The anti-ulcer activity ofF. religiosawas evaluated with thehelp of ulcer area and histopatholgical examination. Preliminary phyto-chemical screeningand acute toxicity studies ofF. religiosaalso carried out.Results:Results showed that theextract treatments prevented ulcer area and gastric secretion in a dose-dependent manner.Administration of 2000 mg/kg extract did not show any acute toxicity in albino mice. Preliminaryphytochemical analysis identified the presence of flavonoids in the ethanolic extract ofF. religiosa.Conclusions:The extract is non-toxic even at relatively high concentrations. The anti-ulceractivity is probably due to the presence of flavanoids. 展开更多

OBJECTIVE: Use of cisplatin, a conventional anticancer drug, is restricted because it generates strong hepatotoxicity by accumulating in liver. Therefore its anticancer potential can only be fully exploited if its own toxicity is considerably reduced. Towards this goal, ethanolic extract of the plant, Boldo (Peumus boldus), known for its antihepatotoxic effects, was used simultaneously with cisplatin, to test its ability to reduce cisplatin's cytotoxicity without affecting its anticancer potential. METHODS: The cytotoxicity of Boldo extract (BE) and cisplatin, administered alone and in combination, was determined in three cancer cell lines (A549, HeLa, and HepG2) and in normal liver cells (WRL-68). Drug-DNA interaction, DNA damage, cell cycle, apoptosis, reactive oxygen species (ROS) and mitochondrial membrane potential (MMP, Aψ) were also studied. Hepatotoxicity and antioxidant activity levels were determined by alanine aminotransferase, aspartate aminotransferase, lactate dehydrogenase and glutathione assays in mice. The cytotoxicity of related proteins was tested by Western blotting. RESULTS: Co-administration of BE and cisplatin increased viability of normal cells, but had no effect on the viability of cancer cells. Boldo protected liver from damage and normalized different antioxidant enzyme levels in vivo and also reduced ROS and re-polarized MMP in vitro. Bax and cytochrome c translocation was reduced with caspase 3 down-regulation. Further, a drug- DNA interaction study revealed that BE reduced cisplatin's DNA-binding capacity, resulting in a reduction in DNA damage. CONCLUSION: Results indicated that a low dose of BE could be used beneficially in combination with cisplatin to reduce its toxicity without hampering cisplatin's anticancer effect. These findings siqnify a potential future use of BE in cancer therapy. 展开更多

Blackberry (Rubus fructicosus) is one of the fruit with the highest concentration of anthocyanins;however, its use is limited for making jams, jellies and liqueur, and recently, fruit concentrates in combination with pomegranate, blueberry and grape. One of the main problems with these pigments is their poor stability in solution, mainly in beverages as liqueur and juices, which depends on factors such as chemical structure, pH, temperature, light, water activity, and presence of oxygen. The effect of light on total monomeric anthocyanins content as well as the degradation rates and browning of two blackberry ethanolic extracts is to establish the conditions for storage of liqueur without adding artificial food coloring. The initial content of anthocyanins on extract without storage was 106 mg?l-1. The study assessed the light irradiation effect on the anthocyanins of blackberry ethanolic extract. The anthocyanins degradation followed the second order reaction kinetics with respect to illuminance of the light source. The t1/2 value at high illuminance (3968.30 lx) was 28.20 hours. 展开更多

Hypoglycemic effects were investigated in the ethanolic extract of fruits of Phyllanthus emblica Linn. with long evans rats (100-120 g). Residual gut sucrose content, gut perfusion and disaccharidase activity studies were carried out to assess these activities. The residual gut sucrose content study was carried out to reveal the amount of sucrose remaining in the gastro intestinal tract (GIT) at six different segments. The ethanolic fruit extract of Phyllanthus emblica Linn. significantly increases the residual sucrose content throughout the gut after sucrose ingestion. Study on intestinal enzyme activity showed that the extract caused a significant (p 0.05), dose dependent inhibition of intestinal disaccharidase enzyme activity. Evaluation of the effect of the plant extract on intestinal enzyme activity showed significant inhibition of intestinal disaccharidase activity, suggesting that reduction of sucrose absorption may be partly related to the inhibition of disaccharidase activity in the gut. The gut perfusion analysis showed that the extract significantly reduced intestinal glucose absorption. The results of the present study suggest that ethanolic extract of Phyllanthus emblica Linn. has significant antidiabetic effects in a dose dependent manner and this may be effective in the treatment of diabetes. 展开更多

