Pseudomonas aeruginosa,recognized for its biofilm production and secretion of virulence factors,posing a severe threat in areas such as clinical infections,food contamination,and marine biofouling.To address this,a ne...Pseudomonas aeruginosa,recognized for its biofilm production and secretion of virulence factors,posing a severe threat in areas such as clinical infections,food contamination,and marine biofouling.To address this,a new type of zinc-binding peptide(CSSP-Zn)was prepared from crimson snapper scales peptides(CSSP)and goslarite,and its antibacterial and anti-quorum-sensing activities toward P.aeruginosa PAO1 were exploited.Results indicated that CSSP-Zn induced planktonic strain PAO1 membrane injury via inhibiting expression levels of cell integrity genes,targeting microbial-specific membrane constituents,disrupting proton motive force,and causing metabolic disturbances.Meanwhile,CSSP-Zn decreased virulence factors pyocyanin,protease and rhamnolipid secretion,while considerably inhibiting quorum sensing-related genes(las,pqs and rhl)expression and decreasing bacterial abundance and pathogenicity in fish models.Moreover,CSSP-Zn not only effectively hindered biofilm formation but also disassembled preformed ones,thus disrupting biofilm topology.Taken together,utilizing food byproducts to obtain CSSP-Zn could help recycle food resources and provide insight into controlling planktonic and biofilm strain PAO1 contamination.展开更多
Perfluorobutanesulfonate(PFBS)is an emerging pollutant capable of potently disrupting the sex and thyroid endocrine systems of teleosts.However,the hypoxic and temporal variation in PFBS endocrine disrupting toxicity ...Perfluorobutanesulfonate(PFBS)is an emerging pollutant capable of potently disrupting the sex and thyroid endocrine systems of teleosts.However,the hypoxic and temporal variation in PFBS endocrine disrupting toxicity remain largely unknown.In the present study,adult marine medaka were exposed to environmentally realistic concentrations of PFBS(0 and 10μg/L)under normoxia or hypoxia conditions for 7 days,aiming to explore the interactive behavior between PFBS and hypoxia.In addition,PFBS singular exposure was extended till 21days under normoxia to elucidate the time-course progression in PFBS toxicity.The results showed that hypoxia inhibited the growth and caused the suspension of egg spawn regardless of PFBS exposure.With regard to the sex endocrine system,7-day PFBS exposure led to an acute stimulation of transcriptional profiles in females,which,subsequently,recovered after the 21-day exposure.The potency of hypoxia to disturb the sex hormones was much stronger than PFBS.A remarkable increase in estradiol concentration was noted in medaka blood after hypoxia exposure.Changes in sex endocrinology of coexposed fish were largely determined by hypoxia,which drove the formation of an estrogenic environment.PFBS further enhanced the endocrine disrupting effects of hypoxia.However,the hepatic synthesis of vitellogenin and choriogenin,two commonly used sensitive biomarkers of estrogenic activity,failed to initiate in response to the estrogen stimulus.Compared to sex endocrine system,disturbances in thyroidal axis by PFBS or hypoxia were relatively mild.Overall,the present findings will advance our toxicological understanding about PFBS pollutant under the interference of hypoxia.展开更多
Tetrabromobisphenol A(TBBPA)is a widely used brominated flame retardant.There is evidence showing that TBBPA can exert thyroid disrupting effects in mammals,but different results were also reported,along with inconsis...Tetrabromobisphenol A(TBBPA)is a widely used brominated flame retardant.There is evidence showing that TBBPA can exert thyroid disrupting effects in mammals,but different results were also reported,along with inconsistent reports regarding its neurotoxicity.Here,we investigated thyroid disrupting effects and neurotoxicity of TBBPA(5,50,500μg/(kg·day))to male mice following maternal and direct exposure through drinking water,with the antithyroid drug propylthiouracil(PTU)as the positive control.On postnatal day(PND)15,we expectedly observed severe thyroid compensatory hyperplasia and cerebellar developmental retardation in PTU-treated pups.The highest dose of TBBPA also caused thyroid histological alteration but had no effects on cerebellar development in terms of Purkinje cell morphology and the thickness of the internal granular layer and the molecular layer of the cerebellum.During puberty and adulthood,the thyroid morphological alterations became more pronounced in the TBBPA-treated animals,accompanied by decreased serum thyroid hormone levels.Furthermore,the 50 and 500μg/(kg·day)TBBPA groups showed a significant decrease in the serum level of serotonin,a neurotransmitter associated with anxiety behaviors.Correspondingly,the highest dose group displayed anxiety-like behaviors in the elevated plus-maze test on PND 35,but this neurobehavioral alteration disappeared on PND 56.Moreover,no changes in neurobehavioral parameters tested were found in TBBPAtreated animals at puberty and adulthood.Altogether,all observations show that TBBPA can exert thyroid disrupting effects but has little overt impact on brain development and neurobehaviors in mice,suggesting that thyroid disruption does not necessarily cause overtly adverse neurodevelopmental outcomes.展开更多

