[Result] The full length of ADH cDNA is 1 088 bp containing a 771 bp of ORF, encoding 256 amino acids, with a calculated relative molecular weight of 30.80 kDa and a theoretical isoelectric point of 8.22. The deduced amino acid sequence shares the highest homology with Glossina morsitans morsitans based on homological analysis and phylogenetic analysis. All 22 tRNA genes show typical clover structures, with the exception of the tRNAser(AGn) which lacks the dihydrouridine (DHU) stem and is instead replaced by a simple loop. Thirteen intergenic spacers totaling 153 bp, and 13 overlapping regions totaling 46 bp are scattered throughout the whole genome. The 377 bp long olD. hyparete A+T-rich region is not comprised of large repetitive sequences, but harbors several features characteristic of the lepidopteran insects, including the motif ATAGA followed by an 18 bp poly-T stretch, a microsatellite-like (AT)s element preceded by the ATTTA motif, an 10 bp polyA-like stretch (AAAAATAAAA) present immediately upstream tRNAMet.展开更多

[ Method ] The full-length cDNA of Dorsal in D. antiqua was cloned through RACE. The similarity among deduced amino acid sequence of Dorsal cDNA and the Dorsals of other 14 insect species were compared, and the phylogenetic analysis of these Dorsals was conducted. The expression level of Dorsal gene in winter-, summer- and non-diapausing pupae was analyzed. [ Result] The full-length of Dorsal cDNA sequence, 2 412 bp, was obtained with ORF 1974 bp, which coded 657 amino acids with predicted Mw 72.9 kDa and PI 8.5. The result of similarity comparison indicated that the DaDorsal was most related to those of Drosophlia melanogaster and Drosophlia pseudoobscura. The result of semi-quantitative RT-PCR analysis showed the expression level of Dorsal gene increased in characteristic duration of winter-, summer- and non- diapansing pupae, especially at the late diapanse, which might imply its relationship to D. antiqua diapause and development. 【Aim】Insect diapause is a critical eco-physiological adaptation for seasonal survival,but little is known about the molecular mechanisms of summer and winter diapause of Delia antiqua.Here,our main goal is to identi...【Aim】Insect diapause is a critical eco-physiological adaptation for seasonal survival,but little is known about the molecular mechanisms of summer and winter diapause of Delia antiqua.Here,our main goal is to identify candidate genes linked to pupal diapause of D.antiqua.【Methods】A straightforward in silico approach was performed to predict the secreted diapause-related proteins based on the RNA-seq and digital gene expression datasets.Amino acid sequence similarity and basic physiochemical characteristics analysis were conducted.Gene expression levels of 12 selected genes from RNA-seq data were further validated in non-diapause pupae,and different developmental stages of summer and winter diapause pupae by qRT-PCR.【Results】A total of 38 putative secreted proteins were differentially regulated between the summer and winter diapause pupae of D.antiqua.Gene Ontology analysis revealed differentially expressed genes to be associated with odorant binding,chitin and lipid metabolic processes,innate immunity and development,etc.Among them,regulation of lipid mobilization and chitin-related metabolism was found to be important during diapause development.qRT-PCR analyses showed that the expression profiles of the 12 selected genes were in good agreement with results from RNA-seq(R=0.862,P=0.001).【Conclusion】This study establishes a robust pipeline for the discovery of diapause-related genes by secretome mining based on the high-throughput sequencing.展开更多

The onion fly,Delia antiqua,is a major underground agricultural pest that can enter pupal diapause in the summer and winter seasons.However,little is known about its molecular regulation due to the lack of genomic res...The onion fly,Delia antiqua,is a major underground agricultural pest that can enter pupal diapause in the summer and winter seasons.However,little is known about its molecular regulation due to the lack of genomic resources.To gain insight into the possible mechanism of summer diapause(SD),high-throughput RNA-Seq data were generated from non-diapause(ND)and SD(initial,maintenance and quiescence phase)pupae.Three pair-wise comparisons were performed and identified,1380,1471 and 435,and were significantly regulated transcripts.Further analysis revealed that the enrichment of several functional terms related to juvenile hormone regulation,cell cycle,carbon hydrate and lipid metabolism,innate immune and stress responses,various signalling transductions,ubiquitin-dependent proteosome,and variation in cuticular and cytoskeleton components were found between ND and SD and between different phases of SD.Global characterization oftranscriptome profiling between SD and ND contributes to the in-depth elucidation of the molecular mechanism of SD.Our results also offer insights into the evolution of insect diapause and support the importance of using the onion fly as a model to compare the molecular regulation events of summer and winter diapauses.展开更多
