Natural colored cotton(NCC)offers a sustainable,dye-free,and eco-friendly alternative for producing colored textiles.Carotenoids,a group of important natural liposoluble pigments,are known for their diverse color spectrum.In this study,we successfully engineered the carotenoid biosynthesis pathway specifically in cotton fibers by utilizing a fiber-specific GbEXPA2 promoter and a CaMV 35S promoter to drive the expression of two key carotenoid biosynthesis genes,CrtB and CrtI,respectively.This approach resulted in the development of a golden fiber cotton germplasm enriched withβ-carotene.Notably,the pigmentation was predominantly observed during the early developmental stages of the fiber(5–20 d post-anthesis).While the presence of carotenoids had no significant effect on plant architecture and growth,it positively influenced the fiber elongation rate,albeit with a slight reduction in fiber length and strength.This study represents a pioneering strategy for the future development of NCCs through carotenoid biofortification. 展开更多

Background:The golden Syrian hamster is a valuable animal model for studying carcinogenesis,metabolic disorders,cardiovascular diseases,and viral infections due to its biological and pathological similarities to humans.However,the development of genetically engineered hamsters has lagged behind that of mice and rats,largely because of an embryonic development block at the two-cell stage in vitro.Although CRISPR/Cas9-mediated gene knockout has been achieved in hamsters,precise DNA fragment insertion or conditional knockout(cKO)models have not previously been reported,likely due to technical limitations in embryo manipulation and insufficient efficiency of homology-directed repair(HDR).Methods:In this study,we generated conditional alleles of the ApoF gene in golden Syrian hamsters.A two-cut strategy was applied using Cas9 protein,two sgRNAs,and a single donor plasmid containing exon 2 flanked by loxP sites and two~0.8 kb homology arms.A mixture of Cas9 protein,sgRNAs,and the donor plasmid was microinjected into the pronuclei of one-cell stage hamster embryos.Results:The efficiency of CRISPR/Cas9-mediated loxP knock-in reached up to 27%,and the genetically modified floxed alleles were successfully transmitted through the germline.The functionality of the inserted loxP sites was validated by in vivo Cremediated recombination following local administration of AAV vectors,including AAV-cTnT-Cre in the heart and AAV-CMV-Cre in the brain.Conclusions:To our knowledge,this work represents the first successful establishment of a conditional knockout model in the golden Syrian hamster,providing a valuable tool for mechanistic studies of gene function and disease modeling. 展开更多

Ming-style furniture represents the pinnacle of Chinese traditional furniture,with the four-head official’s cap chair standing out as a representative category due to its classic form and cultural connotations.This paper takes a specific rosewood four-head official’s cap chair inscribed by Wang Shixiang as the research subject,employing a tripartite approach of form analysis,material examination,and inscription interpretation.The chair strictly adheres to the classic form of Ming-style four-head official’s cap chairs,embodying the Ming Dynasty design philosophy of“the unity of function and aesthetics”.The scarcity and unique grain pattern of the golden-flecked rosewood(Zitan)endow it with the status of“gold among woods”.Wang Shixiang’s inscription binds the furniture to the cultural essence of“the charm of the Ming Dynasty”,transforming it into“heritable cultural heritage”.This paper provides a new perspective for the case study of Ming-style furniture and offers theoretical reference for the contemporary inheritance of traditional craftsmanship. 展开更多

Inoculated fermentation enables rapid fermentation of aquatic products.No studies have been conducted on the lipid profiles of inoculated fermented golden pompano(Trachinotus ovatus).In this study,a lipase-producing Bacillus subtilis with salt tolerance was screened from traditionally fermented golden pompano(TF)and used as a starter culture.Whole-genome sequencing analysis revealed it carries 4 clustered regularly interspaced short vpalindromic repeats structures and 2 genes encoding triacylglycerol lipase.Untargeted lipidomics identified lipid molecules(833)in 6 major classes from B.subtilis SCSMX-2 fermented golden pompano(IF).A total of 28 lipid molecules were upregulated in IF,including phosphatidylcholines(PCs),triacylglycerols(TAGs),and lysophosphatidylcholine.B.subtilis supplementation enhanced the production of polyenyl PCs and mediumand long-chain TAGs.The IF rich in linoleic,docosahexaenoic acids(DHA),and eicosapentaenoic acid were primarily distributed in the sn-2 position of DHA,PC and phosphatidylethanolamine.This research revealed the lipid profiles of IF,providing theoretical basis for the application of B.subtilis in the fermented fish industry. 展开更多