Nowadays, ...Environmental endocrine disrupting chemicals (EDCs), commonly found in the environment, come from industry and agriculture, including pesticides, fungicides, insecticides, herbicides, and other chemicals. Nowadays, more and more EDCs were released into the environment. EDCs go into water body via atmosphere sedi-mentation, surface runoff, soil eluviation, etc., so water body becomes the main place for existing. In order to attract scientific and public attention worldwide and to prevent EDCs pol ution, in this study we reviewed the classification of EDCs and their concentrations in natural water bodies, drinking water sources and water plants, and the reproductive toxicity of EDCs to fish were reviewed. EDCs could disturb the endocrine system and make reproductive organs and reproduction abnor-mal, resulting in fertility descending, reproduction function damage, community quan-tity decrease and even species extinction. In addition, EDCs could disrupt the homeostasis maintained by hormones, which would result in defects of neural de-velopment and abnormalities of the endocrine and reproductive systems. The exact molecular mechanisms have not been completely reported, but researches have suggested that multiple mechanisms were involved in the action of EDCs. Although there have been researches on the biohazard of EDCs, there stil exist problems of weakness in fundamental researches, difficulties in recognizing and identifying EDCs and high cost, which restraint the knowledge on them.展开更多
Detection of estrogenic disrupting compounds(EDCs) in drinking waters around China has led to rising concerns about health risks associated with these compounds.There is,however,a paucity of studies on the occurrenc...Detection of estrogenic disrupting compounds(EDCs) in drinking waters around China has led to rising concerns about health risks associated with these compounds.There is,however,a paucity of studies on the occurrence and identification of the main compounds responsible for this pollution in the source waters.To fill this void,we screened estrogenic activities of 23 source water samples from six main river systems in China,using a recombinant two-hybrid yeast assay.All sample extracts induced significant estrogenic activity,with E2 equivalents(EEQ) of raw water ranging from 0.08 to 2.40 ng/L.Additionally,16 samples were selected for chemical analysis by gas chromatography-mass spectrometry.The EDCs of most concern,including estrone(E1),17β-estradiol(E2),17α-ethinylestradiol(EE2),estriol(E3),diethylstilbestrol(DES),estradiol valerate(EV),4-t-octylphenol(4-t-OP),4-nonylphenols(4-NP) and bisphenol A(BPA),were determined at concentrations of up to 2.98,1.07,2.67,4.37,2.52,1.96,89.52,280.19 and 710.65 ng/L,respectively.Causality analysis,involving comparison of EEQ values from yeast assay and chemical analysis identified E2,EE2 and 4-NP as the main responsible compounds,accounting for the whole estrogenic activities(39.74% to 96.68%).The proposed approach using both chemical analysis and yeast assay could be used for the identification and evaluation of EDCs in source waters of China.展开更多
Solid malignancies have to develop their own blood supply for their aggressive growth and metastasis;a process known as tumor angiogenesis.Angiogenesis is largely involved in tumor survival,progression and spread,whic...Solid malignancies have to develop their own blood supply for their aggressive growth and metastasis;a process known as tumor angiogenesis.Angiogenesis is largely involved in tumor survival,progression and spread,which are known to be significantly attributed to treatment failures.Over the past decades,efforts have been made to understand the difference between nor-mal and tumor vessels.It has been demonstrated that tumor vasculature is structurally immature with chaotic and leaky phenotypes,which provides opportunities for developing novel anticancer strategies.Targeting tumor vasculature is not only a unique therapeutic interven-tion to starve neoplastic cells,but also enhances the efficacy of conventional cancer treatments.Vascular dis-rupting agents(VDAs) have been developed to disrupt the already existing neovasculature in actively growing tumors,cause catastrophic vascular shutdown within short time,and induce secondary tumor necrosis.VDAs are cytostatic;they can only inhibit tumor growth,but not eradicate the tumor.This novel drug mechanism has urged us to develop multiparametric imaging biomark-ers to monitor early hemodynamic alterations,cellular dysfunctions and metabolic impairments before tumor dimensional changes can be detected.In this article,we review the characteristics of tumor vessels,tubulin-destabilizing mechanisms of VDAs,and in vivo effects of the VDAs that have been mostly studied in preclinical studies and clinical trials.We also compare the differ-ent tumor models adopted in the preclinical studies on VDAs.Multiparametric imaging biomarkers,mainly diffu-sion-weighted imaging and dynamic contrast-enhanced imaging from magnetic resonance imaging,are evalu-ated for their potential as morphological and functional imaging biomarkers for monitoring therapeutic effects of VDAs.展开更多

Ozonation and ultrafiltration(UF) are powerful technologies...Endocrine disrupting chemicals(EDCs) in the secondary effluent discharged from wastewater treatment plants are of great concern when water reuse is intended. Ozonation and ultrafiltration(UF) are powerful technologies reported to eliminate EDCs. Due to the importance of effluent organic matters(EfOMs) in secondary effluent, the effects of three kinds of EfOM on the treatment of five EDCs using ozonation and UF were investigated. The three kinds of EfOM studied were humic acid sodium salt(NaAH), bovine serum albumin(BSA)and sodium alginate(NaAg); and the five EDCs were estrone, 17β-estradiol, estriol, 17α-ethynyl estradiol and bisphenol A. The results showed that EfOM accelerated the decay rate of ozone and inhibited the degradation efficiency of EDCs by ozonation in the order NaAH > BSA > NaAg.The ultraviolet absorbance at 280 nm(UVA_(280)) has potential for use as a surrogate indicator to assess EDC removal via ozonation without conducting difficult EDC analyses. When the decline in UVA_(280) exceeded 18%, the five EDCs had been completely removed. The UF behavior of NaAH,BSA and NaAg was found to follow the cake filtration law. The fouling potential of EfOM followed the order NaAg > NaAH > BSA; while EfOM on the membrane surface enhanced EDC removal in the order NaAH > BSA > NaAg. The mean retention rate of the membrane was increased by 24%, 10% and 8%, respectively. The properties of EDCs and EfOM cakes both influenced the EDC removal rates due to adsorption, size exclusion and charge attraction.展开更多

