However, the precise mechanism remains poorly understood. In this study, we administered chitosan intragastrically to an MPTP-induced mouse model of Parkinson's disease and found that it effectively reduced dopamine neuron injury, neurotransmitter dopamine release, and motor symptoms. These neuroprotective effects of chitosan were related to bacterial metabolites, specifically shortchain fatty acids, and chitosan administration altered intestinal microbial diversity and decreased short-chain fatty acid production in the gut. Furthermore, chitosan effectively reduced damage to the intestinal barrier and the blood–brain barrier. Finally, we demonstrated that chitosan improved intestinal barrier function and alleviated inflammation in both the peripheral nervous system and the central nervous system by reducing acetate levels. Based on these findings, we suggest a molecular mechanism by which chitosan decreases inflammation through reducing acetate levels and repairing the intestinal and blood–brain barriers, thereby alleviating symptoms of Parkinson's disease.展开更多

Cellulose, cellulose triacetate, chitin and chitosan were modeled as trimeric form of the linear chain of4 C1 chair conformation of β-D-glucopyranos, its triacetate form, β-N-acetylglucosamine and D-glucosamine, respectively, in the 1 ? 4 linkage. Geometries were optimized at the M062 X functional level of the density functional theory(DFT) using the 6-31 G(d,p) basis set in the gas phase and in the bulk water solution using the conductor-like polarizable continuum model(CPCM) approach. The nature of potential energy surfaces of the optimized geometries were ascertained through the harmonic vibrational frequency analysis. The basis set superposition error(BSSE) corrected interaction energies were obtained using the 6-311 G(d,p)basis set at the same theoretical level. The computed BSSE in the gas phase was used to correct interaction energy in the bulk water solution. Computed and experimental results regarding the ability of considered surfaces in adsorbing the insensitive munitions compounds are discussed.展开更多

Here, the chitin nanocrystal( Chi NC) was introduced into the aforementioned system, and its effects on solution behavior and mechanical properties was investigated. The results showed the incorporation of Chi NC complicated sol-to-gel transition process; a higher loading ratio( 20%) speeded up sol-to-gel transition rate, reduced the solto-gel transition temperature, while still maintained shear-thinning behavior or injectable feature. Moreover, the mechanical properties of gels were significantly enhanced by Chi NC, accompanied by decreased water uptake. The above mentioned behavior favored better applications as injectable tissue-repair implants.展开更多

Then, a three factors (NaOH solution concentration, reaction time, reaction temperature) and three levels (35, 45, 55; 2, 6, 10; 70, 105, 140) L 9(3 4) orthogonal experiment design is further adopted to conduct a de acetyl treatment to prepare chitosan by considering the viscosity and de acetyl degree of the chitosan as the main performance indexes. Determination of de acetyl degree of chitin complys with the procedures given by the reference and the viscosity meter was used for determination of viscosity of chitosan. The results show that the extraction of chitin shall use pulverized crab shell as the raw material and such raw material shall be immersed in 10% HCl solution for 6 hours and washed with water for one time in every 2 hours, then heated in boiled water for 2 hours by the use of 10% thin NaOH solution. Afterwards, the said material shall be washed with water to become a neutral solution and dried over a stove. When chitin is mixed with 55% NaOH solution in a proportion of 1∶10 (W/V, g/mL) and the reaction takes place at a temperature of 105℃ for 6 hours, chitosan having a de acetyl percentage of 94% and viscosity of >200 cps can be available.展开更多

Metallic ions if used for a prolonged time may lead to toxicity. Alternatively chitin,a natural polysaccharide found in nature, is utilized. It is found in fungi, crabs, mushrooms, squids, octopus, and many other living organisms. Chitin has similar structure to cellulose but its deacetylated derivate chitosan has amine groups that provide potential antibacterial properties along with a number of other advantages. Chitin in its natural form is found in three different structural forms, namely α,β,and γ.The β-chitin and chitosan are mostly found in the exoskeleton of squids. Loligo and Humboldt squids were studied. It is anticipated that Humboldt chitin is more effective in serving as antibacterial material and can be utilized for wound care. Differences in steriochemical structure were observed among β-chitin structures obtained and amine group's presences were found along with ability of materials to swell.展开更多

Recombinant chitinase (rChi-Ah) was produced in?Escherichia coli. Purified rChi-Ah had optimal activity at temperature of 42℃?and pH 6.5. The affinity (Km) for chitosan was 4.18 mg·ml-1?with?Vmax?of 202.5 mg·min-1·mg-1. With colloidal chitin as substrate, rChi-Ah generated N,N’-diacetyl-glucosamine predominantly. Conversion of chitosan (≥75% deacetylated) by rChi-Ah revealed five major products: 2 to 4 units of glucosamine, all of which had at least one acetyl group. It was determined that N-acetylated glucosamine was the recognition and cleavage site of rChi-Ah;the minimal and maximal cleavages were two and four glucosamine units, respectively. Functional analysis of rChi-Ah suggests that?A. hydrophilachitinase is a bioactive chitinolytic enzyme, which may benefit the pathogen for survival and/or infection.展开更多

