Compartmentalization of hepatitis B virus: Looking beyond the liver 被引量:2

Hepatitis B virus(HBV) is classically considered to be hepatotropic, but accumulating evidences strongly support its extra-hepatotropic nature too. HBV nucleicacids and proteins have long been reported in a variety of extra-hepatic tissues. Of these, HBV has been studied in details in the peripheral blood mononuclear cells(PBMCs), due to its accessibility. From these studies, it is now well established that PBMCs are permissive to HBV infection, replication, transcription and production of infective virions. Furthermore, molecular evolutionary studies have provided definite evidences towards evolution of HBV genome in PBMCs, which is independent of evolution occurring in the liver, leading to the emergence and selection of compartment specific escape variants or drug resistant strains. These variants/resistant strains of HBV remain restricted within the PBMCs and are rarely detected in the serum/plasma. In addition, HBV infected PBMCs have been reported to be directly transmitted through intrauterine modes, and this infection does not correlate significantly with serum HBV surface antigen or HBV DNA markers. This editorial briefly reviews the current knowledge on this topic, emphasizes and delineates the gaps that are required to be filled to properly understand the biological and clinical relevance of extrahepatic tropism of HBV. 展开更多

The Pacific oyster Crassostrea gigas,one of the most exploited molluscs in the world,has suffered from massive mortality in recent decades,and the occurrence mechanisms have not been well characterized.In this study,to reveal the relationship of associated microbiota to the fitness of oysters,temporal dynamics of microbiota in the gill,hemolymph,and hepatopancreas of C.gigas during April 2018-January 2019 were investigated by 16 S rRNA gene sequencing.The microbiota in C.gigas exhibited tissue heterogeneity,of which Spirochaetaceae was dominant in the gill and hemolymph while Mycoplasmataceae enriched in the hepatopancreas.Co-occurrence network demonstrated that the gill microbiota exhibited higher inter-taxon connectivity while the hemolymph microbiota had more modules.The richness(Chao 1 index)and diversity(Shannon index)of microbial community in each tissue showed no significant seasonal variations,except for the hepatopancreas having a higher richness in the autumn.Similarly,beta diversity analysis indicated a relatively stable microbiota in each tissue during the sampling period,showing relative abundance of the dominant taxa exhibiting temporal dynamics.Results indicate that the microbial community in C.gigas showed a tissue-specific stability with temporal dynamics in the composition,which might be essential for the tissue functioning and environmental adaption in oysters.This work provides a baseline microbiota in C.gigas and is helpful for the understanding of host-microbiota interaction in oysters. 展开更多

Soil salinity significantly affects plant survival and limits crop productivity.Under salt stress,plants can transport sodium ions(Na+)out of cells and sequester them into vacuoles for detoxification.The salt excretion process is governed by the SALT OVERLY SENSITIVE(SOS)pathway,which involves the calcium sensors SOS3 and SOS3-LIKE CALCIUM BINDING PROTEIN 8,the protein kinase SOS2,and the plasma membrane Na+/H+antiporter SOS1.While previous studies have provided insights into Na+transport through the SOS system,the role of this pathway in Na+compartmentalization within vacuoles remains poorly understood.In this study,we demonstrate that SOS1 partially internalizes to the tonoplast under salt stress,which is crucial for Na+compartmentalization in vacuoles in Arabidopsis(Arabidopsis thaliana).We show that SOS2 phosphorylates the endosomal sorting complex required for transport-I(ESCRT-I)component FYVE DOMAIN PROTEIN REQUIRED FOR ENDOSOMAL SORTING 1(FREE1),which disrupts its interaction with VPS23A,an ESCRT-I component.This phosphorylation event inhibits the formation of intraluminal vesicles(ILVs)in prevacuolar compartments and multivesicular bodies(PVCs/MVBs),thereby remodeling endosomal sorting during salt stress.Additionally,our previous research indicated that SOS2-mediated phosphorylation of FREE1leads to vacuole fragmentation by altering endomembrane fusion,thereby regulating intracellular Na+homeostasis.Taken together,our findings reveal how the SOS2-FREE1 module orchestrates both endomembrane fusion and endosome sorting processes to enhance plant salt tolerance,providing novel insights into the cellular mechanisms underlying salt stress adaptation. 展开更多

A fundamental challenge for cells is how to coordinate various biochemical reactions in space and time. To achieve spatiotemporal control, cells have developed organelles that are surrounded by lipid bilayer membranes. Further, membraneless compartmentalization, a process induced by dynamic physical association of biomolecules through phase transition offers another efficient mechanism for intracellular organization. While our understanding of phase separation was predominantly dependent on yeast and animal models, recent findings have provided compelling evidence for emerging roles of phase separation in plants. In this review, we first provide an overview of the current knowledge of phase separation, including its definition, biophysical principles, molecular features and regulatory mechanisms. Then we summarize plant-specific phase separation phenomena and describe their functions in plant biological processes in great detail. Moreover, we propose that phase separation is an evolutionarily conserved and efficient mechanism for cellular compartmentalization which allows for distinct metabolic processes and signaling pathways, and is especially beneficial for the sessile lifestyle of plants to quickly and efficiently respond to the changing environment. 展开更多