Methods A battery of in vitro recombinant yeast bioassays incorporated with exogenous metabolic activation system (rat liver preparation, S9 mix) was conducted to assess the estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PR), and thyroid receptor (TR) ant/agonistic activities of effluents collected from Datansha WWTP. Results The indirect estrogenic, anti‐androgenic, anti‐progesteronic, and anti‐thyroidic activities were observed in the influent. The removal efficiencies of EDCs were above 74%, suggesting that the present wastewater treatment processes were good enough to remove most of these indirect endocrine disrupting chemicals. Conclusion The incorporation of exogenous metabolic capacity into the test system was valid for the study of indirect effects on ER, AR, PR, and TR.展开更多
Endocrine disrupting compounds(EDCs) and pharmaceuticals pose a challenge for water and wastewater treatment because they exist at very low concentrations in the presence of substances at much higher concentrations ...Endocrine disrupting compounds(EDCs) and pharmaceuticals pose a challenge for water and wastewater treatment because they exist at very low concentrations in the presence of substances at much higher concentrations competing for adsorption sites.Sub-micron sized resin particles(approximately 300 nm in diameter)(SMR) were tested to evaluate their potential as a treatment for EDCs including:17-β estradiol(E2),17-α ethinylestradiol(EE2),estrone(E1),bisphenol A(BPA),and diethylstilbestrol(DES) as well as 12 pharmaceuticals.SMR were able to remove 98%of spiked E2,80%of EE2,87%of BPA,and up to 97%of DES from water.For a 0.5 ppm mixture of E2,EE2,E1,BPA and DES,the minimum removal was24%(E2) and the maximum was 49%(DES).They were also able to remove the pharmaceuticals from deionized water and wastewater.Overall,SMR are a promising advanced treatment for removal of both EDCs and pharmaceuticals.展开更多
Obesity and associated metabolic disorders represent a major societal challenge in health and quality of life with large psychological consequences in addition to physical disabilities. They are also one of the leadin...Obesity and associated metabolic disorders represent a major societal challenge in health and quality of life with large psychological consequences in addition to physical disabilities. They are also one of the leading causes of morbidity and mortality. Although, different etiologic factors including excessive food intake and reduced physical activity have been well identified, they cannot explain the kinetics of epidemic evolution of obesity and diabetes with prevalence rates reaching pandemic proportions. Interestingly, convincing data have shown that environmental pollutants, specifically those endowed with endocrine disrupting activities, could contribute to the etiology of these multifactorial metabolic disorders. Within this review, we will recapitulate characteristics of endocrine disruption. We will demonstrate that metabolic disorders could originate from endocrine disruption with a particular focus on convincing data from the literature. Eventually, we will present how handling an original mouse model of chronic exposition to a mixture of pollutants allowed demonstrating that a mixture of pollutants each at doses beyond their active dose could induce substantial deleterious effects on several metabolic end-points. This proof-of-concept study, as well as other studies on mixtures of pollutants, stresses the needs for revisiting the current threshold model used in risk assessment which does not take into account potential effects of mixtures containing pollutants at environmental doses, e.g., the real life exposure. Certainly, more studies are necessary to better determine the nature of the chemicals to which humans are exposed and at which level, and their health impact. As well, research studies on substitute products are essential to identify harmless molecules.展开更多

The dynamic adsorption of four representative EDCs, namely estriol(E3), 17β-estradiol(E2), 17α-ethynylestradiol(EE2), and 4-nonylphenol(4-NP) in a microfiltration system was investigated using the Thomas' model.The product of the equilibrium constant and the total adsorption capacity of the membrane,Ka, for E3, E2, EE2, and 4-NP were 4.91, 9.78, 15.6, and 826, respectively, strongly correlating with the compound octanol–water partition coefficient(KOW). Adsorption appeared to be enhanced when organic fouling formed on the surface of membrane, indicating the role of an additional adsorption column for EDCs acted by a fouling layer in microfiltration. Results of a comparison between the Ka values for clean membrane and fouled membrane illustrated that the significant contribution made by fouling layers may be attributed to the foulant layer's hydrophobicity(in the case of calcium humate layer) and thickness(in the case of calcium alginate layer). This study provided a novel perspective to quantitatively analyze the dynamic adsorption behavior of trace pollutants in membrane process.展开更多
Endocrine disrupting chemical(EDC) pollution in river-based artificial groundwater recharge using reclaimed municipal wastewater poses a potential threat to groundwater-based drinking water supplies in Beijing, Chin...Endocrine disrupting chemical(EDC) pollution in river-based artificial groundwater recharge using reclaimed municipal wastewater poses a potential threat to groundwater-based drinking water supplies in Beijing, China. Lab-scale leaching column experiments simulating recharge were conducted to study the adsorption, biodegradation, and transport characteristics of three selected EDCs: 17β-estradiol(E2), 17α-ethinylestradiol(EE2) and bisphenol A(BPA). The three recharge columns were operated under the conditions of continual sterilization recharge(CSR), continual recharge(CR), and wetting and drying alternative recharge(WDAR). The results showed that the attenuation effect of the EDCs was in the order of WDAR 〉 CR 〉 CSR system and E2 〉 EE2 〉 BPA, which followed first-order kinetics. The EDC attenuation rate constants were 0.0783, 0.0505, and 0.0479 m-1 for E2, EE2 and BPA in the CR system, respectively. The removal rates of E2, EE2, and BPA in the CR system were 98%, 96% and 92%, which mainly depended on biodegradation and were affected by water temperature.In the CR system, the concentrations of BPA, EE2, and E2 in soil were 4, 6 and 10 times higher than in the WDAR system, respectively. According to the DGGE fingerprints, the bacterial community in the bottom layer was more diverse than in the upper layer, which was related to the EDC concentrations in the water-soil system. The dominant group was found to be proteobacteria, including Betaproteobacteria and Alphaproteobacteria, suggesting that these microbes might play an important role in EDC degradation.展开更多

