Multifunctional tri-layer wound dressing containing ZnO nanoparticles and IGF-1 as an efficient biomaterial for healing of full thickness skin injuries

Mimicking the hierarchical structure of the skin is one of the most important strategies in skin tissue engineering.Monolayer wound dressings are usually not able to provide several functions at the same time and cannot meet all clinical needs.In order to maximize therapeutic efficiency,herein,we fabricated a Tri-layer wound dressing,where the middle layer was fabricated via 3D-printing and composed of alginate,tragacanth and zinc oxide nanoparticles(ZnO NPs).Both upper and bottom layers were constructed using electrospinning technique;the upper layer was made of hydrophobic polycaprolactone to mimic epidermis,while the bottom layer consisted of Soluplus■ and insulin-like growth factor-1(IGF-1)to promote cell behavior.Swelling,water vapor permeability and tensile properties of the dressings were evaluated and the Tri-layer dressing exhibited impressive antibacterial activity and cell stimulation following by the release of ZnO NPs and IGF-1.Additionally,the Tri-layer dressing led to faster healing of full-thicknesswound in ratmodel compared to monolayer and Bilayer dressings.Overall,the evidence confirmed that the Trilayer wound dressing is extremely effective for full-thickness wound healing. 展开更多

Effective application of an innovative acemannan-enriched glycolipid sphere dressing in diabetic foot ulcer wound healing:A case report and review of literature

BACKGROUND Aloe vera has been used as a traditional herbal therapy for wound management and dermatological conditions worldwide for thousands of years.Scientific evidence has confirmed that acemannan,the bioactive compound in aloe vera gel,exhibits significant anti-inflammatory and immunomodulatory properties that enhance tissue regeneration.This case report describes the successful application of an innovative acemannan-enriched glycolipid sphere dressing derived from aloe vera gel in diabetic foot ulcer(DFU)treatment,which achieved a clinically remarkable outcome.CASE SUMMARY An 80-year-old female patient with a 20-year history of type 2 diabetes mellitus experienced recurrent diabetic foot pain for 15 years.She had multiple hospitalizations due to acute infections and poorly controlled hyperglycemia.Long-term treatments included metformin and gliclazide.Upon presentation,she had a nonhealing wound on her left dorsal foot,diagnosed as a severe DFU(Texas classification:Grade II,stage D).She declined amputation and opted for conservative treatment.The medical team applied an acemannan-enriched glycolipid sphere dressing five times daily to the left calf and foot,avoiding the wound area.Frequency was reduced to three times daily after scab formation.Weight-bearing on the injured foot was avoided.Through in-person and online consultations,the team managed her lifestyle and diet,emphasizing natural foods.After 5 months,the DFU healed without significant scarring or functional loss.No recurrence was observed during the 2-year follow-up.Acemannan-enriched glycolipid sphere dressings promote DFU healing.This suggests the potential of these dressings for treating other refractory wounds. 展开更多

The healing of diabetic wounds poses a significant healthcare burden due to persistent inflammation,M1 macrophage aggregation,and high glucose levels in the microenvironment.Previous studies have demonstrated that immunomodulatory hydrogel dressings can facilitate diabetic wound healing.However,current immunomodulatory hydrogels require costly and complex treatments such as cell therapy and cytokines.Herein,a hierarchical hydrogel dressing with continuous biochemical gradient based on glycyrrhizic acid(GA) was constructed to modulate immunomodulatory processes in diabetic wounds.The hydrogels present many desirable features,such as tunable mechanical properties,broad antibacterial ability,outstanding conductive,transparent,and self-adhesive properties.The resultant hydrogel can promote diabetic wound healing by preventing bacterial infection,promoting macrophage polarization,improving the inflammatory microenvironment,and inducing angiogenesis and neurogenesis.Furthermore,electrical stimulation(ES) can further promote the healing of chronic diabetic wounds,providing valuable guidance for relevant clinical practice. 展开更多

