Objective:Angiogenesis is a critical target for hepatocellular carcinoma(HCC)treatment The previous studies indicated that Jiedu Fang(JDF)could inhibit hypoxia-induced angiogenesis through interleukin-8(IL-8).Therefor...Objective:Angiogenesis is a critical target for hepatocellular carcinoma(HCC)treatment The previous studies indicated that Jiedu Fang(JDF)could inhibit hypoxia-induced angiogenesis through interleukin-8(IL-8).Therefore,the present study further explores the mechanisms behind JDF's inhibition of HCC angiogenesis.Methods:Angiogenesis was assessed with the capillary-like tube formation assay in vitro and the matrigel plug angiogenesis assay in vivo.A liver cancer-related gene set and genes associated with angiogenesis and the hypoxic microenvironment were analyzed using a bioinformatics platform.Real-time reverse transcription-polymerase chain reaction and Western blotting assays were used to assess the targeted mRNA and protein levels,respectively.The Transwell assay was used to assess the migration and invasion potential of EA.hy 926 cells.The orthotopic tumor xenograft model was established,and immunohistochemistry and immunofluorescence assays were used to detect cluster of differentiation 31 and angiopoietin 2 expression,while an enzyme-linked immunosorbent assay was used to detect vascular endothelial growth factor and IL-8 protein levels.Results:In vitro and in vivo assays showed that IL-8 promoted angiogenesis,and JDF could antagonize this effect.Bioinformatics analysis indicated that aurora kinase A(Aurora A)was an important candidate,which can promote IL-8 expression through activation of signal transducer and activator of transcription3(STAT3).The overexpression of Aurora A increased IL-8 secretion and promoted HCC migration,invasion,and angiogenesis,which was partly inhibited by JDF.Such effects were validated by in vivo assays.Further validation using the STAT3 inhibitor S3I-201 demonstrated that STAT3 was regulated by Aurora A.Conclusion:JDF exhibits efficacy in reducing hypoxia-induced angiogenesis in HCC through a mechanism involving the Aurora A/STAT3/IL-8 signaling pathway.Therefore,JDF holds promise as a potential therapeutic approach for targeting HCC angiogenesis.展开更多

OBJECTIVE:To elucidate the potential mechanisms of Shizhi Fang(SZF,矢志方)in the treatment of uric acid nephropathy(UAN).METHODS:SZF-containing serum was prepared from six male rats and analyzed using ultra-high-perfo...OBJECTIVE:To elucidate the potential mechanisms of Shizhi Fang(SZF,矢志方)in the treatment of uric acid nephropathy(UAN).METHODS:SZF-containing serum was prepared from six male rats and analyzed using ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS).Network pharmacology was employed was integrated with UPLC-Q-TOF-MS to predict SZF targets for the treatment of UAN,which were subsequently validated through in vivo experiments.Sixty male Bagg Albino Laboratory-Bred Mouse,substrain c mice were randomly allocated into six groups:Normal,Model,Febuxostat,and three SZF dosage groups.Except for the Normal group,all mice were administered potassium oxonate(250 mg/kg)and adenine(50 mg/kg)via gavage to induce UAN.Four hours postadministration,the Febuxostat group received Febuxostat(6 mg/kg),while the SZF groups received low(0.234 g/kg),medium(0.468 g/kg),or high(0.936 g/kg)doses of SZF.The Normal and Model groups were given an equivalent volume of saline.All treatments were conducted over a period of four weeks.Urine and blood samples were collected for biochemical analysis,and kidney tissues were subjected to histopathological examination and Western blot analysis.RESULTS:Nine prototype compounds and 30 metabolites were identified in SZF serum.Network pharmacology analysis revealed 195 drug targets and 1608 disease targets,with 76 common drug-disease targets,including signal transducer and activator of transcription 3(STAT3),proto-oncogene tyrosine-protein kinase Src(SRC),matrix metalloproteinase-9(MMP9),Caspase 3,and toll-like receptor 4(TLR4)as key targets.Gene Ontology analysis identified 325 biological processes,48 cellular components,and 72 molecular functions,while Kyoto Encyclopedia of Genes and Genomes analysis identified 113 pathways.Molecular docking demonstrated strong binding affinities between active compounds and their targets.In the animal study,SZF treatment alleviated pathological damage and improved serum and urine biochemical markers compared to the Model group(P<0.05,P<0.01,P<0.001).Western blot analysis showed a significant reduction in phosphorylated-STAT3,phosphorylatedSRC,MMP9,TLR4,and Caspase3 expression in renal tissues of SZF-treated mice(P<0.001).CONCLUSION:SZF may exert therapeutic effects on UAN through multiple targets and pathways.展开更多