The rational for the study was to assess the levels of endocrine disrupting trace metals in River Samre. The levels of Mercury ranged from 0.01 to 0.02 mg/l (mean of 0.006 mg/l), whiles cadmium levels ranged from 0.00...The rational for the study was to assess the levels of endocrine disrupting trace metals in River Samre. The levels of Mercury ranged from 0.01 to 0.02 mg/l (mean of 0.006 mg/l), whiles cadmium levels ranged from 0.002 to 0.011 mg/l (mean of 0.01 mg/l). The high levels of Hg and Cd may have adverse effects on the endocrine system of inhabitants who drink directly from the river without treatment. High levels of mercury and cadmium might be caused by the activities of a Timber and Plywood Company located close to the river and the underlying bedrocks of the area exposed as a result of human activities such as farming. The concentration of lead was below detection limit ( mg/l) but that of Arsenic ranged from 0.001 to 0.007 mg/l (mean of 0.005). Health risk assessment conducted shows that the risk associated with exposure to these metals for now are low. Continuous water quality monitoring is recommended to help protect the resource and also to safeguard human health.展开更多

Diisononylphthalate (DINP), diethylhexylphthalate (DEHP), dibutylphthalate (DBP), diisododecylphthalate (DIDP) and 4-nonylphenol (4-NP) were oestrogenic in the yeast estrogen screen (YES) assay and potently oestrogenic in the MVLN and E-SCREEN assays at environmentally relevant concentrations. DINP and 4-NP were mutagenic in the Ames assay and also induced significant levels of unscheduled DNA synthesis and DNA strand breakage. Significant induction in the percentage of cells containing micronuclei was observed after treatment with DINP, DEHP and 4-NP. In addition, sewage effluents from sewage treatment plants (STPs) in the Border, Midlands and Western (BMW) region of Ireland were significantly oestrogenic in the YES assay. Moreover, analysis of levels of phthalates and alkylphenol identified in Irish rivers receiving treated effluent showed potent oestrogenicity in the YES assay. The proliferative and genotoxic ability of the phthalates and alkylphenol, and the oestrogenicity of the treated effluents reported here, is significant as these EDCs and EDCs within the effluent may play a role in the etiology of human abnormalities.展开更多

The initiation stage of carcinogenesis is a genotoxic process. 4-Octylphenol (alkylphenol), bisphenol A (plasticiser), coumestrol and genistein (phytoestrogens), 2,4-dichlorophe- noxyacetic acid and toxaphene (pesticides) and ethinylestradiol (synthetic hormone) were investigated for potential mutagencicity, DNA strand breakage, clastogenicity and DNA repair. Significant induction in the percentage of cells containing micronuclei was observed for all the EDCs. Toxaphene and coumestrol were mutagenic in the Ames assay. They also induced significant levels of unscheduled DNA synthesis and DNA strand breakage. Bisphenol A induced low level DNA strand breakage in HepG2 cells in the comet assay. The EDCs, with the exception of toxaphene, induced transcriptional activation in the yeast estrogen screen (YES) assay. They were potently oestrogenic in the mammalian based MVLN (transactivation) and E-SCREEN (proliferation) assays. This report on the transactivational, proliferative and genotoxic ability of the EDCs suggests that these chemicals may play a role in the etiology of male and female reproductive cancers.展开更多

However, comparative studies are still lacking on the TH disruption. We investigated the interaction of [125I] ioxynil with serum proteins from rainbow trout, bullfrog, chicken, pig, rat, and mouse, using native polyacrylamide gel electrophoresis. Candidate ioxynil-binding proteins, which included lipoproteins, albumin and transthyretin (TTR), differed among the vertebrates tested. Rainbow trout and bullfrog tadpole serum had the lowest binding activity for ioxynil, whereas the eutherian serum had the highest binding activity. The cellular uptake of, and response to, ioxynil were suppressed by rat serum greater than by tadpole serum. The cellular uptake of [125I]ioxynil competed strongly with phenols with a single ring, but not with THs. Our results suggested that ioxynil interferes with TH homeostasis in plasma and with a step of cellular TH-signaling pathway other than TH-uptake system, in a species-specific manner.展开更多

The results indicated that the food chain processes affected significantly the growth, slightly reduced gonadosomatic indices, and elevated hepatosomatic indices. There was an obvious vitellogenin(VTG) induction, which generally only occurred in mature female, in the serum of juvenile rare minnow and mature male when fed with Limnodrilus sp. In addition, the rare minnow feeding on Limnodrilus sp. had significantly high renal indices, it meant obvious renal hyperplasia. The present work suggested that Limnodrilus sp. from field water may contain toxic pollutants and could lead to endocrine disruption effects to the predators. It was concluded that endocrine disruptors may not only be assimilated through water, but also be bioconcentrated through food web. The results also suggested the importance of food selection in conducting the study of endocrine disruption effects using sensitive species.展开更多

Particularly vulnerable to chemical assaults are the hypothalamic-pituitary-gonadal (HPG) axis and the hypothalamic-pituitary-thyroid (HPT) axis of the endocrine system. While the effects of endocrine disrupting chemicals (EDCs) on the HPG axis have been the subject of intense research efforts, with comprehensive elucidation, a lot remains to be clarified on the effects of EDCs on thyroid functions. For instance, there are no clear-cut biomarkers of exposure and effects of thyroid disrupting chemicals (TDCs) in intact organisms. Consequently, a number of in vitro assays have been developed, and are particularly useful for the identification and mechanistic characterization of potential TDCs considering the increasing number of EDCs that are being released into the environment. However, with the in vitro assays, studies suggest that a plausible major mode of action of TDCs, thyroid hormone receptor (THR) agonist activity, is not environmentally relevant. Here, we reviewed in vitro detection of TDCs activities in wastewater and surface waters. Data strongly suggest that cell lines may be responsible for the less frequent detection of THR agonist activity in wastewater and surface water. 文摘Pseudomonas aeruginosa,recognized for its biofilm production and secretion of virulence factors,posing a severe threat in areas such as clinical infections,food contamination,and marine biofouling.To address this,a new type of zinc-binding peptide(CSSP-Zn)was prepared from crimson snapper scales peptides(CSSP)and goslarite,and its antibacterial and anti-quorum-sensing activities toward P.aeruginosa PAO1 were exploited.Results indicated that CSSP-Zn induced planktonic strain PAO1 membrane injury via inhibiting expression levels of cell integrity genes,targeting microbial-specific membrane constituents,disrupting proton motive force,and causing metabolic disturbances.Meanwhile,CSSP-Zn decreased virulence factors pyocyanin,protease and rhamnolipid secretion,while considerably inhibiting quorum sensing-related genes(las,pqs and rhl)expression and decreasing bacterial abundance and pathogenicity in fish models.Moreover,CSSP-Zn not only effectively hindered biofilm formation but also disassembled preformed ones,thus disrupting biofilm topology.Taken together,utilizing food byproducts to obtain CSSP-Zn could help recycle food resources and provide insight into controlling planktonic and biofilm strain PAO1 contamination.

