Vuill and Colletotrichum dematium (Pers.) Grove). Among these three elicitors, V. dahliae had the highest inducing efficiency, but none of them manifests any noticeable effects on the cell growth of the hairy root cultures. The artemisinin content of the hairy root cultures treated with V. dahliae elicitor was 1.12 mg/g DW, which was 45% higher than the control (0.77 mg/g DW). The results showed that elicitation was dependent on the elicitor concentration, the incubation period and the physiological stage at which the hairy root cultures were treated. In addition, the authors found that for V. dahliae , the optimum concentration was 0.4 mg carbohydrate per millilitre medium, the strongest response of A. annua hairy root cultures to the elicitation was at the late exponential growth stage, and the highest artemisinin content of the hairy root cultures was on the 4th day post treatment.展开更多

Based on NMR and MS analyses, and comparison with literature data and standards, their structures were determined to be 2α,5α,10β_triacetoxy_14β_propionyloxy_4(20),11_taxadiene (1), 2α,5α,10β_triacetoxy_14β_(2′_methyl)_butyryloxy_4(20),11_taxadiene (2), 2α,5α,10β_14β_tetra_acetoxy_4 (20),11_taxadiene (3, taxuyunnanine C), 2α,5α,10β_triacetoxy_14β_(2′_methyl_3′_hydroxy)_butyryloxy_4(20),11_taxadiene (4, yunnanxane) and its 3′_epimer (5), baccatin Ⅳ (6), baccatin Ⅲ (7) and taxol (8), respectively. Among those compounds, 3, 5, 6 and 7 were reported to be isolated from the suspension cell cultures of T. yunnanensis for the first time. TLC and HPLC analyses indicated that the chemical constituents of the culture solution were similar to those of cultured cells. Ethyl acetate extract of the cultures showed inhibitory activities against Penicillium avellaneum UC-4376 which was sensitive to maytansinoids. Exhaustive isolation of natural products from a large scale of suspension cell cultures did not yield maytansine instead of affording nine compounds including one novel triterpenoid, named 2, 3-diacetoxyl maytenusone (1), and eight known ones including squalene (2), beta-sitosterol (3), 2', 3', 4-triacetyl-sitoindoside I (4), salaspermic acid (5), maytenonic acid (6), 2alpha-hydroxy-maytenonic acid (7), 6, 11,12-trihydroxy-8, 11, 13-abietrien-7-one (8) and 11, 12-dihydroxy-8, 11, 13-abietatrien-7-one (9) elucidated on the basis of 1D and 2D NMR data. The H-1-NMR and C-13-NMR assignments were made for 1, 5, 6 and 7, while the C-13-NMR assignments for 5 and 6 were revised. This paper considers the notions of common sense and interobjectivity to articulate an understanding of how different cultural realities give rise to different construals of scientific phenomena across distinct cultur...This paper considers the notions of common sense and interobjectivity to articulate an understanding of how different cultural realities give rise to different construals of scientific phenomena across distinct cultures. Our main focus in this paper is on the social sciences. We propose a quadrant of different cultural–scientific stances from which the study of social phenomena is possible, based on the emic–etic dimension pertaining to the study of culture from contrasting perspectives. Although the emic–etic distinction is normal y applied in fields within the science of culture, it is proposed here that the distinction is in some ways germane to scientific practice in general, making it amenable for use in a culture of science(CoS) programme. The four perspectives that emerge from the quadrant are illustrated using exemplars. Different aspects of CoS—that is, scientific practice, scientific conventions and representations of science—are then discussed in further detail, including in two tables illustrating points of convergence and divergence between the East and West when it comes to different aspects of CoS.展开更多

