The root-knot nematode disease(RKN)caused by Meloidogyne incognita is a devastating threat to the production of crops,especially greenhouse vegetables.Fluopyram,a new nematicide,has been widely used by root drenching ...The root-knot nematode disease(RKN)caused by Meloidogyne incognita is a devastating threat to the production of crops,especially greenhouse vegetables.Fluopyram,a new nematicide,has been widely used by root drenching one-by-one plants for the management of RKN.However,as a common non-fumigation nematicide,its distribution in soil was often the key limiting factor that determined the efficacy and efficiency.To optimize the utilization of fluopyram against RKN,the control efficacy of application methods including blending-of-soil,rootirrigation,drip-irrigation,and furrow-irrigation and the correlation between nematicide distribution and efficacy were studied.The toxicities of fluopyram and abamectin to nematodes collected from different sources and backgrounds exhibited limited differences from crop and region.Compared to the control,all nematicide treatments reduced root galling by M.incognita in cucumber and increased the yield of cucumber.Blending-of-soil treatment and root-irrigation treatment were more beneficial for increasing the distribution area of nematicides in soil,then promoting the control efficacy of nematicides against RKN and improving cucumber production.This work showed that the application of fluopyram by blending-of-soil has the same efficacy as by rootirrigation in controlling RKN in cucumber production,moreover is more efficient and can be an alternative option.展开更多
Sea cucumber derived sulfated sterols significantly ameliorated insulin resistance and decreased lipid accumulation compared to plant sterols.Interestingly,our recent study found that intervention with sea cucumber su...Sea cucumber derived sulfated sterols significantly ameliorated insulin resistance and decreased lipid accumulation compared to plant sterols.Interestingly,our recent study found that intervention with sea cucumber sulfated sterols could significantly increase blood glucose levels of healthy mice in the presence of glucose,while cholesterol sulfate,as one of sulfated sterols,did not have the same effect.However,the exact mechanism of sulfated sterols on glucose metabolism is still unknown.In the present study,we investigated the potential mechanism by which sulfated sterols influenced blood glucose homeostasis in healthy mice.Results showed that intervention with sea cucumber sulfated sterols did not affect the levels of hormones related to glucose metabolism,while led to a significant decrease in the synthesis of liver glycogen and muscle glycogen.Besides,the expression of proteins associated with the promotion of gluconeogenesis dramatically increased in the mice intervened with sea cucumber sulfated sterols.These findings suggested that sea cucumber sulfated sterols might change blood glucose metabolism in healthy mice by reducing glycogenesis and promoting gluconeogenesis.展开更多
Fruit spine density is an important commercial trait for cucumber(Cucumis sativus L.).Most North China-type cucumbers that are grown over large areas have a dense-spine phenotype,which directly affects the appearance ...Fruit spine density is an important commercial trait for cucumber(Cucumis sativus L.).Most North China-type cucumbers that are grown over large areas have a dense-spine phenotype,which directly affects the appearance quality,storage,and transportation of the fruits.Here,we isolated a novel few spines mutant(fs2)from the wild-type(WT)inbred line WD1,a North China-type cucumber with high density fruit spines,by an ethyl methanesulfonate(EMS)mutagenesis treatment.Genetic analysis revealed that the phenotype of fs2 is controlled by a single recessive nuclear gene.We fine-mapped the fs2 locus using F_(2) and BC_(1) populations(1,802 and 420 individuals,respectively),which showed that the candidate gene of FS2(Csa4G652850)encodes an ARID-HMG transcription factor containing an AT-rich interaction domain(ARID)and a high mobility group box domain(HMG).One SNP(C to T)and one InDel(a 40-bp deletion)in the coding region of FS2 result in amino acid variation and premature translation termination in the fs2 mutant,respectively.FS2 was found to be highly expressed in the apical buds and young ovaries.In addition,experiments suggest that FS2 participates in the regulation of fruit spine initiation by activating the expression of the Tril gene in cucumber.This work provides not only an important reference for understanding the molecular mechanisms of fruit spine development but also an important resource for fruit appearance quality breeding in cucumber.展开更多
Plant biomass is an important agronomic trait that has been subjected to intense human selection for yield improvement.The underlying mechanism regulating biomass formation is currently gaining increasing attention,bu...Plant biomass is an important agronomic trait that has been subjected to intense human selection for yield improvement.The underlying mechanism regulating biomass formation is currently gaining increasing attention,but it remains unexplored.In this study,we isolated a cucumber(Cucumis sativus L.)minicuke mutant with remarkably reduced biomass.The causative gene was identified as CsNMT1,a homologue of the Arabidopsis thaliana N-myristoyltransferase1.Our clustered regularly interspaced shot palindromic repeat-based genome editing confirmed the key role of CsNMT1 in biomass regulation.Multi-omics analyses integrating metabolomic and transcriptomic analyses revealed the suppression of a very early step of lignin biosynthesis and the corresponding down-regulation of genes involved in lignin biosynthesis in the minicikue mutant,suggesting an unexpected pathway for regulating biomass accumulation through lignin sink strength.Our findings demonstrate the function of NMT1 in regulating plant biomass and its potential application value for biomass improvement in cucurbits.展开更多
It has been reported that sea cucumber intestine hydrolysates(SCIH)could promote glutamine metabolism in mice,while there is a close connection between glutamine metabolism and insulin sensitivity.However,the effect o...It has been reported that sea cucumber intestine hydrolysates(SCIH)could promote glutamine metabolism in mice,while there is a close connection between glutamine metabolism and insulin sensitivity.However,the effect of SCIH on insulin resistance is still unclear.The results showed that SCIH hydrolyzed by flavor protease had significant activity using the insulin-induced Hep G2 cell model.Animal experiments exhibited that SCIH supplementation significantly improved the high-fat and high-sucrose diet-induced impaired glucose tolerance,reduced fasting serum glucose and glycosylated serum protein.Besides,SCIH ameliorated islet vacuolization and decreased the pancreas TNF-αand IL-6 by 32.1%and 36.2%,respectively.Immunofluorescence staining results showed that SCIH promoted insulin secretion.Interestingly,SCIH significantly increased the liver glutamine levels and upregulated the expression of glutaminase1(GLS1)and glutamate dehydrogenase 1(GLUD1).Furthermore,SCIH increased liver acetyl-Co A levels to enhance histone acetylation and activate the gene transcription and translation on glucose metabolism-related IRS/PI3K/AKT signaling pathway,thereby attenuating insulin resistance.The present findings proposed the potential value for developing functional foods in SCIH utilization.展开更多

