Hewei Jiangni granule(和胃降逆颗粒)alleviates visceral hypersensitivity of non-erosive reflux disease via stromal interaction molecule 1/transient receptor potential vanilloid subfamily member 1 pathway

OBJECTIVE:To explore if Hewei Jiangni granule(和胃降逆颗粒,HWJNG)could regulate esophageal hypersensitivity via stromal interaction molecule 1(STIM1)/transient receptor potential vanilloid subfamily member 1(TRPV1)pathway.METHODS:Qualitative analysis of HWJNG was analysis by high performance of liquid and gas chromatography.In vivo,animal model of non-erosive reflux disease(NERD)was established by fructose intake and restraint stress.HWJNG and Omeprazole were administered by gavage to the drug intervention group.Reflux and visceral hypersensitivity were analyzed by pathological changes,PH value test,mechanical paw withdrawal threshold,thermal withdrawal latency and mast cells(MCs)degranulation.In vitro,substance P(SP)-induced P815 cells and dorsal root ganglion(DRG)cells were cocultured.Expression in both mice and cells of STIM1,TRPV1,and esophageal visceral hypersensitivity-related gastrointestinal neurochemicals were validated by enzyme linked immunosorbent assays,quantitative realtime polymerase chain reaction(qRT-PCR)and Western blot.Moreover,overexpression and small interfering RNA against STIM1 were utilized to verify of the role of HWJNG in DRG cells.RESULTS:HWJNG significantly suppressed intercellular space widening,injury of mitochondrial,MCs degranulation,mechanical allodynia and heat neuropathic sensory and increased pH value of esophageal mucosa in NERD mice.HWJNG inhibited expression of visceral hypersensitivityrelated gastrointestinal neurochemicals in esophageal mucosa and activated P815 cells,and expression of the STIM1,TRPV1 and related neurotransmitters in DRG and DRG cells.STIM1 siRNA and HWJNG both reduced P815 cells adhesion to DRGs cells and Ca2+flow into the cytoplasmic space of DRG cells.Furthermore,HWJNG could reversed STIM1 overexpression induced upregulation of TRPV1.CONCLUSION:HWJNG suppressed intercellular space widening in NERD mice,stabilized MCs and restored neuronal hyperexcitability by regulating visceral hypersensitivity via STIM1/TRPV1 pathway. 展开更多

Objective:To observe the clinical efficacy and safety of Wenzhong Hewei Formula in treating spleen and stomach qi deficiency syndrome in internal medicine diseases.Methods:Sixty patients with spleen and stomach qi deficiency syndrome admitted to the hospital from April 2022 to June 2023 were randomly divided into observation and control groups,with 30 patients in each group.The control group received conventional internal medicine treatment,while the observation group was additionally treated with Wenzhong Hewei Formula on the basis of conventional treatment.Both groups were treated for 4 weeks.Results:The total effective rate of treatment in the observation group was higher than that of the control group(P<0.05).After treatment,the traditional Chinese medicine syndrome scores of both groups were significantly lower than before treatment,with the observation group showing a more pronounced reduction(P<0.05).There was no statistically significant difference in the incidence of adverse reactions between the two groups(P>0.05).Conclusion:Wenzhong Hewei Formula can effectively improve clinical symptoms in patients with spleen and stomach qi deficiency syndrome,enhance clinical efficacy,and have a high level of safety,making it worthy of clinical promotion. 展开更多

