AIM To detect abnormal micro RNA(miRNA) expression in type 1 gastric neuroendocrine neoplasms(g-NENs) and find potential target genes.METHODS Tumour tissues from patients with type 1 g-NENs were used as experimental s...AIM To detect abnormal micro RNA(miRNA) expression in type 1 gastric neuroendocrine neoplasms(g-NENs) and find potential target genes.METHODS Tumour tissues from patients with type 1 g-NENs were used as experimental samples, and gastric mucosal tissues from the same patients obtained during gastroscopy review after several months were used as control samples. miRNA expression was examined with Agilent human miRNA chips and validated via RTPCR. Three types of target gene prediction software(TargetScan, PITA, and microRNAorg) were used to predict potential target genes of the differentially expressed miRNAs, and a dual-luciferase reporter assay system was used for verification. RESULTS Six miRNAs were significantly upregulated or downregulated in the tumours compared to the control samples. Among them, miR-202-3 p was extraordinarily upregulated. RT-PCR of seven sample sets confirmed that miR-202-3 p was upregulated in tumour tissues. In total, 215 target genes were predicted to be associated with miR-202-3 p. Among them, dual-specificity phosphatase 1(DUSP1) was reported to be closely related to tumour occurrence and development. The dual-luciferase reporter assay showed that miR-202-3 p directly regulated DUSP1 in 293 T cells. CONCLUSION miR-202-3 p is upregulated in type 1 g-NEN lesions and might play important roles in the pathogenesis of type 1 g-NENs by targeting DUSP1.展开更多

The solutions of the problem in term...The propagation of a Bleustein-Gulyaev (B-G) type wave in a structure consisting of multiple layers and a half-space of porous piezoelectric materials is theoreti- cally studied. The solutions of the problem in terms of the mechanical displacements and electric potential functions are obtained for each layer and the half-space. The dispersion equation is obtained for electrically open and shorted boundary conditions by use of the transfer matrix method. A peculiar kind of B-G waves is investigated, which can propagate only in the layer over the half-space. The relationship between the piezoelectric constants and the dielectric constants is found for the existence of a peculiar kind of propagation modes. The numerical results in terms of the phase velocity and the electromechanical coupling factor with different thicknesses of the layer stack are presented.展开更多

Inside the Earth, the density and rigidity of the crustal layer and the mantle of the Earth vary exponentially and periodically along the depth. The displacements of the wave are found in the individual medium followed by a dispersion equation using a suitable analytic approach and a boundary condition. Background The association of E670G polymorphism in the proprotein convertase subtilisin/kexin type 9 (PCSK9) gene and serum lipid profiles is inconsistent in dif- ferent ethnic groups.Bai Ku Yao is a special subgroup...Background The association of E670G polymorphism in the proprotein convertase subtilisin/kexin type 9 (PCSK9) gene and serum lipid profiles is inconsistent in dif- ferent ethnic groups.Bai Ku Yao is a special subgroup of the Yao minority in China.The present study was undertaken association of PCSK9 E670G polymorphism and several environmental factors with serum lipid levels in the Guangxi Bai Ku Yao and Han populations.Methods A total of 649 subjects of Bai Ku Yao and 646 participants of Han Chinese were randomly selected from our previous stratified randomized cluster samples.Genotyping of the PCSK9 E670G polymorphism was performed by polymerase chain reaction and restriction fragment length polymorphism combined with gel electrophoresis,and then confirmed by direct sequencing. Results The levels of serum total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C) and apolipoprotein(Apo) AI were lower in Bai Ku Yao than in Han(P【0.01 for all).The frequency of A and G alleles was 98.00%and 2.00%in Bai Ku Yao,and 95.20%and 4.80%in Han(P【0.01);respectively. The frequency of AA,AG and GG genotypes was 95.99%,4.01%and 0%in Bai Ku Yao,and 91.02%, 8.36%and 0.62%in Han(P【0.01);respectively.There