Establishment of human cerebral organoid systems to model early neural development and assess the central neurotoxicity of environmental toxins 被引量:1

Human brain development is a complex process,and animal models often have significant limitations.To address this,researchers have developed pluripotent stem cell-derived three-dimensional structures,known as brain-like organoids,to more accurately model early human brain development and disease.To enable more consistent and intuitive reproduction of early brain development,in this study,we incorporated forebrain organoid culture technology into the traditional unguided method of brain organoid culture.This involved embedding organoids in matrigel for only 7 days during the rapid expansion phase of the neural epithelium and then removing them from the matrigel for further cultivation,resulting in a new type of human brain organoid system.This cerebral organoid system replicated the temporospatial characteristics of early human brain development,including neuroepithelium derivation,neural progenitor cell production and maintenance,neuron differentiation and migration,and cortical layer patterning and formation,providing more consistent and reproducible organoids for developmental modeling and toxicology testing.As a proof of concept,we applied the heavy metal cadmium to this newly improved organoid system to test whether it could be used to evaluate the neurotoxicity of environmental toxins.Brain organoids exposed to cadmium for 7 or 14 days manifested severe damage and abnormalities in their neurodevelopmental patterns,including bursts of cortical cell death and premature differentiation.Cadmium exposure caused progressive depletion of neural progenitor cells and loss of organoid integrity,accompanied by compensatory cell proliferation at ectopic locations.The convenience,flexibility,and controllability of this newly developed organoid platform make it a powerful and affordable alternative to animal models for use in neurodevelopmental,neurological,and neurotoxicological studies. 展开更多

The organotypic retinal explant culture has been established for more than a decade and offers a range of unique advantages compared with in vivo experiments and cell cultures.However,the lack of systematic and continuous comparison between in vivo retinal development and the organotypic retinal explant culture makes this model controversial in postnatal retinal development studies.Thus,we aimed to verify the feasibility of using this model for postnatal retinal development studies by comparing it with the in vivo retina.In this study,we showed that postnatal retinal explants undergo normal development,and exhibit a consistent structure and timeline with retinas in vivo.Initially,we used SOX2 and PAX6 immunostaining to identify retinal progenitor cells.We then examined cell proliferation and migration by immunostaining with Ki-67 and doublecortin,respectively.Ki-67-and doublecortin-positive cells decreased in both in vivo and explants during postnatal retinogenesis,and exhibited a high degree of similarity in abundance and distribution between groups.Additionally,we used Ceh-10 homeodomain-containing homolog,glutamate-ammonia ligase(glutamine synthetase),neuronal nuclei,and ionized calcium-binding adapter molecule 1 immunostaining to examine the emergence of bipolar cells,Müller glia,mature neurons,and microglia,respectively.The timing and spatial patterns of the emergence of these cell types were remarkably consistent between in vivo and explant retinas.Our study showed that the organotypic retinal explant culture model had a high degree of consistency with the progression of in vivo early postnatal retina development.The findings confirm the accuracy and credibility of this model and support its use for long-term,systematic,and continuous observation. 展开更多

With the increasing severity of arsenic(As)pollution,quantifying the environmental behavior of pollutant based on numerical model has become an important approach to determine the potential impacts and finalize the precise control strategies.Taking the industrialintensive Jinsha River Basin as typical area,a two-dimensional hydrodynamic water quality model coupled with Soil andWater Assessment Tool(SWAT)model was developed to accurately simulate the watershed-scale distribution and transport of As in the terrestrial and aquatic environment at high spatial and temporal resolution.The effects of hydro-climate change,hydropower station construction and non-point source emissions on Aswere quantified based on the coupled model.The result indicated that higher As concentration areas mainly centralized in urban districts and concentration slowly decreased from upstream to downstream.Due to the enhanced rainfall,the As concentration was significantly higher during the rainy season than the dry season.Hydro-climate change and the construction of hydropower station not only affected the dissolved As concentration,but also affected the adsorption and desorption of As in sediment.Furthermore,As concentration increased with the input of non-point source pollution,with the maximum increase about 30%,resulting that non-point sources contributed important pollutant impacts to waterways.The coupled model used in pollutant behavior analysis is generalwith high potential application to predict and mitigate water pollution. 展开更多

