Electrochemical carbon dioxide reduction reaction(CO_(2)RR)converts CO_(2) into valuable chemicals by consuming renewable electricity at mild conditions,making it a promising approach to achieving carbon neutrality.Ho...Electrochemical carbon dioxide reduction reaction(CO_(2)RR)converts CO_(2) into valuable chemicals by consuming renewable electricity at mild conditions,making it a promising approach to achieving carbon neutrality.However,the reaction of CO_(2) with hydroxide ions to form carbonates leads to low carbon utilization and energy efficiency in near-neutral or alkaline CO_(2)RR.The high concentration of protons in acidic media can effectively mitigate carbonate formation and deposition,thereby significantly minimizing carbon loss and energy consumption.Unfortunately,hydrogen evolution reaction(HER)is more kinetically favorable than CO_(2)RR in acidic media.Herein,we comprehensively overview recent progress in acidic CO_(2)RR and propose two strategies derived from the competing reaction pathways of HER and CO_(2)RR:one focuses on regulating the H+mass transport,while the other aims to modulate the intrinsic kinetic activity of CO_(2)RR.The two strategies are designed to compete for the limited active sites on the catalyst surface,inhibit side reactions,and enhance the activity and selectivity of CO_(2)RR.The representative approaches include modulating the interface electric field,constructing a local alkaline environment,and regulating competing adsorption sites.Finally,we also review the technical challenges and future perspectives of acidic CO_(2)RR coupled with membrane electrode assemblies(MEAs).展开更多

BACKGROUND As a non-coding RNA molecule,circular RNAs(circRNAs)have significant specificity,and existing data suggest a close relationship between them and the prognosis of patients with gastric cancer(GC).However,thi...BACKGROUND As a non-coding RNA molecule,circular RNAs(circRNAs)have significant specificity,and existing data suggest a close relationship between them and the prognosis of patients with gastric cancer(GC).However,this mechanism has no evidence yet.This article explores the functions of hsa_circRNA_102415 in the malignant behavior and potential downstream signaling of GC cells.The chosen approach is loss of signal and functional gain.AIM To investigate and analyze the relationship between hsa_circRNA_102415 and GC and explore its specific role.Results provide reference for other researchers to develop targeted treatment plans.METHODS The gene expression omnibus(GEO)database can be used to obtain the microarray dataset GSE83521.Data were analyzed using the GEO2R tool to identify differences in circRNAs between normal and GC samples.Quantitative real-time polymerase chain reaction was used to detect differentially expressed genes in GC tissue samples and adjacent cancer tissue samples.GC cells were transfected with small interfering-hsa_circRNA_104415 and plasmid DNA(pcDNA)-hsa_ircRNA_102415.Multiple detection methods,such as Transwell and cell counting kit 8,were used to evaluate cellular physiological activities,including cell invasion and proliferation.The relationship between Wnt family members 2B,microRNA(miR)-4529-5p,etc.,including argonaute 2-RNA immunoprecipitation and luciferase reporter genes was analyzed.Rescue experiments were conducted to analyze and explore the relationship between the malignant behavior of GC cells and hsa_circRNA_102415.RESULTS GEO2R analysis confirmed that hsa_circRNA_102415 had significantly higher expression levels in disease tissues.hsa_circRNA_102415 and miR-4529-5p showed a negative correlation in disease cells,suggesting that hsa_circRNA_102415 upregulated WNT2B expression in GC cells as a competing endogenous RNA for miR-4529-5p.miR-4529-5p mimic or small interfering-WNT2B reversed the effects of pcDNA-hsa_circRNA_102415 or miR-4529-5p inhibitor on cell malignant functions.CONCLUSION miR-4529-5p was used to successfully activate the potential of WNT2B,clarify the role of hsa_circRNA_102415 in GC cells,and provide reference for other researchers to develop targeted treatment plans.展开更多