BACKGROUND Fracture is one of the most pervasive injuries in the musculoskeletal system,and there is a complex interaction between macrophages and adipose tissue-derived stem cells(ADSCs)in fracture healing.However,tw...BACKGROUND Fracture is one of the most pervasive injuries in the musculoskeletal system,and there is a complex interaction between macrophages and adipose tissue-derived stem cells(ADSCs)in fracture healing.However,two-dimensional(2D)coculture of macrophages and ADSCs can not accurately mimic the in vivo cell microenvironment.AIM To establish both 2D and 3D osteogenic coculture models to investigate the interaction between macrophages and ADSCs.METHODS After obtaining ADSCs from surgery and inducing differentiation of the THP1 cell line,we established 2D and 3D osteogenic coculture models.To assess the level of osteogenic differentiation,we used alizarin red staining and measured the relative expression levels of osteogenic differentiation markers osteocalcin,Runt-related transcription factor 2,and alkaline phosphatase through polymerase chain reaction.Verification was conducted by analyzing the expression changes of N-cadherin and the activation of the Wnt/β-catenin signaling pathway using western blotting.RESULTS In this study,it was discovered that macrophages in 3D culture inhibited osteogenic differentiation of ADSCs,contrary to the effect in 2D culture.This observation confirmed the significance of intricate intercellular connections in the 3D culture environment.Additionally,the 3D culture group exhibited significantly higher N-cadherin expression and showed reducedβ-catenin and Wnt1 protein levels compared to the 2D culture group.CONCLUSION Macrophages promoted ADSC osteogenic differentiation in 2D culture conditions but inhibited it in 3D culture.The 3D culture environment might inhibit the Wnt/β-catenin signaling pathway by upregulating N-cadherin expression,ultimately hindering the osteogenic differentiation of ADSCs.By investigating the process of osteogenesis in ADSCs,this study provides novel ideas for exploring 3D osteogenesis in ADSCs,fracture repair,and other bone trauma repair.展开更多
Background The use of conventional two-dimensional(2D)culture of the porcine intestinal epithelial cell(IEC)line IPEC-J2 in animal nutrition research has the disadvantage that IEC function is studied under unphysiolog...Background The use of conventional two-dimensional(2D)culture of the porcine intestinal epithelial cell(IEC)line IPEC-J2 in animal nutrition research has the disadvantage that IEC function is studied under unphysiological conditions,which limits the ability of transferring knowledge to the in vivo-situation.Thus,the aim of the present study was to establish a more convincing and meaningful three-dimensional(3D)culture of IPEC-J2 cells,which allows to study cell function in a more tissue-like environment,and to compare the effect of the endoplasmic reticulum(ER)stress inducer tunicamycin(TM)on ER stress indicators and the expression of tight junction proteins(TJP),inflammatory and apoptosis-related genes and the modulatory role of 1,25-dihydroxy-vitamin D_(3)(1,25D_(3))on these parameters in 2D and 3D cultures of IPEC-J2 cells.Results A published protocol for 3D culture of Caco-2 cells was successfully adopted to IPEC-J2 cells as evident from fully differentiated 3D IPEC-J2 spheroids showing the characteristic spherical architecture with a single layer of IPEC-J2 cells surrounding a central lumen.Treatment of 2D IPEC-J2 cells and 3D IPEC-J2 spheroids with TM for 24 h markedly increased m RNA and/or protein levels of the ER stress target genes,heat shock protein family A(Hsp70)member 5(HSPA5)and DNA damage inducible transcript 3(DDIT3),whereas co-treatment with TM and 1,25D_(3) did not mitigate TM-induced ER stress in IPEC-J2 cells in the 2D and the 3D cell culture.In contrast,TM-induced expression of pro-inflammatory[interleukin-6(IL6),IL8]and pro-apoptotic genes[BCL2 associated X,apoptosis regulator(BAX),caspase 3(CASP3),CASP8]and genes encoding TJP[TJP1,claudin 1(CLDN1),CLDN3,occludin(OCLN),cadherin 1(CDH1),junctional adhesion molecule 1(JAM1)]was reduced by co-treatment with TM and 1,25D_(3) in 3D IPEC-J2 spheroids but not in the 2D cell culture.Conclusions The effect of 1,25D_(3) in the IPEC-J2 cell culture is dependent on the culture model applied.While 1,25D_(3) does not inhibit TM-induced expression of genes involved in inflammation,apoptosis and TJP in conventional 2D cultures of IPEC-J2 cells,TM-induced expression of these genes is abrogated by 1,25D_(3) in the more meaningful 3D IPEC-J2 cell culture model.展开更多

Then the mycoplasma contamination of 25 cell samples was defected by PCR and DNA fluorescence staining. EResultl When these cell samples were detected by DNA fluorescence staining, the positive rate and probable positive rate were respectively 24% and 16%. And when they were detected by PCR, the positive rate was 36%. [ Condusion] The PCR method is more sensitive and specific than the DNA fluorescence staining, and combining these two methods is the optimal way to detect mycoplasma contamination in cell cultures.展开更多

