OBJECTIVE:To explore if Hewei Jiangni granule(和胃降逆颗粒,HWJNG)could regulate esophageal hypersensitivity via stromal interaction molecule 1(STIM1)/transient receptor potential vanilloid subfamily member 1(TRPV1)pat...OBJECTIVE:To explore if Hewei Jiangni granule(和胃降逆颗粒,HWJNG)could regulate esophageal hypersensitivity via stromal interaction molecule 1(STIM1)/transient receptor potential vanilloid subfamily member 1(TRPV1)pathway.METHODS:Qualitative analysis of HWJNG was analysis by high performance of liquid and gas chromatography.In vivo,animal model of non-erosive reflux disease(NERD)was established by fructose intake and restraint stress.HWJNG and Omeprazole were administered by gavage to the drug intervention group.Reflux and visceral hypersensitivity were analyzed by pathological changes,PH value test,mechanical paw withdrawal threshold,thermal withdrawal latency and mast cells(MCs)degranulation.In vitro,substance P(SP)-induced P815 cells and dorsal root ganglion(DRG)cells were cocultured.Expression in both mice and cells of STIM1,TRPV1,and esophageal visceral hypersensitivity-related gastrointestinal neurochemicals were validated by enzyme linked immunosorbent assays,quantitative realtime polymerase chain reaction(qRT-PCR)and Western blot.Moreover,overexpression and small interfering RNA against STIM1 were utilized to verify of the role of HWJNG in DRG cells.RESULTS:HWJNG significantly suppressed intercellular space widening,injury of mitochondrial,MCs degranulation,mechanical allodynia and heat neuropathic sensory and increased pH value of esophageal mucosa in NERD mice.HWJNG inhibited expression of visceral hypersensitivityrelated gastrointestinal neurochemicals in esophageal mucosa and activated P815 cells,and expression of the STIM1,TRPV1 and related neurotransmitters in DRG and DRG cells.STIM1 siRNA and HWJNG both reduced P815 cells adhesion to DRGs cells and Ca2+flow into the cytoplasmic space of DRG cells.Furthermore,HWJNG could reversed STIM1 overexpression induced upregulation of TRPV1.CONCLUSION:HWJNG suppressed intercellular space widening in NERD mice,stabilized MCs and restored neuronal hyperexcitability by regulating visceral hypersensitivity via STIM1/TRPV1 pathway.展开更多
Objective:To observe the clinical efficacy and safety of Wenzhong Hewei Formula in treating spleen and stomach qi deficiency syndrome in internal medicine diseases.Methods:Sixty patients with spleen and stomach qi def...Objective:To observe the clinical efficacy and safety of Wenzhong Hewei Formula in treating spleen and stomach qi deficiency syndrome in internal medicine diseases.Methods:Sixty patients with spleen and stomach qi deficiency syndrome admitted to the hospital from April 2022 to June 2023 were randomly divided into observation and control groups,with 30 patients in each group.The control group received conventional internal medicine treatment,while the observation group was additionally treated with Wenzhong Hewei Formula on the basis of conventional treatment.Both groups were treated for 4 weeks.Results:The total effective rate of treatment in the observation group was higher than that of the control group(P<0.05).After treatment,the traditional Chinese medicine syndrome scores of both groups were significantly lower than before treatment,with the observation group showing a more pronounced reduction(P<0.05).There was no statistically significant difference in the incidence of adverse reactions between the two groups(P>0.05).Conclusion:Wenzhong Hewei Formula can effectively improve clinical symptoms in patients with spleen and stomach qi deficiency syndrome,enhance clinical efficacy,and have a high level of safety,making it worthy of clinical promotion.展开更多
OBJECTIVE:To observe the effects of the Bupi Hewei(BPHW)decoction on diarrhea and intestinal flora disorder induced by 5-fluorouracil(5-FU)and investigate the possible mechanism underlying these actions.METHODS:Thirty...OBJECTIVE:To observe the effects of the Bupi Hewei(BPHW)decoction on diarrhea and intestinal flora disorder induced by 5-fluorouracil(5-FU)and investigate the possible mechanism underlying these actions.METHODS:Thirty-five male Sprague-Dawley rats were randomly divided into four groups:normal control,5-FU,5-FU+BPHW decoction(10.5 g/kg for 5 consecutive days),and 5-FU+Bacillus licheni-formis capsule groups(0.2 g/kg for 5 consecutive days).Animal models were established via the intraperitoneal injection of 5-FU(30 mg/kg for 5 consecutive days).At the end of the treatment period,diarrhea