Two kinds of probabilistic models are constructed, in which the dependent competing failure process is considered. First, based on the assumption of cumulative shock, the probabilistic models for hard failure and soft failure are built respectively. On this basis, the dependent competing failure model involving degradation and shock processes is established. Furthermore, the situation of the shifting-threshold is also considered, in which the hard failure threshold value decreases to a lower level after the arrival of a certain number of shocks. A case study of fatigue crack growth is given to illustrate the proposed models. Numerical results show that shock has a significant effect on the failure process; meanwhile, the effect will be magnified when the value of the hard threshold shifts to a lower level.展开更多
Considering the dependence and competitive relation-ship between traumatic failure and degradation,the reliability assessment of products based on competing failure analysis is studied.The hazard rate of traumatic fai...Considering the dependence and competitive relation-ship between traumatic failure and degradation,the reliability assessment of products based on competing failure analysis is studied.The hazard rate of traumatic failure is regarded as a Weibull distribution of the degradation performance,and the Wiener process is used to describe the degradation process.The parameters are estimated with the maximum likelihood estimation(MLE)method.A reliability model based on competing failure analysis is proposed.A case study of the GaAs lasers is given to validate the effectiveness of the model and its solving method.The results indicate that if only the degradation failure is considered,the estimated result will be comparably optimistic.Meanwhile,the correlation between the degradation and traumatic failure has a great influence on the accuracy of reliability assessment.展开更多

Long noncoding RNAs(lnc R...Recent studies have showed that RNAs regulate each other with micro RNA(mi RNA) response elements(MREs) and this mechanism is known as "competing endogenous RNA(ce RNA)" hypothesis. Long noncoding RNAs(lnc RNAs) are supposed to play important roles in cancer. Compelling evidence suggests that lnc RNAs can interact with mi RNAs and regulate the expression of mi RNAs as ce RNAs. Several lnc RNAs such as H19, HOTAIR and MEG3 have been found to be associated with mi RNAs in gastric cancer(GC), generating regulatory crosstalk across the transcriptome. These MRE sharing elements implicated in the ce RNA networks(ce RNETs) are able to regulate m RNA expression. The ce RNA regulatory networks including m RNAs, mi RNAs, lnc RNAs and circular RNAs may play critical roles in tumorigenesis, and the perturbations of ce RNETs may contribute to the pathogenesis of GC.展开更多

Cell pr...AIM To investigate the role of long noncoding RNA(lnc RNA) RP4 in colorectal cancer.METHODS Lentivirus-mediated lnc RNA RP4 overexpression and knockdown were performed in the colorectal cancer cell line SW480. Cell proliferation, tumor growth, and early apoptosis were evaluated by a cell counting kit-8 assay, an in vivo xenograft tumor model, and annexin V/propidium iodide staining, respectively. Analysis of the lnc RNA RP4 mechanism involved assessment of the association of its expression with mi R-7-5 p and the SH3 GLB1 gene. Western blot analysis was also performed to assess the effect of lnc RNA RP4 on the autophagy-mediated cell death pathway and phosphatidylinositol-3-kinase(PI3 K)/Akt signaling.RESULTS Cell proliferation, tumor growth, and early apoptosis in SW480 cells were negatively regulated by lnc RNA RP4. Functional experiments indicated that lnc RNA RP4 directly upregulated SH3 GLB1 expression by acting as a competing endogenous RNA(ce RNA) for mi R-7-5 p. This interaction led to activation of the autophagy-mediated cell death pathway and de-repression of PI3 K and Akt phosphorylation in colorectal cancer cells in vivo.CONCLUSION Our results demonstrated that lnc RNA RP4 is a ce RNA that plays an important role in the pathogenesis of colorectal cancer, and could be a potential therapeutic target for colorectal cancer treatment.展开更多