Fabrication of multifunctional catalysts has always been the pursuit of synthetic chemists due to their efficiency,cost-effectiveness,and environmental friendliness.However,it is difficult to control multi-step reactions in one-pot,especially the spatial compartmentalization of incompatible active sites.Herein,we constructed metal-organic framework(MOF)composites which regulate the location distribution of metal nanoparticles according to the reaction path and coupled with the diffusion of substrates to achieve tandem reaction.The designed UiO-66-Pt-Au catalyst showed good activity and selectivity in hydrosilylation-hydrogenation tandem reaction,because the uniform microporous structures can control the diffusion path of reactants and intermediates,and Pt and Au nanoparticles were arranged in core-shell spatial distribution in UiO-66.By contrast,the low selectivity of catalysts with random deposition and physical mixture demonstrated the significance of artificial control to the spatial compartmentalization of active sites in tandem catalytic reactions,which provides a powerful approach for designing high-performance and multifunctional heterogeneous catalysts. 展开更多

CoproporphyrinⅢ(CPⅢ),a natural porphyrin derivative,has extensive applications in the biomedical and material industries.S.cerevisiae has previously been engineered to highly accumulate the CPⅢprecursor 5-aminolevulinic acid(ALA)through the C4 pathway.In this study,a combination of cytoplasmic metabolic engineering and mitochondrial compartmentalization was used to enhance CPⅢproduction in S.cerevisiae.By integrating pathway genes into the chromosome,the CPⅢtiter gradually increased to 32.5±0.5 mg/L in shake flask cultivation.Nevertheless,increasing the copy number of pathway genes did not consistently enhance CPⅢsynthesis.Hence,the partial synthesis pathway was compartmentalized in mitochondria to evaluate its effectiveness in increasing CPⅢproduction.Subsequently,by superimposing the mitochondrial compartmentalization strategy on cytoplasmic metabolic engineered strains,the CPⅢtiter was increased to 64.3±1.9 mg/L.Furthermore,augmenting antioxidant pathway genes to reduce reactive oxygen species(ROS)levels effectively improved the growth of engineered strains,resulting in a further increase in the CPⅢtiter to 82.9±1.4 mg/L.Fed-batch fermentations in a 5 L bioreactor achieved a titer of 402.8±9.3 mg/L for CPⅢ.This study provides a new perspective on engineered yeast for the microbial production of porphyrins. 展开更多

Human immunodeficiency virus(HIV)has been shown to evolve independently in different anatomical compartments.Characterizing HIV genetic evolution in different tissues and cells provides insights into the mechanisms that maintain the viral reservoir.HIV compartmentalization has been well documented in the semen but rarely in male genital tract(MGT)organs.The precise mechanisms that result in the development of HIV compartmentalization in multiple genitourinary sites have been poorly explored due to the difficulty in accessing these tissues.Based on evidence from lymph nodes and gut tissues,mechanisms that may influence compartmentalization include immune pressures,local concentrations of antiviral drugs,clonal expansion of different cell types and inflammation that alters the cellular microenvironment.We reviewed phylogenetic evidences supporting viral compartmentalization between the blood and multiple genitourinary sites in HIV-infected people.Characterizing distinct viral subpopulations enhances our overall understanding of the HIV reservoir inMGTand could ultimately lead to the development of novel therapies to eradicate the virus in tissues. 展开更多

Salinity is a major hazard to crop plant growth and significantly reduces grain yield.When subjected to high salinity,plants maintain ion homeostasis through Na^(+)compartmentalization and exclusion,enabling them to mitigate salt stress.In this study,we revealed the role of rice(Oryza sativa)MYB9(OsMYB9)in regulating salt stress tolerance.OsMYB9 was expressed in the vascular bundles of roots and leaves,particularly in the parenchyma cells.The null mutation of OsMYB9 resulted in increased sodium ion accumulation in shoot tissues under salt stress,leading to salt-sensitive phenotypes.Real-Time Quantitative Reverse Transcription PCR analysis indicated the OsMYB9 mutation led to decreased expression of several genes associated with vacuolar Na^(+)/H^(+)antiporters(OsNHX1,OsNHX2,OsNHX3,OsNHX4,and OsNHX5),high-K^(+)affinity transporters(OsHKT1;5),and SALT OVERLY SENSITIVE proteins(OsSOS2 and OsSOS3).Among these,OsMYB9 upregulated the expression of OsNHX1 and OsNHX2 by directly binding to their promoter regions.Furthermore,GIGANTEA(OsGI)interacted with OsMYB9,suggesting that OsGI negatively acts upstream of OsMYB9 and regulates the expression levels of OsNHX1.Collectively,OsMYB9 alleviates the excess accumulation of Na^(+)ions in the xylem by retrieving Na^(+)ions from xylem parenchyma cells and compartmentalizing them into vacuoles.These regulatory mechanisms mediated by OsMYB9 are crucial for minimizing Na^(+)toxicity in photosynthetic tissues and enhancing salt stress tolerance in rice. 展开更多