<strong>Objective: </strong>The present was initiated to study the antibacterial properties of the <i>Crateva adansonii</i> DC extract on germs commonly identified in skin and digestive infecti... <strong>Objective: </strong>The present was initiated to study the antibacterial properties of the <i>Crateva adansonii</i> DC extract on germs commonly identified in skin and digestive infections in Benin as well as the reversion of resistance to these aforementioned germs. <strong>Method: </strong>The bacteria’s sensitivity test to extracts was carried out by the microdilution method in liquid medium as well as the MIC and the reversion of bacterial resistance. For the determination of the MBC, this technique is used coupled with spreading on agar medium. <strong>Results: </strong>The results show an antibacterial activity of the extract with MICs between 0.625 - 5 mg/ml. The CMB of <i>Enterococcus faecalis</i> ATCC 29212 is 2.5 mg/ml while that of Methicillin-resistant <i>Staphylococcus aureus</i> and <i>Staphylococcus epidermidis</i> CIP 8039 is 5 mg/ml. The reversion of bacterial resistance has shown a synergy of action between our extract and conventional antibiotics. 展开更多

Objective: To explore the antidiabetic and the antidiarrhoeal effects of ethanolic extracts of Cynodon dactylon Pers. aerial parts(EECA) in Wistar rats.Methods: To assess the antidiabetic activity of EECA, oral glucose tolerance test(OGTT) model and alloxan induced diabetic test(AIDT) model were performed. The EECA was used at the doses of 2 g/kg, 1 g/kg and 500 mg/kg body weight in OGTT model and 1.5 g/kg was used for AIDT model. Castor oil-induced diarrhoeal model and gastrointestinal motility test with barium sulphate milk model were performed for evaluating the antidiarrhoeal effects at doses of 1 g/kg, 750 mg/kg respectively.Results: The dose 2 g/kg in OGTT and 1.5 g/kg in AIDT model blood glucose levels decreased significantly(P < 0.01) in Wistar rats that showed antidiabetic effect of EECA. After administration of EECA at the dose of 1 g/kg, the extract showed significant(P < 0.05) antidiarrhoeal activity in castor oil-induced diarrhoeal model. The results were also significant(P < 0.01) in barium sulphate milk model for the same dose by using above mentioned animals.Conclusions: It is concluded that EECA contains both antidiabetic and the antidiarrhoeal properties. 展开更多

Objective:To select the appropriate concentrations of methanolic pomegranate extract supplemented in rams’semen extender for obtaining the best-cryopreserved semen quality.Methods:Tris-based semen extender was supplemented with 0.0,0.40,0.48,and 0.56 mg/mL pomegranate peel methanolic extract to extend semen collected from five native rams twice weekly for two months(n=80).Pooled(n=16)post-thaw semen characteristics were determined.Thawed seminal plasma of all supplemented and control groups were used to measure malondialdehyde(MDA),nitric oxide(NO),glutathione peroxidase(GPx),superoxide dismutase(SOD),ascorbic acid,zinc,copper,total cholesterol,low-density lipoproteins(LDL),lactate dehydrogenase(LDH),and alkaline phosphatase(ALP).Results:The supplementation of Tris-based semen extender with 0.48 mg/mL semen extender resulted in the highest post-thaw sperm total motility(P<0.001),sperm progressive motility(P<0.001),live sperm(P<0.001),sperm plasma membrane integrity(P<0.001),acrosome integrity(P<0.001),SOD(P<0.05),zinc(P<0.001),total cholesterol(P<0.001),and LDL(P<0.001)with the lowest percentage of abnormal sperm morphology(P<0.001),the lowest lipid peroxidation(MDA,P<0.01),ascorbic acid(P>0.05),and LDH(P>0.05).Conclusions:Pomegranate peel methanolic extract 0.48 mg/mL supplemented to Tris-based semen extender of rams is the best enrichment in preserving the sperm post-thaw characteristics via improving biochemical profiles and antioxidant capacity. 展开更多