The incompleteness is mainly due to censori...Data obtained from accelerated life testing (ALT) when there are two or more failure modes, which is commonly referred to as competing failure modes, are often incomplete. The incompleteness is mainly due to censoring, as well as masking which might be the case that the failure time is observed, but its corresponding failure mode is not identified. Because the identification of the failure mode may be expensive, or very difficult to investigate due to lack of appropriate diagnostics. A method is proposed for analyzing incomplete data of constant stress ALT with competing failure modes. It is assumed that failure modes have s-independent latent lifetimes and the log lifetime of each failure mode can be written as a linear function of stress. The parameters of the model are estimated by using the expectation maximum (EM) algorithm with incomplete data. Simulation studies are performed to check'model validity and investigate the properties of estimates. For further validation, the method is also illustrated by an example, which shows the process of analyze incomplete data from ALT of some insulation system. Because of considering the incompleteness of data in modeling and making use of the EM algorithm in estimating, the method becomes more flexible in ALT analysis.展开更多
A series of experiments were conducted to examine the interactive effects of an organic ligand,a competing cation, and pH on the dissolution of zinc (Zn) from three California soils,Maymen sandy loam,Merced clay,and Y...A series of experiments were conducted to examine the interactive effects of an organic ligand,a competing cation, and pH on the dissolution of zinc (Zn) from three California soils,Maymen sandy loam,Merced clay,and Yolo clay loam.The concentrations of soluble Zn of the three soils were low in a background solution of Ca(NO3)2.Citric acid,a common organic ligand found in the rhizosphere,was effective in mobilizing Zn in these soils;its presence enhanced the concentration of Zn in soil solution by citrate forming a complex with Zn.The ability of Zn to form a complex with citric acid in the soil solution was dependent on the concentration of citric acid,pH,and the concentration of the competing cation Ca^(2+).The pH of the soil solution determined the extent of desorption of Zn in solid phase in the presence of citric acid.The amounts of Zn released from the solid phase were proportional to the concentration of citric acid and inversely proportional to the concentration of Ca(NO3)2 background solution,which supplied the competing cation Ca^(2+) for the formation of a complex with citrate.When the soil suspension was spiked with Zn,the adsorption of Zn by the soils was retarded by citric acid via the formation of the soluble Zn-citrate complex.The dissolution of Zn in the presence of citric acid was pH dependent in both adsorption and desorption processes.展开更多