Diabetic wounds(DWs)are a major complication of diabetes mellitus,characterized by a complex patho-physiological microenvironment that is associated with elevated morbidity and mortality.Conventional management strategies often fail to address the multifaceted nature of these wounds effectively.Recent advancements in understanding the mechanisms of DW healing have spurred the development of a plethora of bioactive dressings designed to interact with and modulate the DW microenvironment.These innovations have culminated in the introduction of the“microenvironment-sensitive with on-demand management”paradigm aimed at delivering precision therapy responsive to dynamic changes within DW.Despite these advancements,the current literature lacks a comprehensive review that cate-gorizes and evaluates active,passive,and on-demand approaches that address the DW microenviron-ment.Herein,we describe the unique pathogenic mechanisms and microenvironmental characteristics that distinguish DW from normal acute wounds.This review provides an extensive overview of contem-porary active and passive management strategies incorporating on-demand management principles designed for DW microenvironments.Furthermore,it addresses the principal challenges faced in this therapeutic domain and outlines the potential innovations that can enhance the efficacy and specificity of bioactive dressings.The insights presented here aim to guide further research and development in the on-demand management of DW to improve patient outcomes by aligning personalized therapy modali-ties with the pathophysiological realities of DW. 展开更多

Diabetic wounds represent a significant challenge in the medical field,significantly impacting patient quality of life and imposing a heavy burden on healthcare systems.Intelligent hydrogel dressings have attracted significant attention in diabetic wound treatment due to their unique properties.This review systematically explores the three main categories of intelligent hydrogels(natural,synthetic,and composite),dissecting their composition,structure,and the mechanisms that enable their intelligent responses.The crucial roles of these dressings in maintaining a moist wound environment,efficiently absorbing exudate,and precisely delivering drugs are expounded.Moreover,their application advantages in combating bacteria and infections,regulating inflammation and immunity,promoting angiogenesis and tissue regeneration,as well as enabling real-time monitoring and personalized treatment,are explored in depth.Additionally,we discuss future research directions and the prospects for personalized precision medicine in diabetic wound care,aiming to inspire innovation and provide a comprehensive theoretical basis for the development of nextgeneration intelligent dressings. 展开更多

A hydrogel dressing based on bacterial cellulose(BC),which is grafted with quaternary ammonium func-tional and crosslinked with the gelatin-heparin system,is prepared to provide the features mainly con-cerning softness,high swelling ratio,antibacterial property,and biocompatibility.An innovation of prepa-ration is that the BC is beaten into short-chain scaffolds to improve the efficiency of grafting,which not only simplifies the preparation process but also avoids the biotoxicity caused by the introduction of toxic catalyst such as dimethyl sulfoxide(DMSO)or uncertain toxic side products in long-chain graft-ing.Scanning electron microscopy(SEM)shows that the QBC/Hep/Gel composite hydrogel possesses a three-dimensional mesh structure with high porosity.The hydrogel shows outstanding water manage-ment performance indicated by the swelling ratio of 1476%,water retention ratio of more than 90%at 120 h,and moisture permeability of 3296 g m^(-2) 24 h^(-1).The antibacterial experiment is implemented with staphylococcus aureus,and the antibacterial effect is represented by an inhibition zone of 3 cm in diameter.In vivo animal experiments suggested that QBC/Hep/Gel could effectively promote epithelial reconstruction,collagen deposition,and angiogenesis in normal wounds,reduce inflammation,and ac-celerate wound healing.All these results indicate that the proposed QBC/Hep/Gel hydrogel is a potential composite for antibacterial dressing. 展开更多

Cubic boron nitride(cBN)grinding wheels play a pivotal role in precision machining,serving as indispensable tools for achieving exceptional surface quality.Ensuring the sharpness of cBN grains and optimizing the grinding wheel’s chip storage capacity are critical factors.This paper presents a study on the metal-bonded segments and single cBN grain samples using the vacuum sintering method.It investigates the impact of blasting parameters-specifically silicon carbide(SiC)abrasive size,blasting distance,and blasting time-on the erosive wear characteristics of both the metal bond and abrasive.The findings indicate that the abrasive size and blasting distance significantly affect the erosive wear performance of the metal bond.Following a comprehensive analysis of the material removal rate of the metal bond and the erosive wear condition of cBN grains,optimal parameters for the working layer are determined:a blasting distance of 60 mm,a blasting time of 15 s,and SiC particle size of 100#.Furthermore,an advanced simulation model investigates the dressing process of abrasive blasting,revealing that the metal bond effectively inhibits crack propagation within cBN abrasive grains,thereby enhancing fracture toughness and impact resistance.Additionally,a comparative analysis is conducted between the grinding performance of porous cBN grinding wheels and vitrified cBN grinding wheels.The results demonstrate that using porous cBN grinding wheels significantly reduces grinding force,temperature,and chip adhesion,thereby enhancing the surface quality of the workpiece. 展开更多