This study systematically reviews the development history and key technological breakthroughs of large gas fields in the Ordos Basin,and summarizes the development models of three gas reservoir types,low-permeability carbonate,low-permeability sandstone and tight sandstone,as well as the progress in deep coal-rock gas development.The current challenges and future development directions are also discussed.Mature development models have been formed for the three representative types of gas reservoirs in the Ordos Basin:(1)Low-permeability carbonate reservoir development model featuring groove fine-scale characterization and three-dimensional vertical succession between Upper and Lower Paleozoic formations.(2)Low-permeability sandstone reservoir development model emphasizing horizontal well pressure-depletion production and vertical well pressure-controlled production.(3)Tight sandstone gas reservoir development model focusing on single-well productivity enhancement and well placement optimization.In deep coal-rock gas development,significant progress has been achieved in reservoir evaluation,sweet spot prediction,and geosteering of horizontal wells.The three types of reservoirs have entered the mid-to-late stages of the development,when the main challenge lies in accurately characterizing residual gas,evaluating secondary gas-bearing layers,and developing precise potential-tapping strategies.In contrast,for the early-stage development of deep coal-rock gas,continuous technological upgrades and cost reduction are essential to achieving economically viable large-scale development.Four key directions of future research and technological breakthroughs are proposed:(1)Utilizing dual-porosity(fracture-matrix)modeling techniques in low-permeability carbonate reservoirs to delineate the volume and distribution of remaining gas in secondary pay zones,supporting well pattern optimization and production enhancement of existing wells.(2)Integrating well-log and seismic data to characterize reservoir spatial distribution of successive strata,enhancing drilling success rates in low-permeability sandstone reservoirs.(3)Utilizing the advantages of horizontal wells to penetrate effective reservoirs laterally,achieving meter-scale quantification of small-scale single sand bodies in tight gas reservoirs,and applying high-resolution 3D geological models to clarify the distribution of remaining gas and guide well placement optimization.(4)Further strengthening the evaluation of deep coal-rock gas in terms of resource potential,well type and pattern,reservoir stimulation,single-well performance,and economic viability. 展开更多

The generalized rheological tests on sandstone were conducted under both dynamic stress and seepage fields.The results demonstrate that the rheological strain of the specimen under increased stress conditions is greater than that under creep conditions,indicating that the dynamic stress field significantly influences the rheological behaviours of sandstone.Following the rheological tests,the number of small pores in the sandstone decreased,while the number of medium-sized pores increased,forming new seepage channels.The high initial rheological stress accelerated fracture compression and the closure of seepage channels,resulting in reduction in the permeability of sandstone.Based on the principles of generalized rheology and the experimental findings,a novel rock rheological constitutive model incorporating both the dynamic stress field and seepage properties has been developed.Numerical simulations of surrounding rock deformation in geotechnical engineering were carried out using a secondary development version of this model,which confirmed the applicability of the generalized rheological numerical simulation method.These results provide theoretical support for the long-term stability evaluation of engineering rock masses and for predicting the deformation of surrounding rock. 展开更多