The callus was then transferred aseptically to liquid MSmedium exoge-nously supplemented with appropriate concentration of 6-BA, NAA and 2,4-D to establishsuspension cell culture system. Resibufogenin was administered into the well-grown cell cultures andincubated for 4 d. The products dissolved in the liquid phase of the cultures were extracted andpurified by silica gel column chromatography gradiently eluted with petroleum ether and acetonesystem. Results One transformed product was obtained in 40% yield after 4 d incubation, which wasidentified as 3-epi-resibufogenin on the basis of FAB MS, ~1H NMR and ^(13)C NMR spectroscopicanalysis and corresponding data reported in literature. 文摘BACKGROUND Fracture is one of the most pervasive injuries in the musculoskeletal system,and there is a complex interaction between macrophages and adipose tissue-derived stem cells(ADSCs)in fracture healing.However,two-dimensional(2D)coculture of macrophages and ADSCs can not accurately mimic the in vivo cell microenvironment.AIM To establish both 2D and 3D osteogenic coculture models to investigate the interaction between macrophages and ADSCs.METHODS After obtaining ADSCs from surgery and inducing differentiation of the THP1 cell line,we established 2D and 3D osteogenic coculture models.To assess the level of osteogenic differentiation,we used alizarin red staining and measured the relative expression levels of osteogenic differentiation markers osteocalcin,Runt-related transcription factor 2,and alkaline phosphatase through polymerase chain reaction.Verification was conducted by analyzing the expression changes of N-cadherin and the activation of the Wnt/β-catenin signaling pathway using western blotting.RESULTS In this study,it was discovered that macrophages in 3D culture inhibited osteogenic differentiation of ADSCs,contrary to the effect in 2D culture.This observation confirmed the significance of intricate intercellular connections in the 3D culture environment.Additionally,the 3D culture group exhibited significantly higher N-cadherin expression and showed reducedβ-catenin and Wnt1 protein levels compared to the 2D culture group.CONCLUSION Macrophages promoted ADSC osteogenic differentiation in 2D culture conditions but inhibited it in 3D culture.The 3D culture environment might inhibit the Wnt/β-catenin signaling pathway by upregulating N-cadherin expression,ultimately hindering the osteogenic differentiation of ADSCs.By investigating the process of osteogenesis in ADSCs,this study provides novel ideas for exploring 3D osteogenesis in ADSCs,fracture repair,and other bone trauma repair.
文摘Background The use of conventional two-dimensional(2D)culture of the porcine intestinal epithelial cell(IEC)line IPEC-J2 in animal nutrition research has the disadvantage that IEC function is studied under unphysiological conditions,which limits the ability of transferring knowledge to the in vivo-situation.Thus,the aim of the present study was to establish a more convincing and meaningful three-dimensional(3D)culture of IPEC-J2 cells,which allows to study cell function in a more tissue-like environment,and to compare the effect of the endoplasmic reticulum(ER)stress inducer tunicamycin(TM)on ER stress indicators and the expression of tight junction proteins(TJP),inflammatory and apoptosis-related genes and the modulatory role of 1,25-dihydroxy-vitamin D_(3)(1,25D_(3))on these parameters in 2D and 3D cultures of IPEC-J2 cells.Results A published protocol for 3D culture of Caco-2 cells was successfully adopted to IPEC-J2 cells as evident from fully differentiated 3D IPEC-J2 spheroids showing the characteristic spherical architecture with a single layer of IPEC-J2 cells surrounding a central lumen.Treatment of 2D IPEC-J2 cells and 3D IPEC-J2 spheroids with TM for 24 h markedly increased m RNA and/or protein levels of the ER stress target genes,heat shock protein family A(Hsp70)member 5(HSPA5)and DNA damage inducible transcript 3(DDIT3),whereas co-treatment with TM and 1,25D_(3) did not mitigate TM-induced ER stress in IPEC-J2 cells in the 2D and the 3D cell culture.In contrast,TM-induced expression of pro-inflammatory[interleukin-6(IL6),IL8]and pro-apoptotic genes[BCL2 associated X,apoptosis regulator(BAX),caspase 3(CASP3),CASP8]and genes encoding TJP[TJP1,claudin 1(CLDN1),CLDN3,occludin(OCLN),cadherin 1(CDH1),junctional adhesion molecule 1(JAM1)]was reduced by co-treatment with TM and 1,25D_(3) in 3D IPEC-J2 spheroids but not in the 2D cell culture.Conclusions The effect of 1,25D_(3) in the IPEC-J2 cell culture is dependent on the culture model applied.While 1,25D_(3) does not inhibit TM-induced expression of genes involved in inflammation,apoptosis and TJP in conventional 2D cultures of IPEC-J2 cells,TM-induced expression of these genes is abrogated by 1,25D_(3) in the more meaningful 3D IPEC-J2 cell culture model.