AIM: To investigate whether the Chinese medicine Tong Xie Yao Fang(TXYF) improves dysfunction in an irritable bowel syndrome(IBS) rat model. METHODS: Thirty baby rats for IBS modeling were separated from mother rats(1...AIM: To investigate whether the Chinese medicine Tong Xie Yao Fang(TXYF) improves dysfunction in an irritable bowel syndrome(IBS) rat model. METHODS: Thirty baby rats for IBS modeling were separated from mother rats(1 h per day) from days 8 to 21, and the rectum was expanded by angioplasty from days 8 to 12. Ten normal rats were used as normal controls. We examined the effects of TXYF on defection frequency, colonic transit function and smooth muscle contraction, and the expression of 5-hydroxytryptamine(5-HT) and substance P(SP) in colonic and hypothalamus tissues by Western blot and RT-PCT techniques in both normal rats and IBS model rats with characterized visceral hypersensitivity.in normal rats and 4.5 ± 1.58 in IBS model rats(P < 0.001). However, the defecation frequency was significantly decreased(3.0 ± 1.25 vs 4.5 ± 1.58, P < 0.05), while the time(in seconds) of colon transit function was significantly increased(256.88 ± 20.32 vs 93.36 ± 17.28, P < 0.001) in IBS + TXYF group rats than in IBS group rats. Increased colonic smooth muscle tension and contract frequency in IBS model rats were significantly decreased by administration of TXYF. Exogenous agonist stimulants increased spontaneous activity and elicited contractions of colon smooth muscle in IBS model rats, and all of these actions were significantly reduced by TXYF involving 5-HT and SP down-regulation. CONCLUSION: TXYF can modulate the activity of the enteric nervous system and alter 5-HT and SP activities, which may contribute to the symptoms of IBS.展开更多
BACKGROUND Tong Xie Yao Fang is a representative traditional Chinese prescription for the treatment of liver and spleen deficiency,abdominal pain and diarrhea.It has a unique function in the treatment of gastrointesti...BACKGROUND Tong Xie Yao Fang is a representative traditional Chinese prescription for the treatment of liver and spleen deficiency,abdominal pain and diarrhea.It has a unique function in the treatment of gastrointestinal dysfunction including irritable bowel syndrome(IBS),is a common functional bowel disease.Its main symptoms are recurrent abdominal pain,diarrhea,constipation or alternations between diarrhea and constipation.There are obvious differences in metabolites between TCM syndromes.By comparing the body fluid metabolism maps of model animals,metabolomics can discover disease biomarkers,analyze the differences in metabolic pathways and understand the pathological process and the metabolic pathways of substances in the body.Thus,the evaluation of animal models tends to be comprehensive and objective.This may provide further understanding between the interaction between Tong Xie Yao Fang and the IBS model.AIM To evaluate the effect of Tong Xie Yao Fang on IBS rats by using metabolomics method.METHODS Wistar rats were used to establish IBS models,and then randomly divided into four groups:A model control group and three Tong Xie Yao Fang treatment groups(high,medium and low doses).A normal,non-IBS group was established.The rats were treated for 2 wk.On days 0 and 14 of the experimental model,urine was collected for 12 h and was analyzed by ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry.Nine potential biomarkers were identified,and six major metabolic pathways were found to be related to IBS.RESULTS In the study of metabonomics,nine potential biomarkers including L-serine,4-methylgallic acid,L-threonine,succinylacetone,prolyl-hydroxyproline,valylserine,acetyl citrate,marmesin rutinoside and 5-hydroxy-L-tryptophan were identified in urine,which were assigned to amino acids,organic acids,succinyl and glycosides.Furthermore,the metabolic pathway of L-serine,L-threonine and 5-hydroxy-L-tryptophan was found in the Kyoto Encyclopedia of Genes and Genomes,which mainly involved the metabolism of cysteine and methionine,vitamin B6 metabolism,serotonin synapse,tryptophan metabolism,sphingolipid metabolism,digestion,absorption of protein and amino acid metabolism.These pathways are related to intestinal dysfunction,inflammatory syndrome,nervous system dysfunction and other diseases.CONCLUSION Tong Xie Yao Fang has pharmacological effects on IBS,and its mechanism may be related to the metabolism of the nine potential biomarkers identified above in urine.展开更多