The antioxidant activity of their ethanolic extracts, as well as their effects on the oxidation of edible corn oil during accelerated ageing, was studied. Maceration of taxonomically identified commercial vegetable samples, previously peeled and thinly sliced, was carried out at ambient temperature, out of direct light, with occasional agitation and ultrasound, employing 95% ethyl alcohol as the extraction solvent, allowing them to stand for 7 days. The total polyphenolic contents were determined on the filtrated extracts using the Folin-Ciocalteau method. The antioxidant activity was evaluated on emulsions of ethanolic extracts of onion prepared in edible commercial corn oil, using sorbitan monooleate as emulsifying agent. The peroxide values were analyzed using the iodometric method;oxidation induction times were obtained from the peroxide evolution graphs, using the tangent method. Oil samples emulsified with ethanolic onion extracts showed an extension of the induction period. A 7 day ageing study at 45℃ was additionally performed to determine the conjugated dienes on pure commercial corn oil and its emulsions by visible spectrophotometry. The spectral analysis showed an increase of the measured absorbances in oil samples without additives and no change for the oils emulsified with onion extract. An increasing of diene values was observed for corn oil without additives during ageing;no changes in the value were observed in oils emulsified with onion extracts. 展开更多

The present study was undertaken to investigate the effect of the ethanolic extract of Syzygium aromaticum to albino rats. Forty eight Albino rats were employed to test the safety and the anti-inflammatory effect of the extract. Safety of the extract was examined on experimental animal’s model at three dose levels of the extract orally in daily doses for three weeks. Effects of S. aromaticum on rats revealed no significant effect on biochemical or haematological parameters. The anti-inflammatory effect of the extract was tested in four equal groups;groups 1 and 2 were treated with 250 and 500 mg/kg of the extract, respectively, group 3 was treated with indomethacine and group 4 was the untreated control. Carrageenan was used as an acute form inducer of inflammation. Indomethacine, the non-steroidal anti-inflammatory drugs (NSAIDs), was used as a reference compound. Oedema size was monitored at the 1st, 2nd, 4th, 6th and after 24 hours. The ethanolic extract of S. aromaticum showed significant (P 0.001) decreased in the oedema size at efficacy rates of 79.41%, 82.39% and 63.92% for the dose, 500 mg/kg body weight at the 2nd, 4th and 6th hour respectively higher than that produced by indomethacine. 展开更多

Nigella sativa, belonging to the Ranunculacea family, is a versatile phytochemical mine bestowed with multi-dimensional medicinal effects. The current study was performed to investigate the chemopreventive efficacy, if any, of the ethanolic extract of indigenous Nigella sativa seeds (ENS) and to evaluate its potentials on tumor progression during the initiation and post initiation phases of Dimethylhydrazine-induced colon carcinogenesis in a rat model. ENS treatment during the initiation phase unveiled chemopreventive effect manifested by significant reductions in tumor incidence, multiplicity and tumor volumes. Histopathological findings and modified Duke’s classification of tumors provided evidence that ENS, administered in the initiation phase, is capable of delaying progression, restricting invasion and attenuating aggressiveness of colon tumors. These results imply that ENS may be a promising candidate in the primary prevention of colon cancer. On the contrary, we demonstrated that ENS lacked chemopreventive and tumorigenesis inhibitory effects in the post initiation phase. We speculate that the chemopreventive effect of ENS might be due to the synergistic actions of various constituents present in the extract. However, extensive studies are warranted and more efforts need to be dedicated to fractionate, analyze and to further appraise the anticancer effect of ENS before any definitive conclusions can be drawn. 展开更多