BACKGROUND The discrepancy between endoscopic biopsy pathology and the overall pathology of gastric low-grade intraepithelial neoplasia(LGIN)presents challenges in developing diagnostic and treatment protocols.AIM To develop a risk prediction model for the pathological upgrading of gastric LGIN to aid clinical diagnosis and treatment.METHODS We retrospectively analyzed data from patients newly diagnosed with gastric LGIN who underwent complete endoscopic resection within 6 months at the First Medical Center of Chinese People’s Liberation Army General Hospital between January 2008 and December 2023.A risk prediction model for the pathological progression of gastric LGIN was constructed and evaluated for accuracy and clinical applicability.RESULTS A total of 171 patients were included in this study:93 patients with high-grade intraepithelial neoplasia or early gastric cancer and 78 with LGIN.The logistic stepwise regression model demonstrated a sensitivity and specificity of 0.868 and 0.800,respectively,while the least absolute shrinkage and selection operator(LASSO)regression model showed sensitivity and specificity values of 0.842 and 0.840,respectively.The area under the curve(AUC)for the logistic model was 0.896,slightly lower than the AUC of 0.904 for the LASSO model.Internal validation with 30%of the data yielded AUC scores of 0.908 for the logistic model and 0.905 for the LASSO model.The LASSO model provided greater utility in clinical decision-making.CONCLUSION A risk prediction model for the pathological upgrading of gastric LGIN based on white-light and magnifying endoscopic features can accurately and effectively guide clinical diagnosis and treatment. 展开更多

Research on the distribution and development of black shales in the Lianggaoshan Formation has been deficient,which has hindered exploration for lacustrine shale oil in the Sichuan Basin.Our study characterized the well logging data,core samples,outcrops,and geochemistry of black shales in the Lianggaoshan Formation in the Sichuan Basin.Our analysis focused on the lake basin evolution and the migration characteristics,paleoenvironmental features,formation mechanisms,and developmental model of the black shales.The results indicated that black shales in the Lianggaoshan Formation exhibited significant lateral migration,with an overall thickening trend from east to west.Within the 1st Member of the formation,black shale occurred as a single thick layer in the eastern region that gradually thinned toward the central region.Multiple sets of shale developed within the 2nd and 3rd members,and these had lower thicknesses than the 1st Member and migrated toward central Sichuan.Paleoproductivity and terrigenous input were the main factors controlling the deposition of black shales.A semi-humid climate influenced the deposition of black shales,bringing abundant freshwater,terrigenous debris,and nutrients into the basin.Decomposition of organic matter consumed oxygen in sediment and bottom water,causing localized oxygen deficiency in the strata. 展开更多

This paper focuses on rural music education in the context of rural revitalization,deeply analyzes its current resource situation,and points out that tangible resources face shortages of teachers,inappropriate textbooks,and outdated teaching facilities,while intangible resources are confronted with the loss of folk music culture and the weakening of educational foundations due to cultural ecological changes.Resources are characterized by dispersion,scarcity,and inefficient utilization.In response to these difficulties,this paper proposes integration paths for teachers,textbooks,teaching facilities,and folk music culture resources.Additionally,it constructs a diversified development model that combines rural tourism with music activities,develops characteristic music education industries,and integrates rural cultural construction.Simultaneously,it identifies funding shortages and outdated concepts as major obstacles and proposes targeted strategies such as expanding funding channels and changing inherent concepts.The aim is to promote the development of rural music education and contribute to the revitalization of rural culture. 展开更多

In the current settings of osteosarcoma research and drug screening,in vitro three-dimensional(3D)models,which overcome the limitations of traditional models,are favored.In in vitro 3D models,tumor microenvironment simulation,particularly of the mechanical microenvironment,is crucial for estimating the biological effects of a tumor.However,current in vitro osteosarcoma model construction is often limited to a single mechanical signal,which fails to simulate the diversity of osteosarcoma mechanical stimuli.In this study,we utilized embedded bioprinting technology and the multiple response properties of calcium ions in soft and hard stiffness systems with osteosarcoma cell biological functions to construct an integrated gradient biomechanical signal-tailored osteosarcoma model(IGBSTOM).We achieved this by printing a fibrinogen bioink containing calcium ions and osteosarcoma tumor spheroids within an extracellular matrix composed of methacryloylated alginate,methacryloylated gelatin,thrombin,and transglutaminase,which is rich in polysaccharides and proteins and exhibits self-healing properties.Our in vitro and in vivo studies showed that the IGBSTOM enhanced tumor stemness,proliferation,and migration,and successfully reproduced the nest-like structure of tumors,providing an in vitro research platform that is more similar to the natural tumor than the existing models.This study proposes a novel IGBSTOM construction and provides a new strategy for the clinical understanding of tumor development,drug screening,and exploration of drug resistance and metastasis mechanisms. 展开更多