This study established a rat model of chronic cerebral ischemia-caused white matter damage by ligation of the bilateral common carotid arteries. Rats received daily intragastric administration of a suspension of Kaixin ]ieyu Fang powder. After 3, 7 and 21 days of treatment, the degree of white matter damage in the cerebral ischemia rat model was alleviated, Bcl-2 protein and mRNA expression in brain tissue increased, and Bax protein and mRNA expression decreased. These results indicate that Kaixin Jieyu Fang can alleviate cere- bral white matter damage, and the underlying mechanism is associated with regulation of Bcl-2/ Bax protein and mRNA expression, which is one of possible mechanism behind the protective effect of Kaixin Jieyu Fang against vascular depression.展开更多
OBJECTIVE:To determine the effect of Wenyang Huazhuo Fang(WHF),a Traditional Chinese Medicine decoction,on renal function in a rat model of doxorubicin-induced nephropathy,and to elucidate the underlying mechanism.MET...OBJECTIVE:To determine the effect of Wenyang Huazhuo Fang(WHF),a Traditional Chinese Medicine decoction,on renal function in a rat model of doxorubicin-induced nephropathy,and to elucidate the underlying mechanism.METHODS:Sprague-Dawley rats were randomly divided into six groups:control,doxorubicin-nephropathy,and prednisone-treated(6.45 mg·kg^-1·d^-1)doxorubicin nephropathy groups,as well as high-(7.26 g·kg^-1·d^-1),medium-(2.42 g·kg-1·d-),and low-dose(0.81 g·kg^-1·d^-1)WHF-treated doxorubicin-nephropathy groups.The nephropathy rat model was established by two tail vein injections of doxorubicin,followed by prednisone or WHF treatment for 8 weeks.Body weights were monitored and urinary protein was measured every 2 weeks.After the end of the treatment period,the rats were euthanized.Serum biochemical indicators were determined and renal morphological alterations were assessed using histological staining.The expression of transient receptor potential cation channel subfamily C member 6(TRPC6),stromal interaction molecule 1(STIM1),and calcium release-activated calcium channel protein 1(Orai1)was detected using western blotting,and their mRNA levels were examined using quantitative real-time reverse transcription-polymerase chain reaction.RESULTS:WHF treatment was found to significantly ameliorate weight loss,proteinuria,hypoalbuminemia,and dyslipidemia in doxorubicin-nephropathy rats.The protein and mRNA levels of TRPC6,STIM1,and Orai1 were partially,but significantly suppressed by prednisone or WHF treatment.CONCLUSION:Treatment with WHF significantly ameliorates renal injury in a rat model of doxorubicin-induced nephropathy,which could be at least partially related to repression of the TRPC6 pathway.展开更多
OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carci...OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carcinoma (HCC) nude mice. METHODS: Forty male nude mice were randomly divided into 4 groups. Human HCC tissue was implanted in the livers of three groups. After 24 h, the three groups were treated respectively with JPJDF (37.5 g/kg), saline (20 mL/kg) and Tegafur (FT-207, 160 mg/kg) once a day for 10 weeks. The control group without implanting the tissue was concurrently treated with saline (20 mL/kg). The survival data and body weight of all mice were recorded, and expression levels of PTEN, PI3K and FAK in normal tissue and cancer tissue of the livers were eval-uated with immunohistochemical method. RESULTS: The cumulative survival rate of the mice in the JPJDF group was higher than those of the other groups. The rate of weight loss was the lowest in JPJDF group. The survivability and weight loss rate in FT-207 group were the poorest in all groups. The expression intensity of PTEN was higher in normal tissues than in cancer tissues, and lower in the normal tissues of HCC models than in that of mice without HCC. The PTEN expression intensity in normal tissue and cancer tissue from mice treated with FT-207 were lower than that from the mice treated with JPJDF or saline.The expression intensity of PI3K was higher in cancer tissue than in normal tissue. The PI3K expression intensity was the lowest in normal tissue and cancer tissue from mice treated with JPJDF, and the intensity from mice treated with FT-207 was the highest. In mice treated with JPJDF, the expression intensity of FAK was higher in the normal tissue and lower in the cancer tissue than those of the other treatment groups. CONCLUSION: The mechanism accounting for the prolonged survival of HCC-bearing mice treated with JPJDF might be related to the reduction in weight loss and the benign regulation of PTEN, PI3K, and FAK.展开更多
OBJECTIVE:To investigate the influence and possible targets of Dangua Fang(丹瓜方)on tricarboxylic acid(TCA)cycle and respiratory chain to enrich the prescription’s mechanism of effective intervention on glycolipid m...OBJECTIVE:To investigate the influence and possible targets of Dangua Fang(丹瓜方)on tricarboxylic acid(TCA)cycle and respiratory chain to enrich the prescription’s mechanism of effective intervention on glycolipid metabolic diseases such as type 2 diabetes.METHODS:After interventional rats were fed with high glucose and high fat diet ad libitum for 4 weeks,intraperitoneally injected streptozotocin to induce diabetic model.According to blood glucose level,28 diabetic rats were selected and continued to be fed with high glucose and high fat diet,were stratified by body weight,and divided randomly by blood glucose into Model group(was given sterile water by gastric perfusion and injected aquae pro injection intraperitoneally),Dangua group[Dangua liquor(丹瓜方液)20.5 g·kg^(-1)·d^(-1) by perfusion and aquae pro injection intraperitoneally],Inhibitor group[sterile water by perfusion and nicotinamide phosphoribosyl transferase(Nampt)specific blocker GEN-6171.25 mg/kg intraperitoneally],DanInhit group(Dangua liquor and GEN-617 synchronously).Control group were continuously fed with ordinary diet.The intervention was last for 10 weeks.Body weight(BW),liver index(LI),glycosylated hemoglobin(HbA1c),TC,TG,free fatty acids(FFA),creatinine(Cr),and A-ketoglutarate(α-KG),Iso-citric acid(ICA),oxaloacetic acid(OAA)were tested.The cytochrome C oxidase(COX)and Succinate dehydrogenase(SDH)were evaluated by Colorimetry;Nampt protein,Adenosine triphosphate(ATP)synthase(ATPs),Nicotinamide adenine dinucleotide(NAD^(+))and its reduced(NADH)in liver were measured by enzyme linked immunosorbent assay.The expressions of Nampt and mitochondrialnadhdehydrogenase-1(mt-ND1)gene in liver was assessed by real-time polymerase chain reaction.Hepatic tissue staining was also completed.RESULTS:The levels of BW,ICA,α-KG and Nampt-mRNA in the Model group are lower than that in the Normal group(P<0.05),conversely,liver weight,LI,TC,HbA1c,SDH and ATPs,mt-ND1-mRNA,and Nampt protein in the Model group are higher(P<0.01,P<0.05).Compared with Model group,the levels of ICA,Nampt-mRNA and Nampt in Dangua group are significantly increased,and FFA obviously raised(P<0.01 and P<0.05);liver weight,BW,SDH are obviously lower,and HbA1c decreased significantly(P<0.01,P<0.05).TG,FFA and Nampt protein increased in the DanInhit group,TC,TG,BW obviously increased in the Inhibitor group,but SDH is decreased in both the two groups(P<0.05,P<0.01).Compared with Dangua group,DanInhib group has the lower levels of ICA,mt-ND1-mRNA,Nampt-mRNA,and the higher level of BW,LI and HbA1c.In the Inhibitor group,ICA and Nampt protein decreased,BW and LI,HbA1c and TG increased(P<0.01 or P<0.05).Tissue staining display that,in the model group there is obvious pathologic changes ie:fibrosis,steatosis and inflammatory cell infiltration.Lesions in the Dangua group are mild,and those of Inhibitor group are more obvious than the Model group,and DanInhit group is intermediately affected compared to Dangua group and Inhibitor group.CONCLUSION:Dangua Fang increases the metabolic flux of TCA cycle and optimizes respiratory chain function by up-regulating Nampt expression.展开更多

