METHODS The FD with liver depression-spleen deficiency syndrome rat model was established through classic chronic mild unpredictable stimulation every day. XYS group rats received XYS 1 h before the stimulation. The models were assessed by parameters including state ofthe rat, weight, sucrose test result and open-field test result. After 3 wk, the stools of rats were collected and genomic DNA was extracted. PCR products of the V4 region of 16 S rD NA were sequenced using a barcoded Illumina paired-end sequencing technique. The primary composition of the microbiome in the stool samples was determined and analyzed by cluster analysis.RESULTS Rat models were successfully established, per data from rat state, weight and open-field test. The microbiomes contained 20 phyla from all samples. Firmicutes, Bacteroidetes, Proteobacteria, Cyanobacteria and Tenericutes were the most abundant taxonomic groups. The relative abundance of Firmicutes, Proteobacteria and Cyanobacteria in the model group was higher than that in the normal group. On the contrary, the relative abundance of Bacteroidetes in the model group was lower than that in the normal group. Upon XYS treatment, the relative abundance of all dysregulated phyla was restored to levels similar to those observed in the normal group. Abundance clustering heat map of phyla corroborated the taxonomic distribution. CONCLUSION The microbiome relative abundance of FD rats with liver depression-spleen deficiency syndrome was significantly different from the normal cohort. XYS intervention may effectively adjust the gut dysbacteriosis in FD.展开更多

METHODS: A detailed MEDLINE search was carried out to identify all articles and book chapters in English published from January 1995 to January 2015. RESULTS: Based on the main findings, the prevalence of major depressive disorder(MDD) ranged from 14.0% to 27.2%. Furthermore, the prevalence of suicidal ideation varied from 13.6% to 31.0% whereas, attempted suicides were reported to range from 3.9% to 32.7%. Interestingly, various associated risk factors for both depression and suicide were identified in HIV patients. Finally, consistent associations were reported among MDD, suicidal ideation, and poor quality of life in individuals living with HIV. CONCLUSION: Although additional studies are needed to elucidate this complex association, our results suggest the importance of early detection of both MDD and suicidality in patients living with HIV.展开更多

[Methods] A syndrome score scale that was suitable for evaluating the rat models was established according to the evolvement rules of etiology and pathogenesis of TCM and the modern clinical pathological mechanism. At the same time, bifendate pills and Sanqi Zhigan pills were selected as drug counterevidence for the models. Rats in model groups were given different proportions of high-fat and low-protein fodder and different concentrations of alcohol every day,and intraperitoneally injected with porcine serum twice a week. Drug groups were given with bifendate pills and Sanqi Zhigan pills( 8. 1 mg/kg,and 2. 7 g/kg) respectively for 14 consecutive days.During the experiment, general status, weight, daily fodder intake and daily water intake of the animals were observed, and TCM syndromes were scored. After the experiment, the levels of alanine aminotransferase( ALT),aspartate aminotransferase( AST),γ-glutamine transpeptidase( γ-GT),total cholesterol( TC),triglycerides( TG),low density lipoprotein-cholesterol( LDL-C),and high density lipoprotein-cholesterol( HDL-C) in serum were detected, and the pathological changes in liver tissue were observed. [Results] Compared with the control group, body weight, daily fodder and water intake of rats increased slowly in the model group, and the levels of ALT,AST,γ-GT,TC,TG and LDL-C increased significantly, while the level of HDL-C dropped. Pathological examination showed that steatosis and fat granule were observed in hepatocytes of rats in the model group. Behavioral observation found that main symptoms and minor symptoms of rats in the model group conformed to the syndrome manifestation of liver depression and spleen deficiency, which suggested that the three model groups were established successfully. Among them, three rats died in model group 1,and one rat died in model group 2. The manifestation of all the above lesions can be alleviated by drug counterevidence. [Conclusions]The animal model of fatty liver due to liver depression and spleen deficiency shows an obviously lower mortality and shorter duration and can be used for research of correlation between prescription and syndrome and its mechanism.展开更多

文摘AIM To investigate gut microbial diversity and the interventional effect of Xiaoyaosan(XYS) in a rat model of functional dyspepsia(FD) with liver depression-spleen deficiency syndrome. XYS intervention may effectively adjust the gut dysbacteriosis in FD.

