,对分离获得的内生真菌进行乙... 目的:研究药用植物蛇足石杉内生真菌的分布及其多样性,并筛选具有乙酰胆碱酯酶抑制活性的内生真菌。方法:采用平板分离法分离蛇足石杉内生真菌,结合形态学和ITS-rDNA序列分析鉴定内生真菌;并采用DTNB显色法,对分离获得的内生真菌进行乙酰胆碱酯酶抑制活性的筛选。结果:共分离到127株内生真菌,分属于Penicillium,Aspergillus,Podospora等19个属;根、茎、叶中内生真菌的分布存在一定的组织差异性,茎中内生真菌的分离率和定殖率最高,叶中次之,根中最少。共有来自15个属的39株内生真菌显示出乙酰胆碱酯酶抑制活性,且11株有显著的抑制活性,其中7株来源于叶中。结论:药用植物蛇足石杉内生真菌多样性丰富,不同组织部位内生真菌的数量、种类及分布存在一定差异;同时蛇足石杉蕴含着大量抑制乙酰胆碱酯酶活性的内生真菌,为寻找和开发具有乙酰胆碱酯酶抑制活性的药用资源开辟了一条新的途径。 展开更多
