Evaluation of chemopreventive effect of Fumaria indica against N-nitrosodiethylamine and CCl_4-induced hepatocellular carcinoma in Wistar rats 被引量:3

Objective:To investigation the chemopreventive potential of Fumaria indica(F.indica) extract(FIE) on N-nitrosodiethylamine and CCl<sub>4</sub>-induced hepatocarcinogenesis in Wislar rats.Methods:The experimental animals were divided into six groups(n=6).Hepatocellular carcinoma was induced by single intraperitoneal injection of N-nitrosodiethylamine(NDEA) in normal saline at a dose of 200 mg/kg body weight followed by weekly subcutaneous injections of CCl<sub>4</sub>(3 mL/kg/week) for 6 weeks,as the promoter of carcinogenic effect.After administration of the carcinogen,200 and 400 mg/kg of FIE were administered orally once a day throughout the study.At the end of 20 weeks,the body weight,liver weight and relative liver weight were measured.The percentage of nodule incidence and liver cancer markers such as aspartate transaminase(AST),alanine transaminase(ALT),alkaline phosphatase(ALP),γ-glutamyl transferase(γ-GT),total bilirubin level(TBL),α-feto protein(AFP) and carcinoembryonic antigen were estimated along with histopathological investigation in experimental groups of rats. Results:Obtained results demonstrated that the cotreatment with FIE significantly prevented the decrease of the body weight and also increased in relative liver weight caused by NDEA. The treatment with FIE significantly reduced the nodule incidence and nodule multiplicity in the rats after NDEA administration.The levels of liver cancer markers such as AST,ALT,ALP,γ-glutamyl transferase,TBL,AFP and carcinoembryonic antigen were substantially increased by NDEA treatment.However,FIE treatment significantly reduced the liver injury and restored the entire liver cancer markers.Histological observations of liver tissues too correlated with the biochemical observations.Conclusions:These finding powerfully supports that F.indica exert chemopreventive effect by suppressing the tumor burden and restoring the activities of hepatic cancer marker enzymes on NDEA and CCl4-induced hepatocarcinogenesis in Wistar rats. 展开更多

Pugsley(Fumariaceae),known as "Fumitory",is an annual herb found as a common weed all over the plains of India and Pakistan.The whole plant is widely used in traditional and folkloric systems of medicine.In traditional systems of medicine,the plant is reputed for its anthelmintic,diuretic,diaphoretic,laxative,cholagoguc,stomachic and sedative activities and is used to purify blood and in liver obstruction in ethnopharmacology. The whole plant is ascribed to possess medicinal virtues in Ayurvedic and Unani systems of medicine and is also used in preparation of important Ayurvedic medicinal preparations and polyhedral liver formulations.The review reveals that phytochemical constituents of wide range have been separated from the plants and it possesses important pharmacological activities like smooth muscle relaxant,spasmogenic and spasmolytic,analgesic,anti-inflammatory, neuropharmacological and antibacterial activities.The separation of hepatoprotective and antifungal constituents from this plant was also reported newly.This review highlights the traditional,ethnobotanical,phytochemical,pharmacological information available on Fumaria indica,which might be helpful for scientists and researchers to find out new chemical entities responsible for its claimed traditional uses. 展开更多

Beneficial Effects of <i>Fumaria indica</i>on Chronic Stress-Induced Neurobehavioral and Biochemical Perturbations in Rats 被引量:3

Objective: To evaluate the anti-stress activity of standardized extract of Fumaria indica (FI) through validated beha-vioral models of rodents followed by estimation of biochemical changes associated with chronic stress. Methods: Fifty percent ethanolic extract of FI used in this study was standardized on its contents of fumaric acid and its conjugates (0.45% and 0.35% respectively). Stressed Charles Foster rats received unpredictable foot shocks (2 mA, 1 hr, 14 days) through electric grid. FI was given orally as 0.3% carboxymethyl cellulose (CMC) suspension in 100, 200 and 400 mg/kg doses. For comparison, Panax ginseng (PG) extract (100 mg/kg, p.o.) was used as standard adaptogen. Incidence of gastric ulceration, changes in weight of adrenal and spleen, behavioral depression, cognitive dysfunction test and suppression of sexual behavior in male rats were used as validated behavioral models. Plasma corticosterone, brain levels of lipid peroxides (LPO), superoxide dismutase (SOD), catalase (CAT) and reduced glutathione (GSH), and expression of cytokines IL-1β, TNF-α and IL-10 in circulating white blood cells (WBC) were quantified for ascertaining biochemical changes accompanying stress. Results: As compared to vehicle treated stressed rats, the FI and PG treated rats showed fewer incidence of gastric ulceration, reversal of changes in weight of adrenal gland and spleen, reversal of behavioral depression, better performance in passive and active avoidance tests for cognitive function and increased sexual activity. FI and PG significantly decreased chronic unpredictable stress induced elevation of corticosterone, and both extracts normalized also the abnormal oxidative status of the brain observed in stressed rats. FI treatment also suppressed the elevated level of IL-1β, TNF-α and IL-10 in stressed animals. Conclusions: FI could be another adaptogenic herb and fumaric acid and its conjugates are possibly involved in observed bioactivity of its extract. 展开更多