And finally, on the fifth experiment we researched the mechanism of Dangua Fang improving mitochondrial function by comparing the Na~+-k~+-ATPase and peroxisome proliferator-activated receptor-gamma coactivator-1alpha(PGC-1α) in the Dangua group with the diazoxide group and Co Q+Vit C group. RESULTS: We compared the control group in the first experiments and the OD values in DZ1 group was the most significant in all intervening groups. The recipe of DZ1(5% serum containing Dangua Fang) was used in the following experiments. Compared with the control group, cell viability, cell cycle(G2 + S), cytochrome c oxidase(COX), R3 red/green, R2 red/green, R1 red/green decreased and apoptosis, succinate dehydrogenase(SDH), green(R2 + R3), Na+-k+-ATPase, PGC-1α increased in the model group. Compared with the model group, cell viability, G2+S, COX, R3 red/green, R2 red/green, R1 red/green raised and apoptosis, green(R2 + R3), Na-K-ATPase decreased in the Dangua group;G2 + S, R3 red/green, R2 red/green, R1 red/green raised and green(R2 + R3) decreased in the Co Q + Vit C group. Na-K-ATPase increased in the combined group(P < 0.05 or < 0.01). CONCLUSIONS: Dangua Fang protects oxidative stressinduced endothelial cells damaged by promotion of mitochondrial biogenesis, reduction of Na~+-K~+-ATPase activity and regulation of mitochondrial respiratory chain function restoring mitochondrial membrane potential.展开更多