OBJECTIVE:To observe the effects of the Bupi Hewei(BPHW)decoction on diarrhea and intestinal flora disorder induced by 5-fluorouracil(5-FU)and investigate the possible mechanism underlying these actions.METHODS:Thirty-five male Sprague-Dawley rats were randomly divided into four groups:normal control,5-FU,5-FU+BPHW decoction(10.5 g/kg for 5 consecutive days),and 5-FU+Bacillus licheni-formis capsule groups(0.2 g/kg for 5 consecutive days).Animal models were established via the intraperitoneal injection of 5-FU(30 mg/kg for 5 consecutive days).At the end of the treatment period,diarrhea was assessed,and the change of the intestinal flora was examined using 16 S r DNA highthroughput sequencing.Interleukin(IL)-17,IL-21,IL-6,IL-10,RAR-related orphan receptor gamma(RORγt),and forkhead box P3(Foxp3)expression in the jejunum was detected using immunohistochemistry,quantitative real-time polymerase chain reaction(PCR),Western blotting,and enzymelinked immuno sorbent assay.RESULTS:In this study,the BPHW decoction effectively lowered the diarrhea score,increased the proportions of Bacteroidetes and Prevotellaceae-Alloprevotella species,and reduced the proportions of Proteobacteria,Escherichia-Shigella,Ruminococcaceae NK4 A214,and Ruminococcaceae UCG-005 species in the rat intestine after 5-FU chemotherapy.In addition,the BPHW decoction significantly suppressed the expression of IL-17,IL-21,IL-6,IL-10,RORγt,and Foxp3 in the jejunum.CONCLUSION:Our findings suggest that the BPHW decoction can improve the intestinal immune balance and reduce intestinal inflammation by targeting T helper cell/T regulatory cell-associated factors. 展开更多

Objective:Shugan Hewei therapy(SHT)acts to soothe the liver and harmonize the stomach.It is a classical traditional Chinese medicine method widely used in China to treat chronic atrophic gastritis(CAG)due to liver qi invading the stomach.However,the clinical effects of SHT remain unclear.We aimed to evaluate the overall clinical effective rate and safety of SHT in treating CAG.Methods:We used the Jadad scale and Cochrane Collaboration risk of bias tool to evaluate the methodological quality of studies investigating SHT.Eight medical databases were searched to identify relevant studies.After data extraction and quality evaluation,27 randomized controlled trials,including 2,441 patients,were considered eligible for analysis.No serious heterogeneity or publication bias was observed across the included studies.We used Revman 5.3 statistical software to evaluate the general clinical effective rate and safety of SHT.Results:The results showed that SHT was more effective(RR=1.25;95%CI[1.20,1.29];P<0.01)and safer(MD=0.24,95%CI[0.08,0.75];P<0.01)than control interventions comprising western medicine,Chinese patent medicine,and/or western medicine+Chinese patent medicine.Compared with the control interventions,SHT resulted in greater improvements in the symptom scores for stomach distension and stomachache,serum gastrin level,histopathologic changes,Helicobacter pylori(HP)inhibition rate,and gastric mucosal inflammation.Conclusion:SHT was more effective and safer than control interventions for CAG. 展开更多

Objective:To uncover the underlying mechanism of Hewei Jiangni granule(HWJNG)on non-erosive reflux disease(NERD)treatment by examining histological changes,gastrointestinal neurochemicals release and visceral hypersensitivity-related receptor expression in NERD model rats.Methods:A NERD rat model was established via a combination of basal sensitization and acid perfusion.HWJNG treatments at different doses were then administered.Pathological changes to tissues,mast cell(MC)activation,serum levels of esophageal visceral hypersensitivity-related neurochemicals,and transient receptor potential(TRP)receptor mRNA and protein levels were investigated.Results:Compared with the control group,the expression of tryptase in MCs,the changes of intercellular space,and the serum levels of substance P(SP),calcitonin gene-related peptides(CGRP)and proteinaseactivated receptor 2(PAR2)increased in the model group(all P<.05).The expression of TRP vanilloid 1(Trpv1)mRNA decreased in esophagus and dorsal root ganglia(DRG)of the model group(P=.030&P=.013),and the expression of TRP melastatin channel subfamily member 8(Trpm8)mRNA decreased in the esophagus of model group(P<.01).The level of esophageal TRPV1 protein increased in the model group(P<.01)and the level of TRPM8 protein decreased in esophagus and DRG of the model group(both P<.05).Compared with the model group,the serum levels of SP,CGRP,and PAR2 in the mediumdose HWJNG group showed significant decreases(all P<.05).The expression of Trpv1 mRNA in esophagus and DRG of the HWJNG groups and the Omeprazole group remarkably decreased(all P<.05),as was the expression of Trpm8 mRNA in esophagus of the HWJNG groups(all P<.05).Conclusion:HWJNG alleviated visceral hypersensitivity in NERD model rats by regulating TRP-mediated signaling.Our results indicate that HWJNG has potential as a therapeutic agent for NERD. 展开更多

