In this study we investigated the rooting ability and the growth performance of juvenile single-node leafy stem cuttings of Litsea monopetala (Roxb) Pers. collected from two mature mother trees preserved in the hill forest of Chittagong district, Bangladesh. The rooting ability of cuttings was studied under 0%, 0.1%, 0.2% and 0.4% indole-3-butyric acid (IBA) treatments. Significantly better rooting response (p ≤ 0.05) was observed with 0.1% IBA compared to control (0% IBA). The mean number of roots and the length of the longest root of cuttings in different treatments showed no significant difference (p≤0.05). After transfer into polythene bags from non-mist propagator, rooted cuttings treated with 0%, 0.1% and 0.2% IBA demonstrated the highest (100 ± 0.00%) survival capacity. The mean number of shoots developed in cuttings in the polythene bags in first three weeks varied significantly (p≤0.05) among the treatments. Effects of three fertilizer treatments, viz. T0 (no fertilizer), T1 (10g Urea, 20g TSP, 10g MOP dissolved in 1 L water) and T2 (10g Urea, 20g TSP, 10g MOP dissolved in 2 L water) on initial growth ofstecklings were also measured over a 90-days period. The increment of leaf area of stecklings was significantly higher (p≤0.05) under T0 compared with that under T1 and T2 while the increment of stem length, collar diameter and root biomass varied insignificantly among different fertilizer treatments. The results suggest that rooting juvenile single-node leafy stem cuttings could be an effective mean of regenerating L. monopetala. The application of 0.1% IBA concentration is recommended for rooting of juvenile leafy stem cuttings and application of fertilizer appeared unnecessary for the subsequent growth of stecklings in polythene bags. 展开更多

The highest rooting rate (50%) was recorded in the cuttings with the application of IBA @ 4000 ppm. The cuttings treated with IBA @ 2000 ppm had 25% rooting rate. All other treatments along with control (talc powder) failed to induce rooting. It was concluded that IBA @ 4000 ppm was a better-applied concentration for vegetative propagation ofA. indica under Kashmir conditions. 展开更多

Techniques have been developed for the vegetative propagation of Tetraena mongolica,a rare and endangered species endemic to the western Ordos Plateau in the northcentral portion of China.The purpose of the study was to investigate the rooting characteris-tics of the plant species from the aspects of length and ages of cuttings,external hormone concentration,leaf retention,cutting orientations,and cutting positions.The results obtained from the experiments indicated that the rooting ratio of 2-year or 3-year old cuttings was larger than more than of 4-year old cuttings.The influence of external hormone concentration on rooting was ob-vious;the rooting ratio of cuttings was increased by external hormone treatment,especially when treated by ABT1(ABT No.1 Rooting Powder) 50 mg/L for 3 hours,in which case,the rooting ratio reached 91 percent.Leaf retention influenced rooting sig-nificantly;the rooting rate of cuttings became greater as the leaf retention increased.There was no significant difference in rooting capability between south and north orientations of the ortets.The rooting ratio of the cuttings obtained from the distal portions of shoots was significantly larger than those obtained from the middle and the basal portions of the shoots.On day three after treat-ment,calluses started appearing at the incisions of some cuttings dipped in 500 mg/L IAA(indole acetic acid).Most cuttings were healed or swelled,and 6 percent of cuttings rooted on day 6 after treatment.Higher rooting period occurred 7-10 days after the experiment was set up. 展开更多

The present study investigated the rooting ability and growth performance of juvenile single-node leafy stem cuttings of Holarrhena pubescens, collected from two mature mother trees preserved in the hill forests of Chittagong District, Bangladesh. The rooting ability of cuttings was studied with 0, 0.1%, 0.2% and 0.4% indole-3-butyric acid (IBA) treatments. The better rooting response was observed with 0.1% IBA, but was insignificant compared to other treatments. The mean number of roots and length of the longest root of cuttings in different treatments showed no significant difference. After transfer to the polythene bags from a nonmist propagator, rooted cuttings treated with 0.4% IBA demonstrated the highest (50.00± 0.78%) survival capacity. Regarding initial growth of established rooted cuttings (stecklings) over a 90 d period, the increment of stem length of stecklings under T2 (10 g Urea, 20 g TSP, 10 g MOP dissolved in 2 L water) and leaf area expansion of stecklings under T1 (10 g Urea, 20 g TSP, 10 g MOP dissolved in 1 L water) was significantly higher (p 〈 0.05) compared with that under TO (no fertilizer). The increment of collar diameter and root biomass production varied insignificantly among different fertilizer concentrations, although stecklings under T2 showed the better performance. The results suggest that rooting juvenile single-node leafy stem cuttings can be an effective mean of regenerating H. pubescens. The application of 0.1% IBA or without IBA is recommended for rooting juvenile leafy stem cuttings. Application of fertilizer appears necessary for subsequent growth of stecklings in polythene bags. 展开更多