文摘Objective:Angiogenesis is a critical target for hepatocellular carcinoma(HCC)treatment The previous studies indicated that Jiedu Fang(JDF)could inhibit hypoxia-induced angiogenesis through interleukin-8(IL-8).Therefore,the present study further explores the mechanisms behind JDF's inhibition of HCC angiogenesis.Methods:Angiogenesis was assessed with the capillary-like tube formation assay in vitro and the matrigel plug angiogenesis assay in vivo.A liver cancer-related gene set and genes associated with angiogenesis and the hypoxic microenvironment were analyzed using a bioinformatics platform.Real-time reverse transcription-polymerase chain reaction and Western blotting assays were used to assess the targeted mRNA and protein levels,respectively.The Transwell assay was used to assess the migration and invasion potential of EA.hy 926 cells.The orthotopic tumor xenograft model was established,and immunohistochemistry and immunofluorescence assays were used to detect cluster of differentiation 31 and angiopoietin 2 expression,while an enzyme-linked immunosorbent assay was used to detect vascular endothelial growth factor and IL-8 protein levels.Results:In vitro and in vivo assays showed that IL-8 promoted angiogenesis,and JDF could antagonize this effect.Bioinformatics analysis indicated that aurora kinase A(Aurora A)was an important candidate,which can promote IL-8 expression through activation of signal transducer and activator of transcription3(STAT3).The overexpression of Aurora A increased IL-8 secretion and promoted HCC migration,invasion,and angiogenesis,which was partly inhibited by JDF.Such effects were validated by in vivo assays.Further validation using the STAT3 inhibitor S3I-201 demonstrated that STAT3 was regulated by Aurora A.Conclusion:JDF exhibits efficacy in reducing hypoxia-induced angiogenesis in HCC through a mechanism involving the Aurora A/STAT3/IL-8 signaling pathway.Therefore,JDF holds promise as a potential therapeutic approach for targeting HCC angiogenesis.

文摘OBJECTIVE:To elucidate the potential mechanisms of Shizhi Fang(SZF,矢志方)in the treatment of uric acid nephropathy(UAN).METHODS:SZF-containing serum was prepared from six male rats and analyzed using ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS).Network pharmacology was employed was integrated with UPLC-Q-TOF-MS to predict SZF targets for the treatment of UAN,which were subsequently validated through in vivo experiments.Sixty male Bagg Albino Laboratory-Bred Mouse,substrain c mice were randomly allocated into six groups:Normal,Model,Febuxostat,and three SZF dosage groups.Except for the Normal group,all mice were administered potassium oxonate(250 mg/kg)and adenine(50 mg/kg)via gavage to induce UAN.Four hours postadministration,the Febuxostat group received Febuxostat(6 mg/kg),while the SZF groups received low(0.234 g/kg),medium(0.468 g/kg),or high(0.936 g/kg)doses of SZF.The Normal and Model groups were given an equivalent volume of saline.All treatments were conducted over a period of four weeks.Urine and blood samples were collected for biochemical analysis,and kidney tissues were subjected to histopathological examination and Western blot analysis.RESULTS:Nine prototype compounds and 30 metabolites were identified in SZF serum.Network pharmacology analysis revealed 195 drug targets and 1608 disease targets,with 76 common drug-disease targets,including signal transducer and activator of transcription 3(STAT3),proto-oncogene tyrosine-protein kinase Src(SRC),matrix metalloproteinase-9(MMP9),Caspase 3,and toll-like receptor 4(TLR4)as key targets.Gene Ontology analysis identified 325 biological processes,48 cellular components,and 72 molecular functions,while Kyoto Encyclopedia of Genes and Genomes analysis identified 113 pathways.Molecular docking demonstrated strong binding affinities between active compounds and their targets.In the animal study,SZF treatment alleviated pathological damage and improved serum and urine biochemical markers compared to the Model group(P<0.05,P<0.01,P<0.001).Western blot analysis showed a significant reduction in phosphorylated-STAT3,phosphorylatedSRC,MMP9,TLR4,and Caspase3 expression in renal tissues of SZF-treated mice(P<0.001).CONCLUSION:SZF may exert therapeutic effects on UAN through multiple targets and pathways.

文摘Jing Fang granules(JFG)are a modern Chinese medicinal formulation derived from Jing Fang Defeating Toxin San(JFDS).JFG primarily contains bioactive compounds such as flavonoids/flavonoid glycosides,coumarins and other components,which have various activities:anti-inflammatory,immune system regulation,antiviral,anti-aging,detoxification,and protection of the liver,lungs,and kidneys.JFG has broad potential therapeutic applications.The mechanisms of JFG’s actions are primarily associated with signaling pathways such as TLR4/NF-κB,PI3K/AKT,and MAPK.This article reviews the research progress on JFG,including chemical composition,pharmacologic effects,mechanism of action,safety,clinical application,etc.,serving as a foundation for future research and clinical applications.

文摘The mechanism of antithrombotic of Dahuangzhechong Fang separated and purified by Ti-Al intermetallic compound porous material (TAICPM) was researched. Dahuangzhechong Fang, which was isolated and screened by TAICPM, was used to oral rats. At the end of study, their blood and thrombus were collected. The results show that TAICPM with the pore size of 1-5 μm can screen Dahuangzhechong Fang well. Dahuangzhechong Fang can increase 6-keto-PGF1α, lower content of TXD2 and platelet. Dahuangzhechong Fang has good effect to resist arterial thrombosis.