A complex geological environment with faults can be encountered in the process of coal mining.Fault activation can cause instantaneous structure slipping,releasing a significant amount of elastic strain energy during underground coal mining.This would trigger strong rockburst disasters.To understand the occurrence of fault-slip induced rockbursts,we developed a physical model test system for fault-slip induced rockbursts in coal mine drifts.The boundary energy storage(BES)loading apparatus and bottom rapid retraction(BRR)apparatus are designed to realize energy compensation and continuous boundary stress transfer of the surrounding rocks for instantaneous fault slip,as well as to provide space for the potential fault slip.Taking the typical fault-slip induced rockburst in the Xinjulong Coal Mine,China,as the background,we conducted a model test using the test system.The deformation and stress in the rock surrounding the drift and the support unit force during fault slip are analyzed.The deformation and failure characteristics and dynamic responses of drifts under fault-slip induced rockbursts are obtained.The test results illustrate the rationality and effectiveness of the test system.Finally,corresponding recommendations and prospects are proposed based on our findings. 展开更多

Based on the comprehensive analysis of data from petrology and mineralogy,well logging,seismic surveys,paleontology,and geochemistry,a detailed research was conducted on paleoenvironmental and paleoclimatic conditions,and modeling of the source rocks in the second member of the Eocene Wenchang Formation(Wen 2 Member)in the Northern Shunde Subsag at the southwestern margin of the Pearl River Mouth Basin.The Wen 2 Member hosts excellent,thick lacustrine source rocks with strong longitudinal heterogeneity and an average total organic carbon(TOC)content of over 4.9%.The Wen 2 Member can be divided into three units(I,II,III)from bottom to top.Unit I features excellent source rocks with Type I organic matters(average TOC of 5.9%)primarily sourced from lake organisms;Unit II hosts source rocks dominated by Type II2 organic matters(average TOC of 2.2%),which are originated from mixed sources dominated by terrestrial input.Unit III contains good to excellent source rocks dominated by Type II1 organic matters(average TOC of 4.9%),which are mainly contributed by lake organisms and partially by terrestrial input.Under the background of rapid subsidence and limited source supply during intense rifting period in the Eocene,excellent source rocks were developed in Wen 2 Member in the Northern Shunde Subsag under the coordinated control of warm and humid climate,volcanic activity,and deep-water reducing conditions.During the deposition of Unit I,the warm and humid climate and volcanic activity promoted the proliferation of lake algaes,primarily Granodiscus,resulting in high initial productivity,and deep-water reducing conditions enabled satisfactory preservation of organic matters.These factors jointly controlled the development and occurrence of excellent source rocks.During the deposition of Unit II,a transition from warm to cool and semi-arid paleoclimatic conditions led to a decrease in lake algaes and initial productivity.Additionally,enhanced terrestrial input and shallow-water,weakly oxidizing water conditions caused a significant dilution and decomposition of organic matters,degrading the quality of source rocks.During the deposition of Unit III,when the paleoclimatic conditions are cool and humid,Pediastrum and Botryococcus began to thrive,leading to an increase in productivity.Meanwhile,the reducing environment of semi-deep water facilitated the preservation of excellent source rocks,albeit slightly inferior to those in Unit I.The study results clarify the differential origins and development models of various source rocks in the Shunde Sag,offering valuable guidance for evaluating source rocks and selecting petroleum exploration targets in similar marginal sags. 展开更多

Development and Validation of a Postoperative Recurrence Prediction Model for Pancreatic Cancer: A Multicenter Study