The present study, conducted during 2016 and 2017 seasons, aimed to investigate the effect of IBA on rooting of Piper betle L. stem cuttings (softwood and semi-hardwood). The experiment was undertaken in misting house field 2 UPM using the sand media to determine the adventitious roots initiation and development using the histological method. The cuttings were treated with different IBA concentrations (0, 500, 1000, 1500 and 2000 mg/L). The nodes explants were used in the development of a protocol for in vitro propagation of P. betle L., with different concentrations of Clorox with different times of immersion (20% Clorox 10 minutes, 30% Clorox 10 minutes, 20% Clorox 20 minutes, and 30% 20 minutes). In multiplication of the plantlets, Murashige and Skoog (MS) medium with different concentrations of BAP (0, 0.5, 1.0, 2.0 mg/L) were used to investigate the rooting of the explants. The results indicated that the types of the cuttings were different in the rooting capacity and the length of the roots. Moreover, it was found that in comparison with the control treatment, by a rise in the concentrations of the IBA, there was a significant upsurge in the rooting percentage, the root diameter, and the number of the roots. The results indicated that the types of cutting with 1000, 1500 and 2000 mg/L IBA perform better in the root percentage (100%) in the semi hardwood cuttings. The best results, however, were 2000 mg/L IBA in the semi hardwood cuttings, with the number of the roots to be 35.05, and the fresh weight of the roots to be 3.94 g, the dry weight of the roots to be 0.33 g, the length of the roots to be 391.88 cm, the roots diameter to be 1.21 mm, the surface area of the roots to be 121.83 cm2, and the root volume to be 2.99 cm3. Nonetheless, the optimal concentration of Clorox with the time immersion was 20% with the 20-minute immersion time, which produced a shoot induction percentage of 30% dead explants and a mean number of 70.00 shoots per explant and the optimal concentration of benzylaminopurine (BAP) at 1.0 mg/L. It is of note that a shoot induction percentage of 22.29% and a mean number of 4.1% number of auxiliary bud per treatment. P. betle shoots in MS medium without PGR MS (0.0) yielded a good rooting. 展开更多

The study describes the scope of clonal propagation of guava (Psidium guajava Linn.) by stem cutting collected from mature stockplants. Cuttings were treated with 0, 0.2%, 0.4% and 0.8% IBA solution and rooted in the non-mist propagator. Rooted cuttings were allowed to grow in the polybags filled with soil and cow-dung mixed in the ratio of 3: 1 (by volume) for three months to assess the steckling capacity and initial growth performance. The study reveals that the species is amenable for clonal propagation by mature stem cutting. The highest rooting percentage (60%) was observed in the cuttings treated with 0.4% IBA solution followed by 0.2% IBA and the lowest was in controlled cuttings. The maximum number of primary root (32.7) was developed in the cuttings treated with 0.8% IBA solution followed by 0.4% IBA and the lowest was in the cuttings without IBA treatment. The highest survival percentage (70.9) was observed in the cuttings rooted with 0.4% IBA treatment and the lowest (58.3) was in the cuttings without any treatment. However, there was no significant variation in height growth of cutlings due to IBA treatments in rooting. 展开更多

The use of stem cutting in vegetative propagation and grafting is as indispensable tools for mass multiplication of superior genotypes and helps in improve planting yield and quality. The study was aimed to investigate the effect of different diameters (10 - 15, 16 - 25 and 26 - 35 mm) and different hormone concentrations of indo-butyric acid (0, 1.5, 3, 6 and 10 g/L), in the rooting of <i>Jatropha</i> <i>platyphylla</i> and to obtain axillary buds to performed grafts. Rooting efficiency was 80% in greenhouse conditions. Hormone concentration and diameter significantly affected the rooting and shooting ability of <i>Jatropha</i> <i>platyphylla</i> stem cuttings. Stem cuttings of 26 - 35 mm have the greatest number, length and dry root weight. 76% survival of the grafted plants was obtained. <span>This demonstrates the necessity to improve the conventional propagation methods</span> with appropriate scientific inputs for more economical and time efficient techniques for standard propagation protocols. 展开更多