DC proton conductivity σ is obtained from Nyquist plot of impedance measurement data, and the relationship between σ and mixing ratio x has been made clear. It was found that the x dependence of σ is non-monotonous. That is, σ shows the anomalous behavior, and has peaks around x = 0.4 and 0.75. This result indicates that there exist optimal conditions for the realization of high-proton conductivity in the chitin-chitosan mixed compound in which the number of acetyl groups is different. From the FT-IR measurement, we have found that the behavior of proton conductivity in (chitin)x(chitosan)1-x is determined by the amount of water content changed by x. Using these results, proton conductivity, which is important for the application of conducting polymers in chitin-chitosan mixed compounds, will be able to be easily controlled by adjusting the mixing ratio x.展开更多

The quaternization of NBCS was carried out by using ethyl iodide to produce water-soluble cationic polyelectrolytes. The degree of the substitution of the products was measured by means of the electron spectroscopy for a successful chemical analysis. The quaternary ammonium chitosan salt(QACS) was proved to be a novel cholesteric liquid crystalline chitin, by means of the polarized optical microscopy and the circular dichroism spectropolarimetry. The critical mass fraction of the QACS/formic acid solution forming a lyotropic liquid crystal phase was 50%, which was almost the same as that of the NBCS/formic acid solution, but much higher than that of the chitosan in the same solvent.展开更多

文摘This study presents a thorough investigation into the use of single and twin-tailed cationic and anionic surfactant-modified chitosan(SMCS)hydrogel beads as effective adsorbents for the elimination of hazardous polycyclic aromatic hydrocarbons(PAHs)from aqueous solutions. The Chitosan(CS)hydrogel beads were modified with single/twin-tailed anionic surfactants, sodium dodecyl sulfate(SDS)and sodium bis(2-ethylhexyl)sulfosuccinate(AOT),and cationic surfactants, dodecyltrimethylammonium bromide(DTAB)and didodecyldimethylammonium bromide(DDAB),to enhance their adsorption capacity of PAHs.The CS and SMCS beads were evaluated for their structural, mechanical, and adsorption properties using a range of techniques, including infrared spectroscopy(IR),energy-dispersive X-ray spectroscopy(EDX),rheometry, and field emission scanning electron microscopy(FESEM).Adsorption experiments of naphthalene(Nap),acenaphthene(Ace),and phenanthrene(Phe)on SMCS beads demonstrate that they have significantly higher adsorption capacities than CS beads, due to increase in hydrophobic interactions.Adsorption capacity followed the trend,Phen>Ace>Nap for all the beads revealing that twin-tailed SMCS bead possess much higher adsorption capacities(Qmax)compared to single-tailed SMCS beads. For twin tailed surfactants, the maximum adsorption capacities for Nap,Ace and Phe varied as CS-AOT(CS-DDAB):430.0(323.8)611.60(538.18)633.39(536.99)mg/g respectively, outperforming other reported hydrogel beads. The study highlights the simplicity,eco-friendliness,and enhanced performance of surfactant modification for developing high-efficiency adsorbents, paving the way for cost-effective solutions in water re-mediation.

文摘In this study, chitosan(CS)was combined with microcrystalline cellulose(MCC)to fabricate composite hydrogel beads. These beads were further modified through blending and grafting with polyethyleneimine(PEI)to develop chitosan/microcrystalline cellulose@polyethyleneimine(CS/MCC@PEI)composite gel spheres for the efficient adsorption of diclofenac sodium(DS)from aqueous solutions. The adsorbent was characterized using scanning electron microscopy(SEM),X-ray diffraction(XRD),Fourier-transform infrared spectroscopy(FTIR),X-ray pho-toelectron spectroscopy(XPS),and thermogravimetric analysis(TGA).The CS/MCC@PEI composite exhibited a spherical morphology with a porous structure, abundant surface functional groups, and a high adsorption capac-ity of 274.84 mg/g for DS.Kinetic studies revealed that the adsorption process followed the pseudo-second-order model, dominated by physical adsorption, with both surface and internal diffusion influencing the adsorption rate. The Freundlich isotherm model best described the adsorption behavior, indicating multilayer adsorption on heterogeneous surfaces.Environmental adaptability tests demonstrated minimal interference from co-existing anions and humic acid, while regeneration experiments confirmed excellent reusability(>77%removal after five cycles). The adsorption mechanism involved electrostatic interactions and hydrogen bonding between the hydroxyl/amino groups of the composite and DS.These findings highlight the potential of CS/MCC@PEI as a cost-effective and sustainable adsorbent for DS removal from water.

