Non- denatured basic proteins, suc...This work presents an approach to build a high-performance, low-viscous and replaceable separation matrix, semi-crosslinked polyacrylamide (semi-CPA) capillary gel electrophoresis. Non- denatured basic proteins, such as lysozyme, cytochrome C, ribonuclease A and trypsin were separa- ted. The impacts of monomer and cross-linker concentrations on protein separation were studied, and the ability of dynamic capillary inner wall coating was demonstrated. The UV absorption interfer- ence by semi-CPA gel matrix was successfully overcome by a partial filling technique, which results in sensitivity 20 times higher than other protein separation method. The excellent separation ability, reproducibility and dynamic coating ability made semi-CPA an ideal separation media in both capillar- y electrophoresis and microfluidic chip separation scheme.展开更多

Objective: Based on liver cancer model built in SD rats, the contents of trace elements (Cu, Fe, Zn, Ca and Mg), AFP, CEA, SF, TH and IGF-II in serum were measured at different stages to explore the molecular changes ...Objective: Based on liver cancer model built in SD rats, the contents of trace elements (Cu, Fe, Zn, Ca and Mg), AFP, CEA, SF, TH and IGF-II in serum were measured at different stages to explore the molecular changes during the rat liver cancer development. Methods: The SD rat liver cancer model was built by using diethylnitrosamine (DENA) as the mutagen. During 16 weeks of DENA gavage, blood samples were taken in the 14th, 28th, 56th, 77th, 105th and 112th days respectively after the first day of gavage with DENA, then the contents of five trace elements (Cu, Fe, Zn, Ca and Mg), T3, T4, IGF-II, AFP, CEA and SF in serum were determined. Results: During the development of the rat liver cancer, in the test group, the Cu content significantly increased in serum, while the contents of Fe, Zn and Ca significantly decreased. The content of Mg showed no significant change. AFP and CEA of the test group showed same expression level with the control group; while the content of SF was lower than that of the control group when cancerization appeared. T3 and T4 increased at the first stage and then went down, and the content of IGF-II was always high. Conclusion: Cu, Fe, Zn, Ca, T3, T4, SF and IGF-II are closely related to the development of liver cancer. The changes of their contents in the development of cancer could enlighten the researches on cancer pathogenesis and prevention.展开更多
In this study, we investigated the potential nitrification and community structure of soil-based ammonia-oxidizing bacteria (AOB) in apple orchard soil during different growth periods and explored the effects of env...In this study, we investigated the potential nitrification and community structure of soil-based ammonia-oxidizing bacteria (AOB) in apple orchard soil during different growth periods and explored the effects of environmental factors on nitrification activity and AOB community composition in the soil of a Hanfu apple orchard, using a culture-dependent technique and denaturing gradient gel electrophoresis (DGGE). We observed that nitrification activity and AOB abundance were the highest in November, lower in May, and the lowest in July. The results of statistical analysis indicated that total nitrogen (N) content, NH4+-N content, NO3-N content, and pH showed significant correlations with AOB abundance and nitrification activity in soil. The Shannon-Winner diversity, as well as species richness and evenness indices (determined by PCR-DGGE banding patterns) in soil samples were the highest in September, but the lowest in July, when compared to additional sampled dates. The DGGE fingerprints of soil-based 16S rRNA genes in November were apparently distinct from those observed in May, July, and September, possessing the lowest species richness indices and the highest dominance indices among all four growth periods. Fourteen DGGE bands were excised for sequencing. The resulting analysis indicated that all AOB communities belonged to the 13-Proteobacteria phylum, with the dominant AOB showing high similarity to the Nitrosospira genus. Therefore, soil-based environmental factors, such as pH variation and content of NHa+-N and NO3--N, can substantially influence the abundance of AOB communities in soil, and play a critical role in soil-based nitrification kinetics.展开更多
We present a study of the electric field effect on electrochemically grown ultrathin, straight platinum nanowires with minimum diameter of 15 nm and length in the micrometer range, synthesized on a silicon oxide subst...We present a study of the electric field effect on electrochemically grown ultrathin, straight platinum nanowires with minimum diameter of 15 nm and length in the micrometer range, synthesized on a silicon oxide substrate between metal electrodes in H2PtC16 solution. The influence of the concentration of the platinum- containing acid and the frequency of the applied voltage on the diameter of the nanowires is discussed with a corresponding theoretical analysis. We demonstrate for the first time that the electric field profile, provided by the specific geometry of the metal electrodes, dramatically influences the growth and morphology of the nanowires. Finally, we provide guidelines for the controlled fabrication and contacting of straight, ultrathin metal wires, eliminating branching and dendritic growth, which is one of the main shortcomings of the current bottom-up nanotechnology. The proposed concept of self-assembly of thin nanowires, influenced by the electric field, potentially represents a new route for guided nanocontacting via smart design of the electrode geometry. The possible applications reach from nanoelectronics to gas sensors and biosensors.展开更多
Hofmeister series ranks the ability of salt ions in influencing a variety of properties and processes in aqueous solutions.In this review,we reexamine how these ions and some other small molecules affect water structu...Hofmeister series ranks the ability of salt ions in influencing a variety of properties and processes in aqueous solutions.In this review,we reexamine how these ions and some other small molecules affect water structure and thermodynamic properties,such as surface tension and protein backbone solvation.We illustrate the difficulties in interpreting the thermodynamic information based on structural and dynamic arguments.As an alternative,we show that the solvation properties of ions and proteins/small molecules can be used to explain the salt effects on the thermodynamic properties of the solutions.Our analysis shows that the often neglected cation-anion cooperativity plays a very important role in these effects.We also argue that the change of hydrogen donor/acceptor equilibrium by added cosolutes/cosolvents can be used to explain their effects on protein secondary structure denaturation/protection:those increase hydrogen donor concentrations such as urea and salts with strongly solvated cations/weakly hydrated anions tend to dissolve protein backbone acting as secondary structure denaturants,whereas those lack of hydrogen donors but rich in acceptors have the opposite effect.展开更多
