Photodynamic therapy is an"old"strategy for cancer therapy featuring clinical safety and rapid working,but suitable photosensitizers for colorectal cancer therapy remain lacking.This study synthesized a nove...Photodynamic therapy is an"old"strategy for cancer therapy featuring clinical safety and rapid working,but suitable photosensitizers for colorectal cancer therapy remain lacking.This study synthesized a novel photosensitizer termed Ce6-GFFY based on a self-assembling peptide GFFY and a photo-responsive molecule chlorin e6(Ce6).Ce6-GFFY forms macroparticles with a diameter of~160 nm and possesses a half-life of 10 h,as well as an ideal tumor-targeting ability in mouse models.Ce6-GFFY effectively penetrates cells and generates numerous reactive oxygen species upon 660 nm laser irradiation.The reactive oxygen species promotes the accumulation of cytotoxic T cells and decrease of myeloid-derived suppressor cells in the tumor microenvironment through immunogenic cell death,thus prohibiting the growth of both primary and metastatic tumors after once treatment.This study not only provides a strategy for photosensitizer development but also confirms a promising application of Ce6-GFFY for colorectal cancer therapy.展开更多
Hepatocellular carcinoma(HCC)has been known as the second common leading cancer worldwide,as it responds poorly to both chemotherapy and medication.Triptolide(TP),a diterpenoid triepoxide,is a promising treatment agen...Hepatocellular carcinoma(HCC)has been known as the second common leading cancer worldwide,as it responds poorly to both chemotherapy and medication.Triptolide(TP),a diterpenoid triepoxide,is a promising treatment agent for its effective anticancer effect on multiple cancers including HCC.However,its clinical application has been limited owing to its severe systemic toxicities,low solubility,and fast elimination in the body.Therefore,to overcome the above obstacles,photo-activatable liposomes(LP)integrated with both photosensitizer Ce6 and chemotherapeutic drug TP(TP/Ce6-LP)was designed in the pursuit of controlled drug release and synergetic photodynamic therapy in HCC therapy.The TP encapsulated in liposomes accumulated to the tumor site due to the enhanced permeability and retention(EPR)effect.Under laser irradiation,the photosensitizer Ce6 generated reactive oxygen species(ROS)and further oxidized the unsaturated phospholipids.In this way,the liposomes were destroyed to release TP.TP/Ce6-LP with NIR laser irradiation(TP/Ce6-LP+L)showed the best antitumor effect both in vitro and in vivo on a patient derived tumor xenograft of HCC(PDXHCC).TP/Ce6-LP significantly reduced the side effects of TP.Furthermore,TP/Ce6-LP+L induced apoptosis through a caspase-3/PARP signaling pathway.Overall,TP/Ce6-LP+L is a novel potential treatment option in halting HCC progression with attenuated toxicity.展开更多

In this study,a batch of φ12 mm Cs_(2)LiYCl_(6):Ce crystals codoped with different contents of Cu^(+)and Sc^(3+)was successfully grown using the Multi-ampule Bridgeman method.A new emission peaking at 418 nm is found...In this study,a batch of φ12 mm Cs_(2)LiYCl_(6):Ce crystals codoped with different contents of Cu^(+)and Sc^(3+)was successfully grown using the Multi-ampule Bridgeman method.A new emission peaking at 418 nm is found in the photoluminescence spectra of CLYC:Ce codoped with Cu^(+)ion.Codoping Cu^(+)or Sc^(3+)both increases the proportion of intrinsic self-trapped exciton(STE)luminescence,and extends the excitation band of Ce^(3+),especially in Cu^(+)codoped samples,where a new absorption peak at 248 nm can be identified.The light yield of Cu^(+)codoped samples remains largely unchanged,but the energy resolution shows a slight deterioration.Both light yield and energy resolution degrade after Sc^(3+)codoping,and the effect is much severe than that of Cu^(+)codoped samples.X-ray induced afterglow can be suppressed after Cu^(+)codoping and low content of Sc^(3+)codoping.The scintillation decay variation also depends on the codoping ions and their contents.展开更多