文摘Objective:To investigate the biological basis of“depression with liver-qi stagnation and spleen deficiency syndrome”.Methods:A digital gene expression profiling method was conducted to explore global changes in the mRNA transcriptome in a rat model of depression with liver-qi stagnation and spleen deficiency syndrome.Real-time quantitative polymerase chain reaction(q-PCR)was performed to verify the five genes most interest based on the Kyoto Encyclopedia of Genes and Genome(KEGG)analysis.Sini San,which disperses stagnated liver qi and strengthens the spleen, was administered to the model rats to observe whether it could reverse these genetic changes in the liver.Results:Forty-six differentially expressed genes were identified.Three of the five genes of most interestdHnf4a,Hnf4g and Cyp1a1dbased on KEGG analysis, were confirmed by realtime q-PCR.Sini San reduced the gene expression changes of Hnf4a,Hnf4g and Cyp1a1 in the rat model.Conclusions:Hnf4a,Hnf4g and Cyp1a1 are involved in“depression with liver-qi stagnation and spleen deficiency syndrome”.These findings indicate that depressed rats with liver-qi stagnation and spleen deficiency syndrome are at risk of liver diseases.Furthermore, our results will inform exploration of the etiology of depression and help in the development of effective therapeutic strategies.
文摘AIM: To investigate the potential associations among major depression, quality of life, and suicidal behavior in human immunodeficiency virus(HIV) patients. CONCLUSION: Although additional studies are needed to elucidate this complex association, our results suggest the importance of early detection of both MDD and suicidality in patients living with HIV.

文摘[Objectives] To establish animal models of fatty liver due to liver depression and spleen deficiency that were suitable for activity discovery of traditional Chinese medicine( TCM) compounds, efficacy evaluation, new drug research and development and correspondence between prescription and syndrome of TCM. [Conclusions]The animal model of fatty liver due to liver depression and spleen deficiency shows an obviously lower mortality and shorter duration and can be used for research of correlation between prescription and syndrome and its mechanism.

