Sixty adult mice were randomly divided into four groups (mild, moderate and severe salt deficiency groups and control) during the seasonal splitting from winter to spring. The mice in the control group were fed with standard ordinary diet (salt content 0.26%), while in the mild, moderate and severe groups, the mice were fed with 10%, 30% and 50% salt deficient feedstuff, lasting for approximately 3 months. The results showed that the salt content of feed was negatively correlated with the food intake and body weight of mice. The sucrose preference test found that only the mild salt deficiency group had no differ-ence between the beginning and the end of the experiment, and the other three groups in-cluding the control, showed a significant decrease. These results suggest that dietary salt content has an impact on the food intake and body weight of mice and is associated with the emergence of depressive-like behavior. Furthermore, the seasonal splitting from winter to spring may also have a differential synergistic effect on the change of depression-like behavior associated with low salt intake in mice.展开更多

Rats were shamoperated (Sh) or pinealectomized (Px) and following subgroups were selected 1) Px/NaCl (0.9%) and Sh/NaCl (0.9%) : rats injected subcutaneously, once daily for 8 weeks, with saline solution NaCl (0.9%) as vehicle;2) Px/Mel (4 mg/Kg) and Sh/Mel (4 mg/Kg): Rats similarly injected with 4 mg/Kg of Mel. All animals were housed under a photoperiod of (LD:16/8). After different treatments animals were tested in the open-field test (OFT), elevated plus maze test (EPM) to determine anxiety-like behavior, and forced swimming test (FST) to evaluate depressive-like level. Our results revealed that level of anxiety-like and depressive-like behavior are significantly higher in Px/NaCl (0.9%) when compared to Sh/NaCl (0.9%) group, suggesting that pinelectomy induced an anxiogenic and depressant effects. The Px effects would be due to the absence of endogenous Mel synthesis and release. Additionally, we clearly demonstrated that the level of anxiety-like and depressive-like behavior are higher in Px/Mel (4 mg/Kg) and Sh/Mel when compared respectively to Px/NaCl (4 mg/Kg) and Sh/NaCl groups suggesting an anxiolytic and antidepressant effects of exogenous Mel. Behavioral responses were sex dependent since the difference between females and males, especially, after melatonin administration, were statistically significant. These experiments provide evidence that pinealectomy and Mel regulated emotionally behavior in male and female rats.展开更多

Methods In the present study, a chronic cerebral hypoperfusion model was established by perma- 2VO) in rats. Two weeks after 2VO, GAB- nent bilateral common carotid arteries occlusion (two-vessel occlusion, AB receptor agonist baclofen (25 mg · kg^-1 . d^-1 i p ) was administrated for 21 days. The FST was performed to evaluate depressive-like behavior in which the immobility time was recorded. In addition, the expression of brain derived neurotrophic factor (BDNF) in hippocampal CA1 was measured by Western blot. Results The immobility time of 2VO group was significantly prolonged and the expression of BDNF was decreased by 28.95% compared with sham group. After activation of GABAB receptors by baclofen, the immobility time was significantly reduced and the expression of BDNF was increased by 47.91% compared with 2VO group. Conclusion 2VO induced ob- vious depressive-like behavior in rats. Activation of GABAB receptors alleviates the depressive-like behavior in- duced by chronic cerebral ischemia via upredulation of BDNF in hippocampal CA1 in rats.展开更多

文摘Objective Long noncoding RNAs(lncRNAs)and microRNAs(miRNAs)are widely expressed in the brain and are associated with the development of neurological and neurodegenerative diseases. However, their roles and molecular mechanisms in major depressive disorder(MDD)remain largely unknown. This study aimed to identify lncRNAs and miRNAs involved in the development of MDD and elucidate their molecular mechanisms. Methods Transcriptome and bioinformatic analyses were performed to identify miRNAs and lncRNAs related to MDD.C57 mice were subjected to chronic unpredictable mild stress(CUMS)to establish a depression model.Lentiviruses containing either lncRNA NPTN-IT1-201 or miR-142-5p were microinjected into the hippocampal region of these mice.Behavioral tests including the sucrose preference test(SPT),tail suspension test(TST),and forced swim test(FST)were conducted to evaluate depressive-like behaviors. Results The results revealed that overexpression of lncRNA NPTN-IT1-201 or inhibition of miR-142-5p significantly ameliorated depressive-like behaviors in CUMS-treated mice.Dual-luciferase reporter assays confirmed interactions between miR-142-5p with both brain-derived neurotrophic factor(BDNF)and NPTN-IT1-201.ELISA analysis revealed significant alterations in relevant biomarkers in the blood samples of MDD patients compared to healthy controls.Histological analyses, including HE and Nissl staining, showed marked structural changes in brain tissues following CUMS treatment, which were partially reversed by lncRNA NPTN-IT1-201 overexpression or miR-142-5p inhibition.Immunofluorescence imaging demonstrated significant differences in the levels of BAX,Bcl2,p65,Iba1 among different treatment groups.TUNEL assays confirmed reduced apoptosis in brain tissues following these interventions.Western blotting showed the significant differences in BDNF,BAX,and Bcl2 protein levels among different treatment groups. Conclusion NPTN-IT1-201 regulates inflammation and apoptosis in MDD by targeting BDNF via miR-142-5p,making it a potential therapeutic target for MDD.