Inspired by the skin structure,an asymmetric wettability tri-layer nanofiber membrane(TNM)consisting of hydrophilic inner layer loaded with lidocaine hydrochloride(LID),hydrophobic middle layer with ciprofloxacin(CIP)and hydrophobic outer layer has been created.The hydrophobic outer layer endows the TNM with waterproof function and anti-adhesion from contaminants.The hydrophobic middle layer with CIP preserves long-term inhibition of bacteria growth and the hydrophilic inner layer with LID possesses optimal waterabsorbing capacity and air permeability.The TNM dramatically elevates the water contact angles from 10°(inner layer)to 120(outer layer),indicating an asymmetric wettability,which could directionally transport wound exudate within the materials and meanwhile maintain a comfortable and moist environment to promote wound healing.Furthermore,the sequential release of LID and CIP could relieve pain rapidly and achieve antibacterial effect in the long run,respectively.In addition,the TNM shows superior biocompatibility towards L929 cells.The in vivo results show the TNM could prevent infection,accelerate epithelial regeneration and significantly accelerate wound healing.This study indicates the developed TNM with asymmetrical wettability and synergetic drug release shows great potential as a wound dressing in clinical application. 展开更多

Combinations of metal and lanthanide oxides have been done through casted films for potential medical applications. In this regard, samarium(Ⅲ) oxide/chromium(Ⅲ) oxide/graphene oxide(GO)/polycaprolactone(PCL) based films nano-composites(NCs) were fabricated, pointing their utilization as a biological scaffold for wound dressing purposes. Also, samarium(Ⅲ) oxide and chromium(Ⅲ) oxide have been merged as promising optical constituents due to their unique optical behavior. The structural and compositional examination of the studied NCs was executed by X-ray diffraction(XRD), Raman, and field emission scanning electron microscopy(FESEM). The Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL NC exhibits a surface with a lower roughness degree owing to the presence of GO. Cr_(2)O_(3)shows size reduction upon GO insertion to reach 1.2 μm as the average grain size, whilst Sm_(2)O_(3)records an average grain size of less than 1 μm. As well, the polymeric nano-compositions exhibit variation in contact angle values that hit 29.76°± 3.52°for Sm_(2)O_(3)/PCL, and 48.62°± 1.37°for Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL as the second lowest contact angle. The optical behavior contributes to absorption edge relocation along the x-axis from 1.7 eV for pure PCL, to 2.65 eV for Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL. Regarding biological responses, the cell exposed to 2.5 μg/m L of Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL shows cell viability of 119.31%, while 5 μg/m L hits 99.6%. Additionally, the resulting cell attachment micrographs show layers of fibroblast tissue, besides the proliferation and growth of cultivated cells. Thus, the Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL scaffold provides 3D proliferation of fibroblast cells endorsing the wound healing process. 展开更多

Background:Excessive exudate secreted from chronic wounds often leads to overhydration and infection.Although a variety of dressings are currently available in clinical applications,they frequently fail to provide multifunction to promote chronic wound healing.The dressings with a Janus structure,featuring distinct properties on each side,are potential to improve wound healing.Methods:Composite dressings with a Janus structure were fabricated,comprising freeze-dried polycaprolactone(PCL)electrospun membrane and alginate-based hydrogel.The PCL fibrous membrane provided air permeability,while the hydrogel loaded with Deferoxamine,composed of alginate and poly(N-isopropylacrylamide)(PNIPAM),exhibited hygroscopic properties.The inclusion of PNIPAM imparted thermo-responsivity.Results:The hydrogel(thickness of 2.778±0.082 mm)exhibited a robust adhesion to the fiber membrane(thickness of 0.261±0.041 mm).For ANDC(Alginate-PNIPAM hydrogel with Deferoxamine/PCL membrane)samples,the water vapor transmission rate(WVTR)was measured to be 3364.80±23.23 g∙m−2∙day−1 and the swelling ratio at 2 h was determined to be 1179±125%.The thermo-responsivity of ANDC samples manifested in an increased swelling rate,escalating from 797±189%at 37°C to 1132±147%at 4°C.The elastic modulus was assessed for lyophilized and rehydrated ANDC sample.When theωof the rheometer rotor was decreased from 10 rad/s to 0.1 rad/s,the lyophilized dressing exhibited a decrease from 2.65±0.01 MPa to 1.80±0.90 MPa,while the rehydrated dressing demonstrated an increase from 133.65±55.68 Pa to 264.23±141.71 Pa.The pro-healing properties of the dressings were evaluated using full-thickness skin defect model on SD rats,and a circular wound of diameter 10 mm healed completely by day 12.Conclusion:The dressings not only protected the wound and absorbed excess exudate,but also demonstrated nondestructive peelability upon cooling,providing a novel approach for accelerating wound healing and management. 展开更多