Objective:Functional dyspepsia(FD)is a widely prevalent gastrointestinal disorder throughout the world,whereas the efficacy of current treatment in the Western countries is limited.The traditional Chinese herbal formula Xiaopi Hewei capsule(XHC)is a clinically validated remedy in treating FD,but there is no literature expounds the underlying therapeutic mechanism of XHC so far.Methods:In the present study,the network pharmacology technology was used to explore the therapeutic mechanism of XHC in treating FD.We obtained relative compounds of XHC,potential targets of these compounds and FD-related targets by retrieving particular websites.Based on the matching results between XHC potential targets and disease targets,Protein-Protein Interaction(PPI)network was constructed to screen the hub targets by topology.Furthermore,DAVID bioinformatics resources were utilized for the enrichment analysis on GO and KEGG.Results:A total of 62 active compounds and 547 putative identified targets were screened from XHC,of which 241 overlapped with the targets of FD and were considered potential therapeutic targets.14 hub genes were recognized as potential targets of treatments.Moreover,the results of DAVID enrichment analysis indicated that XHC participated in the complex treating effects associated with anti-depression,inflammatory reaction and eradicating Helicobacter Pylori(HP).Molecular docking stimulation results showed that most bioactive compounds of XHC had a strong binding efficiency with hub genes.Conclusions:This study demonstrated that XHC has the characteristics of multi-compounds,multi-targets and multi-pathways in treating FD,which provides the theoretical basis for further research of XHC. 展开更多

OBJECTIVE:To evaluate the safety and efficacy of Hewei Jiangni recipe(和胃降逆方,HWJNR)for treating nonerosive gastroesophageal reflux(NERD)with cold-heat complex syndrome and to clarify its mechanism based on correlation analyses of intestinal flora and metabolites.METHODS:Seventy-two patients with NERD and the Traditional Chinese Medicine(TCM)syndrome of intermingled heat and cold were randomly assigned to either the TCM group or the Western Medicine group,each receiving 8 weeks of treatment.The primary outcome was the score of the gastroesophageal reflux disease questionnaire(GERD-Q).Additionally,10 healthy individuals were recruited.Mechanistic outcomes included correlation analyses of intestinal flora and metabolites in healthy individuals and NERD participants before and after treatment.RESULTS:After 8 weeks,the effectiveness rate was 90%in the TCM group and 86.67%in the Western Medicine group(P>0.05).Compared with omeprazole,the TCM group significantly improved quality of life and alleviated symptoms such as loss of appetite,fatigue,bowel sounds,and coldness in the hands and feet(P<0.05).Dysregulation of intestinal flora and metabolic pathways in NERD patients was restored to balance after TCM treatment,which appeared related to the TCM regulation of"cold and heat disorders."CONCLUSION:HWJNR was clinically as effective as omeprazole and demonstrated advantages in improving quality of life. 展开更多