文摘Perfluorobutanesulfonate(PFBS)is an emerging pollutant capable of potently disrupting the sex and thyroid endocrine systems of teleosts.However,the hypoxic and temporal variation in PFBS endocrine disrupting toxicity remain largely unknown.In the present study,adult marine medaka were exposed to environmentally realistic concentrations of PFBS(0 and 10μg/L)under normoxia or hypoxia conditions for 7 days,aiming to explore the interactive behavior between PFBS and hypoxia.In addition,PFBS singular exposure was extended till 21days under normoxia to elucidate the time-course progression in PFBS toxicity.The results showed that hypoxia inhibited the growth and caused the suspension of egg spawn regardless of PFBS exposure.With regard to the sex endocrine system,7-day PFBS exposure led to an acute stimulation of transcriptional profiles in females,which,subsequently,recovered after the 21-day exposure.The potency of hypoxia to disturb the sex hormones was much stronger than PFBS.A remarkable increase in estradiol concentration was noted in medaka blood after hypoxia exposure.Changes in sex endocrinology of coexposed fish were largely determined by hypoxia,which drove the formation of an estrogenic environment.PFBS further enhanced the endocrine disrupting effects of hypoxia.However,the hepatic synthesis of vitellogenin and choriogenin,two commonly used sensitive biomarkers of estrogenic activity,failed to initiate in response to the estrogen stimulus.Compared to sex endocrine system,disturbances in thyroidal axis by PFBS or hypoxia were relatively mild.Overall,the present findings will advance our toxicological understanding about PFBS pollutant under the interference of hypoxia.

文摘Tetrabromobisphenol A(TBBPA)is a widely used brominated flame retardant.There is evidence showing that TBBPA can exert thyroid disrupting effects in mammals,but different results were also reported,along with inconsistent reports regarding its neurotoxicity.Here,we investigated thyroid disrupting effects and neurotoxicity of TBBPA(5,50,500μg/(kg·day))to male mice following maternal and direct exposure through drinking water,with the antithyroid drug propylthiouracil(PTU)as the positive control.On postnatal day(PND)15,we expectedly observed severe thyroid compensatory hyperplasia and cerebellar developmental retardation in PTU-treated pups.The highest dose of TBBPA also caused thyroid histological alteration but had no effects on cerebellar development in terms of Purkinje cell morphology and the thickness of the internal granular layer and the molecular layer of the cerebellum.During puberty and adulthood,the thyroid morphological alterations became more pronounced in the TBBPA-treated animals,accompanied by decreased serum thyroid hormone levels.Furthermore,the 50 and 500μg/(kg·day)TBBPA groups showed a significant decrease in the serum level of serotonin,a neurotransmitter associated with anxiety behaviors.Correspondingly,the highest dose group displayed anxiety-like behaviors in the elevated plus-maze test on PND 35,but this neurobehavioral alteration disappeared on PND 56.Moreover,no changes in neurobehavioral parameters tested were found in TBBPAtreated animals at puberty and adulthood.Altogether,all observations show that TBBPA can exert thyroid disrupting effects but has little overt impact on brain development and neurobehaviors in mice,suggesting that thyroid disruption does not necessarily cause overtly adverse neurodevelopmental outcomes.

文摘Environmental endocrine disrupting chemicals (EDCs), commonly found in the environment, come from industry and agriculture, including pesticides, fungicides, insecticides, herbicides, and other chemicals. Nowadays, more and more EDCs were released into the environment. EDCs go into water body via atmosphere sedi-mentation, surface runoff, soil eluviation, etc., so water body becomes the main place for existing. In order to attract scientific and public attention worldwide and to prevent EDCs pol ution, in this study we reviewed the classification of EDCs and their concentrations in natural water bodies, drinking water sources and water plants, and the reproductive toxicity of EDCs to fish were reviewed. EDCs could disturb the endocrine system and make reproductive organs and reproduction abnor-mal, resulting in fertility descending, reproduction function damage, community quan-tity decrease and even species extinction. In addition, EDCs could disrupt the homeostasis maintained by hormones, which would result in defects of neural de-velopment and abnormalities of the endocrine and reproductive systems. The exact molecular mechanisms have not been completely reported, but researches have suggested that multiple mechanisms were involved in the action of EDCs. Although there have been researches on the biohazard of EDCs, there stil exist problems of weakness in fundamental researches, difficulties in recognizing and identifying EDCs and high cost, which restraint the knowledge on them.

文摘Detection of estrogenic disrupting compounds(EDCs) in drinking waters around China has led to rising concerns about health risks associated with these compounds.There is,however,a paucity of studies on the occurrence and identification of the main compounds responsible for this pollution in the source waters.To fill this void,we screened estrogenic activities of 23 source water samples from six main river systems in China,using a recombinant two-hybrid yeast assay.All sample extracts induced significant estrogenic activity,with E2 equivalents(EEQ) of raw water ranging from 0.08 to 2.40 ng/L.Additionally,16 samples were selected for chemical analysis by gas chromatography-mass spectrometry.The EDCs of most concern,including estrone(E1),17β-estradiol(E2),17α-ethinylestradiol(EE2),estriol(E3),diethylstilbestrol(DES),estradiol valerate(EV),4-t-octylphenol(4-t-OP),4-nonylphenols(4-NP) and bisphenol A(BPA),were determined at concentrations of up to 2.98,1.07,2.67,4.37,2.52,1.96,89.52,280.19 and 710.65 ng/L,respectively.Causality analysis,involving comparison of EEQ values from yeast assay and chemical analysis identified E2,EE2 and 4-NP as the main responsible compounds,accounting for the whole estrogenic activities(39.74% to 96.68%).The proposed approach using both chemical analysis and yeast assay could be used for the identification and evaluation of EDCs in source waters of China.

