H pylori infection and CagA status were assessed in patients by enzyme-linked immunosorbent assay (EUSA) method. RESULTS: We found a significant increased frequency of HLA-DQBI*0401 allele in Hpylori-positive patients with chronic gastritis when compared with healthy subjects [19 vs 0%, P = 1 × 10^-7, odds ratio (OR) = 4.96; 95% confidence interval (95% CI), 3.87-6.35]. We also found a significant increased frequency of HLA-DQBI*0501 in patients with diffuse-type gastric carcinoma in comparison with healthy individuals (P = 1 × 10^4, OR = 13.07; 95% CI, 2.82-85.14).CONCLUSION: HLA-DQ locus may play a different role in the development of H pylori-related chronic gastritis and difffuse-type gastric adenocarcinoma in the Mexican Mestizo population.展开更多

METHODS: One hundred and sixty H pylori posi...AIM: To investigate the efficacy and tolerability of H pylori eradication in an omeprazole-based triple therapy with high and low dose of clarithromycin and amoxicillin. METHODS: One hundred and sixty H pylori positive patients were randomly assigned to two groups based on the following 2 wk investigation; (1) group A or low-dose regimen received omeprazole 20 mg b.i.d, clarithromycin 250 mg b.i.d and amoxicillin 500 mg b.i.d; and (2) group B or high-dose regimen received omeprazole 20 mg b.i.d, clarithromycin 500 mg b.i.d and amoxicillin 1000 mg b.i.d. During the study Hpylori status was assessed by histology and rapid urease test prior and by 13C-urea breath test 6 wk after the therapy. Standard questionnaires were administered to determine the compliance to treatment and possible adverse events of therapy. Data were subject to x^2 to compare the eradication rates in the two groups. The significant level of 95% (P ≤ 0.05) was considered statistically different. RESULTS: We found that the per-protocol eradication rate was 88% (68/77) in group A, and 89% (67/75) in group B. The intension-to-treat eradication rate was 85% (68/80) in group A and 83.75% (67180) in group B. Overall adverse events were 26% in group A and 31% in group B. The adverse events were generally mild in nature and tolerated well in both groups with a compliance of 98% in group A vs 96% in group B. CONCLUSION: The omeprazole-based low dose regimen of darithromycin and amoxicillin for two weeks in Hpylori eradication is as effective as high dose regimen in Iranian population.展开更多
Histone methylation is involved in a wide range of biological regulation in plants,and is conducted by three major components,including methyltransferases,demethylases,and histone readers.Compared with the other two c...Histone methylation is involved in a wide range of biological regulation in plants,and is conducted by three major components,including methyltransferases,demethylases,and histone readers.Compared with the other two components,research on histone readers is relatively limited.In this study,we demonstrate that OsSHH5 functions as an H3K9me1 reader to regulate rice disease resistance,tillering,and grain yield.Loss of OsSHH5 function significantly enhances both grain yield and disease resistance.Mechanistically,OsSHH5 recruits the H3K9 methyltransferase SGD733 and binds to H3K9me1,thereby maintaining H3K9me1 enrichment and facilitating gene silencing.In leaves,OsSHH5 interacts with the transcriptional factor HPY1 to target the resistance-related genes OsWAKg52 and OsWRKY81,maintaining their H3K9me1 levels and suppressing multiple PAMP-triggered immune responses,which ultimately reduces rice disease resistance.In tiller buds,OsSHH5 interacts with the transcriptional factor TCP19 to target the tillering-related gene OsNGR5,maintaining its H3K9me1 enrichment and inhibition of tillering,leading to reduced yield.Collectively,these findings reveal that OsSHH5 plays a vital role in integrating immune response,tillering,and grain yield in rice,providing new insights into the function of histone readers and offering a new strategy to improve rice yield and disease resistance.展开更多