基于网络药理学联合分子对接技术探究陷胸和胃汤治疗胃癌的作用机制

目的:借助网络药理学和分子对接技术,深入剖析陷胸和胃汤治疗胃癌的作用机制。方法:利用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)、SwissTargetPredi... 目的:借助网络药理学和分子对接技术,深入剖析陷胸和胃汤治疗胃癌的作用机制。方法:利用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)、SwissTargetPrediction数据库对陷胸和胃汤的活性成分及其对应的靶点进行筛选;通过人类基因数据库(the human gene database,GeneCards)、在线人类孟德尔遗传数据库(online mendelian inheritance in man,OMIM).DrugBank数据库收集与胃癌相关的靶点信息,并运用UniProt数据平台对靶点的基因名称进行整理和规范。借助Venny网站对陷胸和胃汤活性成分的靶点与胃癌靶点取交集,运用Cytoscape 3.8.2软件构建“药物-成分-疾病-靶点”网络;通过STRING数据库构建PPI网络、利用R语言进行基因本体论(gene ontology,GO)功能分析以及京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析;选取关键靶点和核心活性成分,使用PyMOL、AutoDock软件进行分子对接验证。结果:经筛选后,得到陷胸和胃汤活性成分72个,其治疗胃癌的交集靶点有105个。通过蛋白-蛋白互作网络(protein-protein interactions,PPI)筛选得出陷胸和胃汤治疗胃癌的前10个关键靶点分别为SRC、STAT3、PIK3CA、PIK3R1、AKT1、KRAS、ESR1、HRAS.CTNNB1、EGFR。GO分析得到2196条生物学过程、75条细胞组分、135条分子功能。KEGG通路主要富集于Rap1信号通路、癌症中的蛋白聚糖、前列腺癌等通路。分子对接结果表明,核心靶点与主要活性成分的结合构象稳定,筛选出陷胸和胃汤治疗胃癌的核心活性成分为黄连碱。结论:陷胸和胃汤能够通过多成分、多靶点、多通路的方式对胃癌发挥治疗作用,充分彰显了中药整体治疗的特色。 展开更多

目的:观察和胃安胎膏外敷神阙穴联合常规疗法治疗脾胃虚弱型妊娠剧吐(HG)的疗效。方法:选取2022年7月—2024年3月浙江中医药大学附属温州市中医院收治的104例脾胃虚弱型HG患者,按随机数字表法分为中药组及对照组各52例。对照组给予现代... 目的:观察和胃安胎膏外敷神阙穴联合常规疗法治疗脾胃虚弱型妊娠剧吐(HG)的疗效。方法:选取2022年7月—2024年3月浙江中医药大学附属温州市中医院收治的104例脾胃虚弱型HG患者,按随机数字表法分为中药组及对照组各52例。对照组给予现代医学常规疗法,中药组在对照组基础上给予和胃安胎膏外敷神阙穴疗法。比较2组临床疗效及不良反应发生率、尿酮体转阴率,比较2组治疗前后中医证候积分、恶心呕吐妊娠专用量化表(PUQE)评分的变化。结果:中药组临床疗效总有效率为96.15%(50/52),对照组临床疗效总有效率为80.77%(42/52),2组比较,差异有统计学意义(P<0.05)。治疗2个疗程后,2组恶心呕吐、疲倦乏力、食欲不振、面色萎黄、舌苔薄白中医证候积分均较治疗前下降(P<0.05),中药组上述5项中医证候积分均低于对照组(P<0.05)。治疗后,2组PUQE量表评分均较治疗前下降,中药组低于同一时间段对照组,差异均有统计学意义(P<0.05)。治疗2个疗程后,对照组尿酮体转阴率为75.00%(39/52),中药组尿酮体转阴率为92.31%(48/52),2组尿酮体转阴率比较,差异有统计学意义(P<0.05)。对照组不良反应发生率为7.69%(4/52),中药组不良反应发生率为17.31%(9/52),2组不良反应发生率比较,差异无统计学意义(P>0.05)。结论:和胃安胎膏外敷神阙穴联合常规疗法治疗脾胃虚弱型HG疗效确切,能缓解恶心呕吐症状,改善胃肠功能,安全性高。 展开更多