This paper aimed to explore the mechanism of tetrapeptide Val-Thr-Pro-Tyr(VTPY)in improving alcoholic gastric injury.VTPY has the potential to enhance the growth and movement of normal human gastric epithelial cells(G...This paper aimed to explore the mechanism of tetrapeptide Val-Thr-Pro-Tyr(VTPY)in improving alcoholic gastric injury.VTPY has the potential to enhance the growth and movement of normal human gastric epithelial cells(GES-1).Following ethanol-induced impairment,VTPY effectively improved migration of GES-1 and human umbilical vein endothelial cells(HUVEC)cells,enhanced angiogenesis,eliminated cellular and mitochondrial reactive oxygen species(ROS),inhibited excessive mitochondrial division,enhanced F-actin polymerization and mitochondrial respiratory capacity.To counteract excessive mitochondrial fission,VTPY primarily restores the mitochondria dynamics by reducing the expression of Drpl and Fis1,while increasing Mfn2.Further studies utilizing inhibitors clarifies that the inhibition of excessive mitochondrial fission can markedly reduce F-actin depolymerization,consequently enhancing cell migration.Additionally,VTPY can inhibit the apoptosis pathway by maintaining potential of mitochondrial membrane,preventing the release of mitochondrial cytochrome C,bolstering the levels of Bcl-XL,while reducing the levels of Bax and cleaved-Caspase-3.Further investigations using inhibitors demonstrates that excessive mitochondrial fission could activate apoptotic pathway.However,VTPY counteracts this effect and enhance cells viability.Further evidence suggests that VTPY effectively improves ulcer index and pathologic changes,relieves inflammation,enhances the balance of oxidation and anti-oxidation,promotes angiogenesis,improves the expression of mitochondrial dynamics factors,blocks apoptotic pathway,and subsequently ameliorates gastric damage in mice through Fis-1/Bcl-2 pathway.展开更多