文摘This article aims to provide some thoughts on the prevention and treatment of Coronavirus Disease 2019(COVID‑19)from the perspective of JingFang Medicine(Classical Chinese Formula).It is believed that the vague theoretical understanding of COVID‑19 in Traditional Chinese Medicine does not hinder the precise treatment of the disease by following the rule of“With this Zheng,prescribe this Fang.”According to the principle of“Fang‑Zheng Correlation”and the knowledge gained from the thousands of years of experience in treating febrile diseases,Xiao Chai Hu Decoction(小柴胡汤)and its modifications are recommended with the emphasis on individualized treatment.As another form of practicing“Fang‑Zheng Correlation,”generalized group treatment should also be paid attention to.Giving considerations to the historical medical data,Jing Fang Bai Du Powder(荆防败毒散)and Shi Shen Decoction(十神汤)are recommended for group prevention treatment.Assisting the Zheng(Upright)Qi and using tonic formulas are two entirely different concepts.According to the principle of“Fang‑Zheng Correlation,”tonics abuse should be avoided in the prevention of COVID‑19,and the using of Huang Qi(黄芪Radix Astragali seu Hedysari)should also be very carefully done.

文摘AIM: To investigate whether the Chinese medicine Tong Xie Yao Fang(TXYF) improves dysfunction in an irritable bowel syndrome(IBS) rat model. METHODS: Thirty baby rats for IBS modeling were separated from mother rats(1 h per day) from days 8 to 21, and the rectum was expanded by angioplasty from days 8 to 12. Ten normal rats were used as normal controls. We examined the effects of TXYF on defection frequency, colonic transit function and smooth muscle contraction, and the expression of 5-hydroxytryptamine(5-HT) and substance P(SP) in colonic and hypothalamus tissues by Western blot and RT-PCT techniques in both normal rats and IBS model rats with characterized visceral hypersensitivity.in normal rats and 4.5 ± 1.58 in IBS model rats(P < 0.001). However, the defecation frequency was significantly decreased(3.0 ± 1.25 vs 4.5 ± 1.58, P < 0.05), while the time(in seconds) of colon transit function was significantly increased(256.88 ± 20.32 vs 93.36 ± 17.28, P < 0.001) in IBS + TXYF group rats than in IBS group rats. Increased colonic smooth muscle tension and contract frequency in IBS model rats were significantly decreased by administration of TXYF. Exogenous agonist stimulants increased spontaneous activity and elicited contractions of colon smooth muscle in IBS model rats, and all of these actions were significantly reduced by TXYF involving 5-HT and SP down-regulation. CONCLUSION: TXYF can modulate the activity of the enteric nervous system and alter 5-HT and SP activities, which may contribute to the symptoms of IBS.

文摘BACKGROUND Tong Xie Yao Fang is a representative traditional Chinese prescription for the treatment of liver and spleen deficiency,abdominal pain and diarrhea.It has a unique function in the treatment of gastrointestinal dysfunction including irritable bowel syndrome(IBS),is a common functional bowel disease.Its main symptoms are recurrent abdominal pain,diarrhea,constipation or alternations between diarrhea and constipation.There are obvious differences in metabolites between TCM syndromes.By comparing the body fluid metabolism maps of model animals,metabolomics can discover disease biomarkers,analyze the differences in metabolic pathways and understand the pathological process and the metabolic pathways of substances in the body.Thus,the evaluation of animal models tends to be comprehensive and objective.This may provide further understanding between the interaction between Tong Xie Yao Fang and the IBS model.AIM To evaluate the effect of Tong Xie Yao Fang on IBS rats by using metabolomics method.METHODS Wistar rats were used to establish IBS models,and then randomly divided into four groups:A model control group and three Tong Xie Yao Fang treatment groups(high,medium and low doses).A normal,non-IBS group was established.The rats were treated for 2 wk.On days 0 and 14 of the experimental model,urine was collected for 12 h and was analyzed by ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry.Nine potential biomarkers were identified,and six major metabolic pathways were found to be related to IBS.RESULTS In the study of metabonomics,nine potential biomarkers including L-serine,4-methylgallic acid,L-threonine,succinylacetone,prolyl-hydroxyproline,valylserine,acetyl citrate,marmesin rutinoside and 5-hydroxy-L-tryptophan were identified in urine,which were assigned to amino acids,organic acids,succinyl and glycosides.Furthermore,the metabolic pathway of L-serine,L-threonine and 5-hydroxy-L-tryptophan was found in the Kyoto Encyclopedia of Genes and Genomes,which mainly involved the metabolism of cysteine and methionine,vitamin B6 metabolism,serotonin synapse,tryptophan metabolism,sphingolipid metabolism,digestion,absorption of protein and amino acid metabolism.These pathways are related to intestinal dysfunction,inflammatory syndrome,nervous system dysfunction and other diseases.CONCLUSION Tong Xie Yao Fang has pharmacological effects on IBS,and its mechanism may be related to the metabolism of the nine potential biomarkers identified above in urine.

