In order to reveal the influence of different plant configurations on the microbial community structure and diversity in rhizosphere soil of Cotinus coggygria in Fragrant Hills park,the ITS+5.8S rDNA gene and 16S rDNA gene V3-V4 region sequencing analysis for fungi and bacteria,respectively,were conducted by high throughput sequencing(Illumina MiSeq).The results showed that the fungal diversity in the rhizosphere soil samples of C.coggygria in Fragrant Hills park in 2018 was significantly higher than that in 2016,and it was higher in the rhizosphere soil of healthy C.coggygria in Xunlupo than that in diseased ones in 2018.Verticillium dahliae,which is the causal agent of C.coggygria wilt,was detected in five soil samples.In 2018,the bacterial diversity in the rhizosphere soil of diseased C.coggygria in Xunlupo was the lowest,while it was the highest in the rhizosphere soil of healthy C.coggygria under Platycladus orientalis in Langfengting. 展开更多

Objective:To assess the analgesic and anti-inflammatory effects of standardized extract of Cotinus coggygria(C.coggygria)in different animal models.Methods:C.coggygria extracts(25,50,and 100 mg/kg)were administered to rats and mice(n=6)during hot plate,tail-flick,acetic acid-induced writhing,and formalin tests to determine its analgesic efficacy.The anti-inflammatory activity of C.coggygria extracts was evaluated by histamine and carrageenan-induced paw edema,cotton pellet-induced granuloma,and acetic acid-induced peritoneal capillary dye leakage tests.Results:C.coggygria extracts(50 and 100 mg/kg)significantly alleviated thermal and chemical-induced pain in rodents(P<0.05).It also demonstrated notable anti-inflammatory properties by mitigating histamine and carrageenan-induced paw edema,granuloma deposits,and vascular permeability(P<0.05).Moreover,C.coggygria extracts remarkably reduced TNF-α,IL-1β,IL-6,COX-2,and oxidative stress in rat paws(P<0.05).Carrageenan-induced histological aberrations in hind paw tissues were effectively(P<0.05)mitigated by treatment with C.coggygria extracts.Conclusions:C.coggygria Scop.extracts show analgesic and anti-inflammatory effects via inhibition of COX-2 and inflammatory and oxidative mediators. 展开更多

We analyzed the inhibition rates of eight kinds of fungicides to the pathogen of verticillium wilt of smoke tree (Cotinus coggygria) by measuring mycelium growth. Results show that four fungicides (Weijunjing, thiophanate-methyl 70% WP (wettable powder), carbendazim 50% WP and Junxianwei) have the best antifungal effects. Three fungicides, Weijunjing, carbendazim 50% WP and Junxianwei, were selected to determine the control of verticillium wilt in potted smoke tree seedlings. We found that the relative efficacy of Weijunjing was up to 74%. Based on the results, Weijunjing and carbendazim 50% WP were prepared for field trials. The results indicate that the relative efficacy of Weijunjing (300 times diluted) and carbendazim 50% WP (400 times diluted) were 66.2% and 48.9% in plot 1 and 26.4% and 31.8% in plot 2 after soil disinfection, and were 224% and 61.8% in plot 3 without soil disinfection in 2007. The relative efficacy of Weijunjing and carbendazim 50% WP were 49.6% and 45.4% in plot 2 in 2008. 展开更多

Objective:To investigate the therapeutic effects of aqueous infusion of Cotinus coggygria Scop.leaves(CCLAI)on the dextran sulphate sodium(DSS)-induced ulcerative colitis(UC)in mice.Methods:A total of 56 C57BL16J mice were divided into 7 groups,including control,DSS model,CCLAI4,CCLAI6,DSS+CCLAI4,DSS+CCLAI6 and DSS+mesalamine groups(n=8).Induction of UC was conducted by 7-day treatment of 3%DSS in mice.CCLAI in 2 different doses(4%and 6%)or mesalamine(250 mg/kg)was administered to C57BL/6J mice orally for 7 consecutive days.The mice in the control group received only tap water.Clinical parameters,antioxidant and anti-inflammatory activities,and histopathological changes in the colon mucosa were examined.Cell proliferatipn index in colon section was determined by immunonistochemical analysis.Colonic cyclooxygenase(COX)-2 and inducible nitric oxide synthase(i NOS)expression levels were detected by Western blot.Results:CCLAI treatment inhibited DSS-induced colon shortening and significantly improved the Disease Activity Index score(P<0.05 or P<0.01).We also found a significant decrease in myeloperoxidase levels,oxidative stress parameters,and neutrophil infiltration index in the experimental groups receiving CCLAI compared to the control group(all P<0.01).In addition,it was observed that CCLAI administration for 7 days decreased the pro-inflammatory cytokines levels including interleukin(IL)-1β,IL-6,interferon-γ,and tumor necrosis factor-αin the colon tissues of mice receiving DSS(P<0.05).Further,CCLAI treatment reduced the COX-2 and i NOS levels with an accompanying decrease in prostaglandin E2 and nitric oxide levels,respectively.Additionally,CCLAI treatment markedly decreased the histological lesion score,which was consistent with the reduction of the pro-inflammatory cytokine levels.Conclusion:CCLAI treatment significantly improved the DSS-induced UC in mice by diminishing histopathological damage in the colonic mucosa and markedly decreased the inflammatory response through inhibiting the formation of inflammatory cytokines and markers of oxidative stress. 展开更多

以紫霞黄栌(Cotinus coggygria var.‘Zixia’)为材料,组培紫霞黄栌外植体,获得最适宜紫霞黄栌的组培条件,得到一套紫霞黄栌苗木资源大规模生产的组织培养技术规范。最适宜的诱导分化培养基为MS基础培养基添加1.0 mg/L 6-BA和0.1 mg/L... 以紫霞黄栌(Cotinus coggygria var.‘Zixia’)为材料,组培紫霞黄栌外植体,获得最适宜紫霞黄栌的组培条件,得到一套紫霞黄栌苗木资源大规模生产的组织培养技术规范。最适宜的诱导分化培养基为MS基础培养基添加1.0 mg/L 6-BA和0.1 mg/L NAA,扦插苗、一年生枝条和二年生枝条作为外植体的诱导率分别达到96.7%、93.3%和73.3%。外植体材料在5~8月适宜采集。增殖培养最适宜条件为以改良MS培养基作为基础培养基,pH调至5.4,添加100μmol/L 6-BA、1μmol/L NAA、10μmol/L IAA并添加1 mg/L维生素C,评价得分4.23分,增殖系数最高达到8.97。生根培养使用MS改良培养基,添加0.3 mg/L IBA和0.2 mg/L NAA时生根率达到53%。移栽以消毒灭菌后的珍珠岩作为基质,温度控制为25℃,相对湿度保持在90%,移栽后组培苗成活率达到70%。使用该方法可大幅提高紫霞黄栌增殖数量。 展开更多