文摘Solid malignancies have to develop their own blood supply for their aggressive growth and metastasis;a process known as tumor angiogenesis.Angiogenesis is largely involved in tumor survival,progression and spread,which are known to be significantly attributed to treatment failures.Over the past decades,efforts have been made to understand the difference between nor-mal and tumor vessels.It has been demonstrated that tumor vasculature is structurally immature with chaotic and leaky phenotypes,which provides opportunities for developing novel anticancer strategies.Targeting tumor vasculature is not only a unique therapeutic interven-tion to starve neoplastic cells,but also enhances the efficacy of conventional cancer treatments.Vascular dis-rupting agents(VDAs) have been developed to disrupt the already existing neovasculature in actively growing tumors,cause catastrophic vascular shutdown within short time,and induce secondary tumor necrosis.VDAs are cytostatic;they can only inhibit tumor growth,but not eradicate the tumor.This novel drug mechanism has urged us to develop multiparametric imaging biomark-ers to monitor early hemodynamic alterations,cellular dysfunctions and metabolic impairments before tumor dimensional changes can be detected.In this article,we review the characteristics of tumor vessels,tubulin-destabilizing mechanisms of VDAs,and in vivo effects of the VDAs that have been mostly studied in preclinical studies and clinical trials.We also compare the differ-ent tumor models adopted in the preclinical studies on VDAs.Multiparametric imaging biomarkers,mainly diffu-sion-weighted imaging and dynamic contrast-enhanced imaging from magnetic resonance imaging,are evalu-ated for their potential as morphological and functional imaging biomarkers for monitoring therapeutic effects of VDAs.

文摘Endocrine disrupting chemicals(EDCs) in the secondary effluent discharged from wastewater treatment plants are of great concern when water reuse is intended. Ozonation and ultrafiltration(UF) are powerful technologies reported to eliminate EDCs. Due to the importance of effluent organic matters(EfOMs) in secondary effluent, the effects of three kinds of EfOM on the treatment of five EDCs using ozonation and UF were investigated. The three kinds of EfOM studied were humic acid sodium salt(NaAH), bovine serum albumin(BSA)and sodium alginate(NaAg); and the five EDCs were estrone, 17β-estradiol, estriol, 17α-ethynyl estradiol and bisphenol A. The results showed that EfOM accelerated the decay rate of ozone and inhibited the degradation efficiency of EDCs by ozonation in the order NaAH > BSA > NaAg.The ultraviolet absorbance at 280 nm(UVA_(280)) has potential for use as a surrogate indicator to assess EDC removal via ozonation without conducting difficult EDC analyses. When the decline in UVA_(280) exceeded 18%, the five EDCs had been completely removed. The UF behavior of NaAH,BSA and NaAg was found to follow the cake filtration law. The fouling potential of EfOM followed the order NaAg > NaAH > BSA; while EfOM on the membrane surface enhanced EDC removal in the order NaAH > BSA > NaAg. The mean retention rate of the membrane was increased by 24%, 10% and 8%, respectively. The properties of EDCs and EfOM cakes both influenced the EDC removal rates due to adsorption, size exclusion and charge attraction.

文摘Objective To measure the endocrine disrupting chemicals (EDCs) in wastewater and evaluate the EDCs removal efficiencies in the municipal wastewater treatment plants (WWTP). Methods A battery of in vitro recombinant yeast bioassays incorporated with exogenous metabolic activation system (rat liver preparation, S9 mix) was conducted to assess the estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PR), and thyroid receptor (TR) ant/agonistic activities of effluents collected from Datansha WWTP. Results The indirect estrogenic, anti‐androgenic, anti‐progesteronic, and anti‐thyroidic activities were observed in the influent. The removal efficiencies of EDCs were above 74%, suggesting that the present wastewater treatment processes were good enough to remove most of these indirect endocrine disrupting chemicals. Conclusion The incorporation of exogenous metabolic capacity into the test system was valid for the study of indirect effects on ER, AR, PR, and TR.

文摘Endocrine disrupting compounds(EDCs) and pharmaceuticals pose a challenge for water and wastewater treatment because they exist at very low concentrations in the presence of substances at much higher concentrations competing for adsorption sites.Sub-micron sized resin particles(approximately 300 nm in diameter)(SMR) were tested to evaluate their potential as a treatment for EDCs including:17-β estradiol(E2),17-α ethinylestradiol(EE2),estrone(E1),bisphenol A(BPA),and diethylstilbestrol(DES) as well as 12 pharmaceuticals.SMR were able to remove 98%of spiked E2,80%of EE2,87%of BPA,and up to 97%of DES from water.For a 0.5 ppm mixture of E2,EE2,E1,BPA and DES,the minimum removal was24%(E2) and the maximum was 49%(DES).They were also able to remove the pharmaceuticals from deionized water and wastewater.Overall,SMR are a promising advanced treatment for removal of both EDCs and pharmaceuticals.