观察针刀联合和胃降气汤对难治性胃食管反流病患者疗效的影响 被引量:3

目的探究针刀联合和胃降气汤对难治性胃食管反流病(RGERD)患者疗效的影响和安全性分析。方法选取2020年1月—2023年1月在医院接受治疗的RGERD患者120例。根据随机数字表将患者分为对照组与研究组,每组各60例。其中对照组患者接受常规治... 目的探究针刀联合和胃降气汤对难治性胃食管反流病(RGERD)患者疗效的影响和安全性分析。方法选取2020年1月—2023年1月在医院接受治疗的RGERD患者120例。根据随机数字表将患者分为对照组与研究组,每组各60例。其中对照组患者接受常规治治疗方案。研究组患者在对照组的基础上接受针刀联合和胃降气汤治疗。比较两组的临床效果,统计患者使用治疗前后的反流性疾病问卷(RDQ)评分,中医证候评分,GERD健康相关生活质量评分(GERD-HRQL)。统计患者治疗后6个月复发情况,记录患者治疗过程中的不良反应。比较患者治疗前后肿瘤坏死因子(TNF)-α,超敏C反应蛋白(hs-CRP)和白细胞介素(IL)-6,胃泌素(GAS)和胃动素(MOT)水平变化。结果研究组患者疗效显著高于对照组患者(P=0.045),并且研究组患者临床症状改善率显著高于对照组患者(P=0.003)。治疗后研究组患者与对照组患者相比烧心、反酸、反流和胸痛明显降低(P<0.05)。治疗后研究组患者与对照组患者相比肝胃郁热、气郁痰阻、胆热犯胃和中虚气逆明显降低,存在差异有统计学意义(P<0.05)。治疗后研究组患者与对照组患者相比GERD-HRQL评分明显降低,存在差异有统计学意义(P<0.05)。治疗后研究组患者与对照组患者相比TNF-α,hs-CRP和IL-6水平明显降低,研究组患者与对照组患者相比GAS和MOT水平明显升高,存在差异有统计学意义(P<0.05)。研究组患者复发率相较对照组明显降低(P=0.030)。对照组患者出现不良反应症状的总发生率要显著高于研究组患者(P=0.031)。结论和胃降气汤联合针刀治疗RGERD具有显著效果,两者结合可提高患者疗效,降低患者炎症反应,改善胃动力和生活质量,并具有降低复发率和不良反应的作用。 展开更多

目的基于前额叶皮层(PFC)-伏隔核(NAc)-腹侧被盖区(VTA)神经环路代谢组学探讨疏肝和胃汤(SGHWT)的抗抑郁作用机制。方法将雄性SD大鼠随机分为空白组,模型组,SGHWT低、中、高剂量组[3.67、7.34、14.68 g/(kg·d),以生药量计]以及氟... 目的基于前额叶皮层(PFC)-伏隔核(NAc)-腹侧被盖区(VTA)神经环路代谢组学探讨疏肝和胃汤(SGHWT)的抗抑郁作用机制。方法将雄性SD大鼠随机分为空白组,模型组,SGHWT低、中、高剂量组[3.67、7.34、14.68 g/(kg·d),以生药量计]以及氟西汀组[1.58 mg/(kg·d),阳性对照],每组12只。除空白组外,其余各组大鼠均采用慢性不可预知温和应激结合单笼饲养的方式构建抑郁模型,并于造模同时灌胃相应药液或生理盐水,每天1次,持续6周。末次给药后,检测各组大鼠的体重、糖水偏好率、总移动距离、穿越中心次数和不动时间。收集空白组、模型组、SGHWT中剂量组、氟西汀组大鼠的脑组织PFC、NAc、VTA区样本,观察其组织形态学特征,进行非靶向代谢组学分析(氟西汀组除外)并验证。结果与模型组相比,各药物组大鼠3个脑区细胞溶解、结构受损等病理损伤均明显改善,其体重、糖水偏好率、总移动距离、穿越中心次数均显著升高或延长(P<0.05),不动时间均显著缩短(P<0.05);非靶向代谢组学结果显示,共鉴定出78个内源性差异代谢物,PFC、NAc、VTA区分别有40、35、24个,主要涉及氨基酸、脂质、鞘脂代谢等;代谢通路富集分析结果显示,SGHWT主要通过鞘脂代谢,丙氨酸、天冬氨酸和谷氨酸代谢,不饱和脂肪酸的生物合成3条代谢通路对抑郁大鼠的神经环路产生影响,且以丙氨酸、天冬氨酸和谷氨酸代谢通路为主;验证实验结果显示,SGHWT可显著升高大鼠NAc区蛋白激酶B(Akt)、哺乳动物雷帕霉素靶蛋白(mTOR)的磷酸化水平,显著下调N-甲基-D-天冬氨酸受体1(NMDAR1)蛋白的表达。结论SGHWT可显著改善模型大鼠的抑郁样行为,减轻其PFC-NAc-VTA神经环路组织的病理损伤,上述作用可能与抑制NMDAR1蛋白表达、激活Akt/mTOR信号通路有关。 展开更多