Natural hydrogels have emerged as a pivotal innovation in wound care,offering a unique combination of high absorbency,biocompatibility,and versatility.However,due to the complexity of wound healing,the physiological state of the wound varies dynamically,and the mechanism of natural hydrogels that boost wound healing is still unclear.In this review,we firstly provide a comprehensive introduction to the bio-logical process of wound healing,emphasizing the critical stages and factors affecting healing.This work concludes the composition and properties of natural hydrogels,including collagen,gelatin,hyaluronic acid,chitosan,alginates,cellulose,and fibroin,highlighting their biocompatibility and biodegradability.The focus shifts to the various crosslinking strategies employed to enhance the structural integrity and functionality of natural hydrogels.This review further investigates the biological effects of natural hydro-gels in wound healing,detailing their antibacterial,antioxidant,anti-inflammatory,adhesive,and hemo-static functions.Furthermore,we propose the challenges and future perspectives of natural hydrogels in practical applications.This review offers a comprehensive overview of the current state and poten-tial future advancements in natural hydrogel dressings for wound care,highlighting their critical role in addressing complex and hard-to-heal wounds. 展开更多

The utilization of arsenic-containing gold dressing tailings is an urgent issue faced by gold production companies worldwide.The thermodynamic analysis results indicate that ferrous arsenate(FeAsO_(4)),pyrite(FeS_(2))and sodium cyanide(NaCN)in the arsenic-containing gold metallurgical tailings can be effectively removed using straight grate process,and the removal of pyrite and sodium cyanide is basically completed during the preheating stage,while the removal of ferrous arsenate requires the roasting stage.The pellets undergo a transformation from magnetite to hematite during the preheating process,and are solidified through micro-crystalline bonding and high-temperature recrystallization of hematite(Fe_(2)O_(3))during the roasting process.Ultimately,pellets with removal rates of 80.77% for arsenic,88.78% for sulfur,and 99.88% for cyanide are obtained,as well as the iron content is 61.1% and the compressive strength is 3071 N,meeting the requirements for blast furnace burden.This study provides an industrially feasible method for treating arsenic-containing gold smelting tailings,benefiting gold production enterprises. 展开更多

Improving pressure ulcer care in intensive care units:Evaluating the impact of bundled care and silver nanoparticle dressings

BACKGROUND Pressure ulcer(PU)are prevalent among critically ill trauma patients,posing substantial risks.Bundled care strategies and silver nanoparticle dressings offer potential solutions,yet their combined effectiveness and impact on patient satisfaction remain insufficiently investigated.AIM To assess the impact of bundled care along with silver nanoparticle dressing on PUs management and family satisfaction in critically ill trauma patients.METHODS A total of 98 critically ill trauma patients with PUs in intensive care unit(ICU)were included in this study.Patients were randomly assigned to either the control group(conventional care with silver nanoparticle dressing,n=49)or the intervention group(bundled care with silver nanoparticle dressing,n=49).The PU Scale for Healing(PUSH)tool was used to monitor changes in status of pressure injuries over time.Assessments were conducted at various time points:Baseline(day 0)and subsequent assessments on day 3,day 6,day 9,and day 12.Family satisfaction was assessed using the Family Satisfaction ICU 24 ques-tionnaire.RESULTS No significant differences in baseline characteristics were observed between the two groups.In the intervention group,there were significant reductions in total PUSH scores over the assessment period.Specifically,surface area,exudate,and tissue type parameters all showed significant improvements compared to the control group.Family satisfaction with care and decision-making was notably higher in the intervention group.Overall family satisfaction was significantly better in the intervention group.CONCLUSION Bundled care in combination with silver nanoparticle dressings effectively alleviated PUs and enhances family satisfaction in critically ill trauma patients.This approach holds promise for improving PUs management in the ICU,benefiting both patients and their families. 展开更多