文摘Low salt intake is associated with depression, but the experimental evidence is not clear. Furthermore, the seasonal splitting from winter to spring may also have a differential synergistic effect on the change of depression-like behavior associated with low salt intake in mice.
文摘The main objective of this work was to 1) study the influence of endogenous melatonin (Mel) abolishment via pinealectomy and 2) explore exogenous Mel effect on anxiety-like and depressive-like behavior in male and female rats. These experiments provide evidence that pinealectomy and Mel regulated emotionally behavior in male and female rats.
文摘Aim To investigate the mechanisms underlying depressive-like behavior induced by chronic cerebral is- chemia in rats. Activation of GABAB receptors alleviates the depressive-like behavior in- duced by chronic cerebral ischemia via upredulation of BDNF in hippocampal CA1 in rats.
文摘OBJECTIVEα-Dstroglycan(α-DG)is a predominant component in the dystrophin-glycoprotein complex(DGC)and a recently char⁃acterized receptor for several extracellular matrix components with high affinity.Recent research⁃es have reported that hypoglycosylation ofα-DG is associated with the pathophysiology of diseas⁃es,especially muscular dystrophy, but little is known about major depressive disorder(MDD).Like-acetylglucosaminyl transferase(Large)is a key enzyme for glycosylation ofα-DG,which mainly modifies two points in the middle domain ofα-DG:Thr-317 and Thr-319.Glycosylatedα-DG(GLY-α-DG)can bind with high affinity to extracellular matrix(ECM)molecules that con⁃tain laminin globular(LG)domains, including per⁃lecan, agrin and neurexin.Agrin is mainly derived from neurons rather than glial cells. In cultured hippocampal neurons,it was found that agrin could regulate the homeostatic plasticity of inhibi⁃tory neurons by acting on GLY-α-DG.Mdx mice are transgenetic models for the investigation of Duchenne muscular dystrophy. Many studies have shown that the expression of GLY-α-DG in the peripheral and brain tissues of Mdx mice is significantly down-regulated.Mdx mice show cognitive impairment and high levels of anxiety. In this study,we employed chronic social defeat stress(CSDS)to establish an animal model of depression and detected the expression of GLY-α-DG among the brain areas associated with the pathophysiology of depression.METHODS So⁃cial interaction test(SIT)and sucrose preference test(SPT)were used to evaluate depressive-like behavior.Open field(OF)and elevated plus maze(EPM)test were used to determine the anxiety-like behavior of Mdx mice.Novelty-sup⁃pressed feeding test(NSFT)forced swim test(FST)and tail suspension test(TST)were used to detect the depressive-like behavior of Mdx mice.Novel object recognition test(NOR)was applied to evaluate the cognition of Mdx mice.Subthreshold social defeat stress was used to explore the susceptibility to stress in Mdx mice.Stereotactic infusion of agrin into the ventral hippocampus(vHip),FST and TST were used to investigate the antidepressant effects of agrin.Adeno-associated virus(AAV)-mediated overex⁃pression techniques, behavior tests and whole-cell path-clamp technique were conducted to determine the impact of Large overexpression on CSDS susceptible mice.RESULTS The expres⁃sion ofα-DG and GLY-α-DG were significantly decreased in the vHip of CSDS susceptible mice.Mdx mice showed decreased expression of GLY-α-DG and increased anxiety-like behav⁃iors.Mdx mice displayed some depressive-like behaviors, and the susceptibility to stress was significantly increased.Downregulation of the expressionα-DG in the vHip by lentivirus increased the susceptibility to stress.Administra⁃tion of agrin to CSDS susceptible mice exerted antidepressant effects, and this effect could par⁃tially sustain for a week. The expression of Large was decreased in vHip.Overexpression of Large through AAV-Large reversed the depressive-like behaviors and restored the decreased frequency and amplitude of mIPSC.CONCLUSION GLY-α-DG and its glycosylase are significantly decreased in CSDS susceptible mice.Adminis⁃tration of agrin and overexpression of Large displays antidepressant effect, which may be related to its promotion of inhibitory synaptic transmission.
