This paper attempts to make a tentative probe into the translator's subjectivity manifested in the translation of Fu Sheng Liu Ji by Shen Fu on the basis of George Steiner's fourfold translation motion theory, namely"trust", "aggression","incorporation"and"compensation". t(16;21)(p11;q22)/FUS∷ERG阳性急性髓系白血病的基因突变谱和预后分析

目的:旨在探讨携带t(16;21)(p11;q22)/FUS∷ERG融合基因的急性髓系白血病(acute myeloid leukemia,AML)患者的临床生物学及预后特征。方法:回顾性分析2015年12月—2023年8月在河南省人民医院收治的12例t(16;21)(p11;q22)/FUS∷ERG阳性AM... 目的:旨在探讨携带t(16;21)(p11;q22)/FUS∷ERG融合基因的急性髓系白血病(acute myeloid leukemia,AML)患者的临床生物学及预后特征。方法:回顾性分析2015年12月—2023年8月在河南省人民医院收治的12例t(16;21)(p11;q22)/FUS∷ERG阳性AML患者的临床和实验室数据,并进行文献复习。结果:t(16;21)(p11;q22)/FUS∷ERG阳性AML占同期初诊AML患者的1.28%(12/935)。患者中男9例,女3例;FAB分型为M2型9例,M4/5型3例。7例患者在诊断时骨髓原始细胞可见空泡形成。所有患者的免疫表型均表达CD56和CD123。染色体核型分析均显示t(16;21)易位,其中7例伴有附加染色体异常。FUS∷ERG融合基因均检测阳性,基因突变情况:PTPN1150.0%(3/6)、NRAS 36.4%(4/11)、BCOR 22.2%(2/9)、RUNX118.2%(2/11)、TET218.2%(2/11)、WT111.1%(1/9)。汇总国内外文献中基因突变信息,突变率最高为PTPN11,占26.7%(12/45),其次为NRAS,占23.5%(8/34),其他常见突变基因为TET2、RUNX1、ASXL1、DNMTA、BCOR等。研究中11例患者诱导化疗后完全缓解率为72.7%,复发率为37.5%;3例患者接受了造血干细胞移植(hematopoietic stem cell transplantation,HSCT),其中2例移植后复发/死亡。11例患者的中位生存期为15个月(95%CI 9.092~20.908)。结论:t(16;21)(p11;q22)/FUS∷ERG阳性AML是一类罕见的具有特定遗传学特征的AML亚型,具有独特的临床生物学和基因表达谱特征,总体预后较差。HSCT效果可能有限,亟需探索新的靶向治疗方法。 展开更多

BACKGROUND Liver fibrosis is a common health problem worldwide and there is still a lack of effective medicines.The Chinese herbal medicine,Gan Shen Fu Fang(GSFF)is composed of salvianolic acid B and diammonium glycyrrhizinate.In this study,we observed the effects of GSFF on liver fibrosis in vivo and in vitro in an attempt to provide some hope for the treatment.AIM To observe the effects of GSFF on liver fibrosis in vivo and in vitro and investigate the mechanism from the perspective of the inflammatory response and extracellular signal-regulated kinase(ERK)phosphorylation.METHODS Common bile duct-ligated rats were used for in vivo experiments.Hepatic stellate cells-T6(HSC-T6)cells were used for in vitro experiments.Hematoxylin and eosin staining and Masson staining,biochemical assays,hydroxyproline(Hyp)assays,enzyme-linked immunoasorbent assay and western blotting were performed to evaluate the degree of liver fibrosis,liver function,the inflammatory response and ERK phosphorylation.The CCK8 assay,immunofluorescence and western blotting were applied to test the effect of GSFF on HSC-T6 cell activation and determine whether GSFF had an effect on ERK phosphorylation in HSC-T6 cells.RESULTS GSFF improved liver function and inhibited liver fibrosis in common bile ductligated rats after 3 wk of treatment,as demonstrated by histological changes,hydroxyproline assays and collagen I concentrations.GSFF alleviated inflammatory cell infiltration and reduced the synthesis of pro-inflammatory cytokines[tumor necrosis factor-α(TNF-α)and interlukin-1β]and NF-κB.In addition,GSFF decreased ERK phosphorylation.In vitro,GSFF inhibited the viability of HSC-T6 cells with and without transforming growth factorβ1(TGF-β1)stimulation and decreased the synthesis of collagen I.GSFF had the greatest effect at a concentration of 0.5μmol/L.GSFF inhibited the expression ofα-smooth muscle actin(α-SMA),a marker of HSC activation,in HSC-T6 cells.Consistent with the in vivo results,GSFF also inhibited the phosphorylation of ERK and downregulated the expression of NF-κB.CONCLUSION GSFF inhibited liver fibrosis progression in vivo and HSC-T6 cell activation in vitro.These effects may be related to an alleviated inflammatory response and downregulated ERK phosphorylation. 展开更多

