目的探讨曲妥珠单抗(trastuzumab,TRZ)在异丙基肾上腺素(isoproterenol,ISO)引起的心脏纤维化中的作用。方法以C57BL/6N雄性小鼠为研究对象,建立ISO(10 mg·kg^(-1))和TRZ(10 mg·kg^(-1))诱导心脏纤维化动物模型。采用小动物... 目的探讨曲妥珠单抗(trastuzumab,TRZ)在异丙基肾上腺素(isoproterenol,ISO)引起的心脏纤维化中的作用。方法以C57BL/6N雄性小鼠为研究对象,建立ISO(10 mg·kg^(-1))和TRZ(10 mg·kg^(-1))诱导心脏纤维化动物模型。采用小动物心脏超声仪检测小鼠心功能,HE染色观察心脏组织形态,天狼猩红染色法检测心脏的纤维化面积,Western blot法检测TGF-β1、α-SMA和FIbronectin的蛋白表达。分离培养原代乳小鼠心脏成纤维细胞,予以TRZ刺激24 h后再予以ISO刺激24 h。Western blot法检测α-SMA和Fibronectin的蛋白表达。结果1.在心脏纤维化动物模型中,与对照和IgG组相比,ISO+TRZ组的心脏射血分数(EF)和左室短轴缩短率(FS)降低(P<0.05,P<0.01);与TRZ组相比,ISO+TRZ组的小鼠心脏纤维化面积显著增加(P<0.05);分别与TRZ、ISO、ISO+IgG组相比,ISO+TRZ组的TGF-β1,α-SMA和FIbronectin蛋白表达均显著升高(P<0.05,P<0.01,P<0.001)。2.在乳鼠原代心脏成纤维细胞中,与ISO+IgG组相比,ISO+TRZ组Fibronectin的蛋白表达明显提高(P<0.05);与ISO组相比,ISO+TRZ组α-SMA的蛋白表达有所提高(P<0.05)。结论TRZ能够加重ISO诱导的小鼠心脏纤维化。 展开更多

Neural EGFL-like 2(NELL2)is a secreted protein known for its regulatory functions in the nervous and reproductive systems,yet its role in bone biology remains unexplored.In this study,we observed that NELL2 was diminished in the bone of aged and ovariectomized(OVX)mice,as well as in the serum of osteopenia and osteoporosis patients.In vitro loss-of-function and gain-offunction studies revealed that NELL2 facilitated osteoblast differentiation and impeded adipocyte differentiation from stromal progenitor cells.In vivo studies further demonstrated that the deletion of NELL2 in preosteoblasts resulted in decreased cancellous bone mass in mice.Mechanistically,NELL2 interacted with the FNI-type domain located at the C-terminus of Fibronectin 1(Fn1).Moreover,we found that NELL2 activated the focal adhesion kinase(FAK)/AKT signaling pathway through Fn1/integrinβ1(ITGB1),leading to the promotion of osteogenesis and the inhibition of adipogenesis.Notably,administration of NELL2-AAV was found to ameliorate bone loss in OVX mice.These findings underscore the significant role of NELL2 in osteoblast differentiation and bone homeostasis,suggesting its potential as a therapeutic target for managing osteoporosis. 展开更多

Irisin is a myokine that is generated by cleavage of the membrane protein fibronectin type Ⅲ domain-containing protein 5(FNDC5) in response to physical exercise. Studies reveal that irisin/FNDC5 has neuroprotective functions against Alzheimer's disease, the most common form of dementia in the elderly, by improving cognitive function and reducing amyloid-β and tau pathologies as well as neuroinflammation in cell culture or animal models of Alzheimer's disease. Although current and ongoing studies on irisin/FNDC5 show promising results, further mechanistic studies are required to clarify its potential as a meaningful therapeutic target for alleviating Alzheimer's disease. We recently found that irisin treatment reduces amyloid-β pathology by increasing the activity/levels of amyloid-β-degrading enzyme neprilysin secreted from astrocytes. Cognitive dysfunction often occurs in Alzheimer’s disease,Parkinson’s disease,cerebrovascular disease,or other neurodegenerative diseases,and can significantly impact the life quality of patients and create serious social,psychological,and economic burdens for individuals and their families.Numerous studies have confirmed that exercise can slow the decline in cognitive function through multiple pathways,in which fibronectin type III domain-containing protein 5(FNDC5)plays an important role.However,the current research on the modulation of FNDC5 by exercise and its ability to improve hippocampal cognitive function lacks a systematic and comprehensive understanding.Therefore,this review focuses on the latest research progress regarding the role of exercise-induced FNDC5 in cognitive function,systematically reviews the positive effects of FNDC5 on cognitive function impairment caused by various factors,and clarifies the specific mechanisms by which exerciseinduced FNDC5 improves cognitive function by inhibiting neuroinflammation and improving hippocampal neurogenesis and hippocampal synaptic plasticity.Based on the existing literature,we also identify the areas that require further research in this field.Overall,this review provides a theoretical basis for exercise-based prevention and improvement of cognitive function impairment. 展开更多