文摘Studies have shown that chitosan protects against neurodegenerative diseases. Based on these findings, we suggest a molecular mechanism by which chitosan decreases inflammation through reducing acetate levels and repairing the intestinal and blood–brain barriers, thereby alleviating symptoms of Parkinson's disease.
文摘This manuscript reports results of combined computational chemistry and batch adsorption investigation of insensitive munition compounds, 2,4-dinitroanisole(DNAN), triaminotrinitrobenzene(TATB), 1,1-diamino-2,2-dinitroethene(FOX-7) and nitroguanidine(NQ), and traditional munition compound 2,4,6-trinitrotoluene(TNT) on the surfaces of cellulose, cellulose triacetate, chitin and chitosan biopolymers. Computed and experimental results regarding the ability of considered surfaces in adsorbing the insensitive munitions compounds are discussed.

文摘Chitin was first discovered by its name from the Greek word“chiton”,which means“mail coat”.It is indeed a polysaccharide made up of naturally occurring acetyl-D-glucosamine monomers.Hatchett was the first researcher who extracted chitin from the shells of mollusks(crabs and lobsters),prawns, and crayfish in 1799.Later in 1811,Henri Braconnot discovered chitin in the cell walls of mushrooms and called it“fungine”.Chitin and chitosan are abundant in the biosphere as essential components of many organisms’exoskeletons and as by-products of the global seafood industry. The biopolymer must be deacetylated before chitosan can be produced. It can also be extracted using microbes in a biological extraction procedure. The development of products that take advantage of the bioactivities of the existing primary commercial source of chitin(crustacean)has lagged expectations. Also, the disadvantages of the present commercial source such as seasonality and competition for other uses among others has been one of the driving forces towards seeking alternative sources of chitin and chitosan in nature. This review highlights some of the efforts made by environmental scholars to locate possible commercial sources of chitin and chitosan in nature over time.

文摘The chitosan/β-glycerophosphate( CS/β-GP),a physical hydrogel system with thermosensitive and injectable features combined with biocompatibility and biodegradability, has great potentials as matrices for drug or cell encapsulation and delivery,or as in situ gel-forming materials for tissue repair. The above mentioned behavior favored better applications as injectable tissue-repair implants.
文摘A combination of both acid and alkali treatments was used to extract chitin from crab shell in this study. When chitin is mixed with 55% NaOH solution in a proportion of 1∶10 (W/V, g/mL) and the reaction takes place at a temperature of 105℃ for 6 hours, chitosan having a de acetyl percentage of 94% and viscosity of >200 cps can be available.
文摘A number of materials are utilized to develop wound care dressing materials with metallic treatments such as ionic silver and zinc. Differences in steriochemical structure were observed among β-chitin structures obtained and amine group's presences were found along with ability of materials to swell.
文摘A chitinase was identified in extracellular products of a virulent?Aeromonas hydrophila?isolated from diseased channel catfish (Ictalurus punctatus). Functional analysis of rChi-Ah suggests that?A. hydrophilachitinase is a bioactive chitinolytic enzyme, which may benefit the pathogen for survival and/or infection.
文摘In order to investigate a key factor for the appearance of proton conductivity in chitin-chitosan mixed compounds, the chitin-chitosan mixed compounds (chitin)x(chitosan)1-x were prepared and these proton conductivities have been investigated. Using these results, proton conductivity, which is important for the application of conducting polymers in chitin-chitosan mixed compounds, will be able to be easily controlled by adjusting the mixing ratio x.