文摘By studying typical "Top Eight Views" in cities like Xining,Lanzhou and Yinchuan in the upper reaches of the Yellow River,mountain and water landscapes in the study area were analyzed in terms of natural ecology;bridges,canals,ferries,pavilions,mansions,towers,temples,historical sites,production and living landscapes were analyzed in terms of humanistic ecology.On this basis,"Top Eight Views" in the upper reaches of the Yellow River were compared and the following conclusions obtained:mountain landscapes were distributed in the whole region,waterscapes unevenly valued in different cities,bridges,canals and ferries mostly located in Ningxia,temples commonly found in all cities,garden arts introduced into cities,traditional cultures carried forward by historical relics.Then the relationship between "Top Eight Views" and regional eco-cultures was analyzed,and it was proposed that humanistic and cultural connotations of these "Top Eight Views" contributed a lot to beautifying urban environment,satisfying needs of the locals' spiritual life,carrying forward local history and culture,enriching urban cultural connotations,expanding living spaces of local residents and improving integrated functions of cities."Top Eight Views" culture complied with the construction gist of urban ecological cultures,and was of great referential value for the construction of urban ecological cultures in the upper reaches of the Yellow River,and also the healthy,scientific and sustainable development of local cities.

文摘[ Objective ] To develop a rapid efficient method for detecting mycoplasma contamination in cell cultures. [ Method] A pair of primers was designed according to two highly conserved nucleotide sequences of the 16S RNA from six kinds of mycoplasma that commonly contaminated cells. Then the mycoplasma contamination of 25 cell samples was defected by PCR and DNA fluorescence staining. EResultl When these cell samples were detected by DNA fluorescence staining, the positive rate and probable positive rate were respectively 24% and 16%. And when they were detected by PCR, the positive rate was 36%. [ Condusion] The PCR method is more sensitive and specific than the DNA fluorescence staining, and combining these two methods is the optimal way to detect mycoplasma contamination in cell cultures.
文摘By analyzing influence of Confucian and Taoist cultures on vernacular dwellings in Jiaodong area,especially the influence of "the doctrine of the mean" on architectural layouts of local dwellings,influence of benevolence and social institutions on the spatial sequence of local dwellings,it was pointed out that Confucian cultures enhanced the hierarchy of spaces,boundaries between internal and external spaces,symmetric layouts and orderly rhythm of courtyards;and meanwhile,the influence of Taoist cultures on the formation of villages in Jiaodong area was also analyzed,the natural philosophy implied in local dwellings was highlighted,such as "Dealing with the nature with natural laws","Going in a natural way","Governing by doing nothing that goes against the nature" and "Respecting natural properties".Based on field investigation of existing vernacular dwellings,common and plain cultural value,realistic significance and sustainability of these dwellings were explored to provide theoretical and practical foundation for the design of new folk houses.

文摘Aim To modify the structure of resibufogenin by using Ginkgo bilobasuspension. Methods Young leaves of Ginkgo biloba were differentiated into callus in MS medium withonly 2,4-D as plant growth regulator. The callus was then transferred aseptically to liquid MSmedium exoge-nously supplemented with appropriate concentration of 6-BA, NAA and 2,4-D to establishsuspension cell culture system. Resibufogenin was administered into the well-grown cell cultures andincubated for 4 d. The products dissolved in the liquid phase of the cultures were extracted andpurified by silica gel column chromatography gradiently eluted with petroleum ether and acetonesystem. Results One transformed product was obtained in 40% yield after 4 d incubation, which wasidentified as 3-epi-resibufogenin on the basis of FAB MS, ~1H NMR and ^(13)C NMR spectroscopicanalysis and corresponding data reported in literature. Conclusion G. biloba suspension cultures canbe used as an enzyme system to biotransform resibufogenin, an animal-originated bufadienolide, into3-epi-resibufogenin.