文摘OBJECTIVE Alcohol dependence is not only the most common public health prob⁃lem worldwide, but also the cause of many alco⁃hol-related diseases, such as anxiety and depres⁃sion. However, the mechanism of these comor⁃bidities is not clear.Recently, the role of phos⁃phodiesterases(PDEs)in anxiety-and depres⁃sion-like behavior induced by alcohol withdrawal has received increasing attentions.PDEs are a superfamily of enzymes that catalyze the hydroly⁃sis of intracellular cyclic AMP(cAMP)and(or)cyclic GMP(cGMP).There are 11 families(PDE1-PDE11),each of which has 1 to 4 sub⁃types encoded by different genes. In the present study,we identified the key PDEs involved in anxiety-and depression-like behavior induced by alcohol withdrawal.METHODS After withdrawal from short-term or chronic intermittent ethanol vapor exposure(STEVE and CIEVE,respectively),adult male C57BL/6J(B6)mice were tested for anxiety-and depression-like behavior using the sucrose preference, open field, elevated plus-maze,tail suspension, forced swimming, hole board and light-dark tests. In addition, the expression of PDE subtypes(PDE2,PDE4,and PDE7)in the hippocampus, amygdala, and stria⁃tum, which are considered as anxiety-and(or)depression-related brain regions, was detected by Western blotting.RESULTS①Differences in anxiety-and depression-like behaviors between STEVE and CIEVE mice.Compared to STEVE mice,CIEVE mice showed greater anxiety-and depression-like behavior in the tests described above.②The expression of PDE2,PDE4 and PDE7 in the anxiety-and depression-related brain regions of CIEVE mice.Compared to the STEVE mice,CIEVE mice showed significant higher expression of PDE4A and PDE7A in the hippo⁃campus(P<0.05)and PDE4D and PDE7A in the amygdala(P<0.05).However,the expres⁃sion of PDE2A,PDE4B,PDE4D and PDE7B in the hippocampus,PDE2A,PDE4A,PDE4B and PDE7B in the amygdala,PDE2A,PDE4A,PDE4B,PDE4D,PDE7A and PDE7B in the striatum was not significantly changed.CONCLUSION The model of CIEVE showed significant anxiety-and depression-like behavior, with concurrent increases in the expression of PDE4A and PDE7A in the hippocampus and PDE4D and PDE7A in the amygdala. These results suggest that PDE4A,PDE4D,and DE7A may play an important role in the regulation of anxiety-and depression-like behavior induced by alcohol withdrawal.