Amyotrophic lateral sclerosis(ALS)is a progressive neurodegenerative disorder characterized by the degeneration of motor neurons in the brain and spinal cord,leading to muscle weakness,paralysis,and ultimately death(Cleveland and Rothstein,2001).Frontotemporal lobar degeneration(FTLD)is a neurodegenerative disease affecting the frontal and temporal lobes of the brain,leading to changes in behavior,personality,and language(Van Langenhove et al.,2012).Both ALS and FTLD are classified as proteinopathies in which abnormal protein aggregation and accumulation in neurons contribute to the disease pathogenesis.Fused in sarcoma(FUS)is a DNA/RNA-binding protein involved in various cellular processes,including transcriptional regulation,RNA splicing,and DNA repair.Mutations in the FUS gene have been linked to familial ALS,highlighting the importance of FUS in the disease pathogenesis(Vance et al.,2009).In ALS and FTLD,aberrant post-translational modifications(PTMs)of FUS,such as phosphorylation,acetylation,and methylation,have been implicated in the promotion of FUS aggregation and neurotoxicity(Choi et al.,2023).Therefore,understanding the regulatory mechanisms of FUS PTMs is crucial for developing targeted therapies against these diseases. 展开更多

Objective Brown and beige adipocytes dissipate energy through thermogenesis,and the impaired thermogenic function of these adipocytes is a key driver of obesity and related metabolic disorders.However,the molecular mechanisms governing adipocyte thermogenesis are not fully understood.This study investigated the role of inorganic pyrophosphatase 1(PPA1)in regulating adipocyte thermogenesis and assessed its potential as a therapeutic target for obesity and metabolic disorders.Methods To investigate the function of PPA1 in adipose tissue thermogenesis,we generated adipose-specific heterozygous PPA1 knockout mice via the Cre-loxP system and established cold exposure models.PPA1 deletion effects were assessed by hematoxylin and eosin(H&E)staining,immunofluorescence,quantitative polymerase chain reaction(qPCR),and immunoblotting.Mitochondrial changes during browning were further characterized via transmission electron microscopy(TEM),mitochondrial DNA(mtDNA)quantification,qPCR,and Western blotting.The molecular mechanisms involved were subsequently dissected via mass spectrometry,coimmunoprecipitation(Co-IP),and immunofluorescence colocalization.Results Adi-PPA1^(fl/+)mice presented impaired adipose tissue thermogenesis upon cold exposure.Mechanistically,PPA1 deficiency impaired adipose browning in an enzyme activity-independent manner.PPA1 knockdown promoted the aberrant translocation and accumulation of fused in sarcoma(FUS)to mitochondria,which triggered mitochondrial dysfunction and suppressed browning.Crucially,silencing FUS effectively rescued the mitochondrial defects caused by PPA1 depletion.Conclusion PPA1 functions as a nonenzymatic positive regulator of adipocyte thermogenesis by interacting with FUS to prevent its mitochondrial mislocalization,thereby maintaining mitochondrial function and promoting adipose browning.These findings highlight PPA1 as a potential therapeutic avenue for obesity and metabolic disorders. 展开更多