The transformed plants...Tobacco ( Nicotiana tabacum L.) “NC89” plants were transformed with deletion mutant of cucumber mosaic virus (CMV) movement protein (MP) gene and full_length CMV MP gene, respectively. The transformed plants were analyzed with polymerase chain reaction (PCR), PCR_Southern, Southern and Western blots. R 0 generation of the transgenic plants were inoculated with CMV. Five out of 10 lines of tobacco plants (BMPK) transformed with CMV MP deletion mutant gene showed high resistance to CMV infection and remained symptomless for up to 50 days post_inoculation. In contrast, tobacco plants (BMPR) transformed with full_length CMV MP gene did not show resistance to CMV infection. However, most of the infected full_length CMV MP gene transgenic plants recovered by showing none or very mild mosaic symptoms in 40 days post_inoculation. The results of R 1 generation of the BMPK transgenic plants tested under field conditions showed that all 5 lines of transgenic plants could delay the virus disease development.展开更多

The results showed that there were significant differences in seed oil content between different cucumber varieties with the amplitude of variation rangeing from 31.03% to 40.59%, among which the seed oil content of south China cucumber was the highest of 40.59%, while the seed oil content of north China cucumber was the lowest of 31.03%. At the same time, inoculation test was also conducted after induced treatment, and the disease condi- tion was coumed. [ Result ] These chemical treatments promoted protective enzyme activities (POD, SOD, PAL, PPO) in different degree, improved the ultra-weak luminescence, and reduced the disease index of cucumber leaves. [ Conclusion] Five chemical treatments expressed good effects on induced resistance. Among the treatments, oxalate had the best induced effect, followed by salicylic acid, chitosan, Na2 HPO4 and calcium nitrate.展开更多

[Method] Cucumber cultivar Zhongnong No.8 was treated with 0, 5, 10 and 15 mmol/L GB solutions for 15 min and stored at 4 ℃. Changes in oxidative metabolism-related parameters were observed. [Result] Increasing exogenous GB concentration could enhance GB content in cucumbers, decline lipoxygenase(LOX) activity, improve peroxidase(POD) and catalase(CAT) activities, remove effectively hydrogen peroxide(H2O2) and reduce the accumulation of malondialdehyde(MDA). Per fruit weighs about 230 g. lt is dark green, densely covered with thorns. lt tastes crisp, containing 167.7 mg/kg Vc, and it is resistant to downy mildew, pow-dery mildew and bacterial angular leaf spot disease. lt is yielding and tolerant to low temperature and weak luminance. [Method] Nutrient solution containing low concentrations of Mn2+, Cu2+ or Zn2+ were used for cultivating the grafted cucumber seedlings for the cold treatment. [Result] The results indicated that reducing the concentrations of Mn2+, Cu2+ or Zn2+ in nutrient solution had declined the SOD activities in grafted cucumber leaves and increased the rates of electrolytic leakage. Mn2+ had shown the maximum influences, followed by Cu2+, and Zn2+ had shown the minimum influences. 文摘Fruit spine density is an important commercial trait for cucumber(Cucumis sativus L.).Most North China-type cucumbers that are grown over large areas have a dense-spine phenotype,which directly affects the appearance quality,storage,and transportation of the fruits.Here,we isolated a novel few spines mutant(fs2)from the wild-type(WT)inbred line WD1,a North China-type cucumber with high density fruit spines,by an ethyl methanesulfonate(EMS)mutagenesis treatment.Genetic analysis revealed that the phenotype of fs2 is controlled by a single recessive nuclear gene.We fine-mapped the fs2 locus using F_(2) and BC_(1) populations(1,802 and 420 individuals,respectively),which showed that the candidate gene of FS2(Csa4G652850)encodes an ARID-HMG transcription factor containing an AT-rich interaction domain(ARID)and a high mobility group box domain(HMG).One SNP(C to T)and one InDel(a 40-bp deletion)in the coding region of FS2 result in amino acid variation and premature translation termination in the fs2 mutant,respectively.FS2 was found to be highly expressed in the apical buds and young ovaries.In addition,experiments suggest that FS2 participates in the regulation of fruit spine initiation by activating the expression of the Tril gene in cucumber.This work provides not only an important reference for understanding the molecular mechanisms of fruit spine development but also an important resource for fruit appearance quality breeding in cucumber.

