目的:探究One cut结构域家族成员2(ONECUT2)是否可以作为对5-FU耐药的胃癌复发患者的治疗靶点。方法:利用癌症基因组图谱(TCGA)数据库分析ONECUT2基因在胃癌患者及胃癌复发患者的表达情况。利用基因集富集分析(GSEA)评估TCGA数据库中ONE... 目的:探究One cut结构域家族成员2(ONECUT2)是否可以作为对5-FU耐药的胃癌复发患者的治疗靶点。方法:利用癌症基因组图谱(TCGA)数据库分析ONECUT2基因在胃癌患者及胃癌复发患者的表达情况。利用基因集富集分析(GSEA)评估TCGA数据库中ONECUT2表达量与药物代谢相关生物学过程的相关性。利用CCK-8细胞毒性实验评估胃癌细胞敲低或过表达ONECUT2对5-FU的耐药情况。分析30例胃癌术后复发患者ONECUT2表达量与5-FU治疗效果的相关性。结果:TCGA数据库分析显示,在经受过化疗且复发的胃癌患者中,ONECUT2高表达的患者无病间隔期(disease-free interval,DFI)更短。GSEA分析结果表明,ONECUT2高表达与异生药物代谢通路呈正相关,这提示可能与胃癌患者的耐药相关。体外实验中,敲低ONECUT2降低胃癌细胞对5-FU的半数抑制浓度(IC_(50))值;反之,过表达ONECUT2增加细胞对5-FU的IC_(50)值。30例胃癌复发患者对5-FU的药物敏感性与ONECUT2表达量相关。结论:胃癌复发患者的ONECUT2表达量更高,ONECUT2高表达与胃癌复发患者的5-FU耐药相关,可能是潜在的治疗靶点。 展开更多

In recent decades,many additives with difierent characteristics have been applied to strengthen and toughen Al_2O_(3)一based ceramic cutting tool materials.Among them,SiC whiskers and SiC nanoparticles showed excellent performance in improving the material properties.W hile no attempts have been made to add SiC whiskers and SiC nanoparticles together into the ceramic matrix and the synergistically toughening efects of them have not been studied.An Al_2O_(3)一SiCw-SiCno advanced ceramic cutting tool material is fabricated by adding both one--dimensional SiC whiskers and zero--dimensional SiC nanoparticles into the Al_2O_(3)matrix with an efective dispersing and mixing process.The composites with 25 vo1%SiC whiskers and 25 vo1%SiC nanoparticles alone are also investegated for comparison purposes.Results show that the Al_2O_(3)一SiCw-SiCno composite with both 20 vo1%SiC whiskers and 5 vo1%SiC nanoparticles additives have much improved mechanical properties.The flexural strength of Al_2O_(3)-SiCw-SiCⅡD is 730~95 MPa and fracture toughness is 5.6±0.6 MPa’m.The toughening and strengthening mechanisms of SiC whiskers and nanoparticles are studied when they are added either individually or in combination.It is indicated that when SiC whiskers and nanoparticles are added together,the grains are further refined and homogenized,so that the microstructure and fracture mode ratio is modified.The SiC nanoparticles are found helpful to enhance the toughening efects of the SiC whiskers.The proposed research helps to enrich the types of ceramic cutting too1 and is benefit to expand the application range of ceramic cutting too1. 展开更多

胃癌根治术联合术后洛铂腹腔热灌注对进展期胃癌的治疗效果研究

目的探究胃癌根治术联合术后洛铂腹腔热灌注对进展期胃癌的治疗效果及对One Cut同源盒蛋白2(ONECUT2)基因、Yes相关蛋白1(YAP1)mRNA表达的影响。方法选取2021年6月至2023年6月某医院收治的120例进展期胃癌患者为研究对象,采用随机数字... 目的探究胃癌根治术联合术后洛铂腹腔热灌注对进展期胃癌的治疗效果及对One Cut同源盒蛋白2(ONECUT2)基因、Yes相关蛋白1(YAP1)mRNA表达的影响。方法选取2021年6月至2023年6月某医院收治的120例进展期胃癌患者为研究对象,采用随机数字表法将患者分为联合组和对照组,每组60例。联合组采用胃癌根治术联合术后洛铂腹腔热灌注治疗方案,对照组采用胃癌根治术联合常规化疗(卡培他滨+顺铂)方案。比较两组进展期胃癌患者的疗效、手术相关指标,治疗前后血清相关肿瘤指标、并发症及随访1年的预后情况。结果联合组进展期胃癌患者治疗总有效率高于对照组,部分缓解(PR)率高于对照组,进展(PD)率低于对照组(P<0.05)。两组术中出血量、术后48 h视觉模拟疼痛(VAS)评分、手术切口长度和术后肛门排气时间比较,差异无统计学意义(P>0.05),但联合组手术时间比对照组长(P<0.05)。与治疗前相比,联合组治疗后血清ONECUT2基因和YAP1 mRNA表达均下降,血清癌胚抗原(CEA)和糖类抗原199(CA199)水平均降低(P<0.05);治疗后,联合组血清ONECUT2基因和YAP1 mRNA表达、CEA和CA199水平均低于对照组(P<0.05)。联合组和对照组并发症总发生率分别为25.00%和15.00%,差异无统计学意义(P>0.05)。联合组患者中位生存期>12个月,对照组患者中位生存期为11.18个月(P<0.05)。结论胃癌根治术联合术后洛铂腹腔热灌注用于治疗进展期胃癌能有效提高疗效,下调ONECUT2基因、YAP1 mRNA表达,不增加并发症风险,且有利于延长生存期,但手术时间较长。 展开更多

As a natural and environmentally friendly renewable material,Northeast China ash wood(NCAW)(Fraxinus mandshurica Rupr.)was cut by water-jet assisted CO_(2) laser(WACL),the surface quality was evaluated by surface roughness of cut section.The surface roughness was measured by three-dimensional(3D)profilometry.Furthermore,the micromorphology of machined surface was observed by scanning electronic microscopy(SEM).Carbon content changes of machined surface were measured by energy dispersive spectrometer(EDS).A relationship between surface roughness and cutting parameters was established using response surface methodology(RSM).It is concluded that the cutting speed,laser power and water pressure played an important role in surface roughness of cut section.The surface roughness increased as an increase in laser power.It decreased caused by increasing of cutting speed and water pressure.Measurements revealed that the surface quality of NCAW part was improved using the optimized combination of cutting parameters.The established quadratic mathematical model of a good prediction is helpful for matching suitable cutting parameters to obtain expected surface quality. 展开更多