Plants are sources of medicinal compounds,and they play a crucial role in human health maintenance.Abrus precatorius is one of the important medicinal plants that have been alleged for their medicinal properties.This research unraveled the pharmacological effect of ethanolic extract of Abrus precatorius on lipid peroxidation,liver parameters,and Na^(+)/K^(+)-ATPase activity in HgCl_(2) treated wistar rats.Twenty-four(24)albino wistar rats weighing between 150-200 g were distributed into four groups of 6 animals each.Group A(control)received normal saline(0.9% NaCl),group B received 400 mg/kg of the extract only,group C received 4 mg/kg HgCl_(2) only,and group D received 400 mg/kg of extract+4 mg/kg of HgCl_(2).The treatment lasted for two weeks,and the animals were sacrificed on the 15th day.The blood,brains and livers were collected and used for assay of lipid peroxidation,liver function,and sodium pump activity.The results of liver function test revealed an elevated(P<0.05)level of serum aspartate transaminase,alanine transaminase,alkaline phosphatase,and total bilirubin in the group that received HgCl_(2) only(group C)when compared with the normal control(group A)that received normal saline only.However,the administration of extract in group D led to a marked(P<0.05)reduction in the activities of these enzymes and the level of total bilirubin when compared to the negative control(group C).On the contrary,HgCl_(2) caused a significant(P<0.05)reduction in serum total protein and albumin levels,but the extract reversed the effect of HgCl_(2) by elevating their concentrations.Nonetheless,the effect elicited by this extract is comparable to group A which received normal saline.Moreso,the result of lipid peroxidation revealed that HgCl_(2) treatment caused a marked(P<0.05)increase in the formation of lipid peroxidation adducts in both liver and brain homogenates in group C.On the contrary,administration of Abrus precatorius extract profoundly(P<0.05)inhibited HgCl_(2)-induced lipid peroxidation in group D.In addition,HgCl_(2) inhibited the activity of cerebral Na^(+)/K^(+)-ATPase,but the extract restored normalcy by increasing the activity of the enzyme in group D.Consequently,the results obtained justify the traditional use of Abrus precatorius and suggest that Abrus precatorius leaves may be used for management of liver diseases,oxidative stress-linked diseases and some neurodegenerative ailments. 展开更多

Objective:To investigate the effect of the ethanolic extract of Rosa laevigata Michx.fruit on rats with mesangial proliferative glomerulonephritis based on the NLRP3 inflammasome pathway.Methods:Thirty Wistar rats were divided into three groups,a blank control group,a diabetic nephropathy(DN)model group,and an ethanolic extract intervention group,according to the random number table method,with 10 rats in each group.One day before the experiment,basic feeding was initiated for all the rats;the changes in activity and weight of each group of rats were observed and recorded after 7 d,and a rat model of renal function injury was established after 1 d.Results:Compared with the control group,the model group had significantly higher kidney/body ratio,24 h urine protein,serum creatinine(SCr),blood urea nitrogen(BUN),glomerular mesangial cell(GMC)count,and extracellular matrix(ECM)positive area ratio(P<0.05);the same indicators were significantly lower in the intervention group than in the model group(P<0.05).The NLRP3 inflammasome pathway in renal intrinsic cells was activated in the intervention group.The overactivation of NLRP3 inflammasome is known to promote interleukin(IL)-1βrelease,which was inhibited in the intervention group.Conclusion:The ethanolic extract of Rosa laevigata Michx.fruit has a protective effect on renal intrinsic cells and may be related to NLRP3 inflammasome pathway,suggesting that the fruit of Rosa laevigata Michx.has a potential role in protecting renal intrinsic cells from inflammatory damage.NLRP3 inflammasomes are involved in the development of various chronic inflammatory diseases,such as acute and chronic glomerulonephritis and renal fibrosis. 展开更多