was assessed,and the change of the intestinal flora was examined using 16 S r DNA highthroughput sequencing.Interleukin(IL)-17,IL-21,IL-6,IL-10,RAR-related orphan receptor gamma(RORγt),and forkhead box P3(Foxp3)expression in the jejunum was detected using immunohistochemistry,quantitative real-time polymerase chain reaction(PCR),Western blotting,and enzymelinked immuno sorbent assay.RESULTS:In this study,the BPHW decoction effectively lowered the diarrhea score,increased the proportions of Bacteroidetes and Prevotellaceae-Alloprevotella species,and reduced the proportions of Proteobacteria,Escherichia-Shigella,Ruminococcaceae NK4 A214,and Ruminococcaceae UCG-005 species in the rat intestine after 5-FU chemotherapy.In addition,the BPHW decoction significantly suppressed the expression of IL-17,IL-21,IL-6,IL-10,RORγt,and Foxp3 in the jejunum.CONCLUSION:Our findings suggest that the BPHW decoction can improve the intestinal immune balance and reduce intestinal inflammation by targeting T helper cell/T regulatory cell-associated factors.展开更多
Objective:Shugan Hewei therapy(SHT)acts to soothe the liver and harmonize the stomach.It is a classical traditional Chinese medicine method widely used in China to treat chronic atrophic gastritis(CAG)due to liver qi ...Objective:Shugan Hewei therapy(SHT)acts to soothe the liver and harmonize the stomach.It is a classical traditional Chinese medicine method widely used in China to treat chronic atrophic gastritis(CAG)due to liver qi invading the stomach.However,the clinical effects of SHT remain unclear.We aimed to evaluate the overall clinical effective rate and safety of SHT in treating CAG.Methods:We used the Jadad scale and Cochrane Collaboration risk of bias tool to evaluate the methodological quality of studies investigating SHT.Eight medical databases were searched to identify relevant studies.After data extraction and quality evaluation,27 randomized controlled trials,including 2,441 patients,were considered eligible for analysis.No serious heterogeneity or publication bias was observed across the included studies.We used Revman 5.3 statistical software to evaluate the general clinical effective rate and safety of SHT.Results:The results showed that SHT was more effective(RR=1.25;95%CI[1.20,1.29];P<0.01)and safer(MD=0.24,95%CI[0.08,0.75];P<0.01)than control interventions comprising western medicine,Chinese patent medicine,and/or western medicine+Chinese patent medicine.Compared with the control interventions,SHT resulted in greater improvements in the symptom scores for stomach distension and stomachache,serum gastrin level,histopathologic changes,Helicobacter pylori(HP)inhibition rate,and gastric mucosal inflammation.Conclusion:SHT was more effective and safer than control interventions for CAG.展开更多

Objective:Functional dyspepsia(FD)is a widely prevalent gastrointestinal disorder throughout the world,whereas the efficacy of current treatment in the Western countries is limited.The traditional Chinese herbal formu...Objective:Functional dyspepsia(FD)is a widely prevalent gastrointestinal disorder throughout the world,whereas the efficacy of current treatment in the Western countries is limited.The traditional Chinese herbal formula Xiaopi Hewei capsule(XHC)is a clinically validated remedy in treating FD,but there is no literature expounds the underlying therapeutic mechanism of XHC so far.Methods:In the present study,the network pharmacology technology was used to explore the therapeutic mechanism of XHC in treating FD.We obtained relative compounds of XHC,potential targets of these compounds and FD-related targets by retrieving particular websites.Based on the matching results between XHC potential targets and disease targets,Protein-Protein Interaction(PPI)network was constructed to screen the hub targets by topology.Furthermore,DAVID bioinformatics resources were utilized for the enrichment analysis on GO and KEGG.Results:A total of 62 active compounds and 547 putative identified targets were screened from XHC,of which 241 overlapped with the targets of FD and were considered potential therapeutic targets.14 hub genes were recognized as potential targets of treatments.Moreover,the results of DAVID enrichment analysis indicated that XHC participated in the complex treating effects associated with anti-depression,inflammatory reaction and eradicating Helicobacter Pylori(HP).Molecular docking stimulation results showed that most bioactive compounds of XHC had a strong binding efficiency with hub genes.Conclusions:This study demonstrated that XHC has the characteristics of multi-compounds,multi-targets and multi-pathways in treating FD,which provides the theoretical basis for further research of XHC.展开更多