This study investigated the therapeutic effects on metabolic syndrome(MetS)and the impact on the intestinal barrier and gut microbiota of Fu brick tea aqueous extracts(FTE)on MetS in rats fed with a high-fat diet(HFD).Here,the results showed that FTE supplement significantly reduced HFD-induced weight gain,adiposity,dyslipidemia,fasting blood glucose(FBG)increment,and non-alcoholic fatty liver disease(NAFLD).Moreover,FTE supplement resulted in a decline in lipopolysaccharide(LPS)level and attenuation of colonic inflammation and oxidative stress to protect the intestinal barrier function.FTE supplement also maintained the intestinal barrier integrity by improving histological appearance and promoting ZO-1,Occludin,and Claudin-1 protein expression levels.Meanwhile,FTE supplement alleviated the gut microbiota dysbiosis by enhancing the Firmicutes/Bacteroidetes(F/B)ratio and stimulating the colonization of probiotic bacteria such as Akkermansia,Lactobacillus,Adlercreutzia,and Bacteroides.These findings collectively suggest that Fu brick tea could alleviate MetS and MetS-associated traits with the mechanism relevant to the protection of intestinal barrier and gut microbiota regulation. 展开更多

Objective:To evaluate the efficacy of the modified Gui Fu Ling Wu Decoction in treating diabetic nephropathy(DN)of the spleen-kidney Yang deficiency type.Methods:A total of 100 DN patients admitted to Changyi Traditional Chinese Medicine Hospital between August 2023 and March 2024 were included in the study.Patients were randomly divided into a control group and a study group,each comprising 50 cases,using a computerized random number generator.The control group received kallikrein treatment,while the study group received a combination of kallikrein and the modified Gui Fu Ling Wu Decoction.Blood glucose control,renal function,and inflammatory markers were assessed before and after treatment in both groups.Results:Before treatment,there were no significant differences in blood glucose and renal function indicators between the two groups(P>0.05).After treatment,postprandial blood glucose,fasting blood glucose,24-hour urinary protein,urinary microalbumin,serum creatinine,serum C-reactive protein(CRP),and interleukin-6 levels significantly decreased in both groups,with the study group showing superior results compared to the control group(P<0.05).Conclusion:The combination of Gui Fu Ling Wu Decoction and kallikrein for treating spleen-kidney Yang deficiency type DN significantly improves blood glucose control,enhances renal function,and reduces inflammatory responses. 展开更多

The high incidence of hand,foot,and mouth disease(HFMD)in children,coupled with improper management,can lead to complications,causing significant distress to both patients and their parents.This article reports on the successful treatment of a case of HFMD with dampness-heat accumulation in the spleen type using a therapeutic approach of applying herbal patches to acupoints to disperse dampness,promote heat dissipation,and unblock the fu organs.The herbs selected primarily include honeysuckle,Forsythia,and mint to relieve the exterior and promote heat dissipation;Agastache and magnolia bark to transform and dry dampness;Scutellaria,blackberry lily,and licorice to clear heat,detoxify,and benefit the throat;and magnolia bark and bitter orange to unblock the fu organs and purge heat.The flexible combination of these herbs with acupoints such as Shenque,Zhongwan,Tiantu,and Feishu fully leverages the synergistic effects of both the herbs and acupoints,resulting in a significant therapeutic effect.This approach provides valuable insights and methodologies for the prevention and treatment of HFMD in children. 展开更多

Xin Xiu Ben Cao(《新修本草》Newly Revised Materia Medica)is China’s first national pharmacopoeia and holds significant importance in the history of Chinese herbal medicine.However,it was lost by the time of the Northern Song dynasty.Japanese envoys to the Tang dynasty brought this text back to Japan,where it was preserved in manuscript form at Ninna-ji Temple(仁和寺)in Kyoto,although it gradually fell into obscurity.In the 19th century,Japanese physicians and scholars such as the Fukui family(福井家),the Asai family(浅井家),the Kojima family(小岛家),and Kariya Ekisai(狩谷棭斋)discovered fragments of Xin Xiu Ben Cao,transcribed them,and gradually disseminated them.During his travels to Japan from 1888 to 1889,the Chinese envoy Fu Yunlong(傅云龙),along with Chen Ju(陈榘),acquired and published this text.The reproduced edition shows that the text consists of eleven volumes,eight of which were originally from the Kojima family’s collection.Volume 3 was transcribed by Kojima Naozane(小岛尚真)based on his father’s compiled edition.The Kojima family’s manuscripts were later acquired by Arai Masatake(新井政毅),the owner of the Arai Bunko(新井文库),and after his passing,they were dispersed among booksellers.The seals printed in the book reveal that the Qing Legation in Japan and Chinese literati who traveled to Japan jointly appreciated and studied this text.By comparing it with other scroll and album-format manuscripts,it becomes evident that the layout of this fragment differs from the scroll format but is closer to the album format. 展开更多

