Objective This study aimed to investigate the effect and underlying mechanism of Fructus lycii in improving exercise fatigue.Methods A network pharmacological approach was used to explore potential mechanisms of actio...Objective This study aimed to investigate the effect and underlying mechanism of Fructus lycii in improving exercise fatigue.Methods A network pharmacological approach was used to explore potential mechanisms of action of Fructus lycii.Skeletal muscle C2C12 cells and immunofluorescence were employed to verify the effect and mechanism of the representative components in Fructus lycii predicted by network pharmacological analysis.Results Six potential active components,namely quercetin,β-sitosterol,stigmasterol,7-Omethylluteolin-6-C-beta-glucoside_qt,atropine,and glycitein,were identified to have potency in improving exercise fatigue via multiple pathways,such as the PI3K-Akt,neuroactive ligand-receptor interaction,IL-17,TNF,and MAPK signaling pathways.The immunofluorescence results indicated that quercetin,a significant active component in Fructus lycii,increased the mean staining area of 2-NBDG,TMRM,and MitoTracker,and decreased the area of CellRox compared to the control.Furthermore,the protein expression levels of p-38 MAPK,p-MAPK,p-JNK,p-PI3K,and p-AKT markedly increased after quercetin treatment.Conclusion Fructus lycii might alleviate exercise fatigue through multiple components and pathways.Among these,quercetin appears to improve exercise fatigue by enhancing energy metabolism and reducing oxidative stress.The PI3K-AKT and MAPK signaling pathways also appear to play a role in this process.展开更多

[Objectives]This study was conducted to investigate the effects of Astragalus membranaceus in different groups on energy metabolism and CNTF protein expression in skeletal muscle of exercise-induced fatigue rats.[Meth...[Objectives]This study was conducted to investigate the effects of Astragalus membranaceus in different groups on energy metabolism and CNTF protein expression in skeletal muscle of exercise-induced fatigue rats.[Methods]Thirty-five clean male SD rats were randomly divided into a normal group,and low-,meddle-and high-dose groups of A.membranaceus aqueous solution,with 7 rats in each group.The low-dose,medium-dose and high-dose groups were given by gavage at 0.65,1.3 and 2.6 g/kg,respectively,while the normal group and the model group were given normal food and water.The weight of rats was observed.The contents of serum urea,lactate,muscle glycogen,liver glycogen and CNTF expression were detected.[Results]After modeling,compared with the normal group,the serum lactate and urea contents of rats in the model group significantly increased(P<0.01),while the muscle glycogen content(P<0.01)and liver glycogen content(P<0.05)of the skeletal muscle significantly decreased.Compared with the model group,the low-,meddle-and high-dose groups of A.membranaceus significantly reduced the levels of lactate and urea in serum(P<0.01),while the levels of muscle glycogen and liver glycogen in the skeletal muscle significantly increased(P<0.01,P<0.05).[Conclusions]This study provides a good research foundation for the treatment of exercise-induced fatigue using traditional Chinese herb A.membranaceus in modern clinical practice.展开更多

Exercise intolerance is a common issue upon acute exposure to high altitude when a person enters high-altitude environment before adapted.Oxidative stress and intestinal microbiota imbalance are increasingly recognize...Exercise intolerance is a common issue upon acute exposure to high altitude when a person enters high-altitude environment before adapted.Oxidative stress and intestinal microbiota imbalance are increasingly recognized as important contributors to hypobaric hypoxia-associated fatigue.Selenium is a well-known anti-oxidative nutrient and probiotics are widely accepted as intestinal microbiota regulators.In this study,selenium-enriched Lacti-caseibacillus rhamnosus SHA113(Se-SHA113)was evaluated for its potential to alleviate exercise-related fatigue under simulated high-altitude environment(4500 m).Mice were orally administered Se-SHA113(0.12 mg Se/kg body weight)for 4 days before hypobaric hypoxia exposure.Se-SHA113 supplementation prolonged exercise performance and was associated with an increased abundance of a taxon annotated as Lactobacillus reuteri in the intestinal microbiota.An intervention with Lactobacillus reuteri ATCC 53608(LR;1×10^(8) CFU/day for 4 days)was also conducted.LR supplementation reproduced part of the beneficial phenotype.Both interventions improved fatigue-related biomarkers,including preservation of hepatic and muscle glycogen,enhancement of intestinal antioxidant enzyme activities,and elevation of fecal short-chain fatty acids,particularly acetate and butyrate.Reduced colonic oxidative stress was also observed after Se-SHA113 treatment.Overall,Se-SHA113 showed potential to alleviate hypobaric hypoxia-associated fatigue under simulated high-altitude environ-ment.These effects were associated with selective intestinal microbiota remodeling,improved glycogen storage,and strengthened intestinal antioxidant defenses,supporting Se-SHA113 as a promising candidate for nutritional intervention under limited-oxygen conditions.展开更多
Sea cucumber intestine-ovum,a processing byproduct,is rich in bioactive components but remains underutilized.In this study,an enzymatic hydrolysate of sea cucumber intestine-ovum(SCIOH)was prepared and administered by...Sea cucumber intestine-ovum,a processing byproduct,is rich in bioactive components but remains underutilized.In this study,an enzymatic hydrolysate of sea cucumber intestine-ovum(SCIOH)was prepared and administered by oral gavage to mice.SCIOH significantly prolonged pole-climbing time by 98.79%and exhaustive swimming duration by 93.62%compared with the fatigue group.It increased muscle and liver glycogen levels by 119.82%and 129.10%,reduced serum lactate(26.79%),urea nitrogen(57.77%),and suppressed lactate dehydrogenase activity(10.88%).SCIOH also enhanced antioxidant capacity by elevating superoxide dismutase activity in serum(103.18%)and liver(64.40%)while reducing malondialdehyde levels by 48.07%in serum and 23.40%in liver.16S rRNA sequencing analysis revealed that SCIOH enhanced gut microbial diversity and enriched beneficial genera such as Dubosiella,which correlated positively with glycogen storage and negatively with lactate and malondialdehyde.PICRUSt2 predicted the enrichment of Dubosiella-associated pathways in glycol-ysis,the TCA cycle,and amino acid biosynthesis,though a functional validation is required.qPCR analyses showed that SCIOH upregulated mRNA expression of LDHB and MCT1 suggesting an enhanced potential for lactate clearance;it also increased expression of GLUT4 and PDK4,which indicates an improved glucose utili-zation.Furthermore,SCIOH elevated Nrf2 and HO-1 mRNA levels,indicating activation of the Nrf2-mediated antioxidant pathway at the transcriptional level.Together,these findings demonstrate that SCIOH alleviates exercise-induced fatigue by improving energy metabolism,reducing oxidative stress,modulating gut microbiota,and regulating key metabolic and antioxidant genes.These findings support its potential as a marine-derived therapeutic agent for fatigue management.展开更多