文摘Olive flounder(Paralichthys olivaceus),a marine demersal flatfish species, exhibits obvious timid and depressive-like characteristics.Adult olive flounder bury themselves in the sand and present motionless behavior. However, the detail mechanism of regulating the motionless behavior is not fully understood. In this study, using adult group(AG)and juvenile group(JG)flounder as a model,we compared the locomotor activity of AG and JG flounder, and analyzed the transcriptomic expression profiles in the spinal cord by RNA-seq methods. The behavioral experiment showed that AG flounder displayed lower locomotor activity.Transcriptome analysis found a total of 1711 significantly differential expressed genes(DEGs),among which 818 genes were up-regulated and 893 genes were down-regulated in AG flounder compared with JG flounder.Specifically, locomotor behavior related genes(parvalbumin, urotensin Ⅱ and creatine kinase)were down-regulated.Meanwhile glial fibrillary acidic protein, the biomarkers of astroglial pathology, and glutamatergic synaptic genes were significantly up-regulated in AG flounder. In addition,Gene Ontology(GO)enrichment analysis revealed that DEGs enriched in peptide biosynthetic process, translation procession were down-regulated in the AG flounder. The Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis revealed that ribosome, oxidative phosphorylation and glutamatergic synapse pathway were over-represented.After treatment with antagonist of glutamate receptor LY341495,flounder more often swam out of the sand and displayed significantly more off-bottom swimming behavior, suggesting the anti-depressive effect of LY341495.Taken together,in combination with behavioral tests and transcriptomic analyses,we conclude that the glutamate receptor in spinal cord plays an important role in timid and depressive-like behavior in flounder.

文摘Short-chain fatty acids(SCFAs)have been increasingly evidenced to be important bioactive metabolites of the gut microbiota and transducers in controlling diverse psychiatric or neurological disorders via the microbiota-gut-brain axis. However, the precise mechanism by which brain SCFAs extert multiple beneficial effects is not completely understood. Our previous research has demonstrated that the acetyl-coenzyme A synthetase short-chain family member 2(ACSS2)is a novel target of the rapid and long-lasting antidepressant responses. Here,we show that micromolar SCFAs significantly augment both total cellular and nuclear ACSS2 to trigger tryptophan hydroxylase 2(TPH2)promoter histone acetylation and its transcription in SH-SY5Y cells. In chronic-restraint-stress-induced depression mice, neuronal ACSS2 knockdown by stereotaxic injection of adeno-associated virus in the hippocampus abolished SCFA-mediated improvements in depressive-like behaviors of mice, supporting that ACSS2 is required for SCFA-mediated antidepressant responses.Mechanistically, the peroxisome-proliferator-activated receptor gamma(PPARγ)is identified as a novel partner of ACSS2 to activate TPH2 transcription.Importantly,PPARγis also responsible for SCFA-mediated antidepressant-like effects via ACSS2-TPH2 axis.To further support brain SCFAs as a therapeutic target for antidepressant effects,d-mannose,which is a naturally present hexose, can significantly reverse the dysbiosis of gut microbiota in the chronic-restraint-stress-exposure mice and augment brain SCFAs to protect against the depressive-like behaviors via ACSS2-PPARγ-TPH2 axis.In summary, brain SCFAs can activate ACSS2-PPARγ-TPH2 axis to play the antidepressive-like effects, and d-mannose is suggested to be an inducer of brain SCFAs in resisting depression.

文摘Studies on social isolation have provided evidence for an enhanced anxiety-like phenotype,depressive-like symptoms and higher levels of 22-kHz ultrasonic vocalizations(USVs);whereas sexual experience could induce significant changes in affective states, decrease anxiety-and depressive-like responses. However, the behavioral and emotional alterations in male rats exposed to isolation after sexual experience remain poorly understood. Therefore, the present study examined anxiety-and depressive-like parameters,as well as USV emissions in male rats isolated for 1,2,or 7 d following 1-week sexual(male-female paired housing)or social(male-male paired housing)interaction.Isolation for 1 d resulted in increased depressive-like symptoms, and 2-d isolation induced elevated anxiety-like behavior in sexually inexperienced rats, while rats with sexual experience exhibited comparable levels of emotionality at each isolation time. Moreover, isolated rats without sexual experience emitted less 22-and 50-kHz USVs,but sexually experienced rats exhibited similar affective states compared with control ones. These results suggested that prior sexual experience could have a modulatory effect on experimental rat emotional responses to isolation.