目的观察复方苦参注射液联合靶向及免疫治疗对晚期肝癌患者的临床疗效。方法纳入100例晚期原发性肝癌患者并按治疗方式分为对照组(卡瑞利珠单抗+仑伐替尼治疗,50例)和观察组(卡瑞利珠单抗+仑伐替尼+复方苦参注射液治疗,50例),2组均进行... 目的观察复方苦参注射液联合靶向及免疫治疗对晚期肝癌患者的临床疗效。方法纳入100例晚期原发性肝癌患者并按治疗方式分为对照组(卡瑞利珠单抗+仑伐替尼治疗,50例)和观察组(卡瑞利珠单抗+仑伐替尼+复方苦参注射液治疗,50例),2组均进行3个周期治疗。比较2组患者的临床疗效、治疗前后中医证候积分、T淋巴细胞亚群水平、不良反应发生率及生存时间。结果观察组的客观缓解率、疾病控制率和中医证候积分总有效率高于对照组(P<0.05)。治疗后,观察组胁痛、腹胀、纳差、黄疸、乏力中医证候积分低于对照组;CD4^(+)、CD4^(+)/CD8^(+)较对照组升高,CD8^(+)较对照组降低(P<0.05)。观察组总不良反应发生率显著低于对照组,且无进展生存期和总生存期较对照组明显延长(P<0.05)。结论复方苦参注射液联合靶向及免疫治疗对晚期肝癌患者疗效明显,可有效改善其临床症状,调节免疫功能,延长生存时间,安全性较好。 展开更多

目的 :评价大剂量醛氢叶酸 +5FU持续滴注 48小时 (双周疗法 )治疗晚期大肠癌的客观疗效及毒副反应。方法 :本临床研究采用大剂量醛氢叶酸 +5FU持续滴注 48小时 ,每两周重复一次 (双周疗法 )的方案治疗5 2例晚期大肠癌病人 ,所有病人均接... 目的 :评价大剂量醛氢叶酸 +5FU持续滴注 48小时 (双周疗法 )治疗晚期大肠癌的客观疗效及毒副反应。方法 :本临床研究采用大剂量醛氢叶酸 +5FU持续滴注 48小时 ,每两周重复一次 (双周疗法 )的方案治疗5 2例晚期大肠癌病人 ,所有病人均接受 4疗程化疗。结果 :CR 2例 ,PR 15例 ,NC 18例 ,PD 17例 ,有效率为32 6 9%。病人的主要毒副反应为外周静脉炎、粘膜炎、腹泻、恶心和呕吐、骨髓抑制、脱发等。除外周静脉炎较严重外 ,其余毒性均为Ⅰ~Ⅱ度 ,病人耐受良好。结论 :大剂量醛氢叶酸 +5FU持续滴注 48小时 (双周疗法 )治疗晚期大肠癌疗效较好 ,方案是安全的 ,病人耐受性良好 ,值得在临床上推广使用。 展开更多

目的:根据5FU、CF药代动力学特点,设计5FU连续8小时静滴,CF100mg,在5FU静滴第0、4、8小时3次静推的CF/5FU给药方案,联合卡铂对晚期鼻咽癌患者进行临床研究,重点摸索该联合化疗方案的剂量限制性毒性... 目的:根据5FU、CF药代动力学特点,设计5FU连续8小时静滴,CF100mg,在5FU静滴第0、4、8小时3次静推的CF/5FU给药方案,联合卡铂对晚期鼻咽癌患者进行临床研究,重点摸索该联合化疗方案的剂量限制性毒性及5FU的最大耐受剂量(MTD)。方法:将符合入选条件的病例分组,每组35例。5FU剂量从500mg/(m2·d)开始,以125mg/(m2·d)逐渐递增,直至最大耐受剂量,CF300mg、卡铂300~350mg/m2剂量固定不变。结果:共21例晚期鼻咽癌患者进入该临床试验,5FU的最大耐受剂量为1000mg/(m2·d),口腔粘膜炎是该联合化疗方案的主要剂量限制性毒性。初步观察该方案有较率为762%。结论:5FU连续8小时静滴与CF、卡铂联合化疗可提高5FU剂量强度,推荐Ⅱ期临床试用时5FU剂量为750~875mg/(m2·d),共5天,3~4周重复一次。该方案对晚期鼻咽癌有较高缓解率,但确切疗效尚待进一步证实。 展开更多