BACKGROUND Hepatitis C virus(HCV)infection progresses through various phases,starting with inflammation and ending with hepatocellular carcinoma.There are several invasive and non-invasive methods to diagnose chronic HCV infection.The invasive methods have their benefits but are linked to morbidity and complications.Thus,it is important to analyze the potential of non-invasive methods as an alternative.Shear wave elastography(SWE)is a non-invasive imaging tool widely validated in clinical and research studies as a surrogate marker of liver fibrosis.Liver fibrosis determination by invasive liver biopsy and non-invasive SWE agree closely in clinical studies and therefore both are gold standards.AIM To analyzed the diagnostic efficacy of non-invasive indices[serum fibronectin,aspartate aminotransferase to platelet ratio index(APRI),alanine aminotransferase ratio(AAR),and fibrosis-4(FIB-4)]in relation to SWE.We have used an Artificial Intelligence method to predict the severity of liver fibrosis and uncover the complex relationship between non-invasive indices and fibrosis severity.METHODS We have conducted a hospital-based study considering 100 untreated patients detected as HCV positive using a quantitative Real-Time Polymerase Chain Reaction assay.We performed statistical and probabilistic analyses to determine the relationship between non-invasive indices and the severity of fibrosis.We also used standard diagnostic methods to measure the diagnostic accuracy for all the subjects.RESULTS The results of our study showed that fibronectin is a highly accurate diagnostic tool for predicting fibrosis stages(mild,moderate,and severe).This was based on its sensitivity(100%,92.2%,96.2%),specificity(96%,100%,98.6%),Youden’s index(0.960,0.922,0.948),area under receiver operating characteristic curve(0.999,0.993,0.922),and Likelihood test(LR+>10 and LR-<0.1).Additionally,our Bayesian Network analysis revealed that fibronectin(>200),AAR(>1),APRI(>3),and FIB-4(>4)were all strongly associated with patients who had severe fibrosis,with a 100% probability.CONCLUSION We have found a strong correlation between fibronectin and liver fibrosis progression in HCV patients.Additionally,we observed that the severity of liver fibrosis increases with an increase in the non-invasive indices that we investigated. 展开更多

本文研究除痰解毒方(Chutan Jiedu Decoction,CJD)联合吉非替尼对肺腺癌H1975荷瘤小鼠移植瘤twist、fibronectin表达的影响,从上皮间质转化(Epithelial-mesenchymal transition,EMT)角度探讨其抗肿瘤作用机制。建立人肺腺癌H1975耐药细... 本文研究除痰解毒方(Chutan Jiedu Decoction,CJD)联合吉非替尼对肺腺癌H1975荷瘤小鼠移植瘤twist、fibronectin表达的影响,从上皮间质转化(Epithelial-mesenchymal transition,EMT)角度探讨其抗肿瘤作用机制。建立人肺腺癌H1975耐药细胞荷瘤裸鼠模型,随机分为模型组,吉非替尼组,除痰解毒方低、中、高剂量组,除痰解毒方中剂量联合吉非替尼组,每组10只,各组予以相应的药物灌胃2周。检测瘤体积和瘤质量,计算抑瘤率,采用免疫组织化学法、Western blot法、荧光定量PCR法检测各组肿瘤组织twist、fibronectin蛋白及基因的表达。结果显示,联合用药组抑瘤率为61.92%,明显高于模型组、吉非替尼组,除痰解毒方低、中、高剂量组(P<0.01)。免疫组织化学法、Western blot法及荧光定量PCR法结果均显示除痰解毒方中剂量、高剂量及联合用药组均能下调twist和fibronectin的表达,且联合用药组下调作用高于单独用药组(P<0.01)。研究结果表明除痰解毒方联合吉非替尼对肺腺癌H1975荷瘤小鼠移植瘤生长及twist、fibronectin表达的抑制作用优于除痰解毒方单药及吉非替尼组,其协同增效作用可能与逆转EMT增强吉非替尼的敏感性有关。 展开更多

