Genomic Characterization of BvMLO Genes in Sugar Beet Focusing on BvMLO2 BvMLO7 Responses to Cercospora beticola and Abiotic Stress

Mildew resistance locus O(MLO)proteins are extensively found in various plant species and are essential for multiple biological functions.The characterization and analysis of MLO genes have been conducted across numerous species.However,the functions and features of MLO genes inside sugar beet remain poorly understood.In the present research,we conducted a comprehensive analysis of the structural features of MLO genes,physicochemical characteristics of proteins,evolutionary connections,and expression profiles in sugar beet.A total of 13 BvMLO genes containing MLO structural domains were detected and renamed based on their locations on chromosomes within the sugar beet genome.According to the classification of AtMLO genes,the evolutionary analysis revealed that these 13 BvMLO genes were classified into three subgroups and unevenly located across four chromosomes.Synteny and collinearity analysis confirmed that gene clusters occurred during the evolution of the BvMLO gene family.Examination of cis-regulatory elements revealed specific stress-induced and hormone-associated components within the regulatory regions of BvMLOs.We also found that the expression levels of BvMLO2 and BvMLO7 cloned from sugar beet plants inoculated by Erysiphe betae(Vanha)were significantly regulated by Cercospora beticola Sacc(C.beticola),which indicated that they might both participate in some disease resistance processes.Moreover,quantitative real-time PCR(qRT-PCR)results confirmed that BvMLO2 and BvMLO7 were involved in plant resistance to various biotic and abiotic stress factors.Overall,this research provides a fundamental basis for upcoming studies on the functions and control mechanisms of BvMLO genes within sugar beet.These research findings help advance the progress of disease-resistant breeding in sugar beet and enhance the effectiveness of its resistance breeding. 展开更多

The gray leaf spot caused by Cercospora zeina has become a serious disease in maize in China.The isolates of C.zeina from Yunnan,Sichuan,Guizhou,Hubei,Chongqing,Gansu,and Shaanxi were collected.From those,127 samples were used for genetic diversity analysis based on inter-simple sequence repeat(ISSR)and 108 samples were used for multi-gene sequence analysis based on five gene fragments.The results indicated that populations of C.zeina were differentiated with a relatively high genetic level and were classified into two major groups and seven subgroups.The intra-population genetic differentiation of C.zeina is the leading cause of population variation in China,and interpopulation genetic similarity is closely related to the colonization time and spread direction.The multi-gene sequence analysis of C.zeina isolates demonstrated that there were nine haplotypes.Genetic diversity and multi-gene sequence revealed that Yunnan population of C.zeina,the earliest colonizing in China,had the highest genetic and haplotype diversity and had experienced an expansion event.With the influence of the southwest monsoon in the Indian Ocean,C.zeina from Yunnan gradually moved to Sichuan,Guizhou,Shaanxi,Gansu,and Chongqing.Meanwhile,C.zeina was transferred directly from the Yunnan into the Hubei Province via seed and then came into Shaanxi,Henan,and Chongqing along with the wind from Hubei. 展开更多

Early Detection of <i>Cercospora</i>Species in Soybean Plants: Immunologic and Molecular Methods

Late-cycle diseases (LCD) cause a significant deterioration in quality and reduce yields in soybean crops. In Argentina, in particular, leaf blight and purple seed stain, caused by the agent Cercospora kikuchii, and frog eye spot, caused by C. sojina, are the prevailing sources of diseases. The early, rapid and accurate detection of these phytopathogens becomes essential, and would contribute to preserving both the environment and the health of humans and animals by preventing the wasteful or improper use of chemicals such as pesticides. In order to detect Cercospora species in soybean plants at an early stage, immunochemical and molecular techniques were developed in this work. Strains from the NITE Biological Resource Center collection (Japan): Cercospora kikuchii NBRC 6711 and Cercospora sojina NBRC 6715 and regional isolates of C. kikuchii were used. To develop Dot-Blot and PCR techniques, experiments with plants undergoing different treatments were carried out: those experimentally inoculated with these fungi, those treated with sterile water and healthy plants as well. Both techniques allowed the detection, at early stages, of Cercospora species involved in two of the most frequent LCD in the country, when the cercosporin concentration produced by the fungus was higher than 3.93 ± 0.39 nmol·cyl-1 ±SD. The sensitivity between both techniques was very different. While Dot-Blot allowed the detection of the disease 4 days after inoculation, PCR detected it after 4 hours, even without visible symptoms of the disease. 展开更多