Kuharske were used to develop protocols for rooting reportedly HLB resistance rootstocks under intermittent mist. Investigated were shoot maturity, nodes per cutting, leaves per cutting, effects of buds, auxin concentrations and auxin solvent. Shoot maturity was most influential for success, with cuttings taken below the first 30 cm of active terminal growth producing greater root generation. Use of a thickening agent (Natrosal) to dilute the commercial auxin was second most in importance for rooting success. Root mass increased with increasing number of leaves. Cutting stems between nodes or below the lowest bud were inconsequential. To produce maximum number of viable cuttings, single node-single leaf cuttings were preferred. Single bud cuttings produced one shoot after rooting. This was adventitious since multi-node cuttings usually sprouted new shoots that would need to be removed before budded. Evaluation of the best combination of auxin and cutting-related attributes were evaluated with four additional common rootstocks in June 2016. Rooting was 100% successful. A quick dip (0.5 s) in a 7500 ppm solution of Dip&Gro produced the most root generation in six weeks for all rootstocks. Root quantity varied by rootstock. 展开更多

【目的】探明上部烟叶一次性带茎砍烤对其理化及品质指标的影响规律,为提高上部烟叶可用性提供理论依据。【方法】以云烟87为试材,测定一次性带茎砍烤和一次性采烤上部烟叶的含水率、色素含量、理化品质、感官质量和经济性状,分析不同... 【目的】探明上部烟叶一次性带茎砍烤对其理化及品质指标的影响规律,为提高上部烟叶可用性提供理论依据。【方法】以云烟87为试材,测定一次性带茎砍烤和一次性采烤上部烟叶的含水率、色素含量、理化品质、感官质量和经济性状,分析不同采烤方式对不同叶位上部烟叶变黄失水规律及品质的影响。【结果】相较于一次性采烤,一次性带茎砍烤可延缓上部烟叶水分散失,促进叶绿素和类胡萝卜素分解;烟叶在叶长和叶宽上均小于一次性采烤,且单叶重和叶面密度均显著低于一次性采烤(P<0.05);上部烟叶的上等烟比例与均价均高于一次性采烤;一次性带茎砍烤上三叶和下三叶烤后淀粉含量分别较一次性采烤显著减少2.45百分点和2.61百分点,总氮含量分别显著减少0.19百分点和0.16百分点,总糖含量分别显著增加2.82百分点和2.36百分点,还原糖含量分别显著增加1.73百分点和2.51百分点,糖碱比和钾氯比显著高于一次性采烤(P<0.05)。一次性带茎砍烤上部烟叶在香气质、香气量、吃味、杂气和刺激性等方面均优于一次性采烤。【结论】上部烟叶一次性带茎砍烤有利于促进烟叶变黄失水协调,促进色素、淀粉等大分子分解,提高两糖含量。在实际生产中,从感官质量和经济效益方面考虑,可选择一次性带茎砍烤。 展开更多

柠条茎秆锯切性能参数分析与试验研究

为研究柠条茎秆在锯切过程中的力学特性,并寻求最优锯切参数组合以支持后续平茬装备的优化改进,利用自制的枝条锯切试验台,通过单因素试验研究枝条直径、切割速度、喂入速度、切割倾角和锯片齿数对枝条峰值切割力、切割功率的影响。在... 为研究柠条茎秆在锯切过程中的力学特性,并寻求最优锯切参数组合以支持后续平茬装备的优化改进,利用自制的枝条锯切试验台,通过单因素试验研究枝条直径、切割速度、喂入速度、切割倾角和锯片齿数对枝条峰值切割力、切割功率的影响。在单因素试验基础上,采用Box-Benhnken中心组合试验方法,选取切割速度、喂入速度、切割倾角、锯片齿数为试验因素,以枝条峰值切割力、切割功率为目标值进行多因素试验,并建立回归模型。试验结果表明,峰值切割力随茎秆直径的增大而增大,随着切割速度、喂入速度、切割倾角、锯片齿数的增大先增大后减小;切割功率随茎秆直径的增大而增大,随着切割速度、喂入速度、切割倾角、锯片齿数的增大先减小后增大。对回归模型进行多目标优化分析,得出最优工作参数组合为切割速度45.7 m/s、喂入速度0.32 m/s、切割倾角8.3°、齿数104,该组合下的峰值切割力为7.02 N,切割功率为181.57 W。峰值切割力与切割功率的预测值与实际值的偏差分别为1.9%、2.2%。试验结果为寻求高效、低耗的柠条平茬装备的设计提供参考。 展开更多