目的 通过对冬枳和胃方治疗非糜烂性反流病(NERD)肝胃郁热证患者的临床观察,评价其在该病治疗中的有效性。方法 收集肝胃郁热型NERD患者72例随机分为治疗组与对照组,治疗组采用冬枳和胃方,对照组给予雷贝拉唑肠溶片,治疗6周,观察一般资... 目的 通过对冬枳和胃方治疗非糜烂性反流病(NERD)肝胃郁热证患者的临床观察,评价其在该病治疗中的有效性。方法 收集肝胃郁热型NERD患者72例随机分为治疗组与对照组,治疗组采用冬枳和胃方,对照组给予雷贝拉唑肠溶片,治疗6周,观察一般资料、中医证候积分、反流性疾病问卷(reflux diagnostic questionnaire, RDQ)和焦虑抑郁量表情况,在SPSS 25.0中进行统计学分析。结果 治疗组完成36例,对照组完成34例。治疗组中医证候总有效率为91.67%,对照组为70.59%,差异有统计学意义(P<0.05)。对胃脘灼痛、脘腹胀满、嗳气反食、易怒方面的改善,治疗组优于对照组(P<0.05);在治疗过程中和治疗后,两组患者均未出现不良反应。结论 冬枳和胃方治疗肝胃郁热型NERD,能明显改善中医证候,优于雷贝拉唑;对烧心、反酸等反流性症状,疗效与雷贝拉唑无显著性差异。冬枳和胃方能明显改善肝胃郁热型NERD患者焦虑抑郁状态,疗效优于雷贝拉唑。冬枳和胃方治疗肝胃郁热型NERD远期疗效优于雷贝拉唑。 展开更多

目的:观察和胃解毒汤联合同步放化疗对食管癌骨髓抑制的影响。方法:选取2020年1月—2022年12月梅州市人民医院治疗的100例食管癌患者,采用随机数字表法分为对照组和联合组。对照组50例(脱落2例,最终纳入48例),采用适型调强放射治疗及TP... 目的:观察和胃解毒汤联合同步放化疗对食管癌骨髓抑制的影响。方法:选取2020年1月—2022年12月梅州市人民医院治疗的100例食管癌患者,采用随机数字表法分为对照组和联合组。对照组50例(脱落2例,最终纳入48例),采用适型调强放射治疗及TP方案化疗,联合组50例(脱落3例,最终纳入47例),在对照组基础上同时加用和胃解毒汤治疗,治疗后比较两组临床疗效,治疗前后给予患者中医证候评分比较,通过营养风险筛查量表(NRS2002)评价患者营养状态,检测患者血清白蛋白、前白蛋白、鳞状细胞癌抗原(SCC)、细胞角蛋白19片段(CY-FRA21-1)、NK细胞亚群CD16^(+)CD56^(+)细胞、T淋巴细胞亚群(CD4^(+)细胞、CD8^(+)细胞)、B淋巴细胞亚群CD19^(+)细胞水平,记录两组不良反应发生情况。结果:联合组疾病控制率、有效率分别为97.87%(46/47),82.98%(39/47),对照组疾病控制率、有效率分别为83.33%(40/48)、56.25%(27/48),联合组疾病控制率、有效率高于对照组(P<0.05);联合组中医证候评分较对照组低(P<0.05);联合组NRS2002评分较对照组低(P<0.05),联合组患者血清白蛋白、前白蛋白表达量均高于对照组(P<0.05);两组CD4^(+)细胞水平较治疗前降低(P<0.05),CD16^(+)CD56^(+)细胞、CD19^(+)细胞、CD8^(+)细胞水平较治疗前升高(P<0.05),联合组患者CD4^(+)细胞、CD16^(+)CD56^(+)细胞、CD19^(+)细胞水平均高于对照组(P<0.05),CD8^(+)细胞水平低于对照组(P<0.05);联合组SCC、CY-FRA21-1表达量低于对照组(P<0.05);联合组骨髓抑制、恶心呕吐、脱发发生率低于对照组(P<0.05)。结论:和胃解毒汤联合同步放化疗治疗食管癌患者,可调节TBNK淋巴亚群水平及营养指标,降低肿瘤标志物水平,提升疗效,减少不良反应发生率。 展开更多