文摘Objective:This study was designed to explore the characteristics of gut microbiota in CHB patients with two most common traditional Chinese medicine(TCM)Syndromes—damp heat syndrome and liver depression and spleen deficiency syndrome. Methods:According to the diagnostic criteria of Western medicine,TCM and screening exclusion criteria,65 cases of CHB with damp heat syndrome and 28 cases of CHB with liver depression and spleen deficiency syndrome were finally included in the study. All the basic information was gathered and the fresh fecal samples were collected for 16S rDNA sequencing.16S rDNA of gut microbiota was sequenced using Illumina hiseq 2,500 high-throughput sequencing platform.Based on the optimized sequence,Operational Taxonomic Units(OTU)clustering analysis and taxonomic annotation were carried out. Results:The difference in relative abundance of gut microbiota was significant between damp heat syndrome and liver depression and spleen deficiency syndrome in CHB patients.Cyanobacteria was only found in damp heat syndrome. The relative abundance of Erysipelotrichia and Subdoligranulum were higher in liver depression and spleen deficiency syndrome, while the relative abundance of Rhodospirillales,Alphaproteobacteria and Lachnospira were higher in the damp heat syndrome.LDA Effect Size(LEfSe)analysis showed that Lachnospira,Olsenella and Subdoligranulum had significant difference in species among the two TCM syndromes. Conclusion:The different characteristics of gut microbiota in the two TCM syndromes of CHB patients may play an important role in syndrome formation of TCM,which provides a new field of vision for the accurate diagnosis and treatment of TCM.
文摘BACKGROUND Many human immunodeficiency virus(HIV)infected patients suffer from depression, but a little focus is given to detecting and treating depression in primary health care.Detection of depression can be improved by introducing short, reliable, and valid screening instruments.AIM To determine the psychometric properties of the patient health questionnaire-2(PHQ-2)and patient health questionnaire-9(PHQ-9)for depression screening and diagnosis, and the sensitivity and specificity of the PHQ-2 in HIV infected patients.METHODS A cross-sectional study was conducted on 158 HIV-infected patients aged 18 years and above in Lahore,Pakistan.PHQ-2 was implemented to screen depression.PHQ-9 was implemented to diagnose major depressive disorder as a reference standard.Reliability,Validity tests and receiver operating characteristic curve were computed.RESULTS The Cronbach's alpha of PHQ-2 and PHQ-9 were 0.732 and 0.759,respectively.The study results showed that the score of 2 on PHQ-2 indicates the highest Youden's index of 0.924,with both sensitivity and specificity of 0.96,and the area under the curve for PHQ-2 was 0.98(95%CI:0.953-0.998).CONCLUSION Good psychometric properties for the PHQ-2 and PHQ-9 indicated their significant potential as tools for depression screening and diagnosis in the HIV-infected population.
文摘Antidepressants are the main drugs used to treat depression, but they have not been shown to be effective in the treatment of child and adolescent depression. However, many adolescent depression treatment guidelines still recommend the use of antidepressants, especially specific serotonin re-uptake inhibitors.Previous studies have suggested that antidepressants have little therapeutic effect but many side effects, such as switching to mania, suicide, and non-suicidal self injury(NSSI),in the treatment of child and adolescent depression. In the process of developing guidelines, drug recommendations should not only focus on impro-ving symptoms, but they should also consider potential side effects. This review discusses the serious side effects of antidepressants, including switching to mania, suicide, and NSSI.
文摘BACKGROUND Atypical depression is an important indicator of a high risk of bipolar disorder and a genetic predisposition to immunometabolic traits.AIM To analyze common depression assessment scales for their inclusion of items related to atypical symptoms such as mood reactivity, hypersomnia, increased appetite(or weight gain),leaden paralysis, and interpersonal sensitivity.METHODS A search for English-language articles was conducted without time restrictions in the MEDLINE and Russian Science Citation Index databases using the following keywords:“depression”OR“bipolar depression”AND“scales”OR“questionnaires”.The analytical method used in this review involved a descriptive analysis of the included studies.RESULTS After reviewing studies on the validation of depression assessment scales,we found that only a small number include items addressing both increases and decreases in appetite or weight,as well as variations in sleep duration. Moreover, only a few studies have evaluated mood reactivity, leaden paralysis, and interpersonal sensitivity. The most well-developed scale that considers all aspects of atypical and non-atypical depressions is the Inventory of Depressive Symptomatology.CONCLUSION Ignoring atypical symptoms in common scales can lead to underestimation of depression severity and inaccuracies in evaluating therapy effectiveness in clinical trials, as well as hinder fundamental research aimed at finding biomarkers.
文摘Following an acute stroke, patients often encounter a range of impairments affecting their physical, cognitive, verbal, and social capabilities.Poststroke depression(PSD)has been identified as a significant consequence of stroke and serves as a crucial predictor of patient outcomes. The diagnosis of PSD presents a challenge,as the physical manifestations following a stroke can overlap with certain depressive symptoms, leading to a potential underdiagnosis of this condition. This review employs a narrative approach to synthesize diverse findings within this domain.PSD is categorized as an organic mood disorder, and a more accurate diagnosis may be achieved by considering the location of the stroke, the patient's specific symptoms, and the timeline of depressive symptom onset.Notably,if depressive symptoms emerge at more than one year poststroke, the likelihood of PSD diminishes. The etiology of PSD is currently understood to stem from a combination of physiological and psychosocial factors,as well as their interactions.Several mechanisms associated with PSD have been identified, including inflammation, dysregulation of the hypothalamic-pituitary-adrenal axis, oxidative stress, autophagy, apoptosis, abnormal neurotrophic responses,glutamate-mediated excitotoxicity, dysfunction within brain networks, reduced monoamine levels, locations of brain lesions, and psychosocial influences. This review also outlines potential directions for future research.