The muzzle blast overpressure induces disturbances in the flow field inside the crew compartment(FFICC)of a truck-mounted howitzer during the artillery firing.This overpressure is the primary factor preventing personnel from firing artillery within the cab.To investigate the overpressure characteristics of the FFICC,a foreign trade equipment model was used as the research object,and a numerical model was established to analyze the propagation of muzzle blast from the muzzle to the interior of the crew compartment under extreme firing condition.For comparative verification,the muzzle blast experiment included overpressure data from both the flow field outside the crew compartment(FFOCC)and the FFICC,as well as the acceleration data of the crew compartment structure(Str-CC).The research findings demonstrate that the overpressure-time curves of the FFICC exhibit multi-peak characteristics,while the pressure wave shows no significant discontinuity.The enclosed nature of the cab hinders the dissipation of pressure wave energy within the FFICC,leading to sustained high-amplitude overpressure.The frameskin structure helps attenuate the impact of muzzle blast on the FFICC.Conversely,local high overpressure caused by the convex or concave features of the cab's exterior significantly amplifies the overpressure amplitude within the FFICC. 展开更多

Discussion on the Problem of Salt Gland of Glycine soja 被引量:6

Glycine soja Sieb. et Zucc. plants living in saline soil in three provinces of China were treated with different salinity concentrations under different laboratory culture conditions (including solution, sand and field cultivation). The attachment shape and distribution on the surface of stalk and leaf of G. soja plants were observed with scanning electron microscopy (SEM), and the ultrastructure of glandular hair with transmission electron microscopy (TEM). Na+ and Cl- contents in the secretion of the leaf surface and inside the leaf of G. soja subjected to different treatments were measured. The Na+ relative contents in glandular cells, epidermal cells and mesophyllous cells of leaves under different salinities were determined by X-ray microanalysis. Results show that only glandular and epidermal hair exist on the surface attachments of leaves and stalks of G. soja plants. These glandular hair were similar in shape to some salt glands of Gramineae halophytes, and they attached to the vein on the leaf surface. The cell structure of the glandular hair showed the characteristics of common salt glands, such as big vacuoles, dense cytoplasm, a great deal of mitochondria, chloroplast, plasmodesmata and thicker cell walls, etc. The results of Na+ and Cl- contents in the leaf secretion and inside the leaf showed that the glandular hair executed the function of salt-secretion, and when treated with the salt gland inhibitor the salt-secretion process was inhibited. As a result, Na+ and Cl- were mainly accumulated inside G. soja leaves. The results of Na+ X-ray microanalysis under different salinities proved that the three cells of the glandular hair, especially the top cell, possessed strong competence for Na+ accumulation. Above all, the glandular hair were the salt gland, and no other kind of salt glands were found on G. soja plants. The secreting mechanism of the salt gland was also discussed. 展开更多

Combined with the experimental work, validated mathematical model as well as three-dimensional computational fluid dynamics (CFD) model was established to analyze the flow regions of feedwells on an industrial scale. The concept of RTD, although a well-known method for the characterization of mixing behavior in conventional mixers and reactors, is still a novel measure for the characterization of mixing in feedwells. Numerical simulation results show that the inlet feed rate and the aspect ratio of feedwells are the most critical parameters which affect the RTD of feedwell. Further simulation experiments were then carried out. Under the optimal operation conditions, the volume fraction of dead zone can reduce by10.8% and an increasement of mixing flow volume fraction by 6.5% is also observed. There is an optimum feed inlet rate depending on the feedwell design. The CFD model in conjunction with the RTD analysis then can be used as an effective tool in the design, evaluation and optimization of thickener feedwell in the red mud separation. 展开更多

The clinical pharmacokinetics of ribavirin after a single oral dose of 600 mg ribavirin tablets in healthy Chinese volunteers was studied. A rapid and simple high performance liquid chromatography (HPLC) method was developed to determine the ribavirin concentration in human plasma. C18 column was used for separation with a column temperature of 25℃, the mobile phase was ultrapure water adjusted to pH 3 with acetic acid at the flow rate of 1 mL/min, and the detection wavelength was set at 207 rim. The linear range of the standard curves was 50.4-2016.0 ng/mL and the lower limit of quantification (LLOQ) was 50.4 ng/mL. The relative recoveries of ribavirin were more than 90% in plasma. The RSD of the intra-day precision was less than 10% and that of inter-day was less than 15%. The pharmacokinetic parameters of ribavirin were calculated by WinNonlin. Results indicated that the two-compartment model was a better model for describing the pharmacokinetics profile of ribavirin than one-compartment model. The AUC0-t was 10807.8 h.ng/mL, the CL/F was 64879.5 mL, and the Cmax was 525.1 ng/mL. These results provided the experimental data for the development of ribavirin dosage form. 展开更多