文摘Obesity and associated metabolic disorders represent a major societal challenge in health and quality of life with large psychological consequences in addition to physical disabilities. They are also one of the leading causes of morbidity and mortality. Although, different etiologic factors including excessive food intake and reduced physical activity have been well identified, they cannot explain the kinetics of epidemic evolution of obesity and diabetes with prevalence rates reaching pandemic proportions. Interestingly, convincing data have shown that environmental pollutants, specifically those endowed with endocrine disrupting activities, could contribute to the etiology of these multifactorial metabolic disorders. Within this review, we will recapitulate characteristics of endocrine disruption. We will demonstrate that metabolic disorders could originate from endocrine disruption with a particular focus on convincing data from the literature. Eventually, we will present how handling an original mouse model of chronic exposition to a mixture of pollutants allowed demonstrating that a mixture of pollutants each at doses beyond their active dose could induce substantial deleterious effects on several metabolic end-points. This proof-of-concept study, as well as other studies on mixtures of pollutants, stresses the needs for revisiting the current threshold model used in risk assessment which does not take into account potential effects of mixtures containing pollutants at environmental doses, e.g., the real life exposure. Certainly, more studies are necessary to better determine the nature of the chemicals to which humans are exposed and at which level, and their health impact. As well, research studies on substitute products are essential to identify harmless molecules.

文摘To understand the adsorption behavior of endocrine disrupting chemicals(EDCs) is important for enhancing the treatment performance and preventing potential secondary pollution caused by EDCs desorption in a microfiltration system. The dynamic adsorption of four representative EDCs, namely estriol(E3), 17β-estradiol(E2), 17α-ethynylestradiol(EE2), and 4-nonylphenol(4-NP) in a microfiltration system was investigated using the Thomas' model.The product of the equilibrium constant and the total adsorption capacity of the membrane,Ka, for E3, E2, EE2, and 4-NP were 4.91, 9.78, 15.6, and 826, respectively, strongly correlating with the compound octanol–water partition coefficient(KOW). Adsorption appeared to be enhanced when organic fouling formed on the surface of membrane, indicating the role of an additional adsorption column for EDCs acted by a fouling layer in microfiltration. Results of a comparison between the Ka values for clean membrane and fouled membrane illustrated that the significant contribution made by fouling layers may be attributed to the foulant layer's hydrophobicity(in the case of calcium humate layer) and thickness(in the case of calcium alginate layer). This study provided a novel perspective to quantitatively analyze the dynamic adsorption behavior of trace pollutants in membrane process.

文摘Endocrine disrupting chemical(EDC) pollution in river-based artificial groundwater recharge using reclaimed municipal wastewater poses a potential threat to groundwater-based drinking water supplies in Beijing, China. Lab-scale leaching column experiments simulating recharge were conducted to study the adsorption, biodegradation, and transport characteristics of three selected EDCs: 17β-estradiol(E2), 17α-ethinylestradiol(EE2) and bisphenol A(BPA). The three recharge columns were operated under the conditions of continual sterilization recharge(CSR), continual recharge(CR), and wetting and drying alternative recharge(WDAR). The results showed that the attenuation effect of the EDCs was in the order of WDAR 〉 CR 〉 CSR system and E2 〉 EE2 〉 BPA, which followed first-order kinetics. The EDC attenuation rate constants were 0.0783, 0.0505, and 0.0479 m-1 for E2, EE2 and BPA in the CR system, respectively. The removal rates of E2, EE2, and BPA in the CR system were 98%, 96% and 92%, which mainly depended on biodegradation and were affected by water temperature.In the CR system, the concentrations of BPA, EE2, and E2 in soil were 4, 6 and 10 times higher than in the WDAR system, respectively. According to the DGGE fingerprints, the bacterial community in the bottom layer was more diverse than in the upper layer, which was related to the EDC concentrations in the water-soil system. The dominant group was found to be proteobacteria, including Betaproteobacteria and Alphaproteobacteria, suggesting that these microbes might play an important role in EDC degradation.

文摘The rational for the study was to assess the levels of endocrine disrupting trace metals in River Samre. The levels of Mercury ranged from 0.01 to 0.02 mg/l (mean of 0.006 mg/l), whiles cadmium levels ranged from 0.002 to 0.011 mg/l (mean of 0.01 mg/l). The high levels of Hg and Cd may have adverse effects on the endocrine system of inhabitants who drink directly from the river without treatment. High levels of mercury and cadmium might be caused by the activities of a Timber and Plywood Company located close to the river and the underlying bedrocks of the area exposed as a result of human activities such as farming. The concentration of lead was below detection limit ( mg/l) but that of Arsenic ranged from 0.001 to 0.007 mg/l (mean of 0.005). Health risk assessment conducted shows that the risk associated with exposure to these metals for now are low. Continuous water quality monitoring is recommended to help protect the resource and also to safeguard human health.
文摘A battery of tests was established to determine the oestrogenic, mutagenic and genotoxic potential of two categories of endocrine disrupting chemicals (EDCs), phthalates and alkylphenols. Diisononylphthalate (DINP), diethylhexylphthalate (DEHP), dibutylphthalate (DBP), diisododecylphthalate (DIDP) and 4-nonylphenol (4-NP) were oestrogenic in the yeast estrogen screen (YES) assay and potently oestrogenic in the MVLN and E-SCREEN assays at environmentally relevant concentrations. DINP and 4-NP were mutagenic in the Ames assay and also induced significant levels of unscheduled DNA synthesis and DNA strand breakage. Significant induction in the percentage of cells containing micronuclei was observed after treatment with DINP, DEHP and 4-NP. In addition, sewage effluents from sewage treatment plants (STPs) in the Border, Midlands and Western (BMW) region of Ireland were significantly oestrogenic in the YES assay. Moreover, analysis of levels of phthalates and alkylphenol identified in Irish rivers receiving treated effluent showed potent oestrogenicity in the YES assay. The proliferative and genotoxic ability of the phthalates and alkylphenol, and the oestrogenicity of the treated effluents reported here, is significant as these EDCs and EDCs within the effluent may play a role in the etiology of human abnormalities.
文摘A diverse range of endocrine disrupting chemicals (EDCs) was examined, using an in vitro test system, for critical events required for the onset of carcinogenesis in vivo. The initiation stage of carcinogenesis is a genotoxic process. 4-Octylphenol (alkylphenol), bisphenol A (plasticiser), coumestrol and genistein (phytoestrogens), 2,4-dichlorophe- noxyacetic acid and toxaphene (pesticides) and ethinylestradiol (synthetic hormone) were investigated for potential mutagencicity, DNA strand breakage, clastogenicity and DNA repair. Significant induction in the percentage of cells containing micronuclei was observed for all the EDCs. Toxaphene and coumestrol were mutagenic in the Ames assay. They also induced significant levels of unscheduled DNA synthesis and DNA strand breakage. Bisphenol A induced low level DNA strand breakage in HepG2 cells in the comet assay. The EDCs, with the exception of toxaphene, induced transcriptional activation in the yeast estrogen screen (YES) assay. They were potently oestrogenic in the mammalian based MVLN (transactivation) and E-SCREEN (proliferation) assays. This report on the transactivational, proliferative and genotoxic ability of the EDCs suggests that these chemicals may play a role in the etiology of male and female reproductive cancers.