文摘The Chinese compound Kaixin fieyu Fang can be used to treat vascular depression; however, the underlying mechanism remains unclear. This study established a rat model of chronic cerebral ischemia-caused white matter damage by ligation of the bilateral common carotid arteries. Rats received daily intragastric administration of a suspension of Kaixin ]ieyu Fang powder. After 3, 7 and 21 days of treatment, the degree of white matter damage in the cerebral ischemia rat model was alleviated, Bcl-2 protein and mRNA expression in brain tissue increased, and Bax protein and mRNA expression decreased. These results indicate that Kaixin Jieyu Fang can alleviate cere- bral white matter damage, and the underlying mechanism is associated with regulation of Bcl-2/ Bax protein and mRNA expression, which is one of possible mechanism behind the protective effect of Kaixin Jieyu Fang against vascular depression.

文摘OBJECTIVE:To determine the effect of Wenyang Huazhuo Fang(WHF),a Traditional Chinese Medicine decoction,on renal function in a rat model of doxorubicin-induced nephropathy,and to elucidate the underlying mechanism.METHODS:Sprague-Dawley rats were randomly divided into six groups:control,doxorubicin-nephropathy,and prednisone-treated(6.45 mg·kg^-1·d^-1)doxorubicin nephropathy groups,as well as high-(7.26 g·kg^-1·d^-1),medium-(2.42 g·kg-1·d-),and low-dose(0.81 g·kg^-1·d^-1)WHF-treated doxorubicin-nephropathy groups.The nephropathy rat model was established by two tail vein injections of doxorubicin,followed by prednisone or WHF treatment for 8 weeks.Body weights were monitored and urinary protein was measured every 2 weeks.After the end of the treatment period,the rats were euthanized.Serum biochemical indicators were determined and renal morphological alterations were assessed using histological staining.The expression of transient receptor potential cation channel subfamily C member 6(TRPC6),stromal interaction molecule 1(STIM1),and calcium release-activated calcium channel protein 1(Orai1)was detected using western blotting,and their mRNA levels were examined using quantitative real-time reverse transcription-polymerase chain reaction.RESULTS:WHF treatment was found to significantly ameliorate weight loss,proteinuria,hypoalbuminemia,and dyslipidemia in doxorubicin-nephropathy rats.The protein and mRNA levels of TRPC6,STIM1,and Orai1 were partially,but significantly suppressed by prednisone or WHF treatment.CONCLUSION:Treatment with WHF significantly ameliorates renal injury in a rat model of doxorubicin-induced nephropathy,which could be at least partially related to repression of the TRPC6 pathway.

文摘OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carcinoma (HCC) nude mice. METHODS: Forty male nude mice were randomly divided into 4 groups. Human HCC tissue was implanted in the livers of three groups. After 24 h, the three groups were treated respectively with JPJDF (37.5 g/kg), saline (20 mL/kg) and Tegafur (FT-207, 160 mg/kg) once a day for 10 weeks. The control group without implanting the tissue was concurrently treated with saline (20 mL/kg). The survival data and body weight of all mice were recorded, and expression levels of PTEN, PI3K and FAK in normal tissue and cancer tissue of the livers were eval-uated with immunohistochemical method. RESULTS: The cumulative survival rate of the mice in the JPJDF group was higher than those of the other groups. The rate of weight loss was the lowest in JPJDF group. The survivability and weight loss rate in FT-207 group were the poorest in all groups. The expression intensity of PTEN was higher in normal tissues than in cancer tissues, and lower in the normal tissues of HCC models than in that of mice without HCC. The PTEN expression intensity in normal tissue and cancer tissue from mice treated with FT-207 were lower than that from the mice treated with JPJDF or saline.The expression intensity of PI3K was higher in cancer tissue than in normal tissue. The PI3K expression intensity was the lowest in normal tissue and cancer tissue from mice treated with JPJDF, and the intensity from mice treated with FT-207 was the highest. In mice treated with JPJDF, the expression intensity of FAK was higher in the normal tissue and lower in the cancer tissue than those of the other treatment groups. CONCLUSION: The mechanism accounting for the prolonged survival of HCC-bearing mice treated with JPJDF might be related to the reduction in weight loss and the benign regulation of PTEN, PI3K, and FAK.