文摘Plant biomass is an important agronomic trait that has been subjected to intense human selection for yield improvement.The underlying mechanism regulating biomass formation is currently gaining increasing attention,but it remains unexplored.In this study,we isolated a cucumber(Cucumis sativus L.)minicuke mutant with remarkably reduced biomass.The causative gene was identified as CsNMT1,a homologue of the Arabidopsis thaliana N-myristoyltransferase1.Our clustered regularly interspaced shot palindromic repeat-based genome editing confirmed the key role of CsNMT1 in biomass regulation.Multi-omics analyses integrating metabolomic and transcriptomic analyses revealed the suppression of a very early step of lignin biosynthesis and the corresponding down-regulation of genes involved in lignin biosynthesis in the minicikue mutant,suggesting an unexpected pathway for regulating biomass accumulation through lignin sink strength.Our findings demonstrate the function of NMT1 in regulating plant biomass and its potential application value for biomass improvement in cucurbits.

文摘It has been reported that sea cucumber intestine hydrolysates(SCIH)could promote glutamine metabolism in mice,while there is a close connection between glutamine metabolism and insulin sensitivity.However,the effect of SCIH on insulin resistance is still unclear.The results showed that SCIH hydrolyzed by flavor protease had significant activity using the insulin-induced Hep G2 cell model.Animal experiments exhibited that SCIH supplementation significantly improved the high-fat and high-sucrose diet-induced impaired glucose tolerance,reduced fasting serum glucose and glycosylated serum protein.Besides,SCIH ameliorated islet vacuolization and decreased the pancreas TNF-αand IL-6 by 32.1%and 36.2%,respectively.Immunofluorescence staining results showed that SCIH promoted insulin secretion.Interestingly,SCIH significantly increased the liver glutamine levels and upregulated the expression of glutaminase1(GLS1)and glutamate dehydrogenase 1(GLUD1).Furthermore,SCIH increased liver acetyl-Co A levels to enhance histone acetylation and activate the gene transcription and translation on glucose metabolism-related IRS/PI3K/AKT signaling pathway,thereby attenuating insulin resistance.The present findings proposed the potential value for developing functional foods in SCIH utilization.

文摘This paper aimed to explore the mechanism of tetrapeptide Val-Thr-Pro-Tyr(VTPY)in improving alcoholic gastric injury.VTPY has the potential to enhance the growth and movement of normal human gastric epithelial cells(GES-1).Following ethanol-induced impairment,VTPY effectively improved migration of GES-1 and human umbilical vein endothelial cells(HUVEC)cells,enhanced angiogenesis,eliminated cellular and mitochondrial reactive oxygen species(ROS),inhibited excessive mitochondrial division,enhanced F-actin polymerization and mitochondrial respiratory capacity.To counteract excessive mitochondrial fission,VTPY primarily restores the mitochondria dynamics by reducing the expression of Drpl and Fis1,while increasing Mfn2.Further studies utilizing inhibitors clarifies that the inhibition of excessive mitochondrial fission can markedly reduce F-actin depolymerization,consequently enhancing cell migration.Additionally,VTPY can inhibit the apoptosis pathway by maintaining potential of mitochondrial membrane,preventing the release of mitochondrial cytochrome C,bolstering the levels of Bcl-XL,while reducing the levels of Bax and cleaved-Caspase-3.Further investigations using inhibitors demonstrates that excessive mitochondrial fission could activate apoptotic pathway.However,VTPY counteracts this effect and enhance cells viability.Further evidence suggests that VTPY effectively improves ulcer index and pathologic changes,relieves inflammation,enhances the balance of oxidation and anti-oxidation,promotes angiogenesis,improves the expression of mitochondrial dynamics factors,blocks apoptotic pathway,and subsequently ameliorates gastric damage in mice through Fis-1/Bcl-2 pathway.

