However, few studies have investigated the differences in community structure between indoor and outdoor bacteria. In this work, the characterization of airborne bacteria was studied in a municipal wastewater treatment plant in Beijing, China. Two indoor(i.e., fine screen room and sludge dewatering house) and two outdoor(i.e., aeration tank and control site) sampling sites were selected. An Andersen six-stage impactor was used for collecting culturable airborne bacteria in the air, and Illumina MiSeq sequencing was conducted to track the emission source of the culturable airborne bacteria. The results indicate that, compared with the outdoor aeration tank site, the concentrations of culturable airborne bacteria in the indoor fine screen room with poor ventilation were more than ten times higher and the particle size was about twice as large. The community structures of indoor and outdoor culturable airborne bacteria were obviously different. Enterobacteriaceae and opportunistic pathogens were detected in indoor culturable airborne bacteria, with wastewater and sludge dewatering machine identified as the primary sources. Conversely,Enterobacteriaceae and opportunistic pathogens were not detected in outdoor culturable airborne bacteria. Outdoor high wind speed might have resulted in rapid dilution and mixing of culturable airborne bacteria generated from the aeration tank with the ambient air. The results of the present research suggest that covering pollution sources, increasing ventilation rates, and using protective measures for personnel should be implemented to decrease the exposure risk to indoor culturable airborne bacteria.展开更多

Methods Total soil aerobic bacteria, actinomycetes and fungi were counted by a 10-fold serial dilution plate technique. Numbers of anaerobic fermentative bacteria (AFB), denitrifying bacteria (DNB) and hydrogen-producing acetogenic bacteria (HPAB) were numerated by three-tube anaerobic most-probable-number (MPN) methods with anaerobic liquid enrichment media. The number of methanogenic bacteria (MB) and nitrogen-fixing bacteria (NFB) was determined by the rolling tube method in triplicate. Soil respiration was monitored by a 102G-type gas chromatography with a stainless steel column filled with GDX-104 and a thermal conductivity detector. Results Quinclorac concentration was an important factor affecting the populations of various culturable microorganisms. There were some significant differences in the aerobic heterotrophic bacteria. AFB and DNB between soils were supplemented with quinclorac and non-quinclorac at the early stage of incubation, but none of them was persistent. The number of fungi and DNB was increased in soil samples treated by lower than 1.33μg·g-1 dried soil, while the CFU of fungi and HPAB was inhibited in soil samples treated by higher than 1.33μg·g-1 dried soil. The population of actinomycete declined in negative proportion to the concentrations of quinclorac applied after 4 days. However, application of quinclorac greatly stimulated the growth of AFB and NFB. MB was more sensitive to quinclorac than the others, and the three soil samples with concentrations higher than 1 μg·g-1 dried soil declined significantly to less than 40% of that in the control, but the number of samples with lower concentrations of quinclorac was nearly equal to that in the control at the end of experiments. Conclusion Quinclorac is safe to the soil microorganisms when applied at normal concentrations (0.67μg·g-1).展开更多

文摘Sharp eyespot(Rhizoctonia cerealis)is a widespread soil-borne fungal disease that poses a severe threat to wheat health, and it is one of the main obstacles to achieving stable and high-quality wheat yields in China. Our collaborative team has developed a novel, efficient, and low-toxicity fungicide named Y17991(N-(2-(2,4-bis-(trifluoromethyl)phenoxy)phenyl)-3-(difluoromethyl)-1-methyl-1H-pyrazole-4-carboxamide).Preliminary laboratory tests confirmed the significant inhibitory effect of this agent on R.cerealis.Large-area field trials also demonstrated its efficacy, with a disease prevention index of 83.52%,which is 1.97%greater than that of the widely used thifluzamide, and it significantly increased the wheat yield. Moreover, this study explored the impacts of Y17991 on the structure and function of the microbial community in wheat rhizosphere soil.Bacterial communities were more strongly affected than fungal communities.Y17991 significantly modulated key amino acid metabolic pathways and certain biosynthetic processes in diseased wheat rhizospheres, and it also enhanced certain biosynthetic pathways and metabolic activities in healthy wheat rhizospheres.Additionally, the application of Y17991 regulated rhizosphere metabolites, thus exerting significant control over the microbial community. We identified 15 microbial strains potentially involved in the prevention and treatment of R.cerealis, and Y17991 treatment promoted the growth of Pedobacter and Bacillus strains. These strains not only aid in plant growth but they also have the potential for disease prevention. In summary,Y17991 application at a reasonable dose does not cause significant disruption to nontarget rhizosphere microbial communities. In future studies,we will continue to investigate the impacts of Y17991 on nonmicrobial components in soil ecosystems, such as protozoa and nematodes. Our research provides a theoretical basis for the scientific application and promotion of new fungicides and offers a significant reference for establishing a comprehensive system for assessing the ecological impact of pesticides on the environment.

