METHODS: By using immunohistochemistry, expres-sion of claudin-1 in mucosa and invasive front of 136 gastric adenocarcinoma cases and proliferative index (Ki-67) were detected and analyzed. RESULTS: In mucosa, the claudin-1 over-expression rate of mucinous adenocarcinomas (including signet-ring cell carcinomas) was the highest. It was nega-tively related with the differentiation but positively related with the invasiveness and metastasis of gastric cancer. In invasive front, the claudin-1 over-expression rate was positively related with the differentiation, in-vasiveness and metastasis of gastric carcinoma. The expression transformation of claudin-1 was found in gastric carcinoma. The expression of claudin-1 in inva-sive front was transformed in 28/136 gastric carcinoma cases. The transformation rate in highly differentiated tubular adenocarcinomas was the highest (51.5%, 17/33). The deeper was the invasiveness, the higher was the transformation rate. The claudin-1 expression transformation rate in serosa and omenta was signifi -cantly higher (92.9%) than in tunica muscularis of in-vasive gastric cancer cases, as well as in patients withlymph node metastasis than in those without lymph node metastasis. CONCLUSION: Up-regulation of claudin-1 expres-sion and its transformation in invasive and metastatic gastric carcinoma suggest that claudin-1 participates in the transformation of biological behaviors in neo-plasms. Further study is needed to elucidate the pre-cise mechanism and the relation of claudin-1 expres-sion with the neoplasm progress.展开更多
The utility of artificial microRNAs(amiRNAs) to induce loss of gene function has been reported for many plant species,but expression efficiency of the different amiRNA constructs in different transgenic plants was l...The utility of artificial microRNAs(amiRNAs) to induce loss of gene function has been reported for many plant species,but expression efficiency of the different amiRNA constructs in different transgenic plants was less predictable,In this study,expressions of amiRNAs through the gene backbone of Arabidopsis miR168a were examined by both Agrobacterium-mediated transient expression and stable plant genetic transformation.A corresponding trend in expression of amiRNAs by the same amiRNA constructs between the transient and the stable expression systems was observed in the experiments.Plant genetic transformation of the constructs that were highly expressible in amiRNAs in the transient agro-infiltration assays resulted in generation of transgenic lines with high level of amiRNAs.This provides a simple method for rapid and effective selection of amiRNA constructs used for a time-consuming genetic transformation in plants.展开更多

The oncogene was cloned and identified as a single nucleot...An Ha-ras oncogene was isolated from a cell line of gastric carcinoma called BGE-823 in order to elucidate genetic control and the influence of DNA sequences. The oncogene was cloned and identified as a single nucleotide substitution of thymine for guanine in the 12th codon through the sequencing of its first axon. We compared the differences of expression and regulation between the transformed Ha-ras cells and untransformed parent cells. Data indicated that the expression of Ha-ras in the transformed cells was five-fold higher than in the untransformed cells and that the Ha-ras gene in the former was hypersensitive toward DNase I. In addition, a nuclear protein of 35 kilodaltons bound strongly to the 2.5 Kb fragment located upstream of the 6.6 Kb Ha-ras gene and contained a CC rich region. These results suggest that there might be another mechanism of activation for the ras gene besides point mutation.展开更多
We have demonstrated that the distribution of microtubules (MT), mlcrofilaments (MF) and fibronectin (FN) were diminished, while the gene expression of the calmodulin and c- fos enhanced in the transformed C3H10T1/2 c...We have demonstrated that the distribution of microtubules (MT), mlcrofilaments (MF) and fibronectin (FN) were diminished, while the gene expression of the calmodulin and c- fos enhanced in the transformed C3H10T1/2 cells. After treatment with 1 mM db-cAMP for 1 hour and 2 hours, there was an early and repldly reduced in gene expression of Calmodulin and c-fos respectively. After db-cAMP treatment for 4 -5 days, the number of capping cells of ConA binding decreased significantly and the cell surface microvllll decreased as well. The growth of treated cells was inhibited markedly. By using 4F1 cDNA probe, which is preferentially expressed In G1 phase, we have found that the db- cAMP treated cells were accumulated at G1 phase. Of particular interest is the fact that the distribution of microtubules, mlcrofilaments and fibronectln were recovered after treatment with 1 mM db-cAMP for 6 days. It is suggested that the Inhibition of proliferation, alteration, of phenotype and reco- very of cytoskeleton is transformed cells after treatment with db-cAMP are related to the Inhibition of gene expression of Calmodulin.展开更多

The research on scroll profiles theory at present mainly is aiming at the single profile and its amelioration or parameter optimization,not aiming at the natural characteristic of the profiles equation of scroll profi...The research on scroll profiles theory at present mainly is aiming at the single profile and its amelioration or parameter optimization,not aiming at the natural characteristic of the profiles equation of scroll profiles itself.Thus,the intrinsic differential equation bearing on arc-length and tangent direction angle was studied.Using the inherent differential equation and general functional theory,the conversion ideas between the expression of intrinsic differential equation and the expression in Cartesian coordinate was studied,and the condition of involutes characteristic was presented.The detailed analysis of four typical scroll profiles was given.According to the four typical scroll profiles and based on the method of Taylor series,the unified formation of general functional integration scroll profiles was established,which perfected general theory of scroll profiles increasingly.The envelope method to seek conjugation engagement profiles was studied.The unified general functional equation of scroll profiles represented the admissible function scrolls and the conjugation engagement characteristic of which was studied.The engagement condition of conjugation scroll profiles and the expression in Cartesian coordinate of conjugation scroll profiles of general functional integration scroll profiles were educed.Therefore,the restriction of inherent characteristics of single scroll profiles model is broken through.Solid modeling and machine tool path simulation can bridge the gap between design and manufacture which is essential for the design and manufacture of scroll compressors.With this method a perfect solid model can be achieved,which leads to an optimal design in terms of quality,delivery time and cost.The presentation of unified formation of general functional integration scroll profiles represents foundation factor of instinct characteristics of scroll profiles,which widens the design method of scroll compressor for automobile air conditioning.展开更多
The problems of ITRF2008,the latest International Terrestrial Reference Frame,are pointed out and analyzed as follows:(1) ITRF is not a mm-level Terrestrial Reference Frame;(2) the origin of ITRF is neither the Earth&...The problems of ITRF2008,the latest International Terrestrial Reference Frame,are pointed out and analyzed as follows:(1) ITRF is not a mm-level Terrestrial Reference Frame;(2) the origin of ITRF is neither the Earth's center of mass (CM) nor the center of figure (CF);(3) the scale of ITRF is not a uniform system in the sense of the gravitational theory of relativity.These problems result from the linear hypothesis used in the establishment and maintenance of ITRF,which includes the linear hypothesis of the coordinates definition of the ITRF reference stations,and the seven coordinate transformation parameters (three translation parameters,three rotation parameters,and one scale parameter) when the ITRF combine solution is constructed.The linear hypothesis of the ITRF construction leads to the current terrestrial reference frame only at the cm-level,which cannot satisfy the requirements of monitoring mm-level crust movements as well as the global environment.This article points out that the construction of a mm-level Terrestrial Reference Frame is actually a leap from linear to nonlinear.Therefore,according to the main characteristics of nonlinear changes of the crust's deformation,the geocenter motion and the overall height fluctuation of the Earth,the new ITRF station coordinates definition and the new observation equations of combined solutions are constructed for the realization of a mm-level nonlinear ITRF,which can solve the problems of the current ITRF.展开更多