AIM:To investigate the role of insulin-like growth factor binding protein-7 (IGFBP-7) in the activation and transdifferentiation of hepatic stellate cells (HSC) in vitro.METHODS:Rat HSC-T6 cells were cultured in separate dishes and treated with various concentration of transforming growth factor (TGF)-β1,IGFBP-7 or antiIGFBP-7 antibody for 24 h.The supernatant or a cytoplasm suspension was obtained from cultured HSC,followed by transfer of cells to form cell-coated dishes.Immunocytochemistry and Western blotting were used to analyze the expression of IGFBP-7 induced by TGF-β1 and the level of fibronectin,collagen and α-smooth muscle actin (SMA).The pro-apoptotic effect of antiIGFBP-7 antibody was determined by flow cytometry.RESULTS:Immunocytochemistry and Western blotting revealed that the expression of IGFBP-7 in TGF-β1 treated HSC was significantly up-regulated compared to that in the control group.In addition,fibronectin,collagen and α-SMA also showed enhanced expression in accordance with the transdifferentiation process in a dose-dependent manner to some extent.Moreover,flow cytometry suggested that anti-IGFBP-7 antibody induced apoptosis of activated HSC,which is responsible for the development of liver fibrosis,and may represent a novel pathway and target for therapeutic intervention.CONCLUSION:IGFBP-7 showed increased expression in activated HSC and played an important role in the activation and transdifferentiation process of HSC.AntiIGFBP-7 antibody may ameliorate liver fibrogenesis. 展开更多

OBJECTIVE: To investigate the effect of the clinical effective prescription of Qingre Lishi Yishen decoction(QRLS) on the activation of mesangial cells in immunoglobulin A nephropathy(IgAN) rats.METHODS: IgAN rat’s model was established by combine with intragastric administration of bovine serum albumin(BSA) + intravenous injection of lipopolysaccharide(LPS) by + subcutaneous injection of carbon tetrachloride(CCL4). Then the animals were randomly divided into four groups: control group, IgAN model group, IgAN model with Valsartan(Val) treatment group and IgAN model with QRLS treatment group. To observe the indexes of 24-h urine protein, renal function, deposition of immune complexes, expression of activation factor, fibrosis marker and inflammatory cytokines in four different groups.RESULTS: The Val or QRLS treatment group:(a) it reduced the immune complexes deposition of IgA in glomerular mesangial and inhibited mesangial cell proliferation;(b) it decreased the expression of smooth muscle actin(α-SMA), fibronectin(FN) and tumor necrosis factor alpha(TNF-α).CONCLUSION: The study suggested that QRLS ameliorate renal structure and function in IgAN rat’s model. Furthermore, we also observed that QRLS alleviated mesangial cells activation and matrix accumulation partly by decreasing the α-SMA,then to downregulated the expression of FN and TNF-α. 展开更多

An understanding of osteoblast adhesion and proliferation on biomaterials is crucial to optimizing the surfaces of artificial implants used in clinical practice. Polished, anodic oxidation (AO) and micro-arc oxidation (MAO) treated titanium (Ti) plates were used as model surfaces to study the adhesion of MG-63 cells. Cells were monitored for 0.5 and 4 h; faster adhesion and spreading of MG-63 ceils were observed on the AO and MAO modified samples. Stimulated secretion of fibronectin (FN) influenced the adhesion rates. In addition, AO and MAO modified surfaces promoted cell proliferation through apparent up-regulation of FN and integrin a5 transcription via outside-in signaling. This strongly suggests that FN secretion by osteoblasts plays an essential role in enhanced cell adhesion, spreading and proliferation on these modified Ti surfaces. 展开更多