were also significant differences in the genotypic and allelic frequencies between n and the ratio of ApoAI to ApoB in Han Chinese but not in Bai Ku Yao were different between the AA and AG/GG genotypes(P【0.05 for all).The G allele carriers had higher serum HDL-C and higher ApoAI to ApoB ratio than the G allele noncarriers.When serum lipid parameters in Han were analyzed according to sex,the G allele carriers had higher serum HDL and ApoAI levels in males (P【0.05),and lower ApoB level and higher ApoAI to ApoB ratio in females(P【0.05 for all).Multiple linear regression analysis showed that serum HDL-C levels were correlated with genotypes in both ethnic groups(P【0.05 each).Serum lipid parameters were also correlated with sex,age,body massindex,alcohol consumption,cigarette smoking,and blood pressure in both ethnic groups(P【0.05-0.001).Conclusions These results suggest that the PCSK9 E670G polymorphism is mainly associated with some serum lipid parameters in the Han population,both gender show different relations to different serum lipid parameters.The G allele carriers might have higher serum lipid profiles than the G allele noncarriers. ormal LDL-C(≤3.20 mmol/L) and high LDL-C subgroups (】 3.20 mmol/L,P【0.01;respectively) in Bai Ku Yao, and between normal ApoB(≤1.14 g/L) and high ApoB subgroups(】 1.14 g/L,P 【 0.01;respectively) in Han.展开更多
Free translation caused by attractive field-gradient force was observed for Ni and ferrite(CuFe_2O_4) grains that were released in a diffuse area.In order to exclude the effect of terrestrial gravity,translation was o...Free translation caused by attractive field-gradient force was observed for Ni and ferrite(CuFe_2O_4) grains that were released in a diffuse area.In order to exclude the effect of terrestrial gravity,translation was observed in micro-gravity(μG) condition produced by a compact drop-shaft installed in a ordinary laboratory room.Magnetization Ms per unit mass of the grain is obtained by analyzing the above-mentioned translations in terms of a energy conservation rule;here conservation of the sum of field-induced potential mMsH and kinetic energy 1/2/mv^2 is considered for a particle with mass m.The present method of obtaining Ms is free of measuring the mass of sample;this is because the field-gradient force is a volume force that is proportional to m.The method is also free of an interfering signal emitted from the sample holder.Accordingly,Ms is detectable irrespective of sample size,provided that the field-induced translation is observable.The above-mentioned procedure to estimate Ms from filed-induced translation is a step to obtain a reliable magnetic data from a single nano-sized grain.The efficiency of material identification was recently confirmed on various solid grains,which was based on diamagnetic magnetization data obtained from its translations caused by field-gradient repulsive force.A diamagnetic material generally possesses an intrinsic value of magnetic susceptibility and its anisotropy.The present results on nickel grains indicate that the principle of material identification based on its magnetization data is applicable for the three major category of magnetic materials,namely ferro-(or ferri-) magnetic, paramagnetic and diamagnetic material.展开更多

By employing affymetrix microarray ...The ubiquitin-proteasome proteolysis pathway is responsible for the degradation of abnormal and short-lived proteins to regulate many important biochemical activities in eukaryotes. By employing affymetrix microarray analysis, we have identified a novel ubiquitin ligase E3 gene GhRING2 that is differentially expressed between two Gossypium hirsutum lines-Texas Marker-1 (TM-1) and Chromosome Substitution Line CS-B25. The CS-B25 line has chromosome 25 from G. barbadense substituted into TM-1. The complete nucleotide sequences of GhRING2 along with its 5’-flanking region were obtained by genomic walking. GhRING2 was highly expressed in elongating fiber, and GUS expression directed by the GhRING2 promoter was found in hypocotyls and young stems of transgenic Arabidopsis plants. Using a yeast two-hybrid assay GhRING2 was found to interact with a PROTODERMAL FACTOR1 (GhPDF1) protein. GhPDF1 was expressed preferentially in immature ovules