文摘Vitellogenin(Vtg)is the precursor of egg yolk proteins,and its expression has been used as a reliable biomarker for estrogenic contamination in the aquatic environment.To examine the biomarker potential of the self-fertilizing killifish Kryptolebias marmoratus Vtgs(Km-Vtgs),full genomic DNAs of Km-Vtgs-Aa,Km-Vtgs-Ab,and Km-Vtgs-C were cloned,sequenced,and characterized.Three Vtg genes in K.marmoratus are tandemly placed in a550 kb section of the same chromosome.In silico analysis of promoter regions revealed that both the Km-Vtgs-Aa and Km-Vtgs-Ab genes had an estrogen response element(ERE),but the Km-Vtgs-C gene did not.However,all three Km-Vtgs genes had several ERE-half sites in their promoter regions.Phylogenetic analysis demonstrated that the three deduced amino acid residues were highly conserved with conventional Vtgs protein,forming distinctive clades within teleost Vtgs.Liver tissue showed the highest expression of Km-Vtg transcripts in all tested tissues(brain/pituitary,eye,gonad,intestine,skin,and muscle)in response to endocrine disrupting chemical(EDC)-exposed conditions.Km-Vtg transcripts were significantly increased in response to 17β-estradiol(E2),tamoxifen(TMX),4-n-nonylphenol(NP),bisphenol A(BPA),and octylphenol(OP)over 24 hr exposure.The Km-Vtg-A gene was highly expressed compared to the control in response to NP and OP.EDC-induced modulatory patterns of Km-Vtg gene expression were different depending on tissue,gender,and isoforms.

文摘Ioxynil, a phenolic herbicide, is known to exert thyroid hormone (TH) disrupting activity by interfering with TH-binding to plasma proteins and a step of the cellular TH-signaling pathway in restricted animal species. However, comparative studies are still lacking on the TH disruption. We investigated the interaction of [125I] ioxynil with serum proteins from rainbow trout, bullfrog, chicken, pig, rat, and mouse, using native polyacrylamide gel electrophoresis. Candidate ioxynil-binding proteins, which included lipoproteins, albumin and transthyretin (TTR), differed among the vertebrates tested. Rainbow trout and bullfrog tadpole serum had the lowest binding activity for ioxynil, whereas the eutherian serum had the highest binding activity. The cellular uptake of, and response to, ioxynil were suppressed by rat serum greater than by tadpole serum. The cellular uptake of [125I]ioxynil competed strongly with phenols with a single ring, but not with THs. Our results suggested that ioxynil interferes with TH homeostasis in plasma and with a step of cellular TH-signaling pathway other than TH-uptake system, in a species-specific manner.
文摘The potential endocrine disrupting effects and other toxicity effects on aquatic biota resulted from food uptake was simulated by feeding the laboratory cultured rare minnow(Gobiocypris rarus) with field collected Limnodrilus sp. The results indicated that the food chain processes affected significantly the growth, slightly reduced gonadosomatic indices, and elevated hepatosomatic indices. There was an obvious vitellogenin(VTG) induction, which generally only occurred in mature female, in the serum of juvenile rare minnow and mature male when fed with Limnodrilus sp. In addition, the rare minnow feeding on Limnodrilus sp. had significantly high renal indices, it meant obvious renal hyperplasia. The present work suggested that Limnodrilus sp. from field water may contain toxic pollutants and could lead to endocrine disruption effects to the predators. It was concluded that endocrine disruptors may not only be assimilated through water, but also be bioconcentrated through food web. The results also suggested the importance of food selection in conducting the study of endocrine disruption effects using sensitive species.
文摘The developmental and reproductive effects of endocrine disruption on humans and wildlife rank among the most threatening of all environmental health concerns. Particularly vulnerable to chemical assaults are the hypothalamic-pituitary-gonadal (HPG) axis and the hypothalamic-pituitary-thyroid (HPT) axis of the endocrine system. While the effects of endocrine disrupting chemicals (EDCs) on the HPG axis have been the subject of intense research efforts, with comprehensive elucidation, a lot remains to be clarified on the effects of EDCs on thyroid functions. For instance, there are no clear-cut biomarkers of exposure and effects of thyroid disrupting chemicals (TDCs) in intact organisms. Consequently, a number of in vitro assays have been developed, and are particularly useful for the identification and mechanistic characterization of potential TDCs considering the increasing number of EDCs that are being released into the environment. However, with the in vitro assays, studies suggest that a plausible major mode of action of TDCs, thyroid hormone receptor (THR) agonist activity, is not environmentally relevant. Here, we reviewed in vitro detection of TDCs activities in wastewater and surface waters. Data strongly suggest that cell lines may be responsible for the less frequent detection of THR agonist activity in wastewater and surface water. It was concluded that the development of reporter gene assays with thyroid hormone function related cell lines, is required.