Objective:To explore some Fumaria species which were recorded to be traditionally used against malaria and other protozoal diseases.Methods:Consequently,in the current study,antiprotozoal effect of the ethanol extracts obtained from five Fumaria species(Fumaria densiflora,Fumaria cilicica,Fumaria rostellata,Fumaria kralikii,and Fumaria parviflora)was investigated against the parasites;Plasmodium falciparum(malaria)and Trypanosoma bruceirhodesiense(human African trypanosomiasis)at 0.81 and 4.85μg/mL concentrations.Results:Among them,Fumaria densiflora extract exerted the highest antiplasmodial(93.80%)and antitrypanasomal effect(55.40%),while the ethanol extracts of Fumaria kralikii(43.45%)and Fumaria rostellata(41.65%)showed moderate activity against Plasmodium falciparum.Besides,phenolic acid contents of the extracts were analyzed using high performance liquid chromatography(HPLC)and trans-cinnamic(4.32 mg/g)and caffeic(3.71 mg/g)acids were found to be the dominant phenolic acids in Fumaria densiflora.Conclusions:According to our results,Fumaria densiflora deserve further study for its promising antiprotozoal activity. 展开更多

Objective: The genus fumaria includes more than 40 species in the world. The aim of this study was to quantify the phytochemical constituents of Fumaria vaillantii L. aerial parts and compare the different methods of extraction. Total phenol, total flavonoid, total alkaloid, ascorbic and organic acids (oxalic, maleic, citric, succinic and fumaric acids) yields were evaluated in terms of the temperature effect, type of solvent and maceration time. Methods: Dried plant samples were extracted by different procedures. Total phenolic, total flavonoid, total alkaloid and ascorbic acid yields were determined by spectrophotometric methods. Also, the organic acid yields were analyzed using high performance liquid chromatography method. Results: With the same extraction method, the natural flora extract was showed more yields of oxalic, maleic and citric acids than the commercial one, while the commercial extract was showed more yields of total phenol, ascorbic, succinic and fumaric acids than the natural flora one. The water-boiled extract was showed more yields of total phenol and total flavonoid. The macerated in ethanol 80% extract was also demonstrated more amounts of total alkaloid and ascorbic acid. Among different aqueous macerated extracts of the commercial sample, as the maceration time increased, total phenol, total flavonoid, oxalic, maleic, succinic, fumaric and ascorbic acids yields decreased. Macerated commercial dried fumitory in double-distilled water for 24 hrs resulted in an extract with the highest possible fumaric acid yield. Conclusion: It can be concluded that both water-boiled and macerated in ethanol 80% extracts can be used as rich sources of total phenolic and total flavonoid, which are considered as the important antioxidants. 展开更多

Holistic Psychopharmacology of Fumaria indica(Fumitory)

Fumaria indica is a medicinal plant of the fumitory family wildly growing throughout India. Classical texts of Ayurveda, i.e. the oldest traditionally known health care and medical system originating in Indian subcontinent, mentions diverse medicinal uses of the plant. During more recent decades broad spectrums of therapeutically interesting pharmacological properties of its extracts and secondary metabolites have also been reported. Recent observations made during efforts to define its pharmacological activity profile according to the Ayurvedic concepts of mind body medicine have revealed exceptionally broad spectrums of psychopharmacological activity profiles of diverse types of hydro alcoholic extracts of the plant. These effects of the extracts become apparent after their repeated daily doses only. Taken together with prior preclinical knowledge on the plant, these observations strongly suggest that Fumaria indica could be an easily available source for discovering and developing phyto-pharmaceuticals or drugs potentially useful for treatments of mental health problems commonly associated with numerous physical disorders and chronic diseases. Since several psychoactive and other phytochemical of Fumaria indica are also encountered in other plants commonly used in Chinese and other traditionally known medical systems, observations made and the holistic strategy used for defining its psychopharmacological activity profile could be of interest of others involved in efforts necessary for proper understanding of therapeutic potentials of many plants containing Fumaria alkaloids and other bioactive phytochemicals present in Fumaria indica. 展开更多