目的建立健脾和胃方的高效液相色谱(HPLC)指纹图谱,并进行化学模式识别分析和其中7种成分的含量测定,为健脾和胃方的质量控制及后续制剂开发提供参考。方法以15批健脾和胃方标准汤剂为样品,采用《中药色谱指纹图谱相似度评价系统(2012版... 目的建立健脾和胃方的高效液相色谱(HPLC)指纹图谱,并进行化学模式识别分析和其中7种成分的含量测定,为健脾和胃方的质量控制及后续制剂开发提供参考。方法以15批健脾和胃方标准汤剂为样品,采用《中药色谱指纹图谱相似度评价系统(2012版)》建立HPLC指纹图谱,并进行相似度评价、色谱峰的指认和归属分析。以15批样品的共有峰峰面积为变量,采用层次聚类分析、主成分分析、正交偏最小二乘-判别分析对样品进行化学模式识别分析。采用HPLC法测定15批样品中腺嘌呤、5-羟甲基糠醛、延胡索乙素、柚皮苷、脱氢紫堇碱、新橙皮苷、甘草酸的含量。结果15批样品的HPLC指纹图谱中共标定出19个共有峰,各批次样品的相似度均大于0.95。化学模式识别分析结果显示,15批样品共聚为3类,筛选出的差异性成分有3个[峰7(5-羟甲基糠醛)、峰17(新橙皮苷)和峰15(柚皮苷)]。7种成分在各自进样质量浓度范围内线性关系均良好(R2≥0.9994);精密度、稳定性、重复性试验的RSD均小于2%(n=6);平均加样回收率为98.95%~103.81%,RSD为0.61%~2.75%(n=6);含量分别为0.031~0.106、0.267~0.824、0.089~0.144、1.344~2.091、0.089~0.178、1.328~2.028、0.040~0.150 mg/g。结论所建立的HPLC指纹图谱及多指标成分含量测定方法准确、可靠,结合化学模式识别分析可用于健脾和胃方的质量控制。 展开更多

目的:用无水乙醇诱导大鼠胃损伤模型,验证特殊膳食和渭肽的胃保护效果,并结合网络药理学筛选相关通路靶点,为临床应用提供了实验依据。方法:选用雄性SD大鼠预防性给药,分别给予不同剂量和渭肽及阳性药雷尼替丁,给药结束后除正常对照组... 目的:用无水乙醇诱导大鼠胃损伤模型,验证特殊膳食和渭肽的胃保护效果,并结合网络药理学筛选相关通路靶点,为临床应用提供了实验依据。方法:选用雄性SD大鼠预防性给药,分别给予不同剂量和渭肽及阳性药雷尼替丁,给药结束后除正常对照组外均用无水乙醇造模1h,采用腹主动脉采血并取胃组织,肉眼观察及HE、PAS染色镜下观察胃壁形态特点,检测血清中白细胞介素6(IL-6)和过氧化氢酶(CAT)的表达情况。通过TCMSP和HERB数据库获取和渭肽中各药物的化学成分,在GeneCards和OMIM数据库中搜索相关作用靶点绘制Venn图;通过STRING网站、Cytoscape3.9.1软件和DAVID平台绘制PPI网络图并进行GO及KEGG富集分析;运用CytoScape 3.9.1软件构建和渭肽药物-活性成分-疾病-靶点网络图,预测和渭肽发挥胃保护作用的核心靶点及主要活性成分。结果:动物实验相关生理生化指标及HE、PAS染色结果表明,特殊膳食和渭肽对无水乙醇诱导的大鼠急性胃损伤具有保护作用。网络药理学结果表明,和渭肽可能通过作用于TP53、EGFR、AKT1等核心靶点,调节PI3K-Akt、MAPK等炎症信号通路发挥预防胃损伤的作用。结论:和渭肽通过调节炎症因子水平、减轻氧化应激、维持胃黏液屏障的完整性、调节PI3K-Akt、MAPK等炎症信号通路发挥预防胃损伤的作用。 展开更多
