By gas chromatogram, six crude oils fingerprinting distributed in four oilfields and four oil platforms were analyzed and the corre- sponding normal paraffin hydrocarbon ( including pristane and phytane) concentration was obtained by the internal standard methed. The normal paraffin hydrocarbon distribution patterns of six crude oils were built and compared. The cluster analysis on the normal paraffin hydrocarbon concentration was conducted for classification and some ratios of oils were used for oils comparison. The results indicated: there was a clear difference within different crude oils in different oil fields and a small difference between the crude oils in the same oil platform. The normal paraffin hydrocarbon distribution pattern and ratios, as well as the cluster analysis on the nomad paraffin hydrocarbon concentration can have a better differentiation result for the crude oils with small difference than the original gas chromatogram. 展开更多

To evaluate the quality of Polyporus umbellatus,we established a simple,repeatable and reliable method based on high performance liquid chromatography(HPLC)for specific chromatogram and fingerprint analysis,which was applied to analyze samples of medicinal materials and decoction pieces collected from different regions.Finally,ten characteristic peaks were designated in the specific chromatograms and applied to the authenticate identification of P.umbellatus samples.Nine common peaks were designated in the fingerprints,and then the similarities between 32 batches of samples were calculated.Among them,eight compounds were identified by HPLC-APCI-IT-TOF-MS^(n),four of which were identified in specific chromatograms and four in fingerprints.In the present study,we,for the first time,combined HPLC specific chromatograms and fingerprints for the species identification and quality evaluation of P.umbellatus.Collectively,our findings provided a new method for establishing a comprehensive quality standard of P.umbellatus. 展开更多

DNA barcoding and HPLC specific chromatogram were used to identify three kinds of Plumeria flowers respectively. DNAs extracted from the three Plumeria species were amplified by PCR with universal primers, and the psbA-trnH region was selected. All the amplified products were sequenced and the results were analyzed by MEGA 5.0. Chemometric methods including principal components analysis and hierarchical clustering analysis were conducted on the SAS 9.0 software to demonstrate the variability among samples. In conclusion, the psbA-trnH of all samples were successfully amplified from total DNA and sequenced. These three varieties of Plumeria can be differentiated by the psbA-trnH region and clustered into three groups respectively through building neighbor joining tree, which conformed to their germplasm origins. However, it was hard to distinguish them by HPLC specific chromatograms combined with chemometrics analysis. These indicated that DNA barcoding was a promising and reliable tool for the identification of three kinds of Plumeria flowers compared to HPLC specific chromatogram generally used. It could be treated as a powerful complementary method for traditional authentication, especially for those varieties which are difficult to be identified by conventional chromatography. 展开更多

The fingerprint chromatogram of 10 batches of samples of Tetracera asiatica was determined for similarity comparison. [Results] It established HPLC fingerprint chromatogram of Tetracera asiatica produced in Guangxi,determined a total of 13 common fingerprint peaks. Sweet from different habitats, in order to provide experimental basis for the quality control of A. indicum and the spectrum-efficacy relationship of petroleum ether.[Methods] The HPLC chromatogram of petroleum ether was obtained using Agilent SB-C 18 (4.6 mm×250 mm, 5 μm) and acetine-0.1% phosphoric acid solution for gradient elution at temperature of 30 ℃, flow rate of 1.0 mL/min and detection wavelength of 205 nm.[Results] The typical HPLC chromatogram of petroleum ether in A. indicum was preliminarily established, with 12 characteristic peaks. It was verified by A. indicum from different habitats, characterized by reproducibility and stability.[Conclusions] Typical HPLC chromatogram provides a stable new method for the quality control of petroleum ether in A. indicum . Ganoderma polysaccharides(peptides)are essential bio-macromoleculars that determine the quality of Ganoderma.Their molecular weight,distribution and content are influenced by factors such as origin,strain,cultivation conditions,harvesting period and processing methods.In this study,we established a molecular weight specific chromatogram using high performance gel permeation chromatography(HPGPC)for Ganoderma glycopeptide and assessed its potential applications.The results revealed consistent molecular weight distributions across different cultivation conditions,with 6–8 characteristic peaks showing similarity values exceeding 0.93.The average molecular weight of common peaks was highest during the budding stage and lowest in the mature stage.Ganoderma lucidum(GL)exhibited higher molecular weights compared to Ganoderma sinense(GS).Moreover,both molecular weight and Ganoderma glycopeptides decreased with prolonged growth periods.The content of GL-PPSQ2 was significantly higher in the budding(1.63±0.15%)and cap-opening stages(1.51±0.12%)compared to the mature stage(1.31±0.05%).Chemometrics analysis revealed distinct molecular weight profiles for different growth stages and varieties.Additionally,the HPGPC method was successfully applied to detect adulteration in Ganoderma extracts,identifying excipients such as maltodextrin and dextran.This study underscores the importance of molecular weight distribution and glycopeptide content as key quality control indicators,providing a rapid and reliable tool for ensuring the authenticity and quality of Ganoderma extracts in the market. 展开更多