and fiber initials, and the GhPDF1 gene had been suggested to play a role in cell fate determination and fiber development. Pull down and plasmid swap assays further confirmed the interaction between GhRING2 and GhPDF1. The expression and protein interaction data indicate that GhRING2 may be involved in the turnover of GhPDF1 and participation in the transition from initiation to elongation stages during fiber development. Our data strongly suggest that the ubiquitin-proteasome pathway may regulate cotton fiber growth and development. Abu Garadi area is covered mainly by metasediments, alkali feldspar granites and stream sediments. The alkali feldspar granite is traversed by a major strike-slip fault trending in an N-S direction as well as two subordinate sets of faults trending NW to WNW for the first one and NE for the second one. These faults represent the shear zones affected by magmatic (syngenetic) as well as hydrothermal (epigenetic) activities causing alteration of the granitic rocks. The most common alteration features are albitization, greisenization and koalinitization. The mass balance calculations of the studied altered samples show enrichments in Zr, Y, Ni, U, Th and Ga and depletions in Zn, Sr, Nb, Ba, Pb, Cu and V. Only the greisenized samples exhibit a significant enrichment in Nb, ∑REE budget and pronounced lanthanide tetrad effect (M-type), especially TE1,4, while weakly expressed tetrad effects are for the other albitized and koalinitized samples. Mineralogically, the common accessory minerals in the altered samples include samarskite-(Y), betafite, uranothorite, zircon, fluorite and cassiterite. The greisenized granites contain high eU and eTh than the other altered types, where they are characterized by an assemblage of the radioactive minerals; samarskite-(Y), betafite, uranothorite in addition to zircon. The inter-element relationships between U and Th and also their ratios illustrate that the radioelement distribution in these granites is mainly governed by magmatic processes, in addition to post-magmatic ones. The distribution of chemical elements and the fractionation of some isovalents within the shear zone are largely controlled by the newly formed mineral phases. With respect to uranium mobilization, uranium migrated from the host alkali feldspar granites of G. Abu Garadi, while the shear zones acted as traps for the migrated uranium. Moreover, U migrated in the shear zone during greisenization and albitization, and migrated out during koalinitization.展开更多

AIM:To characterize the regeneration-associated stem cell-related phenotype of hepatocyte-derived growth factor receptor(HGFR)-expressing cells in active ulcerative colitis(UC).METHODS:On the whole 38 peripheral blood...AIM:To characterize the regeneration-associated stem cell-related phenotype of hepatocyte-derived growth factor receptor(HGFR)-expressing cells in active ulcerative colitis(UC).METHODS:On the whole 38 peripheral blood samples and 38 colonic biopsy samples from 18 patients with histologically proven active UC and 20 healthy control subjects were collected.After preparing tissue microarrays and blood smears HGFR,caudal type homeobox 2(CDX2),prominin-1(CD133) and Musashi-1conventional and double fluorescent immunolabelings were performed.Immunostained samples were digitalized using high-resolution Mirax Desk instrument,and analyzed with the Mirax TMA Module software.For semiquantitative counting of immunopositive lamina propria(LP) cells 5 fields of view were counted at magnification x 200 in each sample core,then mean ± SD were determined.In case of peripheral blood smears,30 fields of view with 100 μm diameter were evaluated in every sample and the number of immunopositive cells(mean ± SD) was determined.Using 337 nm UVA Laser MicroDissection system at least 5000 subepithelial cells from the lamina propria were collected.Gene expression analysis of HGFR,CDX2,CD133,leucine-rich repeat-containing G-protein coupled receptor 5(Lgr5),Musashi-1 and cytokeratin20(CK20) were performed in both laser-microdisscted samples and blood samples by using real time reverse transcription polymerase chain reaction(RT-PCR).RESULTS:By performing conventional and double fluorescent immunolabelings confirmed