文摘BACKGROUND Hepatocellular carcinoma(HCC)is the most common subtype of primary liver cancer with varied incidence and epidemiology worldwide.Sorafenib is still a recommended treatment for a large proportion of patients with advanced HCC.Different patterns of treatment responsiveness have been identified in differentiated hepatoblastoma HepG2 cells and metastatic HCC SNU449 cells.AIM To define the long non-codingRNA-microRNA-mRNA(lncRNA-miRNA-mRNA)predicted signatures related to selected hallmarks of cancer(apoptosis, autophagy, cell stress, cell dedifferentiation and invasiveness)in RNAseq studies using Sorafenib-treated HepG2 and SNU449 cells.Various available software analyses allowed us to establish the lncRNA-miRNA-mRNA regulatory axes following treatment in HepG2 and SNU449 cells.METHODS HepG2 and SNU449 cells were treated with Sorafenib(10μmol/L)for 24 hours.Total RNA,including small and long RNA,was extracted with a commercial miRNeasy kit.RNAseq was carried out for the identification of changes in lncRNA-miRNA-mRNA regulatory axes.RESULTS MALAT,THAP9-AS1 and SNGH17 appeared to coordinately regulate miR-374b-3p and miR-769-5p that led to upregulation of SMAD7,TIRARP,TFAP4 and FAXDC2 in HepG2 cells.SNHG12,EPB41 L4A-AS1,LINC01578,SNHG12 and GAS5 interacted with let-7b-3p,miR-195-5p and VEGFA in SNU449 cells. The axes MALAT1/hsamir-374b-3p/SMAD7 and MALAT1/hsa-mir-769-5p/TFAP4 were of high relevance for Sorafenib response in HepG2 cells, whereas PVT1/hsa-miR-195-5p/VEGFA was responsible for the differential response of SNU449 cells to Sorafenib treatment.CONCLUSION Critical lncRNAs acting as sponges of miRNA were identified that regulated mRNA expression, whose proteins mainly increased the antitumor effectiveness of the treatment(SMAD7,TIRARP,TFAP4,FAXDC2 and ADRB2).However,the broad regulatory axis leading to increased VEGFA expression may be related to the side effect of Sorafenib in SNU449 cells.

文摘The organotypic retinal explant culture has been established for more than a decade and offers a range of unique advantages compared with in vivo experiments and cell cultures. However, the lack of systematic and continuous comparison between in vivo retinal development and the organotypic retinal explant culture makes this model controversial in postnatal retinal development studies. Thus,we aimed to verify the feasibility of using this model for postnatal retinal development studies by comparing it with the in vivo retina. In this study,we showed that postnatal retinal explants undergo normal development, and exhibit a consistent structure and timeline with retinas in vivo.Initially,we used SOX2 and PAX6 immunostaining to identify retinal progenitor cells. We then examined cell proliferation and migration by immunostaining with Ki-67 and doublecortin,respectively.Ki-67-and doublecortin-positive cells decreased in both in vivo and explants during postnatal retinogenesis, and exhibited a high degree of similarity in abundance and distribution between groups.Additionally,we used Ceh-10 homeodomain-containing homolog,glutamate-ammonia ligase(glutamine synthetase),neuronal nuclei, and ionized calcium-binding adapter molecule 1 immunostaining to examine the emergence of bipolar cells,Müller glia, mature neurons, and microglia, respectively. The timing and spatial patterns of the emergence of these cell types were remarkably consistent between in vivo and explant retinas. Our study showed that the organotypic retinal explant culture model had a high degree of consistency with the progression of in vivo early postnatal retina development. The findings confirm the accuracy and credibility of this model and support its use for long-term,systematic,and continuous observation.