目的:通过Fibronectin介导进行差别粘附法筛选出人黄韧带干细胞并进行鉴定,探讨其作为组织工程种子细胞的可行性。方法:从椎间孔镜或椎间盘镜微创手术中获取10例人黄韧带标本,机械-酶消化法联合获取原代细胞,扩增第二代后接种于Fibron... 目的:通过Fibronectin介导进行差别粘附法筛选出人黄韧带干细胞并进行鉴定,探讨其作为组织工程种子细胞的可行性。方法:从椎间孔镜或椎间盘镜微创手术中获取10例人黄韧带标本,机械-酶消化法联合获取原代细胞,扩增第二代后接种于Fibronectin包被过的培养瓶。流式细胞仪检测细胞表面特异性标志物。免疫组化方法测定干细胞表达相关特异性蛋白。使用干细胞诱导培养基(实验组)向成骨、成脂、成软骨多向诱导分化,使用常规生长培养基培养者作为阴性对照(对照组),分别行茜素红染色、油红O染色、阿利辛兰染色鉴定。PT-PCR检测成骨、成脂肪、成软骨基因表达。取同一患者的髂骨骨髓提取骨髓间充质干细胞,统计学分析黄韧带干细胞与骨髓间充质干细胞的细胞周期、增殖能力和干性基因表达差异。结果:原代细胞形态呈三角形或多角形,经Fibronectin介导粘附培养后形态比较均一,呈长梭形克隆群。流式细胞仪检测筛选后的黄韧带干细胞CD90、CD73阳性率〉96.8%,CD105阳性率〉95.9%,而stro-1为阴性。实验组茜素红染色呈强阳性,油红O染色可见大小不等脂滴呈红色,阿利辛兰染色大量蛋白聚糖聚集呈蓝色,对照组均为阴性。PT-PCR测定实验组成骨基因(OC、ALP、RUNX-2)、成脂肪基因(LPL、APP、PPAR2)、成软骨基因(COLⅡ、AGG、SOX9)显著上调,其中RUNX-2在对照组有低水平表达。免疫组化显示黄韧带干细胞和骨髓间充质干细胞均表达a-SMA、Ⅰ型胶原、纤连蛋白,但均不表达Ⅱ型胶原。两种干细胞均超过80%的细胞比例处于G0/G1期提示都有很强自我更新能力(P〉0.05)。CCK-8检测不同时间点两种细胞的增殖能力,在第1~10天时OD值逐渐增加,第10~14天达稳定状态,二者有相似的增殖能力(P〉0.05)。两种干细胞均表达干性基因NONAG、OCT-4、SOX2,组间差异无显著性(P〉0.05)。结论:通过Fibronectin差别粘附筛选法可有效筛选纯化出黄韧带干细胞,其可向成骨、成脂、成软骨多向分化,为组织工程技术治疗退变椎间盘提供了新的种子细胞。 展开更多

目的:探讨局部应用Fibronectin(FN)对周围神经再生的影响。方法:将34只SD大鼠分为4组,1~3组每组10只,手术切除双侧10mm坐骨神经并用硅胶管套接,左侧套管内注入FN10μg;右侧注入生理盐水作为对照。术后1、3、4个月分别进行电生... 目的:探讨局部应用Fibronectin(FN)对周围神经再生的影响。方法:将34只SD大鼠分为4组,1~3组每组10只,手术切除双侧10mm坐骨神经并用硅胶管套接,左侧套管内注入FN10μg;右侧注入生理盐水作为对照。术后1、3、4个月分别进行电生理和组织学检查。第4组另4只鼠于术后3个月行辣根过氧化物酶(HRP)逆行示踪检查。结果:FN治疗组术后1、3、4个月时的电生理和形态学检查结果均明显优于对照组。术后HRP逆行示踪检查发现相应节段的治疗组脊髓前角和背根神经节标记神经元明显多于对照组。结论:FN可促进周围神经损伤后的再生。 展开更多

Effects of Huang Qi Wu Wu Decoction on Plasma Proteins in 70 Cases of Chronic Pulmonary Heart Disease

Simple immune agar diffusion test was used to assay the contents of 12 plasma proteins in 70 cases of chronic pulmonary heart disease treated by Huang Qi Wu Wu Decoction (黄芪五物汤), with the other 70 cases who were not given Huang Qi Wu Wu Decoction as the control group. The total clinical effective rate in the treatment group was 90.0%, while that in the control group was 75.7%, with a statistically significant difference between the two groups (P<0.05). In the treatment group, the levels of prealbumin, transferrin and fibronectin elevated obviously after treatment, and the contents of C-reactive protein, ceruloplasmin, haptoglobin, a 1-antitrypsin and a 1-acid glycoprotein decreased markedly (P<0.01). In the control group, only the levels of ceruloplasmin and C-reactive protein decreased significantly (P<0.05). It is shown that Huang Qi Wu Wu Decoction may enhance the therapeutic effects for pulmonary heart disease, regulate the metabolism of plasma proteins, and improve the life quality of the patients. 展开更多