文摘[Objective] This study was to investigate the effects of different irrigation amount on water consumption and water use efficiency of greenhouse cucumber.[Method]Under the condition of drip irrigation with different water amounts in sunlight greenhouse of the arid areas in Ningxia,the soil water was measured and the water consumption of crop was calculated.[Result]When irrigation amount was 563 mm,the water consumption as a whole gradually increased with the delay of growth period,reached peak during the thriving stage of fruit setting,and then gradually declined;in each treatment,the daily water consumption increased with the increasing of irrigation amount during each growth period.However,the consumption of soil moisture reduced with the significant increase of irrigation.563 mm of irrigation amount could meet the water requirements of cucumber and began to add water to soil,and water utilization efficiency could reach 33.4 kg/m3.[Conclusion]The research had provided theoretical basis for water management in the production process of greenhouse cucumber.
文摘Tobacco ( Nicotiana tabacum L.) “NC89” plants were transformed with deletion mutant of cucumber mosaic virus (CMV) movement protein (MP) gene and full_length CMV MP gene, respectively. The transformed plants were analyzed with polymerase chain reaction (PCR), PCR_Southern, Southern and Western blots. R 0 generation of the transgenic plants were inoculated with CMV. Five out of 10 lines of tobacco plants (BMPK) transformed with CMV MP deletion mutant gene showed high resistance to CMV infection and remained symptomless for up to 50 days post_inoculation. In contrast, tobacco plants (BMPR) transformed with full_length CMV MP gene did not show resistance to CMV infection. However, most of the infected full_length CMV MP gene transgenic plants recovered by showing none or very mild mosaic symptoms in 40 days post_inoculation. The results of R 1 generation of the BMPK transgenic plants tested under field conditions showed that all 5 lines of transgenic plants could delay the virus disease development.

文摘[Objective] The research aimed to construct the fusion protein expression vector of α-galactosidase-EGFP (enhanced green fluorescent protein) in cucumber controlled by CaMV35S promoter.[Method] CaMV35S promoter sequence and the coding region of EGFP were amplified by polymerase chain reactions (PCR) with vector pCambia 1303 as the template.Using reverse transcript PCR technology,with total RNAs of cucumber as template,the coding region of acid α-galactosidase Ⅰ in cucumber was amplified.The above three fragments were inserted into the multiple cloning sites of expression vector pCambia 1381c.The fusion expression vector of α-galactosidase-EGFP located at the C-terminal of the target genes was constructed.[Result] After enzyme digestion and sequencing,the fusion expression of α-galactosidase-EGFP in cucumber was constructed successfully.[Conclusion] The research laid the experimental basis for further study on the subcellular localization of α-galactosidase in cucumber.

文摘Present mature plants hydroponic technology was used,combined with some excellent characteristics,such as growth conditions was easy to control and process of root growth was easy to continuously observe,the nematicidal activity of 5 kinds of Chinese herbs extracts and the compound solution of Avermectin,with strong contact toxicity effect indoor,was systematically studied and investigated the affection on the root-knot nematode parasitized on the cucumber seeding stage. It is found that under the premise of no influence on root growth of cucumber,extracts from Picrorhiza scrophulariiflora and Punica granatum showed strong prevention and nematicidal activity,and had the similar efficacy of Avermectin; while the extracts from Cibotium barometz,Aucklandia lappa Decne and Fructus cnidii showed low nematicidal activity and various degrees inhibition effect on plant growth.

文摘With 35 different ecological types of cucumber germplasm resources as test materials, the soxhlet extraction was used to extract the crude fat from the cu- cumber seeds to analyze the seed oil content, with the aim to lay foundations for the application and new variety breeding related with cucumber seed oil. The results showed that there were significant differences in seed oil content between different cucumber varieties with the amplitude of variation rangeing from 31.03% to 40.59%, among which the seed oil content of south China cucumber was the highest of 40.59%, while the seed oil content of north China cucumber was the lowest of 31.03%. Four va.rieties with oil content of greater than 38.50% and four varieties with oil content of less than 32.01% werescreened out.

文摘ln order to explore the design and construction of cucumber powdery mildew warning system in solar greenhouse, internet of things technology was used to conduct the real-time dynamic monitoring of the incidence of cucumber powdery mildew and cucumber growth environment in solar greenhouse. The growth environ-ment included temperature and humidity of air and soil. Logistic regression model was used to construct cucumber powdery mildew warning model. The results showed that humidity characteristic variable (maximum air humidity) and temperature characteristic variable (maximum air temperature) had significant effects on the inci-dence probability of cucumber powdery mildew in solar greenhouse. And it was fea-sible to construct cucumber powdery mildew warning system in solar greenhouse with internet of things.

