In the central nervous system(CNS), the mTOR pathway regulates diverse aspects of neural development and function. Genetic mutations within the m TOR pathway lead to severe neurodevelopmental disorders, collectively known as “mTORopathies”(Crino, 2020). Objectives:Phosphodiesterase 1A(PDE1A)regulates intracellular cyclic nucleotide signaling and has been implicated in tumor progression,but its clinical relevance and functional role in epithelial ovarian cancer(EOC),p...Objectives:Phosphodiesterase 1A(PDE1A)regulates intracellular cyclic nucleotide signaling and has been implicated in tumor progression,but its clinical relevance and functional role in epithelial ovarian cancer(EOC),particularly in relation to the response to platinum remain unclear.This study aimed to evaluate the clinical significance of PDE1A in EOG and to clarify its functional role in tumor progression and response to platinum-based chemotherapy.Methods:PDE1A mRNA and protein levels were analyzed using public databases,RNA sequencing,and immunohistochemistry.Correlations between PDE1A expression,clinicopathological features,and prognosis were assessed.Functional roles were investigated in ovarian cancer cell lines.Results:PDE1A was significantly overexpressed in EOC tissues compared with that in normal ovarian epithelial tissues.Overexpression correlated with advanced International Federation of Gynecology and Obstetrics(FIGO)stage,poor tumor grade,and reduced response to platinum-based chemotherapy.High PDE1A levels were linked to worse disease-free survival and overall survival,and multivariate analysis confirmed PDE1A as an independent prognostic factor.To elucidate its functional role,we performed in vitro experiments showing that PDE1A knockdown suppressed cell proliferation and colony formation,induced G1 arrest,and downregulatedβ-catenin signaling with reduced cyclin D1 and c-Myc expression.Notably,these inhibitory effects were partially rescued by lithium chloride(LiCl),a Wingless-related integration site(Wnt)/β-catenin activator.Conclusions:In conclusion,our findings identify PDE1A as a Wnt/β-catenin-linked biomarker of tumor progression and platinum resistance in EOC and provide a biological rationale for further investigation of PDE1A-targeted strategies in preclinical models.展开更多

The development of skeletal muscle are complicated processes involving genes responsible for proper muscle morphology,contractility,cell proliferation,differentiation,interactions,migration,and death.The three-dimensi...The development of skeletal muscle are complicated processes involving genes responsible for proper muscle morphology,contractility,cell proliferation,differentiation,interactions,migration,and death.The three-dimensional chromatin architecture of skeletal muscle development has not been studied intensively although dynamic transcriptional regulation during differentiation of muscle cells is one of the most deeply studied processes.The RNA-seq was used to analyze the transcriptome pattern during chicken muscle development across 12 stages.Hi-C was used to build chromatin architectures during four representative stages.Ch IP-seq was conducted to identify enhancers and promoters in these four stages,which are occupied by histone H3K27ac and H3K4me3 peaks.Results show that large-scale genome architecture changes are mostly unidirectional,and coupled by complex on/off dynamic patterns of gene expression.Specifically,we observed 258.30 Mb of the genome undergoing A/B compartment switching.Notable alterations(316.57 Mb)of interaction frequencies within TADs were observed.Substantial aging-associated genes exhibited ascending connectivity with the compartment transition from repressive to active status during muscle development.Some muscle-related gene promoters that interacted with active enhancers during development,and some myopathy/aging-associated genes that were activated in aging muscle were founded.These results provide key insights into skeletal muscle development in vivo,and offer a valuable resource that allows in-depth functional characterization of candidate genes.展开更多
AIM:To investigate whether vaccinia-related kinase 1(VRK1)mediates transforming growth factor-beta2(TGF-β2)-caused epithelial-mesenchymal transition(EMT)and inflammatory responses in retinal pigment epithelial(RPE)ce...AIM:To investigate whether vaccinia-related kinase 1(VRK1)mediates transforming growth factor-beta2(TGF-β2)-caused epithelial-mesenchymal transition(EMT)and inflammatory responses in retinal pigment epithelial(RPE)cells through regulating snail family transcriptional repressor 1(SNAI1),and to validate its role in a proliferative vitreoretinopathy(PVR)mouse model.METHODS:Human RPE cell line ARPE-19 cells were treated with TGF-β2 to construct an EMT model.Western blot detected VRK1 level.The effects of VRK1 on SNAI1 expression and biological behavior of ARPE-19 cells were detected by immunofluorescence,ELISA,Transwell,and scratch assay,and the interaction between VRK1 and SNAI1 was confirmed through immunoprecipitation.A PVR mouse model was constructed,and the effects of VRK1 or/and SNAI1 on retinal damage were assessed by pathologic staining.Inflammatory factors and EMT-related proteins were assessed with ELISA and Western blot.RESULTS:VRK1 was upregulated in ARPE-19 cells after TGF-β2 treatment.Overexpression of VRK1 increased cell viability,promoted cell migration and EMT,and the levels of inflammatory factors.Silencing of VRK1 reversed the above indexes.There was a direct interaction between VRK1 and SNAI1,and overexpresssion SNAI1 weakened the impacts of silencing of VRK1.In PVR mice,silencing of VRK1 ameliorated retinal structural damage,decreased proinflammatory factor levels,and suppressed SNAI1 and mesenchymal marker expression.SNAI1 overexpression antagonized the protective effects of silencing VRK1 and exacerbated EMT and inflammatory responses.CONCLUSION:VRK1 plays a key role in retinal structural and inflammatory damage in PVR mice by regulating SNAI1 and mediating TGF-β2-caused EMT and inflammatory responses in RPE cells.展开更多
Against the backdrop of China s accelerated green development and its pursuit of the"dual carbon"goals,green finance,as a crucial instrument for the rational allocation of resources,plays a significant role ...Against the backdrop of China s accelerated green development and its pursuit of the"dual carbon"goals,green finance,as a crucial instrument for the rational allocation of resources,plays a significant role in promoting agricultural green transformation.Taking Henan Province as a case study,this research employs panel data from 18 prefecture-level cities spanning 2014 to 2023,utilizing dynamic panel models and mediating effect models for empirical analysis.The findings reveal that green finance indirectly fosters agricultural green development by driving agricultural technological progress.This promoting effect is significant,with the mediating effect accounting for 3.67%.Based on these results,policy recommendations are proposed:expanding the coverage of green finance and constructing a diversified product system;comprehensively promoting agricultural technological innovation and mechanization development;and coordinating and integrating green financial resources to facilitate agricultural green transformation tailored to local conditions.展开更多