The metabolic enzyme CTP synthase(CTPS) is able to compartmentalize into filaments,termed cytoophidia,in a variety of organisms including bacteria,budding yeast,fission yeast,fruit flies and mammals.A previous study in budding yeast shows that the filament-forming process of CTPS is not sensitive to temperature shift.Here we study CTPS filamentation in the fission yeast Schizosaccharomyces pombe.To our surprise,we find that both the length and the occurrence of cytoophidia in S.pombe decrease upon cold shock or heat shock.The temperature-dependent changes of cytoophidia are fast and reversible.Taking advantage of yeast genetics,we demonstrate that heat-shock proteins are required for cytoophidium assembly in S.pombe.Temperature sensitivity of cytoophidia makes S.pombe an attractive model system for future investigations of this novel membraneless organelle. 展开更多

The gene encoded for tryptophan decarboxylase (TDC), which is the key enzyme in terpenoil indole alkaloids pathway, was targeted to different subcellular compartments and stably expressed in transgenic tobacco (Nicotiana tabacum L.) plants at the levels detected by Western blot and tryptamine accumulation analysis. It was shown that the TDC was located in subcellular compartments, the chloroplasts and cytosol. The recombinant TDC targeted to chloroplasts and cytosol in tobacco plants was effectively expressed as soluble protein by Western blot analysis and enzymatic assay. The level of tryptamine accumulation in chloroplast was higher than that in cytosol and very low in vacuole and endoplasmic reticulum (ER) to be hardly detected by Western blot analysis. It was indicated that the highest amount of tryptamine was in chloroplasts, lower in endoplasmic reticula and the lowest in vacuoles as compared to those in wild type plants. The TDC targeted to different subcellular compartments of tobacco plants and its expression level were studied by different nucleotide sequences coding signal peptides at 5'-end of tdc gene in order to know the effects of the TDC in compartmentation on its functionality. 展开更多

Objective: Kaiso is upregulated in many cancers and proposed to bind with both methylated- and unmethylated-DNA in the nucleus as a transcriptional repressor. The objective is to define its subcellnlar localization in vivo and exact binding DNA sequences in cells. Methods: Compartmentalization of exogenous Kaiso in cells was tracked with enhanced green fluorescence protein (EGFP) tag. The endogenous Kaiso expression in gastric carcinoma tissue was examined with immunohistochemical staining. Kaiso-DNA binding was tested using electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation assay (CHIP). Results: Kaiso mainly localized in the nucleus of cancer and stromal cells in vivo, but remained in the cytoplasm of cultured cells. Most importantly, nuclear Kaiso can bind with the methylated-CGCG- containing sequence in the CDKN2A promoter, but not with the hydroxymethylated-CGCG sequence in HCT116 cells. Conclusions: Kaiso locates mainly in the nucleus in vivo where it binds with the methylated-CGCG sequences, but does not bind with the hydroxymethylated-CGCG sequences. 展开更多

Located in Iranian sector of the Persian Gulf, Foroozan Oilfield has been producing hydrocarbons via seven different reservoirs since the 1970 s. However, understanding fluid interactions and horizontal continuity within each reservoir has proved complicated in this field. This study aims to determine the degree of intra-reservoir compartmentalization using gas geochemistry, light hydrocarbon components, and petroleum bulk properties, comparing the results with those obtained from reservoir engineering indicators. For this purpose, a total of 11 samples of oil and associated gas taken from different producing wells in from the Yammama Reservoir were selected. Clear distinctions, in terms of gas isotopic signature and composition, between the wells located in northern and southern parts of the reservoir(i.e. lighter δ13 C1, lower methane concentration, and negative sulfur isotope in the southern part) and light hydrocarbon ratios(e.g. nC 7/toluene, 2,6-dmC7/1,1,3-tmcyC5 and m-xylene/4-mC8) in different oil samples indicated two separate compartments. Gradual variations in a number of petroleum bulk properties(API gravity, V/Ni ratios and asphaltene concentration) provided additional evidence on the reservoir-filling direction, signifying that a horizontal equilibrium between reservoir fluids across the Yammama Reservoir is yet to be achieved. Finally, differences in water-oil contacts and reservoir types further confirmed the compartmentalization of the reservoir into two separate compartments. 展开更多