Background: Pancreatic cancer is one of the most lethal malignancies, with postoperative recurrence severely affecting patient survival and prognosis. This study aims to develop and validate a clinical prediction model for postoperative recurrence in pancreatic cancer patients, incorporating multiple preoperative, intraoperative, and postoperative factors to assist clinical decision-making. Methods: A retrospective study was conducted on 216 patients who underwent surgical treatment for pancreatic malignancy at the First Affiliated Hospital of Chongqing Medical University between January 2015 and January 2023. An independent external validation cohort of 76 patients from the Second Affiliated Hospital of Chongqing Medical University was used to validate the model. Seven independent risk factors for postoperative recurrence were identified through univariate and multivariate Cox regression analyses. The model’s performance was evaluated using the concordance index (C-index) and ROC curves, and its accuracy and clinical value were assessed using calibration curves and decision curve analysis (DCA). Results: The predictive model demonstrated good discriminatory power, with a C-index of 0.72 in the training cohort and 0.66 in the validation cohort. The ROC curves for predicting recurrence at 3, 6, and 12 months postoperatively showed AUC values ranging from 0.72 to 0.83, indicating strong predictive value. Calibration curves and DCA confirmed the model’s accuracy and clinical utility. Conclusion: This study successfully developed and validated a clinical prediction model that incorporates seven independent risk factors for postoperative recurrence in pancreatic cancer. The model provides a useful tool for predicting recurrence risk, aiding in the identification of high-risk patients, and informing clinical decision-making. 展开更多

BACKGROUND Ischemic heart disease(IHD)impacts the quality of life and has the highest mortality rate of cardiovascular diseases globally.AIM To compare variations in the parameters of the single-lead electrocardiogram(ECG)during resting conditions and physical exertion in individuals diagnosed with IHD and those without the condition using vasodilator-induced stress computed tomography(CT)myocardial perfusion imaging as the diagnostic reference standard.METHODS This single center observational study included 80 participants.The participants were aged≥40 years and given an informed written consent to participate in the study.Both groups,G1(n=31)with and G2(n=49)without post stress induced myocardial perfusion defect,passed cardiologist consultation,anthropometric measurements,blood pressure and pulse rate measurement,echocardiography,cardio-ankle vascular index,bicycle ergometry,recording 3-min single-lead ECG(Cardio-Qvark)before and just after bicycle ergometry followed by performing CT myocardial perfusion.The LASSO regression with nested cross-validation was used to find the association between Cardio-Qvark parameters and the existence of the perfusion defect.Statistical processing was performed with the R programming language v4.2,Python v.3.10[^R],and Statistica 12 program.RESULTS Bicycle ergometry yielded an area under the receiver operating characteristic curve of 50.7%[95%confidence interval(CI):0.388-0.625],specificity of 53.1%(95%CI:0.392-0.673),and sensitivity of 48.4%(95%CI:0.306-0.657).In contrast,the Cardio-Qvark test performed notably better with an area under the receiver operating characteristic curve of 67%(95%CI:0.530-0.801),specificity of 75.5%(95%CI:0.628-0.88),and sensitivity of 51.6%(95%CI:0.333-0.695).CONCLUSION The single-lead ECG has a relatively higher diagnostic accuracy compared with bicycle ergometry by using machine learning models,but the difference was not statistically significant.However,further investigations are required to uncover the hidden capabilities of single-lead ECG in IHD diagnosis. 展开更多

Geared-rotor systems are critical components in mechanical applications,and their performance can be severely affected by faults,such as profile errors,wear,pitting,spalling,flaking,and cracks.Profile errors in gear teeth are inevitable in manufacturing and subsequently accumulate during operations.This work aims to predict the status of gear profile deviations based on gear dynamics response using the digital model of an experimental rig setup.The digital model comprises detailed CAD models and has been validated against the expected physical behavior using commercial finite element analysis software.The different profile deviations are then modeled using gear charts,and the dynamic response is captured through simulations.The various features are then obtained by signal processing,and various ML models are then evaluated to predict the fault/no-fault condition for the gear.The best performance is achieved by an artificial neural network with a prediction accuracy of 97.5%,which concludes a strong influence on the dynamics of the gear rotor system due to profile deviations. 展开更多