To address the problem of high lifespan loss and poor state of charge(SOC)balance of electric vehicles(EVs)participating in grid peak shaving,an improved golden eagle optimizer(IGEO)algorithm for EV grouping control strategy is proposed for peak shaving sce-narios.First,considering the difference between peak and valley loads and the operating costs of EVs,a peak shaving model for EVs is constructed.Second,the design of IGEO has improved the global exploration and local development capabilities of the golden eagle optimizer(GEO)algorithm.Subsequently,IGEO is used to solve the peak shaving model and obtain the overall EV grid connected charging and discharging instructions.Next,using the k-means algorithm,EVs are dynamically divided into priority charging groups,backup groups,and priority discharging groups based on SOC differences.Finally,a dual layer power distribution scheme for EVs is designed.The upper layer determines the charging and discharging sequences and instructions for the three groups of EVs,whereas the lower layer allocates the charging and discharging instructions for each group to each EV.The proposed strategy was simulated and verified,and the results showed that the designed IGEO had faster optimization speed and higher optimization accuracy.The pro-posed EV grouping control strategy effectively reduces the peak-valley difference in the power grid,reduces the operational life loss of EVs,and maintains a better SOC balance for EVs. 展开更多

Molecular cloning remains a cornerstone technique in genetic engineering and synthetic biology.In this study,we conducted a systematic comparative analysis between the classical cloning method and the Golden Gate assembly technique,utilizing Escherichia coli as the model organism.Through polymerase chain reaction(PCR)amplification,restriction enzyme digestion,ligation,transformation,and Sanger sequencing,we assessed the operational efficiency and cloning fidelity of both strategies.Our results demonstrated that Golden Gate assembly,leveraging type IIS restriction enzymes and simultaneous ligation,significantly enhanced cloning efficiency and precision,particularly for seamless multi-fragment assembly.In contrast,the classical cloning approach maintained certain advantages in simplicity and robustness for specific experimental conditions.Challenges encountered during transformation and sequencing highlighted the critical impact of technical accuracy on experimental outcomes.This study underscores the importance of selecting appropriate cloning methodologies tailored to experimental objectives and laboratory capabilities,providing a foundation for optimized molecular cloning workflows in future synthetic biology and biotechnology applications. 展开更多

Reliable Cluster Head(CH)selectionbased routing protocols are necessary for increasing the packet transmission efficiency with optimal path discovery that never introduces degradation over the transmission reliability.In this paper,Hybrid Golden Jackal,and Improved Whale Optimization Algorithm(HGJIWOA)is proposed as an effective and optimal routing protocol that guarantees efficient routing of data packets in the established between the CHs and the movable sink.This HGJIWOA included the phases of Dynamic Lens-Imaging Learning Strategy and Novel Update Rules for determining the reliable route essential for data packets broadcasting attained through fitness measure estimation-based CH selection.The process of CH selection achieved using Golden Jackal Optimization Algorithm(GJOA)completely depends on the factors of maintainability,consistency,trust,delay,and energy.The adopted GJOA algorithm play a dominant role in determining the optimal path of routing depending on the parameter of reduced delay and minimal distance.It further utilized Improved Whale Optimisation Algorithm(IWOA)for forwarding the data from chosen CHs to the BS via optimized route depending on the parameters of energy and distance.It also included a reliable route maintenance process that aids in deciding the selected route through which data need to be transmitted or re-routed.The simulation outcomes of the proposed HGJIWOA mechanism with different sensor nodes confirmed an improved mean throughput of 18.21%,sustained residual energy of 19.64%with minimized end-to-end delay of 21.82%,better than the competitive CH selection approaches. 展开更多