Background:Skin,being a vital organ that regulates physiological responses in the human body,is prone to injury from external environmental factors.Healing full-thickness skin defects becomes especially challenging when infections and vascular injuries are involved.Traditional wound dressings with single functions,such as antibacterial or angiogenic properties,fall short in achieving rapid wound healing.To address this,there is a need to develop wound dressing materials that possess both effective antibacterial and angiogenic properties.Methods:In this study,we utilized electrospinning technology to fabricate hyaluronic acid-cellulose acetate fibrous membrane dressings,incorporating poly(ionic liquid)as an antibacterial polymer and deferoxamine as an angiogenic agent.Results:The resulting fibrous membrane dressing contained poly(ionic liquid)and deferoxamin showcased a microporous structure,drug-releasing capabilities,and excellent air permeability.It not only demonstrated highly effective antibacterial properties but also exhibited remarkable angiogenesis,thereby promoting the healing of full-thickness skin defect wounds in both in vitro and in vivo assays.Conclusion:These findings highlight the immense potential of this wound dressing material for future clinical applications. 展开更多

Silver-containing dressings have gained popularity in clinical applications due to their exceptional antibacterial properties and ability to enhance wound healing.These dressings primarily exert their antibacterial effects through the utilization of silver ions or nano-silver particles.The antimicrobial property of silver ions is a result of multiple mechanisms,with the most significant being the interaction between Ag or Ag+and the bacterial cell wall,as well as the interaction between Ag+and bacterial DNA or RNA,which inhibits replication and division.Silver-containing dressings can effectively reduce inflammation in wound tissue and expedite wound healing.The antibacterial and wound-healing promoting effects of these dressings are closely associated with the controlled release rate of silver ions.Consequently,developing novel silver-containing dressings that accurately regulate this release rate has become a prominent area of research interest.With broad prospects for application in wound care,silver-containing dressings are anticipated to offer innovative solutions for addressing numerous challenges encountered in clinical wound management. 展开更多

The aim of this study was to evaluate the cost and effectiveness of soft silicone foam dressings (SSFD, Mepilex) on the treatment of pressure ulcers. We searched electronic databases and retrieved articles to make a systematic evaluation, and then make a cost-effectiveness analysis by decision tree model combined with data from clinical treatments. The result shows that compared with the common sterile gauze, SSFD possesses an apparent advantage. The effective ratio is 96.3% versus 77.3%, although the cost of SSFD is much higher than that of sterile gauze, Mepilex appears to be more cost-effectiveness for preventive use. 展开更多

Diabetes is one of the most prevalent diseases in the world with high-mortality and complex complications including diabetic foot ulcer(DFU). It has been reported that the difficulties in repairing the wound related to DFU has much relationship with the wound infection,change of inflammatory responses, lack of extracellular matrix(ECM), and the failure of angiogenesis. Following the development of medical materials and pharmaceutical technology, nanofibers has been developed by electrospinning with huge porosity, excellent humidity absorption, a better oxygen exchange rate, and some antibacterial activities. That is to say, as a potential material, nanofibers must be a wonderful candidate for the DFU treatment with so many benefits. Careful selection of polymers from natural resource and synthetic resource can widen the nanofibrous application. Popular methods applied for the nanofibrous fabrication consist of uniaxial electrospinning and coaxial electrospinning. Furthermore, nanofibers loading chemical, biochemical active pharmaceutical ingredient(API)or even stem cells can be wonderful dosage forms for the treatment of DFU. This review summarizes the present techniques applied in the fabrication of nanofibrous dressing(ND)that utilizes a variety of materials and active agents to offer a better health care for the patients suffering from DFU. 展开更多

In order to prepare a novel hemostatic dressing for uncontrolled hemorrhage, a porous chitosan sponge was coated with self-assembled(thrombin/tannic acid)n films, which were based on hydrogen bonding interactions between thrombin and tannic acid at physiologic p H. According to the whole blood clotting test, the coated chitosan sponges showed a significantly high rate of blood clotting due to the addition of thrombin. On the other hand, the storable half-life of immobilized thrombin is extended to 66.9 days at room temperature, which is 8.5 times longer than unfixed thrombin. It is because of the immobilization effect of, not only the porous structure of chitosan sponge but also the interactions between thrombin and tannic acid. In addition, the tannic acid has similar antibacterial effect to chitosan. Therefore, it is an excellent combination of chitosan, thrombin and tannic acid. Besides, all of materials in this research have been approved by the United States Food and Drug Administration(FDA). So the chitosan-based sponge is a promising candidate dressing for uncontrolled hemorrhage due to its storable, bio-safe and highly effective hemostatic properties. 展开更多