文摘Behavior recognition of Hu sheep contributes to their intensive and intelligent farming.Due to the generally high density of Hu sheep farming, severe occlusion occurs among different behaviors and even among sheep performing the same behavior, leading to missing and false detection issues in existing behavior recognition methods.A high-low frequency aggregated attention and negative sample comprehensive score loss and comprehensive score soft non-maximum suppression-YOLO(HLNC-YOLO)was proposed for identifying the behavior of Hu sheep, addressing the issues of missed and erroneous detections caused by occlusion between Hu sheep in intensive farming.Firstly, images of four typical behaviors-standing,lying,eating,and drinking-were collected from the sheep farm to construct the Hu sheep behavior dataset(HSBD).Next,to solve the occlusion issues, during the training phase, the C2F-HLAtt module was integrated, which combined high-low frequency aggregation attention, into the YOLO v8 Backbone to perceive occluded objects and introduce an auxiliary reversible branch to retain more effective features.Using comprehensive score regression loss(CSLoss)to reduce the scores of suboptimal boxes and enhance the comprehensive scores of occluded object boxes. Finally, the soft comprehensive score non-maximal suppression(Soft-CS-NMS)algorithm filtered prediction boxes during the inferencing.Testing on the HSBD,HLNC-YOLO achieved a mean average precision(mAP@50)of 87.8%,with a memory footprint of 17.4 MB.This represented an improvement of 7.1,2.2,4.6,and 11 percentage points over YOLO v8,YOLO v9,YOLO v10,and Faster R-CNN,respectively.Research indicated that the HLNC-YOLO accurately identified the behavior of Hu sheep in intensive farming and possessed generalization capabilities, providing technical support for smart farming.
文摘To investigate the influence of Al-Zn-Mg-Cu alloy with as-homogenized and as-rolled initial microstructures on the tensile flow behavior, isothermal tensile tests were conducted on a GLEEBLE-3500 isothermal simulator at temperatures of 380-440℃and strain rates of 0.05-1 s^(−1).The Johnson-Cook model,Hensel-Spittel model,strain-compensated Arrhenius model, and critical fracture strain model were established. Results show that through the evaluation of the models using the correlation coefficient(R)and the average absolute relative error, the strain-compensated Arrhenius model can represent the flow behavior of the alloy more accurately.Shear bands are more pronounced in the as-homogenized specimens, whereas dynamic recrystallization is predominantly observed in as-rolled specimens.Fracture morphology analysis reveals that a mixed fracture mechanism is prevalent in the as-homogenized specimen, whereas a ductile fracture mechanism is predominant in the as-rolled specimen. The processing maps indicate that the unstable region is reduced in the as-rolled specimens compared with that in the as-homogenized specimens. The optimal hot working windows for the as-homogenized and as-rolled specimens are determined as 410-440℃/0.14-1 s^(−1)and 380-400℃/0.05-0.29 s^(−1),respectively.

文摘Residential energy-use behavior and energy-saving awareness play a crucial role in sustainable urban energy planning and building energy efficiency, particularly under the pressures of climate change. However, existing studies often lack comparative analysis of urban-rural differences and tend to focus excessively on behavior patterns while neglecting the dimension of energysaving awareness. With China’s urbanization rate reaching 66.16%,understanding such regional disparities has become increasingly important. To address these research gaps, this study conducts a large-scale survey on space cooling behaviors among residents in Beijing,a representative Chinese megacity. It should be noted that living standards in such megacities are generally higher than the national average, which may shape distinctive energy-use profiles.Analyzing 1573valid samples(1064 urban/442 rural)in 2024,this study employed K-Prototypes and K-Modes clustering to identify typical cooling behavior and energy-saving awareness pattems, followed by Kendall/Chi-square correlation tests and XGBoost importance analysis to determine key influencing factors, with subsequent urban-rural comparative analysis. Results indicate that urban residents are primarily heat-sensitive or heat-tolerant,with a secondary patten of mid-low temperature preference, and generally exhibit long cooling durations;rural behavior is dominated by heat-tolerant type, followed by heat-sensitive,mid-low temperature preference, and never-on types as secondary patterns;both urban and rural areas exhibit energy-savingawareness characterized by low consumption-lowwillingness,though urban areas show marginally higher motivation;energy-saving awareness correlates with cooling behavior in rural areas, but this relationship weakens significantly in urban contexts.