by RT-PCR,higher number of HGFR(blood:6.7 ± 1.22 vs 38.5 ±3.18;LP:2.25 ± 0.85 vs 9.22 ± 0.65;P < 0.05),CDX2(blood:0 vs 0.94 ± 0.64;LP:0.75 ± 0.55 vs 2.11± 0.75;P < 0.05),CD133(blood:1.1 ± 0.72 vs 8.3± 1.08;LP:11.1 ± 0.85 vs 26.28 ± 1.71;P < 0.05)and Musashi-1(blood and LP:0 vs scattered) positive cells were detected in blood and lamina propria of UC samples as compared to controls.HGFR/CDX2(blood:0 vs 1± 0.59;LP:0.8 ± 0.69 vs 2.06 ± 0.72,P < 0.05)and Musashi-1/CDX2(blood and LP:0 vs scattered) coexpressions were found in blood and lamina propria of UC samples.HGFR/CD133 and CD133/CDX2 coexpressions appeared only in UC lamina propria samples.CDX2,Lgr5 and Musashi-1 expressions in UC blood samples were not accompanied by CK20 mRNA expression.CONCLUSION:In active UC,a portion of circulating HGFR-expressing cells are committed to the epithelial lineage,and may participate in mucosal regeneration by undergoing mesenchymal-to-epithelial transition.展开更多

By employing affymetrix microarray analysis, we have identified a novel ubiquitin ligase E3 gene GhRING2 that is differentially expressed between two Gossypium hirsutum lines-Texas Marker-1 (TM-1) and Chromosome Substitution Line CS-B25. The CS-B25 line has chromosome 25 from G. barbadense substituted into TM-1. The complete nucleotide sequences of GhRING2 along with its 5’-flanking region were obtained by genomic walking. GhRING2 was highly expressed in elongating fiber, and GUS expression directed by the GhRING2 promoter was found in hypocotyls and young stems of transgenic Arabidopsis plants. Using a yeast two-hybrid assay GhRING2 was found to interact with a PROTODERMAL FACTOR1 (GhPDF1) protein. GhPDF1 was expressed preferentially in immature ovules and fiber initials, and the GhPDF1 gene had been suggested to play a role in cell fate determination and fiber development. Pull down and plasmid swap assays further confirmed the interaction between GhRING2 and GhPDF1. The expression and protein interaction data indicate that GhRING2 may be involved in the turnover of GhPDF1 and participation in the transition from initiation to elongation stages during fiber development. Our data strongly suggest that the ubiquitin-proteasome pathway may regulate cotton fiber growth and development. 文摘The G. Abu Garadi area is covered mainly by metasediments, alkali feldspar granites and stream sediments. The alkali feldspar granite is traversed by a major strike-slip fault trending in an N-S direction as well as two subordinate sets of faults trending NW to WNW for the first one and NE for the second one. These faults represent the shear zones affected by magmatic (syngenetic) as well as hydrothermal (epigenetic) activities causing alteration of the granitic rocks. The most common alteration features are albitization, greisenization and koalinitization. The mass balance calculations of the studied altered samples show enrichments in Zr, Y, Ni, U, Th and Ga and depletions in Zn, Sr, Nb, Ba, Pb, Cu and V. Only the greisenized samples exhibit a significant enrichment in Nb, ∑REE budget and pronounced lanthanide tetrad effect (M-type), especially TE1,4, while weakly expressed tetrad effects are for the other albitized and koalinitized samples. Mineralogically, the common accessory minerals in the altered samples include samarskite-(Y), betafite, uranothorite, zircon, fluorite and cassiterite. The greisenized granites contain high eU and eTh than the other altered types, where they are characterized by an assemblage of the radioactive minerals; samarskite-(Y), betafite, uranothorite in addition to zircon. The inter-element relationships between U and Th and also their ratios illustrate that the radioelement distribution in these granites is mainly governed by magmatic processes, in addition to post-magmatic ones. The distribution of chemical elements and the fractionation of some isovalents within the shear zone are largely controlled by the newly formed mineral phases. With respect to uranium mobilization, uranium migrated from the host alkali feldspar granites of G. Abu Garadi, while the shear zones acted as traps for the migrated uranium. Moreover, U migrated in the shear zone during greisenization and albitization, and migrated out during koalinitization.