文摘N -Butyl chitosan(NBCS) derivatives were prepared by introducing butyl groups into the amine groups of chitosan via Schiff base intermediates. The critical mass fraction of the QACS/formic acid solution forming a lyotropic liquid crystal phase was 50%, which was almost the same as that of the NBCS/formic acid solution, but much higher than that of the chitosan in the same solvent.
文摘The potent antibacterial activity of silver nanoparticles is primarily attributed to the release of silver ions, which disrupt cell membranes and inactivate essential enzymes through Ag−S bonding formation.[Objective]To explore silver ion immobilization to minimize silver release.[Methods]A macrocyclic cryptand with nitrogen bridgeheads was prepared and subsequently chelated with silver ions to produce Cage silver(I),which was then coordinated with different ratios of sulfonated chitosan(SCS)to form SCS/Cage Ag(I)complexes(SCA1,SCA2,and SCA3).The antioxidant activities of the complexes were assessed by reducing power and 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radical and hydrogen peroxide scavenging assays. The antibacterial activities of the complexes were evaluated based on the minimum inhibitory concentrations(MICs)and minimum bactericidal concentrations(MBCs)against Staphylococcus aureus ATCC 6538 and Escherichia coli O157:H7 and the inhibition rate on biofilm formation.[Results]Cage silver(I)exhibited strong antibacterial activity, with the MIC of 0.015 mg/mL and MBC of 0.031 mg/mL against S.aureus ATCC 6538,and the MIC of 0.031 mg/mL and MBC of 0.120 mg/mL against E.coli O157:H7.Significant antioxidant properties of Cage silver(I)were also observed,as demonstrated by the DPPH free radical scavenging rates of 42.2%and 53.1%at 326 nm and 517 nm,respectively.Cage silver(I)exhibited the highest antibacterial and antioxidant activities, followed by SCA1,SCA2,SCA3,and SCS,because the content of silver ions in Cage silver(I)was 10-fold higher than that in SCA1.The antibacterial and antioxidant activities of SCA1 were better than those of Cage silver(I),which further indicated that the sulfonic groups of SCS may intensely coordinate with silver ions to exert synergistic effects.[Conclusion]Combining the merits of silver ions and SCS improves the bioavailability of the agent at microbicidal concentrations, minimizes the accumulation in the environment, and reduces treatment costs. The method developed herein offers a sustainable approach to enhance microbial control while minimizing the impact on the environment.

文摘Chitin is an abundant aminopolysaccharide found in insect pests and phytopathogenic microorganisms but absent in higher plants and vertebrates. It is crucial for mitigating threats posed by chitin-containing organisms to human health, food safety, and agriculture. Therefore, targeting the chitin biosynthesisassociated bioprocess holds a promise for developing human-safe and eco-friendly antifungal agents or pesticides. Chitin biosynthesis requires chitin synthase and associated factors, which are involved in the modification, regulation, organization or turnover of chitin during its biosynthesis. A number of enzymes such as chitinases, hexosaminidases, chitin deacetylases are closely related and therefore are promising targets for designing novel agrochemicals that target at chitin biosynthesis. This review summarizes the advances in understanding chitin biology over the past decade by our research group and collaborates, specifically regarding essential proteins linked to chitin biosynthesis that can be exploited as promising pesticide targets. Examples of small bioactive molecules that against the activity of these targets are given.

文摘The combination of solar disinfection and photocatalysis technology presents a viable solution for eliminating harmful pathogenic microorganisms from water. However, some photocatalysts(e.g.,zinc oxide-based composites)are susceptible to pH-dependent dissolution in water, which can result in the loss of photocatalysts and additional environ-mental pollution. To obtain zinc oxide-based composites with low dissolution and high antibacterial efficiency for pho-tocatalytic water disinfection,we prepared MoS_(2)/ZnO@CS composites via a precipitation method to encapsulate chitosan(CS)around MoS_(2)/ZnO.The amino groups in the CS molecules act as storerooms for hydrogen ions, which inhibits the dissolution of zinc oxide. In addition, the MoS_(2)/ZnO@CS composites exhibit high production of reactive oxygen species(ROS)and broad-spectrum antibacterial activity under simulated solar irradiation(0.1 W·cm^(-2)).This makes it an excellent antibacterial agent for solar disinfection in water treatment.

文摘Chitin is an abundant natural biopolymer that plays a crucial role in insect growth and development as a fundamental structural component of the exoskeleton. The membrane-integralβ-glycosyltransferase,chitin synthase, has been identified as the central component in chitin biosynthesis. However, the precise roles of other proteins in facilitating chitin synthase in chitin biosynthesis remain unclear. In this study,we employed split-ubiquitin membrane yeast two-hybrid(MYTH)and pull-down assays to demonstrate the physical interaction between Twinstar(Tsr),a small molecular protein in the actin-depolymerizing factor ADF/Cofilin protein family, and chitin synthase Krotzkopf verkehrt(Kkv)in Drosophila melanogaster in vitro. The RNA interference(RNAi)-mediated global knockdown of Tsr in D.melanogaster resulted in larval lethality.Furthermore, targeted suppression of Tsr in the tracheal and epidermal tissues also led to larval mortality, while knocking down Tsr in the wing tissues led to wrinkled wings.Additionally, silencing Tsr not only reduced the chitin content in the first longitudinal vein of the wings but also led to the absence of the chitin lamellar structure. To validate the functional conservation of Tsr in other insect orders, the two agricultural pests Ostrinia furnacalis and Tribolium castaneum, representing lepidoptera and coleoptera insects, respectively, were investigated.Knockdown experiments targeting the Drosophila Tsr orthologues OfTsr in O.furnacalis and TcTsr in T.castaneum produced abnormal larvae during molting or pupation in O.furnacalis and lethality in T.castaneum.Our findings not only improve ourknowledge of the chitin biosynthesis machinery in insect cuticles but also provide new potential targets for the control of major agricultural pests.