文摘AIM:To investigate whether vaccinia-related kinase 1(VRK1)mediates transforming growth factor-beta2(TGF-β2)-caused epithelial-mesenchymal transition(EMT)and inflammatory responses in retinal pigment epithelial(RPE)cells through regulating snail family transcriptional repressor 1(SNAI1),and to validate its role in a proliferative vitreoretinopathy(PVR)mouse model.METHODS:Human RPE cell line ARPE-19 cells were treated with TGF-β2 to construct an EMT model.Western blot detected VRK1 level.The effects of VRK1 on SNAI1 expression and biological behavior of ARPE-19 cells were detected by immunofluorescence,ELISA,Transwell,and scratch assay,and the interaction between VRK1 and SNAI1 was confirmed through immunoprecipitation.A PVR mouse model was constructed,and the effects of VRK1 or/and SNAI1 on retinal damage were assessed by pathologic staining.Inflammatory factors and EMT-related proteins were assessed with ELISA and Western blot.RESULTS:VRK1 was upregulated in ARPE-19 cells after TGF-β2 treatment.Overexpression of VRK1 increased cell viability,promoted cell migration and EMT,and the levels of inflammatory factors.Silencing of VRK1 reversed the above indexes.There was a direct interaction between VRK1 and SNAI1,and overexpresssion SNAI1 weakened the impacts of silencing of VRK1.In PVR mice,silencing of VRK1 ameliorated retinal structural damage,decreased proinflammatory factor levels,and suppressed SNAI1 and mesenchymal marker expression.SNAI1 overexpression antagonized the protective effects of silencing VRK1 and exacerbated EMT and inflammatory responses.CONCLUSION:VRK1 plays a key role in retinal structural and inflammatory damage in PVR mice by regulating SNAI1 and mediating TGF-β2-caused EMT and inflammatory responses in RPE cells.

文摘Against the backdrop of China s accelerated green development and its pursuit of the"dual carbon"goals,green finance,as a crucial instrument for the rational allocation of resources,plays a significant role in promoting agricultural green transformation.Taking Henan Province as a case study,this research employs panel data from 18 prefecture-level cities spanning 2014 to 2023,utilizing dynamic panel models and mediating effect models for empirical analysis.The findings reveal that green finance indirectly fosters agricultural green development by driving agricultural technological progress.This promoting effect is significant,with the mediating effect accounting for 3.67%.Based on these results,policy recommendations are proposed:expanding the coverage of green finance and constructing a diversified product system;comprehensively promoting agricultural technological innovation and mechanization development;and coordinating and integrating green financial resources to facilitate agricultural green transformation tailored to local conditions.

文摘N6-methyladenosine(m^(6)A)modification,the most abundant internal modification in messenger RNA(mRNA)and long non-coding RNA(lncRNA),has emerged as a critical epitranscriptomic regulatory mechanism in eukaryotes.While the importance of m^(6)A modification in various biological processes has been recognized,a comprehensive understanding of its diverse roles in plant biology and agricultural applications remains fragmented.This review analyzes recent advances inm^(6)A modification's biological functions in plants.m^(6)A modification plays crucial roles in multiple aspects of plant life,including seed germination,organ development,and reproductive structure formation.Furthermore,m^(6)A has been found to significantly influence plant responses to environmental stresses,including salt,drought,temperature,and heavy metal exposure.We also uncover m^(6)A involvement in important agricultural traits.This review provides insights into the mechanistic understanding of m^(6)A modification in plants and highlights its applications in agricultural improvement,offering a foundation for future research in crop enhancement and stress resistance.