文摘Previous studies have demonstrated that a missense single-nucleotide polymorphism variant (2316A〉G;rs2230739)of the adenylate cyclase type IX gene was associated with bipolar disorder and affective disorder.We determined genotype and allele frequencies using a ligase detection reaction method in 315 patients with major depressive disorder and 278 unrelated, sex-matched healthy control subjects.We did not detect any statistically significant differences in genotype and allele frequencies between patients and healthy control subjects.Furthermore,we found no significant difference between genders in major depressive disorder,nor between patients and controls in the same gender.These results suggest that 2316A〉G(rs2230739)may not be a risk factor for increasing susceptibility to major depressive disorder in the Chinese Han population.

文摘AIM:To characterize the regeneration-associated stem cell-related phenotype of hepatocyte-derived growth factor receptor(HGFR)-expressing cells in active ulcerative colitis(UC).METHODS:On the whole 38 peripheral blood samples and 38 colonic biopsy samples from 18 patients with histologically proven active UC and 20 healthy control subjects were collected.After preparing tissue microarrays and blood smears HGFR,caudal type homeobox 2(CDX2),prominin-1(CD133) and Musashi-1conventional and double fluorescent immunolabelings were performed.Immunostained samples were digitalized using high-resolution Mirax Desk instrument,and analyzed with the Mirax TMA Module software.For semiquantitative counting of immunopositive lamina propria(LP) cells 5 fields of view were counted at magnification x 200 in each sample core,then mean ± SD were determined.In case of peripheral blood smears,30 fields of view with 100 μm diameter were evaluated in every sample and the number of immunopositive cells(mean ± SD) was determined.Using 337 nm UVA Laser MicroDissection system at least 5000 subepithelial cells from the lamina propria were collected.Gene expression analysis of HGFR,CDX2,CD133,leucine-rich repeat-containing G-protein coupled receptor 5(Lgr5),Musashi-1 and cytokeratin20(CK20) were performed in both laser-microdisscted samples and blood samples by using real time reverse transcription polymerase chain reaction(RT-PCR).RESULTS:By performing conventional and double fluorescent immunolabelings confirmed by RT-PCR,higher number of HGFR(blood:6.7 ± 1.22 vs 38.5 ±3.18;LP:2.25 ± 0.85 vs 9.22 ± 0.65;P < 0.05),CDX2(blood:0 vs 0.94 ± 0.64;LP:0.75 ± 0.55 vs 2.11± 0.75;P < 0.05),CD133(blood:1.1 ± 0.72 vs 8.3± 1.08;LP:11.1 ± 0.85 vs 26.28 ± 1.71;P < 0.05)and Musashi-1(blood and LP:0 vs scattered) positive cells were detected in blood and lamina propria of UC samples as compared to controls.HGFR/CDX2(blood:0 vs 1± 0.59;LP:0.8 ± 0.69 vs 2.06 ± 0.72,P < 0.05)and Musashi-1/CDX2(blood and LP:0 vs scattered) coexpressions were found in blood and lamina propria of UC samples.HGFR/CD133 and CD133/CDX2 coexpressions appeared only in UC lamina propria samples.CDX2,Lgr5 and Musashi-1 expressions in UC blood samples were not accompanied by CK20 mRNA expression.CONCLUSION:In active UC,a portion of circulating HGFR-expressing cells are committed to the epithelial lineage,and may participate in mucosal regeneration by undergoing mesenchymal-to-epithelial transition.