【目的】分析再造梗丝在制品的化学成分、微观结构和热解特性变化规律。【方法】以关键工序再造梗丝在制品为研究对象,利用成分分析及热重分析等技术研究再造梗丝在加工过程中的理化性质变化规律。【结果】(1)再造梗丝在加工过程中的化... 【目的】分析再造梗丝在制品的化学成分、微观结构和热解特性变化规律。【方法】以关键工序再造梗丝在制品为研究对象,利用成分分析及热重分析等技术研究再造梗丝在加工过程中的理化性质变化规律。【结果】(1)再造梗丝在加工过程中的化学成分含量变化较大,原梗经浸提和气流干燥后,总糖、总氮、烟碱含量显著降低,纤维素含量升高,料液回填后样品的纤维素和半纤维素相对含量均降低,总糖和还原糖含量增加。(2)提取和二级气流干燥后样品较原梗色差值变化明显,但成品再造梗丝能够保持与原始梗丝相似的色泽外观;SEM和BET显示二级气流干燥对在制品的微观孔隙结构有显著的膨胀作用。(3)再造梗丝在制品的热失重过程可分为5个阶段,各样品的阶段失重温度区间和失重率差异明显。提取和二级气流干燥样品在阶段Ⅱ和Ⅲ的失重率均较低,料液回填后样品阶段Ⅱ的失重率与原始梗丝较接近;各样品均在第Ⅳ失重阶段表现出最高的失重速率,提取和二级气流干燥样品阶段Ⅳ的最大失重速率较高,且具有较高的Ti和Tf;成品的综合热解指数高于原始梗丝样品,综合热解特性有所提升。(4)各再造梗丝在制品主要失重阶段的热解过程均符合F1.5级化学反应控制模型,工序加工会影响样品同一失重阶段的热解动力学参数。 展开更多

对从烟田土壤和腐朽烟梗中筛选获得产果胶酶菌株TS-1和产漆酶菌株TS-2进行形态学观察和分子生物学鉴定,通过单因素试验优化二者的产酶条件,并将果胶酶粗酶液与漆酶粗酶液复配酶解膨胀梗丝,研究膨胀梗丝感官品质及致香成分的变化。结果表... 对从烟田土壤和腐朽烟梗中筛选获得产果胶酶菌株TS-1和产漆酶菌株TS-2进行形态学观察和分子生物学鉴定,通过单因素试验优化二者的产酶条件,并将果胶酶粗酶液与漆酶粗酶液复配酶解膨胀梗丝,研究膨胀梗丝感官品质及致香成分的变化。结果表明:菌株TS-1和TS-2分别鉴定为黑曲霉(Aspergillus niger)和黄孢原毛平革菌(Phanerodontia chrysosporium),二者最优产果胶酶和漆酶条件分别为烟梗粉质量浓度50 g/L、发酵温度30℃、发酵pH值6.0及烟梗粉质量浓度50 g/L、(NH4)2SO4质量浓度0.6 g/L、发酵温度30℃。果胶酶粗酶液、漆酶粗酶液酶解膨胀梗丝中果胶和木质素的最优条件分别为果胶酶粗酶液添加量240 U/g、酶解时间4.0 h、液质比1∶2、酶解温度30℃及漆酶粗酶液添加量2.0 U/g、酶解时间3.5 h、液质比1∶2、酶解温度30℃。膨胀梗丝酶解的最优粗酶液复配条件为果胶酶粗酶液添加量170 U/g、漆酶粗酶液添加量1.5 U/g、酶解时间3.0 h,酶解后其感官评吸得分较未酶解膨胀梗丝提高了8.2分,致香成分含量明显增加。 展开更多