文摘OBJECTIVE:To investigate the influence and possible targets of Dangua Fang(丹瓜方)on tricarboxylic acid(TCA)cycle and respiratory chain to enrich the prescription’s mechanism of effective intervention on glycolipid metabolic diseases such as type 2 diabetes.METHODS:After interventional rats were fed with high glucose and high fat diet ad libitum for 4 weeks,intraperitoneally injected streptozotocin to induce diabetic model.According to blood glucose level,28 diabetic rats were selected and continued to be fed with high glucose and high fat diet,were stratified by body weight,and divided randomly by blood glucose into Model group(was given sterile water by gastric perfusion and injected aquae pro injection intraperitoneally),Dangua group[Dangua liquor(丹瓜方液)20.5 g·kg^(-1)·d^(-1) by perfusion and aquae pro injection intraperitoneally],Inhibitor group[sterile water by perfusion and nicotinamide phosphoribosyl transferase(Nampt)specific blocker GEN-6171.25 mg/kg intraperitoneally],DanInhit group(Dangua liquor and GEN-617 synchronously).Control group were continuously fed with ordinary diet.The intervention was last for 10 weeks.Body weight(BW),liver index(LI),glycosylated hemoglobin(HbA1c),TC,TG,free fatty acids(FFA),creatinine(Cr),and A-ketoglutarate(α-KG),Iso-citric acid(ICA),oxaloacetic acid(OAA)were tested.The cytochrome C oxidase(COX)and Succinate dehydrogenase(SDH)were evaluated by Colorimetry;Nampt protein,Adenosine triphosphate(ATP)synthase(ATPs),Nicotinamide adenine dinucleotide(NAD^(+))and its reduced(NADH)in liver were measured by enzyme linked immunosorbent assay.The expressions of Nampt and mitochondrialnadhdehydrogenase-1(mt-ND1)gene in liver was assessed by real-time polymerase chain reaction.Hepatic tissue staining was also completed.RESULTS:The levels of BW,ICA,α-KG and Nampt-mRNA in the Model group are lower than that in the Normal group(P<0.05),conversely,liver weight,LI,TC,HbA1c,SDH and ATPs,mt-ND1-mRNA,and Nampt protein in the Model group are higher(P<0.01,P<0.05).Compared with Model group,the levels of ICA,Nampt-mRNA and Nampt in Dangua group are significantly increased,and FFA obviously raised(P<0.01 and P<0.05);liver weight,BW,SDH are obviously lower,and HbA1c decreased significantly(P<0.01,P<0.05).TG,FFA and Nampt protein increased in the DanInhit group,TC,TG,BW obviously increased in the Inhibitor group,but SDH is decreased in both the two groups(P<0.05,P<0.01).Compared with Dangua group,DanInhib group has the lower levels of ICA,mt-ND1-mRNA,Nampt-mRNA,and the higher level of BW,LI and HbA1c.In the Inhibitor group,ICA and Nampt protein decreased,BW and LI,HbA1c and TG increased(P<0.01 or P<0.05).Tissue staining display that,in the model group there is obvious pathologic changes ie:fibrosis,steatosis and inflammatory cell infiltration.Lesions in the Dangua group are mild,and those of Inhibitor group are more obvious than the Model group,and DanInhit group is intermediately affected compared to Dangua group and Inhibitor group.CONCLUSION:Dangua Fang increases the metabolic flux of TCA cycle and optimizes respiratory chain function by up-regulating Nampt expression.

文摘OBJECTIVE: To evaluate the protective effects of serum containing Dangua Fang( 丹 瓜 方) on vascular endothelium damaged by oxidative stress. METHODS: Five experiments were completed in this paper. In the first experiment, we found the most suitable serum containing Dangua Fang by comparing groups with different serum containing Dangua Fang. In the second experiments we analyzed Dangua Fang influencing endothelial cell viability and apoptosis and cell cycle. The third experiment on Dangua Fang intervention of mitochondrial respiratory chain. The fourth experiment on Dangua Fang intervention of mitochondrial membrane potential. And finally, on the fifth experiment we researched the mechanism of Dangua Fang improving mitochondrial function by comparing the Na~+-k~+-ATPase and peroxisome proliferator-activated receptor-gamma coactivator-1alpha(PGC-1α) in the Dangua group with the diazoxide group and Co Q+Vit C group. RESULTS: We compared the control group in the first experiments and the OD values in DZ1 group was the most significant in all intervening groups. The recipe of DZ1(5% serum containing Dangua Fang) was used in the following experiments. Compared with the control group, cell viability, cell cycle(G2 + S), cytochrome c oxidase(COX), R3 red/green, R2 red/green, R1 red/green decreased and apoptosis, succinate dehydrogenase(SDH), green(R2 + R3), Na+-k+-ATPase, PGC-1α increased in the model group. Compared with the model group, cell viability, G2+S, COX, R3 red/green, R2 red/green, R1 red/green raised and apoptosis, green(R2 + R3), Na-K-ATPase decreased in the Dangua group;G2 + S, R3 red/green, R2 red/green, R1 red/green raised and green(R2 + R3) decreased in the Co Q + Vit C group. Na-K-ATPase increased in the combined group(P < 0.05 or < 0.01). CONCLUSIONS: Dangua Fang protects oxidative stressinduced endothelial cells damaged by promotion of mitochondrial biogenesis, reduction of Na~+-K~+-ATPase activity and regulation of mitochondrial respiratory chain function restoring mitochondrial membrane potential.