文摘BACKGROUND Major depressive disorder(MDD)and bipolar depression(BD-D)are both intricate, enduring, and profound psychiatric conditions characterized primarily by depressive episodes and cognitive dysfunction. However, distinguishing the characteristics and influencing factors of cognitive impairment in unipolar and BD-D is crucial for identification and intervention.AIM To compare neurocognitive characteristics and investigate associations between cognitive function and clinical features in unipolar and BD-D.METHODS The THINC-integrated tool(THINC-it)as a cognitive assessment tool was applied to 295 individuals:75 patients with depressive disorders(MDD),120 individuals with BD-D,and 100 healthy controls. The Hamilton Depression Scale-17(HAMD),Hamilton Anxiety Scale-14(HAMA),and Pittsburgh Sleep Quality Index(PSQI)were employed to assess depression, anxiety, and sleep.Neurocognitive function characteristics and the relationships between cognitive impairment and general clinical attributes were analyzed.RESULTS There were no statistically significant differences in the overall THINC-it with each objective subscale. However, the subjective subscale(Perceived Deficits Questionnaire for Depression-5-item)showed significant differences between MDD and BD-D(P<0.001).Linear regression analyses were explored to determine associations.Age,years of education, age at onset, and HAMD were significantly co-associated with the overall THINC-it and each subscale in both MDD and BD-D(P<0.05).Furthermore,years of education showed a positive correlation with objective cognitive impairment(e.g.,Codebreaker,Trails)(P<0.05).There was a notable difference in that the number of depressive episodes, disease duration, hospitalizations,HAMA,and PSQI were significantly associated with the overall THINC-it with each subscale between MDD and BD-D(P<0.05).CONCLUSION Although both unipolar and BD-D showed similar objective cognitive impairments, there was a significant difference in subjective cognitive impairment. Our findings suggest that factors like age, years of education, age at onset, and depression severity might not be significantly difference in the influence of cognitive impairment.Furthermore,we found that education was a protective factor for cognitive impairment in both unipolar and BD-D.Our analysis revealed that distinct factors including disease duration, number of depressive episodes, hospitalizations, anxiety levels, and sleep quality influenced cognitive impairment between unipolar and BD-D.Therefore,it was important to investigate the specific characteristics of cognitive impairment and influencing factors to identify differentiating unipolar and BD-D.

文摘Zinc(Zn)is an essential micronutrient for both plants and humans.Zn deficiency is common in many paddy fields and reduces yield and grain Zn content(GZC).To accelerate breeding for improved GZC and Zn deficiency tolerance(ZDT)rice varieties,it is important to identify genes regulating Zn homeostasis. In this study,a member of the Ser/Thr protein phosphatase family,LOC_Os07g48840(named as OsGZ7),was found to contributed to ZDT and GZC in rice. The expression of OsGZ7 in roots and shoots was inhibited by Zn deficiency and toxicity, but induced by iron(Fe),manganese(Mn),and copper(Cu)deficiency,as well as chromium toxicity.OsGZ7 localized to the cytoplasm and was expressed in all tissues during the tillering, flowering, and grain-filling stages, particularly in the leaf blade and leaf sheath. At the seedling stage, knockout of OsGZ7 enhanced ZDT and increased Zn accumulation in both roots and shoots. At the maturity stage, knockout of OsGZ7 enhanced GZC,while overexpression of OsGZ7 reduced GZC.RNA-seq analysis suggested that OsGZ7 might regulate Zn homeostasis by affecting metal binding, hormone signal transduction, and oxidoreductase activity.Taken together, our findings indicate that OsGZ7 contributes to ZDT and Zn accumulation in rice.