OBJECTIVE:To evaluate the safety and efficacy of Hewei Jiangni recipe(和胃降逆方,HWJNR)for treating nonerosive gastroesophageal reflux(NERD)with cold-heat complex syndrome and to clarify its mechanism based on correla...OBJECTIVE:To evaluate the safety and efficacy of Hewei Jiangni recipe(和胃降逆方,HWJNR)for treating nonerosive gastroesophageal reflux(NERD)with cold-heat complex syndrome and to clarify its mechanism based on correlation analyses of intestinal flora and metabolites.METHODS:Seventy-two patients with NERD and the Traditional Chinese Medicine(TCM)syndrome of intermingled heat and cold were randomly assigned to either the TCM group or the Western Medicine group,each receiving 8 weeks of treatment.The primary outcome was the score of the gastroesophageal reflux disease questionnaire(GERD-Q).Additionally,10 healthy individuals were recruited.Mechanistic outcomes included correlation analyses of intestinal flora and metabolites in healthy individuals and NERD participants before and after treatment.RESULTS:After 8 weeks,the effectiveness rate was 90%in the TCM group and 86.67%in the Western Medicine group(P>0.05).Compared with omeprazole,the TCM group significantly improved quality of life and alleviated symptoms such as loss of appetite,fatigue,bowel sounds,and coldness in the hands and feet(P<0.05).Dysregulation of intestinal flora and metabolic pathways in NERD patients was restored to balance after TCM treatment,which appeared related to the TCM regulation of"cold and heat disorders."CONCLUSION:HWJNR was clinically as effective as omeprazole and demonstrated advantages in improving quality of life.展开更多
目的:借助网络药理学和分子对接技术,深入剖析陷胸和胃汤治疗胃癌的作用机制。方法:利用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)、SwissTargetPredi...目的:借助网络药理学和分子对接技术,深入剖析陷胸和胃汤治疗胃癌的作用机制。方法:利用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)、SwissTargetPrediction数据库对陷胸和胃汤的活性成分及其对应的靶点进行筛选;通过人类基因数据库(the human gene database,GeneCards)、在线人类孟德尔遗传数据库(online mendelian inheritance in man,OMIM).DrugBank数据库收集与胃癌相关的靶点信息,并运用UniProt数据平台对靶点的基因名称进行整理和规范。借助Venny网站对陷胸和胃汤活性成分的靶点与胃癌靶点取交集,运用Cytoscape 3.8.2软件构建“药物-成分-疾病-靶点”网络;通过STRING数据库构建PPI网络、利用R语言进行基因本体论(gene ontology,GO)功能分析以及京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析;选取关键靶点和核心活性成分,使用PyMOL、AutoDock软件进行分子对接验证。结果:经筛选后,得到陷胸和胃汤活性成分72个,其治疗胃癌的交集靶点有105个。通过蛋白-蛋白互作网络(protein-protein interactions,PPI)筛选得出陷胸和胃汤治疗胃癌的前10个关键靶点分别为SRC、STAT3、PIK3CA、PIK3R1、AKT1、KRAS、ESR1、HRAS.CTNNB1、EGFR。GO分析得到2196条生物学过程、75条细胞组分、135条分子功能。KEGG通路主要富集于Rap1信号通路、癌症中的蛋白聚糖、前列腺癌等通路。分子对接结果表明,核心靶点与主要活性成分的结合构象稳定,筛选出陷胸和胃汤治疗胃癌的核心活性成分为黄连碱。结论:陷胸和胃汤能够通过多成分、多靶点、多通路的方式对胃癌发挥治疗作用,充分彰显了中药整体治疗的特色。展开更多

文摘Objective:Functional dyspepsia(FD)is a widely prevalent gastrointestinal disorder throughout the world,whereas the efficacy of current treatment in the Western countries is limited.The traditional Chinese herbal formula Xiaopi Hewei capsule(XHC)is a clinically validated remedy in treating FD,but there is no literature expounds the underlying therapeutic mechanism of XHC so far.Methods:In the present study,the network pharmacology technology was used to explore the therapeutic mechanism of XHC in treating FD.We obtained relative compounds of XHC,potential targets of these compounds and FD-related targets by retrieving particular websites.Based on the matching results between XHC potential targets and disease targets,Protein-Protein Interaction(PPI)network was constructed to screen the hub targets by topology.Furthermore,DAVID bioinformatics resources were utilized for the enrichment analysis on GO and KEGG.Results:A total of 62 active compounds and 547 putative identified targets were screened from XHC,of which 241 overlapped with the targets of FD and were considered potential therapeutic targets.14 hub genes were recognized as potential targets of treatments.Moreover,the results of DAVID enrichment analysis indicated that XHC participated in the complex treating effects associated with anti-depression,inflammatory reaction and eradicating Helicobacter Pylori(HP).Molecular docking stimulation results showed that most bioactive compounds of XHC had a strong binding efficiency with hub genes.Conclusions:This study demonstrated that XHC has the characteristics of multi-compounds,multi-targets and multi-pathways in treating FD,which provides the theoretical basis for further research of XHC.