目的基于特征图谱探究牛膝盐炙、酒炙前后的差异性特征成分,结合网络药理学预测及分子对接验证,筛选牛膝干预膝骨关节炎(KOA)的潜在质量标志物(Q-Marker)。方法采用HPLC技术建立牛膝、盐牛膝及酒牛膝的特征图谱,结合主成分分析(PCA)与... 目的基于特征图谱探究牛膝盐炙、酒炙前后的差异性特征成分,结合网络药理学预测及分子对接验证,筛选牛膝干预膝骨关节炎(KOA)的潜在质量标志物(Q-Marker)。方法采用HPLC技术建立牛膝、盐牛膝及酒牛膝的特征图谱,结合主成分分析(PCA)与正交偏最小二乘法-判别分析(OPLS-DA)筛选炮制前后的主要差异成分;通过网络药理学预测特征成分治疗KOA的核心靶点与关键通路,绘制“药物-成分-靶点-疾病-通路”网络以筛选抗KOA差异活性成分;结合Q-Marker五原则及分子对接验证结果,筛选潜在Q-Marker。结果15批牛膝、盐牛膝及酒牛膝的特征图谱分别标定27、28、34个共有峰,经对照品比对,3种牛膝共指认14种共有化学成分;化学计量学分析显示,牛膝盐炙前后的主要差异成分为人参皂苷Ro、姜状三七皂苷R1、齐墩果酸、金盏花皂苷E、竹节参皂苷Ⅰ、竹节参皂苷Ⅳa及竹节参皂苷Ⅳa甲酯,酒炙前后的主要差异成分为人参皂苷Ro、竹节参皂苷Ⅰ、姜状三七皂苷R1、金盏花皂苷E、β-蜕皮甾酮、齐墩果酸及竹节参皂苷Ⅳa。网络药理学预测表明,核心成分β-蜕皮甾酮、旌节花甾酮D、竹节参皂苷Ⅳa甲酯、齐墩果酸、人参皂苷Ro,可能通过调控白蛋白(ALB)、原癌基因(JUN)、过氧化物酶体增殖物激活受体γ(PPARG)、半胱氨酸天冬氨酸蛋白酶3(CASP3)、雌激素受体α基因(ESR1)、抗凋亡蛋白B细胞淋巴瘤因子-2(BCL2)6个核心靶标,参与癌症通路、白细胞介素(IL)-17信号通路、前列腺癌等关键信号通路发挥抗KOA作用;分子对接验证进一步证实,上述5个核心成分与JUN、BCL2靶标均具有良好结合活性。结合Q-Marker五原则,最终筛选出β-蜕皮甾酮、人参皂苷Ro、齐墩果酸为牛膝治疗KOA的潜在Q-Marker。结论建立的HPLC特征图谱可有效区分牛膝及其盐炙、酒炙品;经网络药理学预测、分子对接验证及Q-Marker五原则筛选出的差异活性成分,可作为牛膝治疗KOA的潜在Q-Marker。 展开更多

目的建立以对照药材为基准物质的定性和不依赖多种对照品定量的北柴胡药材“质-量”双标控制方法。方法采用HPLC技术,以对照药材为基准物质建立北柴胡特征图谱,通过Q-TOF-MS技术,鉴定共有峰的化学成分,以对照药材和供试药材特征峰的相似... 目的建立以对照药材为基准物质的定性和不依赖多种对照品定量的北柴胡药材“质-量”双标控制方法。方法采用HPLC技术,以对照药材为基准物质建立北柴胡特征图谱,通过Q-TOF-MS技术,鉴定共有峰的化学成分,以对照药材和供试药材特征峰的相似度,明确药材真伪,即“质”;对内标成分柴胡皂苷a进行准确定量,计算不同批次供试品中各特征峰的相对含量,取“平均数-标准差”作为相对含量下限,依据特征峰相对含量下限明确北柴胡药材优劣,即“量”。结果建立的特征图谱及含量测定方法符合方法学考察要求;确定了12个共有色谱峰,各北柴胡供试药材与北柴胡对照药材的相似度均大于0.900;分子对接结果表明柴胡皂苷a与各蛋白靶点结合能力强。结论该方法不依赖多种对照品,能快速地判断药材的真伪优劣,为北柴胡的质量控制方法提供参考。 展开更多