METHODS In the present study, we obtained CRC tissue and normal tissue gene expression pr...AIM To identify and predict the competing endogenous RNA(ce RNA) networks in colorectal cancer(CRC) by bioinformatics analysis. METHODS In the present study, we obtained CRC tissue and normal tissue gene expression profiles from The Cancer Genome Atlas project. Differentially expressed(DE) genes(DEGs) were identified. Then, upregulated and downregulated mi RNA-centered ceRNA networks were constructed by analyzing the DEGs using multiple bioinformatics approaches. DEm RNAs in the ceRNA networks were identified in Kyoto Encyclopedia of Genes and Genomes(KEGG) pathways using KEGG Orthology Based Annotation System 3.0. The interactions between proteins were analyzed using the STRING database. Kaplan-Meier survival analysis was conducted for DEGs and real time quantitative polymerase chain reaction(RT-qPCR) was also performed to validate the prognosis-associated lncRNAs in CRC cell lines.RESULTS Eighty-one DElncRNAs, 20 DEmiRNAs, and 54 DEmRNAs were identified to construct the ceRNA networks of CRC. The KEGG pathway analysis indicated that nine out of top ten pathways were related with cancer and the most significant pathway was "colorectal cancer". Kaplan-Meier survival analysis showed that the overall survival was positively associated with five DEGs(IGF2-AS, POU6 F2-AS2, hsa-miR-32, hsa-miR-141, and SERPINE1) and it was negatively related to three DEGs(LINC00488, hsamiR-375, and PHLPP2). Based on the STRING protein database, it was found that SERPINE1 and PHLPP2 interact with AKT1. Besides, SERPINE1 can interact with VEGFA, VTN, TGFB1, PLAU, PLAUR, PLG, and PLAT. PHLPP2 can interact with AKT2 and AKT3. RT-qPCR revealed that the expression of IGF2-AS, POU6 F2-AS2, and LINC00488 in CRC cell lines was consistent with the in silico results.CONCLUSION Ce RNA networks play an important role in CRC. Multiple DEGs are related with clinical prognosis, suggesting that they may be potential targets in tumor diagnosis and treatment.展开更多
BACKGROUND Colorectal cancer(CRC)is one of the most prevalent tumors worldwide.Recently,long noncoding RNAs(lncRNAs)have been shown to influence tumorigenesis and tumor progression by acting as competing endogenous RN...BACKGROUND Colorectal cancer(CRC)is one of the most prevalent tumors worldwide.Recently,long noncoding RNAs(lncRNAs)have been shown to influence tumorigenesis and tumor progression by acting as competing endogenous RNAs(ceRNAs).It is difficult to extract prognostic lncRNAs and useful bioinformation from most ceRNA networks constructed previously.AIM To construct a prognostic related ceRNA regulatory network and lncRNA related signature based on risk score in CRC.METHODS RNA transcriptome profile and clinical information of 506 CRC patients were downloaded from the Cancer Genome Atlas database.R packages and Perl program were used for data processing.Cox regression analysis was used for prognostic model construction.Quantitative real-time polymerase chain reaction was used to detect the expression of lncRNAs.RESULTS A prognostic-related ceRNA network was constructed,including 9 lncRNAs,44 mRNAs,and 30 miRNAs.In addition,a four-lncRNA model was constructed using multivariate Cox regression analysis,which could be an independent prognostic model in CRC.The risk score for each patient was calculated,and the 506 patients were divided into high and low-risk groups(253 for each group)based on the median risk score.The results of the survival analysis showed that patients with a high-risk score had a poor survival rate.Furthermore,the predictive value of the four-lncRNA model was evaluated in GSE38832.Patient survival probabilities could be better predicted when combing the risk score and clinical features.Gene Set Enrichment Analysis results verified that a number of cancer-related signaling pathways were enriched with a high-risk score in CRC.Finally,we validated a novel lncRNA(LINC00488)using quantitative real-time polymerase chain reaction in 22 paired CRC patient tumor tissues compared to adjacent non-tumor tissues.CONCLUSION The four-lncRNA model could give better predictive value for CRC patients.Our understanding of the lncRNA-related ceRNA regulatory mechanism could provide a potential diagnostic indicator for CRC patients.展开更多

To investigate death for liver failure and for tumor recurrence as competing events after hepatectomy of hepatocellular carcinoma.METHODSData from 864 cirrhotic Child-Pugh class A consecutive patients, submitted to cu...To investigate death for liver failure and for tumor recurrence as competing events after hepatectomy of hepatocellular carcinoma.METHODSData from 864 cirrhotic Child-Pugh class A consecutive patients, submitted to curative hepatectomy (1997-2013) at two tertiary referral hospitals, were used for competing-risk analysis through the Fine and Gray method, aimed at assessing in which circumstances the oncological benefit from tumour removal is greater than the risk of dying from hepatic decompensation. To accomplish this task, the average risk of these two competing events, over 5 years of follow-up, was calculated through the integral of each cumulative incidence function, and represented the main comparison parameter.RESULTSWithin a median follow-up of 5.6 years, death was attributable to tumor recurrence in 63.5%, and to liver failure in 21.2% of cases. In the first 16 mo, the risk of dying due to liver failure exceeded that of dying due to tumor relapse. Tumor stage only affects death from recurrence; whereas hepatitis C infection, Model for End-stage Liver Disease score, extent of hepatectomy and portal hypertension influence death from liver failure (P < 0.05 in all cases). The combination of these clinical and tumoral features identifies those patients in whom the risk of dying from liver failure did not exceed the tumour-related mortality, representing optimal surgical candidates. It also identifies those clinical circumstances where the oncological benefit would be borderline or even where the surgery would be harmful.CONCLUSIONHaving knowledge of these competing events can be used to weigh the risks and benefits of hepatic resection in each clinical circumstance, separating optimal from non-optimal surgical candidates.展开更多