文摘OBJECTIVE:To evaluate the safety and efficacy of Hewei Jiangni recipe(和胃降逆方,HWJNR)for treating nonerosive gastroesophageal reflux(NERD)with cold-heat complex syndrome and to clarify its mechanism based on correlation analyses of intestinal flora and metabolites.METHODS:Seventy-two patients with NERD and the Traditional Chinese Medicine(TCM)syndrome of intermingled heat and cold were randomly assigned to either the TCM group or the Western Medicine group,each receiving 8 weeks of treatment.The primary outcome was the score of the gastroesophageal reflux disease questionnaire(GERD-Q).Additionally,10 healthy individuals were recruited.Mechanistic outcomes included correlation analyses of intestinal flora and metabolites in healthy individuals and NERD participants before and after treatment.RESULTS:After 8 weeks,the effectiveness rate was 90%in the TCM group and 86.67%in the Western Medicine group(P>0.05).Compared with omeprazole,the TCM group significantly improved quality of life and alleviated symptoms such as loss of appetite,fatigue,bowel sounds,and coldness in the hands and feet(P<0.05).Dysregulation of intestinal flora and metabolic pathways in NERD patients was restored to balance after TCM treatment,which appeared related to the TCM regulation of"cold and heat disorders."CONCLUSION:HWJNR was clinically as effective as omeprazole and demonstrated advantages in improving quality of life.
文摘目的:借助网络药理学和分子对接技术,深入剖析陷胸和胃汤治疗胃癌的作用机制。方法:利用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)、SwissTargetPrediction数据库对陷胸和胃汤的活性成分及其对应的靶点进行筛选;通过人类基因数据库(the human gene database,GeneCards)、在线人类孟德尔遗传数据库(online mendelian inheritance in man,OMIM).DrugBank数据库收集与胃癌相关的靶点信息,并运用UniProt数据平台对靶点的基因名称进行整理和规范。借助Venny网站对陷胸和胃汤活性成分的靶点与胃癌靶点取交集,运用Cytoscape 3.8.2软件构建“药物-成分-疾病-靶点”网络;通过STRING数据库构建PPI网络、利用R语言进行基因本体论(gene ontology,GO)功能分析以及京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析;选取关键靶点和核心活性成分,使用PyMOL、AutoDock软件进行分子对接验证。结果:经筛选后,得到陷胸和胃汤活性成分72个,其治疗胃癌的交集靶点有105个。通过蛋白-蛋白互作网络(protein-protein interactions,PPI)筛选得出陷胸和胃汤治疗胃癌的前10个关键靶点分别为SRC、STAT3、PIK3CA、PIK3R1、AKT1、KRAS、ESR1、HRAS.CTNNB1、EGFR。GO分析得到2196条生物学过程、75条细胞组分、135条分子功能。KEGG通路主要富集于Rap1信号通路、癌症中的蛋白聚糖、前列腺癌等通路。分子对接结果表明,核心靶点与主要活性成分的结合构象稳定,筛选出陷胸和胃汤治疗胃癌的核心活性成分为黄连碱。结论:陷胸和胃汤能够通过多成分、多靶点、多通路的方式对胃癌发挥治疗作用,充分彰显了中药整体治疗的特色。