目的:建立祖卡木颗粒的特征图谱和多成分含量测定的高效液相色谱法,采用化学模式识别技术评价祖卡木颗粒的质量,为其质量控制提供依据。方法:运用《中药色谱指纹图谱相似度评价系统(2012版)》建立12批祖卡木颗粒的高效液相色谱(HPLC)特... 目的:建立祖卡木颗粒的特征图谱和多成分含量测定的高效液相色谱法,采用化学模式识别技术评价祖卡木颗粒的质量,为其质量控制提供依据。方法:运用《中药色谱指纹图谱相似度评价系统(2012版)》建立12批祖卡木颗粒的高效液相色谱(HPLC)特征图谱,指认共有峰,并进行相似度评价;建立样品中没食子酸、迷迭香酸、烟花苷、对甲氧基肉桂酸乙酯和甘草酸铵5个有效成分的HPLC含量测定方法,并进行含量测定;采用SPSS 23.0、SIMCA 14.1软件对祖卡木颗粒进行聚类分析以及正交偏最小二乘判别分析,以变量重要性投影(VIP)值> 1.0为标准筛选影响祖卡木颗粒质量的差异性成分。结果:建立的12批样品的特征图谱及18批样品的多成分含量测定方法均经方法学考察。其中,12批祖卡木颗粒特征图谱的相似度为0.993、0.971、0.987、1.000、0.999、0.997、0.994、0.998、0.999、1.000、0.999、0.999,共有10个共有峰,通过对照品指认了其中5个共有峰。此外,测定了18批祖卡木颗粒中没食子酸、迷迭香酸、烟花苷、对甲氧基肉桂酸乙酯和甘草酸铵的平均含量分别为0.506、0.124、0.071、0.003和0.089 mg·g-1。聚类分析和正交偏最小二乘判别分析结果表明,18批祖卡木颗粒可聚为2类,其中ZKM03、ZKM05和ZKM13聚为一类,其余为一类,峰1、7、9的VIP值> 1.0。结论:建立的特征图谱和含量测定方法稳定,可靠,结合化学模式识别技术可用于评价祖卡木颗粒的整体质量,没食子酸、烟花苷和对甲氧基肉桂酸乙酯是影响祖卡木颗粒质量的差异性成分。 展开更多

目的:建立经典名方桔梗汤的高效液相色谱法(HPLC)特征图谱,并基于一测多评法测定其主要成分的含量。方法:采用Phenomenex-C_(18)色谱柱(250 mm×4.6 mm,5μm),以0.05%磷酸水溶液(A)-乙腈(B)为流动相梯度洗脱,采用二极管阵列检测器,... 目的:建立经典名方桔梗汤的高效液相色谱法(HPLC)特征图谱,并基于一测多评法测定其主要成分的含量。方法:采用Phenomenex-C_(18)色谱柱(250 mm×4.6 mm,5μm),以0.05%磷酸水溶液(A)-乙腈(B)为流动相梯度洗脱,采用二极管阵列检测器,检测波长为203 nm,柱温为35℃,流速为1.0 mL·min^(-1),建立桔梗汤的HPLC特征图谱,对其共有峰进行指认、归属,并对15批桔梗汤的HPLC特征图谱与对照图谱间的相似度进行分析。采用一测多评法测定桔梗汤中去芹糖桔梗皂苷E、桔梗皂苷D、甘草苷、甘草酸4个主要成分的含量。结果:建立了桔梗汤的HPLC特征图谱,共标定14个特征峰,指认峰5、峰9、峰12、峰14代表的化学成分分别为甘草苷、甘草酸、去芹糖桔梗皂苷E、桔梗皂苷D,其中甘草苷、甘草酸来自甘草,芹糖桔梗皂苷E、桔梗皂苷D来自桔梗。15批桔梗汤HPLC特征图谱与对照图谱间的相似度均大于0.900。15批桔梗汤中去芹糖桔梗皂苷E、桔梗皂苷D、甘草苷、甘草酸的质量分数分别为0.31~0.35、5.43~6.14、6.31~7.05、10.35~12.45 mg·g^(-1)。结论:建立的HPLC特征图谱和一测多评法高效、可行,为经典名方桔梗汤的质量评价提供参考。 展开更多