Inparticular, some lncRNAs can interact with miRNAs as competing endogenousRNAs (ceRNAs) to modulate mRNA expression. Accordingly, these RNAmolecules are interrelated and coordinate to form a dynamic lncRNA-mediatedceRNA regulatory network. Mounting evidence has revealed that lncRNAs thatact as ceRNAs are closely related to tumorigenesis. To date, numerous studieshave established many different regulatory networks in hepatocellular carcinoma(HCC), and perturbations in these ceRNA interactions may result in the initiationand progression of HCC. 文摘BACKGROUND As a non-coding RNA molecule,circular RNAs(circRNAs)have significant specificity,and existing data suggest a close relationship between them and the prognosis of patients with gastric cancer(GC).However,this mechanism has no evidence yet.This article explores the functions of hsa_circRNA_102415 in the malignant behavior and potential downstream signaling of GC cells.The chosen approach is loss of signal and functional gain.AIM To investigate and analyze the relationship between hsa_circRNA_102415 and GC and explore its specific role.Results provide reference for other researchers to develop targeted treatment plans.METHODS The gene expression omnibus(GEO)database can be used to obtain the microarray dataset GSE83521.Data were analyzed using the GEO2R tool to identify differences in circRNAs between normal and GC samples.Quantitative real-time polymerase chain reaction was used to detect differentially expressed genes in GC tissue samples and adjacent cancer tissue samples.GC cells were transfected with small interfering-hsa_circRNA_104415 and plasmid DNA(pcDNA)-hsa_ircRNA_102415.Multiple detection methods,such as Transwell and cell counting kit 8,were used to evaluate cellular physiological activities,including cell invasion and proliferation.The relationship between Wnt family members 2B,microRNA(miR)-4529-5p,etc.,including argonaute 2-RNA immunoprecipitation and luciferase reporter genes was analyzed.Rescue experiments were conducted to analyze and explore the relationship between the malignant behavior of GC cells and hsa_circRNA_102415.RESULTS GEO2R analysis confirmed that hsa_circRNA_102415 had significantly higher expression levels in disease tissues.hsa_circRNA_102415 and miR-4529-5p showed a negative correlation in disease cells,suggesting that hsa_circRNA_102415 upregulated WNT2B expression in GC cells as a competing endogenous RNA for miR-4529-5p.miR-4529-5p mimic or small interfering-WNT2B reversed the effects of pcDNA-hsa_circRNA_102415 or miR-4529-5p inhibitor on cell malignant functions.CONCLUSION miR-4529-5p was used to successfully activate the potential of WNT2B,clarify the role of hsa_circRNA_102415 in GC cells,and provide reference for other researchers to develop targeted treatment plans.

文摘The reliability assessment problem for products subject to degradation and random shocks is investigated. Two kinds of probabilistic models are constructed, in which the dependent competing failure process is considered. First, based on the assumption of cumulative shock, the probabilistic models for hard failure and soft failure are built respectively. On this basis, the dependent competing failure model involving degradation and shock processes is established. Furthermore, the situation of the shifting-threshold is also considered, in which the hard failure threshold value decreases to a lower level after the arrival of a certain number of shocks. A case study of fatigue crack growth is given to illustrate the proposed models. Numerical results show that shock has a significant effect on the failure process; meanwhile, the effect will be magnified when the value of the hard threshold shifts to a lower level.

文摘Considering the dependence and competitive relation-ship between traumatic failure and degradation,the reliability assessment of products based on competing failure analysis is studied.The hazard rate of traumatic failure is regarded as a Weibull distribution of the degradation performance,and the Wiener process is used to describe the degradation process.The parameters are estimated with the maximum likelihood estimation(MLE)method.A reliability model based on competing failure analysis is proposed.A case study of the GaAs lasers is given to validate the effectiveness of the model and its solving method.The results indicate that if only the degradation failure is considered,the estimated result will be comparably optimistic.Meanwhile,the correlation between the degradation and traumatic failure has a great influence on the accuracy of reliability assessment.