文摘[ Objectlve] The paper was to compare the induced resistance of several chemicals on cucumber against Pseudoperonospora cubensis [ Method ] When the cucumber seedlings grew to 4-leaf stage, the second true leaf was treated with salicylic acid, oxalate, chitosan, calcium nitrate and Na2HPO4, the activity of protective enzymes in the third leaf was determined after 7 d. At the same time, inoculation test was also conducted after induced treatment, and the disease condi- tion was coumed. [ Result ] These chemical treatments promoted protective enzyme activities (POD, SOD, PAL, PPO) in different degree, improved the ultra-weak luminescence, and reduced the disease index of cucumber leaves. [ Conclusion] Five chemical treatments expressed good effects on induced resistance. Among the treatments, oxalate had the best induced effect, followed by salicylic acid, chitosan, Na2 HPO4 and calcium nitrate.
文摘[Objective] This study aimed to analyze the effects of different concentrations of glycine betaine(GB) on oxidation metabolism in cucumbers under low-temperature stress and to investigate the possible mechanism of low-temperature resistance in cucumber during low-temperature storage. [Method] Cucumber cultivar Zhongnong No.8 was treated with 0, 5, 10 and 15 mmol/L GB solutions for 15 min and stored at 4 ℃. Changes in oxidative metabolism-related parameters were observed. [Result] Increasing exogenous GB concentration could enhance GB content in cucumbers, decline lipoxygenase(LOX) activity, improve peroxidase(POD) and catalase(CAT) activities, remove effectively hydrogen peroxide(H2O2) and reduce the accumulation of malondialdehyde(MDA). [Conclusion] Treating cucumbers with10 mmol/L GB exhibited the most remarkable effect.

文摘‘Oriental Show’ is a generation of new cucumber variety, with the superi-or inbred line ‘101’ as the female parent and inbred line ‘102’ as the male par-ent. The growth vigor tends to be strong and the first female blossom produces on the 4th or the 5th internode. Fruit is 34 cm long and 32 cm in transverse diameter. Per fruit weighs about 230 g. lt is dark green, densely covered with thorns. lt tastes crisp, containing 167.7 mg/kg Vc, and it is resistant to downy mildew, pow-dery mildew and bacterial angular leaf spot disease. lt is yielding and tolerant to low temperature and weak luminance. The yield can be as high as over 112 443.778 kg/hm^2, and ‘Oriental Show’ is suitable to be cultivated in greenhouses in early spring or solar greenhouses in north China or Henan province.

文摘[Objective] The study discussed the indoor activity and field control effect of vegetable oil on cucumber powdery mildew.[Method] The cucumber seedlings of Changchunmici were adopted as the material,which were inoculated with the fungus pathogen of cucumber powdery mildew naturally occurred in the field.The indoor protective test and field test were performed respectively to observe the control effects of different plant oils EC on the cucumber powdery mildew.[Result] When six plant oils EC including cottonseed oil,soybean oil,canola oil,corn oil,sesame oil and sunflower oil was diluted into 10 and 5 ml/L,they had better control effect on cucumber powdery mildew.As for the indoor protective effect,the protective effect of the vegetable oil EC was equivalent to the control agent triadimefon EC,the field control effect of vegetable oil EC was in the range of 60%-75%,which was consistent with 69%-70% control effect of contrast agent with 1 ml/L triadimefon EC dilution.[Conclusion] All vegetable oils EC had significant control effect on cucumber powdery mildew in the test.

文摘[Objective] The research aimed to study the effects of reducing the concentrations of Mn2+, Cu2+ or Zn2+ in nutrient solution on the contents of Mn, Cu and Zn of grafted cucumber leaves as well as on the SOD and isozymes activities and the rate of electrolytic leakage under cold stress. [Method] Nutrient solution containing low concentrations of Mn2+, Cu2+ or Zn2+ were used for cultivating the grafted cucumber seedlings for the cold treatment. [Result] The results indicated that reducing the concentrations of Mn2+, Cu2+ or Zn2+ in nutrient solution had declined the SOD activities in grafted cucumber leaves and increased the rates of electrolytic leakage. Mn2+ had shown the maximum influences, followed by Cu2+, and Zn2+ had shown the minimum influences. [Conclusion] Cold resistance of cucumber can be improved by changing the contents of Mn2+, Cu2+ or Zn2+ in nutrient solution.