目的:探讨C-Jun氨基末端激酶(C-Jun N-terminal kinase,JNK)信号转导通路对人肝癌5-氟尿嘧啶(5-fluorouracil,5-FU)耐药细胞系Bel-7402/FU产生多药耐药(multidrug resistance,MDR)的调控作用,为肝癌细胞MDR机制的研究提供新靶点。方法:... 目的:探讨C-Jun氨基末端激酶(C-Jun N-terminal kinase,JNK)信号转导通路对人肝癌5-氟尿嘧啶(5-fluorouracil,5-FU)耐药细胞系Bel-7402/FU产生多药耐药(multidrug resistance,MDR)的调控作用,为肝癌细胞MDR机制的研究提供新靶点。方法:采用蛋白质印迹法检测人肝癌亲本细胞Bel-7402和耐药细胞Bel-7402/FU中JNK及磷酸化-JNK(phospho-JNK,p-JNK)的表达;在Bel-7402/FU细胞中,使用特异性阻断剂SP600125抑制TNK通路后,采用RT-PCR和免疫细胞化学法分别检测MDR1 mRNA和P-糖蛋白(P-glycoprotein,P-gp)的表达;FCM法检测罗丹明123(rhodanmine 123,Rh123)在细胞内的积聚和外排情况;MTT法检测Bel-7402/FU细胞对5-FU的敏感性。结果:耐药细胞Bel-7402/FU组与亲本细胞Bel-7402组相比,JNK蛋白的表达无明显变化,而p-JNK蛋白的表达量明显增加;抑制JNK通路后,Bel-7402/FU细胞中MDR1 mRNA表达水平和P-gP蛋白的表达水平均明显下调,Rh123的蓄积明显增加,外排明显减少,且对5-FU的敏感性明显增加。结论:JNK信号转导通路参与人肝癌耐药细胞株Bel-7402/FU中多药耐药基因MDR1和多药耐约蛋白P-gp的表达,对其耐药起调控作用。 展开更多

目的构建FUS1和人IL-12双基因真核共表达质粒并在肺癌A549细胞中检测基因表达及诱导细胞凋亡作用。方法采用RT-PCR技术从人肺成纤维细胞MRC-5中逆转录扩增全长FUS1基因,通过PCR技术从pORF-hIL-12质粒中扩增含双亚基的全长IL-12基因。FUS... 目的构建FUS1和人IL-12双基因真核共表达质粒并在肺癌A549细胞中检测基因表达及诱导细胞凋亡作用。方法采用RT-PCR技术从人肺成纤维细胞MRC-5中逆转录扩增全长FUS1基因,通过PCR技术从pORF-hIL-12质粒中扩增含双亚基的全长IL-12基因。FUS1和IL-12分别经酶切后定向插入真核细胞双顺反子载体pVITRO2的多克隆位点中,酶切和测序鉴定双基因共表达质粒pVITRO2-FUS1-IL-12的构建。以pVITRO2-FUS1-IL-12转染肺癌A549细胞,通过RT-PCR、ELISA和Western blot方法检测目的基因的表达,以Hoechst33258染色及流式细胞术验证pVITRO2-FUS1-IL-12在体外的促凋亡作用。结果酶切和测序分析表明成功构建了双基因真核共表达质粒pVITRO2-FUS1-IL-12,在转染后的细胞中同时检测到了FUS1和IL-12的表达,两种基因在双基因表达载体上的表达量基本不受影响。双基因共表达质粒pVITRO2-FUS1-IL-12具有明显诱导肺癌细胞凋亡的作用。结论成功构建pVITRO2-FUS1-IL-12双基因真核共表达质粒,为肺癌的多基因联合治疗提供了又一新的思路和方法。 展开更多