文摘Recent studies have showed that RNAs regulate each other with micro RNA(mi RNA) response elements(MREs) and this mechanism is known as "competing endogenous RNA(ce RNA)" hypothesis. Long noncoding RNAs(lnc RNAs) are supposed to play important roles in cancer. Compelling evidence suggests that lnc RNAs can interact with mi RNAs and regulate the expression of mi RNAs as ce RNAs. Several lnc RNAs such as H19, HOTAIR and MEG3 have been found to be associated with mi RNAs in gastric cancer(GC), generating regulatory crosstalk across the transcriptome. These MRE sharing elements implicated in the ce RNA networks(ce RNETs) are able to regulate m RNA expression. The ce RNA regulatory networks including m RNAs, mi RNAs, lnc RNAs and circular RNAs may play critical roles in tumorigenesis, and the perturbations of ce RNETs may contribute to the pathogenesis of GC.

文摘AIM To investigate the role of long noncoding RNA(lnc RNA) RP4 in colorectal cancer.METHODS Lentivirus-mediated lnc RNA RP4 overexpression and knockdown were performed in the colorectal cancer cell line SW480. Cell proliferation, tumor growth, and early apoptosis were evaluated by a cell counting kit-8 assay, an in vivo xenograft tumor model, and annexin V/propidium iodide staining, respectively. Analysis of the lnc RNA RP4 mechanism involved assessment of the association of its expression with mi R-7-5 p and the SH3 GLB1 gene. Western blot analysis was also performed to assess the effect of lnc RNA RP4 on the autophagy-mediated cell death pathway and phosphatidylinositol-3-kinase(PI3 K)/Akt signaling.RESULTS Cell proliferation, tumor growth, and early apoptosis in SW480 cells were negatively regulated by lnc RNA RP4. Functional experiments indicated that lnc RNA RP4 directly upregulated SH3 GLB1 expression by acting as a competing endogenous RNA(ce RNA) for mi R-7-5 p. This interaction led to activation of the autophagy-mediated cell death pathway and de-repression of PI3 K and Akt phosphorylation in colorectal cancer cells in vivo.CONCLUSION Our results demonstrated that lnc RNA RP4 is a ce RNA that plays an important role in the pathogenesis of colorectal cancer, and could be a potential therapeutic target for colorectal cancer treatment.

文摘Data obtained from accelerated life testing (ALT) when there are two or more failure modes, which is commonly referred to as competing failure modes, are often incomplete. The incompleteness is mainly due to censoring, as well as masking which might be the case that the failure time is observed, but its corresponding failure mode is not identified. Because the identification of the failure mode may be expensive, or very difficult to investigate due to lack of appropriate diagnostics. A method is proposed for analyzing incomplete data of constant stress ALT with competing failure modes. It is assumed that failure modes have s-independent latent lifetimes and the log lifetime of each failure mode can be written as a linear function of stress. The parameters of the model are estimated by using the expectation maximum (EM) algorithm with incomplete data. Simulation studies are performed to check'model validity and investigate the properties of estimates. For further validation, the method is also illustrated by an example, which shows the process of analyze incomplete data from ALT of some insulation system. Because of considering the incompleteness of data in modeling and making use of the EM algorithm in estimating, the method becomes more flexible in ALT analysis.