Alcoholic liver injury is a liver disease caused by excessive alcohol consumption,which can lead to chronic liver disease death.Solanum Nigrum Linn taste bitter,cold,has the effect of clearing heat and detoxification,promoting blood and detumescence.Solanum Nigrum Linn fruit contains a variety of antioxidant enzymes,can remove the body produced by aerobic metabolism harmful substances.In this paper,a model of alcohol-induced liver injury in C57BL/6 mice was established to evaluate the protective effect of Solanum Nigrum Linn green fruit(SNGF)ethanolic extract on alcohol-induced liver injury.H&E staining and oil red O(ORO)staining showed that hepatic lobules were clearly demarcated,vacuoles were significantly reduced and lipid droplets were reduced in SNGF ethanolic extract treatment group.Serum levels of TC,TG,LDH,TBA,AKP,ALT and AST were decreased in the SNGF ethanolic extract treatment group,and SNGF ethanolic extract could clear reactive oxygen species(ROS)in time.MDA content was signifi cantly decreased after SNGF ethanolic extract treatment,while superoxide dismutase(SOD)and GSH-Px contents were increased after SNGF ethanolic extract treatment.These results suggest that SNGF ethanolic extract has a protective effect on alcohol-induced liver injury. 展开更多

Background:Artemisia vulgaris,a medicinal aromatic plant,is widely used as a food item,tonic pharmaceutical,and cosmetic industry additive owing to its antibacterial,antihypertensive,hepatoprotective,antioxidant,and antispasmodic properties.But the effect of different geographic locations on the chemical composition and bioactivities of its extracts is unclear.Methods:Biological activities of essential oils and ethanol extracts of three varieties of Artemisia vulgaris leaves,which are grown in Shanxi province China,were studied.Results:Gas chromatography-mass spectrometry analysis revealed that the main components of essential oils were terpenes and ketones.Essential oils and ethanol extract of Artemisia vulgaris leaves possessed good antioxidant activities,and their half maximal inhibitory concentrations determined using 1,1-diphenyl-2-picrylhydrazyl and 2,2’-azinobis-(3-ethylbenzthiazoline-6-sulphonate)assays were 57.0 and 22.9μg/mL,respectively.The essential oils also exhibited remarkable antibacterial activity against three foodborne pathogenic bacterial strains.The ethanol extract presented a high anticancer activity against the MGC-803 human gastric cancer cell line.Conclusion:These biological activities were well correlated with the composition of the extract and EOs,which in turn is affected by the genetic composition of Artemisia vulgaris and geographic location and diverse climatic condition under which it is grown.These findings demonstrate the remarkable potential of Artemisia vulgaris as a valuable source of antioxidant,antibacterial,and anticancer agents. 展开更多

Due to the spontaneity of electron transfer of anions, it has been found clinically that ibuprofen drug cures cancers of colon, protostate, lung and breast. The breast cancer treatment of Matthew Gdovin group in two hours by injection of ethanolic solution of nitrobenzaldehyde in the breast tumor in presence of uv-irradiation has been studied from TD-DFT point of view;the excited states of these molecules in presence of uv-irradiation act as electron donors to the cancerous cells to compensate the electron deficiency. Finally, it has been concluded that the electron transfer is the main cause of the breast cancer treatment which is the most aggressive type of cancers and is one of the hardest to treat. 展开更多

Methods: The free radical scavenging activity was evaluated by in vitro methods like reducing power assay, total antioxidant activity, 2,2-diphenyl-l-picrylhydrazyl (DPPH) reduction, superoxide radical scavenging activity, 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS*) scavenging activity, hydroxyl radical scavenging assay, and nitric oxide radical scavenging assay, which were studied by using ascorbic acid as standard. Results: The extract showed significant activities in all antioxidant assays compared with the reference antioxidant ascorbic acid. The total antioxidant activity as well as the reducing power was also found to increase in a dose-dependent manner. Conclusion: Evolvulus alsinoides may act as a chemopreventive agent, providing antioxidant properties and offering effective protection from free radicals. 展开更多

