Glinus lotoides ethanolic extract alleviates LPS-induced anxiety and depression-like behavior by modulating antioxidant and inflammatory biomarkers in rats 被引量:2

Objective:To explore the protective role of Glinus lotoides ethanolic extract in a depression model through modulating oxidant/antioxidant enzyme system and inflammatory status.Methods:Phytochemical constituents of Glinus lotoides ethanolic extract were evaluated qualitatively and quantitatively along with HPLC.Rats were divided into six groups.The normal control and the intoxicated groups received normal saline,and the standard group received imipramine,while the remaining groups received 100,300,and 500 mg/kg Glinus lotoides ethanolic extract.All groups received treatments for 14 d.Lipopolysaccharides(LPS)were then administered i.p.(0.83 mg/kg)to all groups except the normal control group.After 24 h,anxiety and depression-like behaviors were evaluated by performing behavioral analysis(open field,tail suspension,forced swim,sucrose preference test),and determining total oxidant status,total antioxidant capacity,catalase,and biochemical parameters[malondialdehyde,glutathione,superoxide dismutase,tumor necrosis factor(TNF)-alpha and interleukin(IL)-6].Results:Phytochemical studies confirmed the presence of phenols and flavonoids and HPLC analysis showed the presence of gallic acid,quercetin,chlorogenic,and caffeic acid.Total oxidant status was significantly decreased,while total antioxidant capacity was significantly increased in the Glinus lotoides ethanolic extract treated groups.Moreover,Glinus lotoides ethanolic extract diminished malondialdehyde,IL-6,and TNF-alpha levels,while increasing superoxide dismutase,catalase,and glutathione activities.Conclusions:Glinus lotoides ethanolic crude extract shows significant antidepressant activity by modulating oxidative and biochemical parameters that supports its folkloric use in traditional systems of medicine. 展开更多

G. lotoides L. is a threatened plant that is frequently harvested for medicinal purpose. However, its distribution in the world is limited because of short period of seed viability and poor seed germination. The objective of this study was to develop in vitro propagation protocol for G. lotoides through callus induction. For callus induction, different concentrations of 2,4-D (2,4-dichlorophenoxyacetic acid), NAA (α-naphthalene acetic acid) and BAP (6-benzyl amino purine) were used. Seeds were sown on growth regulator-free MS medium and shoots from the in vitro germinated seedlings were excised and cultured on MS medium containing 0.5 mg/l BAP. Young leaves from these shoots were used as explant for callus induction and shoot regeneration. Maximum callus induction (100%) was observed on medium containing 2,4-D (0.5, 2.0, 3.5 mg/l) or NAA (2.0, 2.5 mg/l) in combination with 0.5 mg/l BAP. However, 2,4-D was the best in overall callus induction. The highest regeneration (20%) frequency was achieved on the medium containing 0.5 mg/l BAP. Highest number of shoot (2.83 ± 1.22) and shoot length (2.16 ± 0.87 cm) per explant were obtained in the presence of 0.25 mg/l BAP + 0.5 mg/l KIN (Kinetin). In shoot multiplication media, maximum mean (6.43 ± 0.87) of shoot was observed on MS medium containing 0.5 mg/l BAP. The best shoot length (1.70 ± 0.14 cm) was recorded on control medium. The highest (95%), maximum root number (14.10 ± 1.47) and root length (1.01 ± 0.10 cm) were obtained on a medium supplemented with 1.5 mg/l Indole-3-butyric acid (IBA). All the plants (100%) were survived after acclimatization in greenhouse. The present study can be useful for callus induction and indirect shoot regeneration form G. lotoides. 展开更多

Phytochemical Constituents and Allelopathic Effects of Some Medicinal Plants Extract on the Soil Algal Diversity

Phytochemical constituents of the shoots and roots extract of chosen eight medicinal plants (Plantago major, Verbesina encelioides, Glinus lotoides, Helotropium supinum, Mentha microphylla, Euphorbia hirta, Juncus subulatus and Convolvulus arvensis) were estimated using standard qualitative analysis. The extract contained alkaloids, glycosides, saponins, phenols, tannins, f[avonoids and terpenoids in some medicinal plants while others contain some only of these phytochemical components. Allelopathic effect of the various concentrations of the extracts on the soil algal diversity was also investigated. Wollea saccata was the only algal species disappeared from all applied investigated plant extracts. Phormidium richardsii, Monoraphidium braunii, Eunotia verneris and Nitzschia bilobata were the highly sensitive algal species to all applied shoot and root extract from all tested plants, they disappeared from most of the studied plants. On the other hand, Phormidium animale and Chlorella neustonice were highly tolerant algal species to all applied extracts. Counts of Cyanophyceae, Chlorophyceae and Bacillariophyceae were decreased by addition of some investigated medicinal plants extracts such as Juncus subulatus, Convolvulus arvensis and Euphorbia hirta. So the authors can use these plants extracts in biocontrol of the nuisance of algal bloom or any other microorganisms. Also, these plants may be useful to recover eutrophic water which needs further investigation. 展开更多