金黄心大白菜和橘红心大白菜都富含类胡萝卜素,颜色亮丽,广受欢迎,因此,内叶颜色成为大白菜新品种选育的重要目标性状之一,但是其调控机制尚不清晰。本研究利用金黄心和橘红心大白菜构建回交群体,并通过开发金黄心性状的BrGOLDEN基因和... 金黄心大白菜和橘红心大白菜都富含类胡萝卜素,颜色亮丽,广受欢迎,因此,内叶颜色成为大白菜新品种选育的重要目标性状之一,但是其调控机制尚不清晰。本研究利用金黄心和橘红心大白菜构建回交群体,并通过开发金黄心性状的BrGOLDEN基因和橘红心性状的Bror基因性状的分子标记,对BrGOLDEN和Bror的互作规律进行初步的分析;对成熟期金黄心大白菜和浅黄心大白菜的内叶进行转录组测序(RNA-seq)分析,了解类胡萝卜素代谢途径相关基因的表达;对BrGOLDEN启动子进行克隆并分析关键顺式作用元件,同时利用5'截短启动子克隆及GUS染色探究BrGOLDEN启动子的核心功能区域。遗传规律结合分子标记结果表明:显性基因BrGOLDEN与隐性基因Bror非独立遗传,Bror基因对BrGOLDEN基因表现为上位效应,即BrGOLDEN基因与Bror同时存在时,仅表现橘红心表型。转录组数据显示:金黄心大白菜中2-C-甲基-D-赤藓糖醇-4-磷酸(MEP)前体合成途径及类胡萝卜素合成途径相关基因的表达量基本都比浅黄心大白菜高;类胡萝卜素合成途径中合成八氢番茄红素聚合酶(PSY)、玉米黄质环氧化酶(ZEP)的基因表达量基本都高于途径上其他的基因,表明BrGOLDEN基因可能通过调控类胡萝卜素合成途径的起始和结束的相关基因来控制金黄心大白菜合成更多的类胡萝卜素。启动子功能分析结果表明:在启动强度上,不同缺失长度的启动子表现不同,并且BrGOLDEN基因启动子的核心作用元件可能在-1111 bp到-601 bp;在该区间内存在着参与水杨酸反应的顺式作用元件、对厌氧诱导必不可少的顺式调节元件以及参与干旱诱导的MYB结合位点,说明该区间可能存在着一些转录因子在上游调控BrGOLDEN基因的表达,从而导致不同类型的大白菜内叶类胡萝卜素的差异积累。这些发现为进一步探究BrGOLDEN的分子调控机制提供了一定依据。 展开更多

A symmetrical quartic polynomial, named golden one, can be connected to coefficients of the icosahedron equation as well as to the gyromagnetic correction of the electron and to number 137. This number is not a mystic one, but is connected with the inverse of Sommerfeld’s fine-structure constant and this way again connected with the electron. From number-theoretical realities, including the reciprocity relation of the golden ratio as effective pre-calculator of nature’s creativeness, a proposed closeness to the icosahedron may point towards the structure of the electron, thought off as a single-strand compacted helically self-confined charged elemantary particle of less spherical but assumed blunted icosahedral shape generated from a high energy double-helix photon. We constructed a chiral Moebius “ball” from a 13 times 180&#730;twisted double helix strand, where the turning points of 12 generated slings were arranged towards the vertices of a regular icosahedron, belonging to the non-centrosymmetric rotation group I532. Mathematically put, we convert the helical motion of an energy quantum into a stationary motion on a Moebius stripe structure. The radius of the ball is about the Compton radius. This chiral closed circuit Moebius ball motion profile can be tentatively thought off as the dominant quantum vortex structure of the electron, and the model may be named CEWMB (Charged Electromagnetic Wave Moebius Ball). Also the gyromagnetic factor of the electron (g<sub>e</sub> = 2.002319) can be traced back to this special structure. However, nature’s energy infinity principle would suggest a superposition with additional less dominant (secondary) structures, governed also by the golden mean. A suggestion about the possible structure of delocalized hole carriers in the superconducting state is given. 展开更多

三维绘图软件Golden Software Voxler已广泛应用于地质勘探、水质分析、空气污染监测等方面,而在土壤污染方面的应用较少。根据对某污染场地前期的调查与研究,确定了该场地的土壤关注污染物为重金属镍。运用Voxler 3.0对污染物监测数据... 三维绘图软件Golden Software Voxler已广泛应用于地质勘探、水质分析、空气污染监测等方面,而在土壤污染方面的应用较少。根据对某污染场地前期的调查与研究,确定了该场地的土壤关注污染物为重金属镍。运用Voxler 3.0对污染物监测数据进行处理,绘制污染物的钻孔轨迹图、三向切片图以及体积渲染图。结果显示镍主要集中分布在场地东北部地表以下50~250 cm范围内,浓度随着深度增加而减小且有向下扩散的趋势。Voxler的应用为污染物在土壤中的空间分布提供了可视化表达,从而为后续的风险评估和污染防治工作提供数据支撑。 展开更多

The water ... The golden apple snail Pomacea canaliculata (Lamarck 1819) was first recorded at Black Dragon Spring, Dianchi Basin, Baiyi Township, Songming County, Kunming City, Yunnan Province, China, in October 2004. The water from the spring flows into the Songhuaba Reservoir, the major drinking water resource for Kunming City, and part of the Dianchi Lake basin. This is the first record of this invasive snail in the Dianchi Lake Basin. Pomacea canaliculata originates from Central and South America, and in Asia the snail has spread through deliberate and accidental introductions to the Philippines, Vietnam, Thailand, Laos, Cambodia, Malaysia, Indonesia, Papua New Guinea, Korea, Japan and South China. It has become a major pest in rice-growing areas, resulting in huge damage to crops. Strict prevention and control measures have to be implemented to prevent the spread of the snail in Yunnan, together with public awareness campaigns to inform the public of the dangers of this invasive snail. 展开更多