Objective: To evaluate the effect of ethanolic extract of Pericampylus glaucus (Lam) Merr (P. glaucus) on biochemical parameters in high fats diets treated rats. Methods: The effect of P. glaucus extract on biochemical parameters was studied in male Sprague Dawley rats. The high-fat diets along with standard normal laboratory rat's diets were given for a period of 28 days along with ethanolic extract of P. glaucus and standard drug. The standard group was treated with simvastatin at dose of 80 mg/kg and the tested groups were treated with ethanol extract of P. glaucus at 400, 600 and 800 mg/kg daily for 28 days of the experiment. The weight of animals in each group was measured weekly and the effect of extract and standard on serum lipids profile was evaluated at the end of the study. Results: The results showed that the ethanolic extract of P. glaucus at different doses of 400, 600 and 800 mg/kg and standard drug induced a significant reduction in the body weight (P< 0.001), total cholesterol, triglycerides (TG), low density lipoproteins (LDL) and uric acid levels and increase (P < 0.001) in high density lipoproteins (HDL) as compared to untreated hyperlipidemic group. Conclusions: The present work indicates that the ethanolic extract significantly suppressed the biochemical parameters in high fats diet treated animals, suggesting the antihyperlipidemic potential of P. glaucus. 展开更多

Objective:To compare the antibacterial activity and antiseptic effect of ethanol extract of Peganum harmala L.(P.harmala)seeds and Betadine on eight reference strains of Streptococcus pneumoniae,Salmonella typhimurium,Escherichia coli,Corynebacterium pseudotuberculosis,Klebsiella pneumoniae,Staphylococcus aureus,Micrococcus luteus which are known to cause different types of skin infections,and Corynebacterium pseudotuberculosis isolated from an abscess on horse's neck.Methods:The antibacterial activity was assessed using a disc diffusion method.The minimum inhibitory concentration was tested by serial dilution method and the percentage of bacterial growth inhibition by absorbance microplate reader device.The minimum bactericidal concentration was then recorded.Clinical outcomes were obtained through washed up healing time of longitudinal and surface skin on the back of 16 rats with concentrations of 10,50,100,150 and 500 mg/mL of P.harmala extract.Results:The results of in vitro experiments showed that the lowest minimum inhibitory concentration(0.68-1.3 mg/mL)and minimum bactericidal concentration(1.3-5 mg/mL)values were observed in the ethanol extract of P.harmala seeds.Also the results of in vivo experiment showed that wound healing in the concentration of 50 mg/mL of this plant extract was better and quicker than Betadine.Conclusions:This study confirmed that ethanol extract of P.harmala has appropriate effect on the microorganisms and the healing of skin wounds in comparison with Betadine. 展开更多

Mesenchymal stem cell-derived extracellular vesicles have emerged as a promising form of regenerative and immunomodulatory therapy;indeed,micro(mi)RNAs contained within mesenchymal stem cell-derived extracellular vesicles modulate target gene expression and impact disease-associated pathways.Chronic alcohol consumption leads to neuroinflammation,brain damage,and impaired cognition.Evidence indicates that females are more vulnerable to alcohol-induced damage than males.While mesenchymal stem cell-derived extracellular vesicles have been studied in various neuroinflammatory conditions,their potential to counteract alcohol-induced brain damage remains unclear.In this study,we investigated whether repeated intravenous administration of mesenchymal stem cell-derived extracellular vesicles could ameliorate neuroinflammation and behavioral impairment induced by chronic alcohol consumption in female mice.Mesenchymal stem cell-derived extracellular vesicles diminished the increased binding of a micro-positron emission tomography tracer(^(18)F-FDG)when analyzing whole-brain 3D images and brain coronal sections of ethanol-treated mice.Mesenchymal stem cell-derived extracellular vesicle administration protected against ethanol-induced proinflammatory gene upregulation,cognitive dysfunction,and the conditioned rewarding effects of cocaine.MiRNA sequencing data from mesenchymal stem cell-derived extracellular vesicles revealed the elevated expression of extracellular vesicle-derived miR-483-5p and miR-140-3p in the brains of ethanol-treated female mice following mesenchymal stem cell-derived extracellular vesicle administration.In addition,mesenchymal stem cell-derived extracellular vesicles modulated the expression of pro-inflammatory-related miRNA target genes(e.g.,Socs3,Tnf,Mtor,and Atf6)in the brains of ethanol-treated female mice.These results suggest that mesenchymal stem cell-derived extracellular vesicles could function as a neuroprotective therapy to ameliorate the neuroinflammation,cognitive dysfunction,and conditioned rewarding effects of cocaine associated with chronic alcohol consumption. 展开更多