文摘A series of experiments were conducted to examine the interactive effects of an organic ligand,a competing cation, and pH on the dissolution of zinc (Zn) from three California soils,Maymen sandy loam,Merced clay,and Yolo clay loam.The concentrations of soluble Zn of the three soils were low in a background solution of Ca(NO3)2.Citric acid,a common organic ligand found in the rhizosphere,was effective in mobilizing Zn in these soils;its presence enhanced the concentration of Zn in soil solution by citrate forming a complex with Zn.The ability of Zn to form a complex with citric acid in the soil solution was dependent on the concentration of citric acid,pH,and the concentration of the competing cation Ca^(2+).The pH of the soil solution determined the extent of desorption of Zn in solid phase in the presence of citric acid.The amounts of Zn released from the solid phase were proportional to the concentration of citric acid and inversely proportional to the concentration of Ca(NO3)2 background solution,which supplied the competing cation Ca^(2+) for the formation of a complex with citrate.When the soil suspension was spiked with Zn,the adsorption of Zn by the soils was retarded by citric acid via the formation of the soluble Zn-citrate complex.The dissolution of Zn in the presence of citric acid was pH dependent in both adsorption and desorption processes.

文摘AIM To identify and predict the competing endogenous RNA(ce RNA) networks in colorectal cancer(CRC) by bioinformatics analysis. METHODS In the present study, we obtained CRC tissue and normal tissue gene expression profiles from The Cancer Genome Atlas project. Differentially expressed(DE) genes(DEGs) were identified. Then, upregulated and downregulated mi RNA-centered ceRNA networks were constructed by analyzing the DEGs using multiple bioinformatics approaches. DEm RNAs in the ceRNA networks were identified in Kyoto Encyclopedia of Genes and Genomes(KEGG) pathways using KEGG Orthology Based Annotation System 3.0. The interactions between proteins were analyzed using the STRING database. Kaplan-Meier survival analysis was conducted for DEGs and real time quantitative polymerase chain reaction(RT-qPCR) was also performed to validate the prognosis-associated lncRNAs in CRC cell lines.RESULTS Eighty-one DElncRNAs, 20 DEmiRNAs, and 54 DEmRNAs were identified to construct the ceRNA networks of CRC. The KEGG pathway analysis indicated that nine out of top ten pathways were related with cancer and the most significant pathway was "colorectal cancer". Kaplan-Meier survival analysis showed that the overall survival was positively associated with five DEGs(IGF2-AS, POU6 F2-AS2, hsa-miR-32, hsa-miR-141, and SERPINE1) and it was negatively related to three DEGs(LINC00488, hsamiR-375, and PHLPP2). Based on the STRING protein database, it was found that SERPINE1 and PHLPP2 interact with AKT1. Besides, SERPINE1 can interact with VEGFA, VTN, TGFB1, PLAU, PLAUR, PLG, and PLAT. PHLPP2 can interact with AKT2 and AKT3. RT-qPCR revealed that the expression of IGF2-AS, POU6 F2-AS2, and LINC00488 in CRC cell lines was consistent with the in silico results.CONCLUSION Ce RNA networks play an important role in CRC. Multiple DEGs are related with clinical prognosis, suggesting that they may be potential targets in tumor diagnosis and treatment.
文摘BACKGROUND Colorectal cancer(CRC)is one of the most prevalent tumors worldwide.Recently,long noncoding RNAs(lncRNAs)have been shown to influence tumorigenesis and tumor progression by acting as competing endogenous RNAs(ceRNAs).It is difficult to extract prognostic lncRNAs and useful bioinformation from most ceRNA networks constructed previously.AIM To construct a prognostic related ceRNA regulatory network and lncRNA related signature based on risk score in CRC.METHODS RNA transcriptome profile and clinical information of 506 CRC patients were downloaded from the Cancer Genome Atlas database.R packages and Perl program were used for data processing.Cox regression analysis was used for prognostic model construction.Quantitative real-time polymerase chain reaction was used to detect the expression of lncRNAs.RESULTS A prognostic-related ceRNA network was constructed,including 9 lncRNAs,44 mRNAs,and 30 miRNAs.In addition,a four-lncRNA model was constructed using multivariate Cox regression analysis,which could be an independent prognostic model in CRC.The risk score for each patient was calculated,and the 506 patients were divided into high and low-risk groups(253 for each group)based on the median risk score.The results of the survival analysis showed that patients with a high-risk score had a poor survival rate.Furthermore,the predictive value of the four-lncRNA model was evaluated in GSE38832.Patient survival probabilities could be better predicted when combing the risk score and clinical features.Gene Set Enrichment Analysis results verified that a number of cancer-related signaling pathways were enriched with a high-risk score in CRC.Finally,we validated a novel lncRNA(LINC00488)using quantitative real-time polymerase chain reaction in 22 paired CRC patient tumor tissues compared to adjacent non-tumor tissues.CONCLUSION The four-lncRNA model could give better predictive value for CRC patients.Our understanding of the lncRNA-related ceRNA regulatory mechanism could provide a potential diagnostic indicator for CRC patients.