文摘This study utilizes wet/dry cyclic corrosion testing combined with corrosion big data technology to investigate the mechanism by which chloride ions(Cl^(-))influence the corrosion behavior of 650 MPa high-strength low-alloy(HSLA)steel in industrially polluted environments. The corrosion process of 650 MPa HSLA steel occurred in two distinct stages:an initial corrosion stage and a stable corrosion stage.During the initial phase, the weight loss rate increased rapidly owing to the instability of the rust layer.Notably, this study demonstrated that 650 MPa HSLA steel exhibited superior corrosion resistance in Cl-containing environments. The formation of a corrosion-product film eventually reduced the weight-loss rate. However, the intrusion of Cl^(-)at increasing concentrations gradually destabilized theα/γ^(*)phases of the rust layer, leading to a looser structure and lower polarization resistance(R_(p)).The application of corrosion big data technology in this study facilitated the validation and analysis of the experimental results, offering new insights into the corrosion mechanisms of HSLA steel in chloride-rich environments.

文摘The volume change behavior of natural gas hydrate-bearing sediment is essential as it influences settlement, strength, and stiffness, which directly affect the stability of hydrate reservoirs during hydrate extraction or in response to environmental changes. The volume change is influenced not only by stress but also by the formation and dissociation of hydrates. This study adopted a customized apparatus for one-dimensional compression tests, allowing independent control of gas pressure and effective stress.Tests were conducted on samples with different hydrate saturations along various temperature-gas pressure-effective stress paths, yielding some conclusions related to compressibility and creep. An unusual phenomenon was observed under low-stress conditions:hydrate formation led to shrinkage rather than expansion.Three potential mechanisms behind this occurrence were discussed. As hydrate saturation increases, the yield stress rises while the compression and swelling indexes remain minimally affected. After hydrate dissociation, the compression curve of hydrate-bearing sediment drops to that of hydrate-free sediment.Once hydrate is formed, the compression curve of hydrate-free sediment gradually approaches that of hydrate-bearing sediment during the subsequent loading.Under low-stress conditions, the creep of both hydrate-free and hydrate-bearing sediments is very weak. However, when stress increases, significantly beyond the yield stress, the creep of both sediments increases significantly, with hydrate-bearing sediment exhibiting much greater creep than hydrate-free sediment.

文摘Drilling and blasting tunneling is a cyclic process in which tunnel rock undergoes repeated blast loading, affecting its dynamic characteristics, energy evolution, and damage progression. To explore the dynamic mechanical properties and damage mechanisms of carbonaceous slate under cyclic impact loads of varying intensities, cyclic dynamic tests are conducted using a triaxial split Hopkinson pressure bar. This study analyzes the stress-strain relationship, energy damage evolution, and macro-to-micro failure characteristics. The results show that peak stress and strain are significantly influenced by impact intensity and the number of impacts. The initial dynamic stress is positively correlated with the impact intensity, but with more impact, the dynamic stress decreases while the peak strain increases.Energy evolution follows a pattern of"slow growthfluctuating growthrapid growth,"with the crack initiation stress and its proportion decreasing.CT and SEM analyses reveal that as the impact intensity increases, failure becomes more chaotic, the fracture volume increases, and the fracture mode shifts from interlayer and intergranular to through-layer and trans-granular fractures. These findings provide an experimental basis for soft rock tunnel stability analysis.