针对梗丝回潮出口含水率受外界因素影响较大,难以确定加工过程中的加水总量,进而会对后续梗丝加料出口含水率稳定性及梗丝质量产生影响的问题,提出基于卷积神经网络(CNN)和长短期记忆(LSTM)网络的深度集成,并结合注意力机制开发梗丝回... 针对梗丝回潮出口含水率受外界因素影响较大,难以确定加工过程中的加水总量,进而会对后续梗丝加料出口含水率稳定性及梗丝质量产生影响的问题,提出基于卷积神经网络(CNN)和长短期记忆(LSTM)网络的深度集成,并结合注意力机制开发梗丝回潮加水量预测模型。首先,通过结合CNN和注意力机制解决影响梗丝回潮加水量的多变量和强耦合特性对预测建模带来的挑战。其次,通过LSTM网络门控单元处理序列数据,有效学习数据的时间特征。试验结果表明,此预测模型在预测梗丝回潮加水量时的决定系数(R^(2))可达0.97,平均绝对误差(MAE)低至5%。实际生产中,在后端工序梗丝加料出口含水率稳定性得到了大幅提高,合格率由72%提高至95%。 展开更多

为研究不同生产工序离线物料及生产批次顺序对梗丝出丝率的影响,以洗梗工序筛分碎梗量、切丝工序切梗机残留量、切梗料头剔除量、风选工序梗头剔除量共4类离线物料以及生产批次顺序为试验因素,进行描述性统计,分析各因素与梗丝出丝率的... 为研究不同生产工序离线物料及生产批次顺序对梗丝出丝率的影响,以洗梗工序筛分碎梗量、切丝工序切梗机残留量、切梗料头剔除量、风选工序梗头剔除量共4类离线物料以及生产批次顺序为试验因素,进行描述性统计,分析各因素与梗丝出丝率的相关性,采用控制变量的方法测定各生产工序离线指标对梗丝出丝率的影响并进行讨论分析。结果表明,生产批次顺序的变异系数最大,为56.42%;梗丝出丝率与筛分碎梗量、切梗料头剔梗量、切梗机残留量和风选梗头量均呈负相关,与生产批次顺序呈正相关;随着筛分碎梗量、切梗机残留量、切梗机料头量和风选梗头量的逐渐增加,梗丝出丝率呈下降趋势;随着生产批次的进行,梗丝出丝率首批次最低、第五批次最高,中间生产批次出丝率趋于稳定;筛分碎梗量、切梗机残留量和切梗机料头量随着生产批次的进行均呈不同程度下降并趋于稳定,而风选梗头剔除量逐渐增加但差异性不明显。以上结果表明,通过优化烟梗配方、减少切丝机残留、降低切梗料头以及控制风选梗头剔除量等措施,有利于提高梗丝出丝率,通过控制不同生产批次顺序间离线物料量,优化相关工艺参数可以提高各工序原料利用率,达到提高梗丝出丝率的目的。 展开更多

【目的】测量切梗丝机压实切面的压实效果,研究其对切梗丝质量的影响。【方法】采用机器视觉技术对切梗丝机压实切面进行图像获取、点云图转换及Grab Cut分割,并进行全变分提取和标准分转换,实现对切梗丝机压实切面的定量测量,同时应用... 【目的】测量切梗丝机压实切面的压实效果,研究其对切梗丝质量的影响。【方法】采用机器视觉技术对切梗丝机压实切面进行图像获取、点云图转换及Grab Cut分割,并进行全变分提取和标准分转换,实现对切梗丝机压实切面的定量测量,同时应用全变分标准分对切后产品梗签量和切梗丝厚度均匀性进行辅助判定与验证。【结果】(1)全变分标准分P与机器视觉测量值满足P=0.05R_(vB)/R_(v)(x)的关系;(2)全变分标准分P在不同位置的计分变异系数为0.27%~1.29%;针对不同压梗尺寸的物料进行测量时,全变分标准分P测量的变异系数为0.25%~1.37%,该方法准确性符合定量分析要求;(3)随着压实切面全变分标准分P递增,压实切面的压实效果变好,梗签剔除量降低47.15%,梗丝宽度合格率提升21.95%,标偏由0.024 mm降低至0.013 mm。【结论】该方法对压实切面具有可靠的描述能力,能有效评价压实效果,可辅助推断切梗丝质量。 展开更多