By building its mathematical model and simulating its evolution process, we have found the statistical regularity for support rates of two different products at a steady stage. The research shows that strength of the public opinion spreading is proportional to the final support rates of a product.

文摘A method for reliability analysis of the competing failure with the probabilistic failure threshold value not the fixed threshold value is presented, which involves the random shocks and the degradation is independent and dependent respectively. Specifically, for the dependent condition, the effect due to the random shocks on the degradation is considered with a damage factor. In addition, the dependent competing failure model is applied to the reliability analysis of the k-out-of-n systems. Finally, two studied cases are presented to illustrate the proposed method, and the results show the proposed method is reasonable.
文摘To investigate death for liver failure and for tumor recurrence as competing events after hepatectomy of hepatocellular carcinoma.METHODSData from 864 cirrhotic Child-Pugh class A consecutive patients, submitted to curative hepatectomy (1997-2013) at two tertiary referral hospitals, were used for competing-risk analysis through the Fine and Gray method, aimed at assessing in which circumstances the oncological benefit from tumour removal is greater than the risk of dying from hepatic decompensation. To accomplish this task, the average risk of these two competing events, over 5 years of follow-up, was calculated through the integral of each cumulative incidence function, and represented the main comparison parameter.RESULTSWithin a median follow-up of 5.6 years, death was attributable to tumor recurrence in 63.5%, and to liver failure in 21.2% of cases. In the first 16 mo, the risk of dying due to liver failure exceeded that of dying due to tumor relapse. Tumor stage only affects death from recurrence; whereas hepatitis C infection, Model for End-stage Liver Disease score, extent of hepatectomy and portal hypertension influence death from liver failure (P < 0.05 in all cases). The combination of these clinical and tumoral features identifies those patients in whom the risk of dying from liver failure did not exceed the tumour-related mortality, representing optimal surgical candidates. It also identifies those clinical circumstances where the oncological benefit would be borderline or even where the surgery would be harmful.CONCLUSIONHaving knowledge of these competing events can be used to weigh the risks and benefits of hepatic resection in each clinical circumstance, separating optimal from non-optimal surgical candidates.
文摘Long noncoding RNAs (lncRNAs) and microRNAs (miRNAs) are noncodingRNAs (ncRNAs) that occupy over 90% of the human genome, and their mainfunction is to directly or indirectly regulate messenger RNA (mRNA) expressionand participate in the tumorigenesis and progression of malignances. Inparticular, some lncRNAs can interact with miRNAs as competing endogenousRNAs (ceRNAs) to modulate mRNA expression. Accordingly, these RNAmolecules are interrelated and coordinate to form a dynamic lncRNA-mediatedceRNA regulatory network. Mounting evidence has revealed that lncRNAs thatact as ceRNAs are closely related to tumorigenesis. To date, numerous studieshave established many different regulatory networks in hepatocellular carcinoma(HCC), and perturbations in these ceRNA interactions may result in the initiationand progression of HCC. Herein, we emphasize recent advances concerning thebiological function of lncRNAs as ceRNAs in HCC, with the aim of elucidating themolecular mechanism underlying these HCC-related RNA molecules andproviding novel insights into the diagnosis and treatment of HCC.