文摘Objectives:24-h movement behaviors(24-HMB),encompassing physical activity, sedentary behavior, and sleep duration, are increasingly regarded as interrelated and important factors for mental health. However, evidence on the comprehensive association of these behaviors with mental health in adults with diabetes in developing countries remains scarce. This study examined the association between 24-HMB guidelines and psychological health among adults with diabetes in developing countries. Methods:Data were retrieved from the World Health Organization’s study on Global Aging and Adult Health Survey dataset.Adults(N=1905)diagnosed with diabetes from five low-and middle-income countries were included. The exposure of interest was adherence to 24-HMB guidelines, depression, cognition, and quality of life(QoL).Multiple logistic and multiple linear regression analyses were used to examine the association between meeting 24-HMB guidelines and depression, cognition, and QoL,respectively. Results:This cross-sectional study revealed that 28.61%complied with all three 24-HMB guidelines.Diabetic patients who met more numbers of 24-HMB guidelines had lower depression risk(OR=0.74,95%CI:0.61 to 0.91,p=0.004),greater cognition(β=0.42,95%CI:0.25 to 0.60,p<0.001),and QoL(β=1.30,95%CI:1.04 to 1.55,p<0.001)with the non-compliant population. For specific combinations, meeting all three guidelines were significantly associated with lower odds of depression, improved cognitive function, and enhanced QoL(all p<0.001).Conclusion:These findings support that meeting 24-HMB guidelines in a single or combined movement behaviors was significantly related to reduced risk of depression, enhanced cognitive function, and improved QoL among individuals with diabetes.

文摘Significant differences exist between deep and medium-shallow coalbed methane(CBM)reservoirs. The unclear understanding of flowback and production behavior severely constrains the development of deep CBM resources. To address this challenge, guided by the gas-liquid two-phase flow theory in ultra-low permeability reservoirs, and integrating theoretical analysis, numerical simulation, and insights from production practices, this study classifies the flowback and production stages of deep CBM well considering the Daning-Jixian Block,Eastern Ordos Basin as a representative case.We summarize the flowback characteristics for each stage and establish a standard flowback production type curve, aiming to guide field operations. The results indicate that:(a)The production process of deep CBM horizontal wells can be divided into five distinct stages:initial single-phase water dewatering stage, initial gas appearance to peak water production stage, gas breakthrough to peak gas production stage, stable production and decline stage, and low-rate production stage.(b)Based on reservoir energy, two standard type curves for horizontal well flowback production are established:the‘Sufficient Reservoir Energy’type and the‘Insufficient Reservoir Energy’type. The former achieves a higher initial gas rate(up to 12×10^(4)m^(3)/d)but exhibits poorer stability, while the latter achieves a lower stable rate(up to 8×10^(4)m^(3)/d)but demonstrates stronger stability.Numerical simulation confirms these behavioral patterns and reveals the underlying mechanisms related to the effectively drained area where pressure is significantly depleted. The findings from this study have guided the flowback production operations in 53 deep CBM wells with positive results, demonstrating high potential for broad application.
文摘BACKGROUND Despite societal guidelines recommending targeted screening for Barrett’s esophagus(BE)and esophageal adenocarcinoma(EAC)in individuals with gastroesophageal reflux symptoms(GERS),screening adherence is suboptimal.Current screening approaches fail to identify individuals not seeking medical consultation for GERS or whose GERS are managed with‘over-the-counter’(OTC)acid suppressant therapies.AIM To assess patients’self-management and help-seeking behavior for GERS.METHODS This cross-sectional study collected data from the Dutch general population aged 18-75 years between January and April 2023 using a web-based survey. The survey included questions regarding self-management(e.g.,use of acid suppressant therapy with or without prescription)and help-seeking behavior(e.g.,consulting a primary care provider)for GERS.Simple random sampling was performed to select individuals within the target age group. In total,18156 randomly selected individuals were invited to participate. The study protocol was registered in ClinicalTrials.gov(identifier:NCT05689918).RESULTS Of the 18156 invited individuals,3214 participants(17.7%)completed the survey,of which 1572 participants(48.9%)reported GERS.Of these,904 participants(57.5%)had never consulted a primary care provider for these symptoms,of which 331 participants(36.6%)reported taking OTC acid suppressant therapy in the past six months and 100 participants(11.1%)fulfilled the screening criteria for BE and EAC according to the European Society of Gastrointestinal Endoscopy Guideline.CONCLUSION The population fulfilling the screening criteria for BE and EAC is incompletely identified, suggesting potential underutilization of medical consultation.Raising public awareness of GERS as a risk factor for EAC is needed.