Cercospora leaf spot is fast turning into a critically important disease in Zimbabwe.The disease is caused by Cercospora coffeicola which significantly reduces productivity and quality of coffee.Disturbingly,optimum sporulation of Cercospora coffeicola in culture remains a limiting factor for microbial analysis and quantitative studies of Cercospora leaf spot.Faced with this challenge,an in-vitro study was conducted at Coffee Research Institute,Manicaland,Zimbabwe to examine growth of Cercospora coffeicola in different nutrient media and to determine the best media for Cercospora coffeicola analysis.Six nutrient media were assessed(corn meal agar,oat meal agar,Czapek Dox agar,malt extract agar,yeast extract agar and potato dextrose agar)for the growth of Cercospora coffeicola.The laboratory-based experiment was duplicated,laid out in a Completely Randomized Design,replicated three times and based on Cercospora coffeicola nutrient inoculation.Data were collected on radial growth,colour and texture of mycelium at 3 and 6 days after inoculation.There were significant differences(p<0.05)in the growth of Cercospora coffeicola in media after 3 and 6 days.Malt extract agar had the greatest radial growth(34 mm and 32 mm)of Cercospora coffeicola for trials 1 and 2 respectively,whilst the least growth was in the oat meal agar(14.2 mm and 15.7 mm)for trials 1 and 2 respectively.There were variations in colour and texture of mycelium with malt extract agar,potato dextrose agar and oat meal agar associated with darker colours and rough texture while smooth white mycelia were found in corn meal agar.After considering all nutrient media,malt extract agar was found to be the best media for the growth of Cercospora coffeicola in-vitro.On the basis of our findings,the authors recommend the use of malt extract agar as the primary media for identification and characterisation of Cercospora coffeicola. 展开更多

Uncovering genes essential in domestication and breeding of sugar beet

The genus Beta encompasses economically important root crops such as sugar and table beet.A Beta diversity set including the wild relative B.vulgaris ssp.maritima was grown in the field,and a large phenotypic diversity was observed.The genomes of 290 accessions were sequenced,and more than 10 million high-quality SNPs were employed to study genetic diversity.A genome-wide association study was performed,and marker-trait associations were found for nine phenotypic traits.The candidate gene within the M locus controlling monogermity on chromosome 4 was previously unknown.The most significant association for monogermity was identified at the end of chromosome 4.Within this region,a non-synonymous mutation within the zinc-finger domain of the WIP2 gene co-segregated with monogermity.This gene plays a regulatory role in AGL8/FUL in Arabidopsis.Intriguingly,commercial hybrids are in a heterozygous state at this position.Thus,the long-sought gene for monogermity was identified in this study.Red and yellow pigmentation due to betalain accumulation in shoots and roots is an important characteristic of table and leaf beets.The strongest associations were found upstream or downstream of two genes encoding Cytochrome P450 and anthocyanin MYB-like transcription factor proteins involved in betalain biosynthesis.Significant associations for Cercospora leaf spot resistance were identified on chromosomes 1,2,7,and 9.The associated regions harbor genes encoding proteins with leucinerich repeats and nucleotide binding sites whose homologs are major constituents of plant-pathogen defense. 展开更多

Resistance to race 7 of C. sojina was found to be due to a single dominant gene through analyzing resistant behavior of the cross NEAU91212 (susceptible to race 7)× NEAU9674 (resistant to all races). 3 RAPD markers linked to the resistant gene Rcsc 7 were identified using the BSA method. 2 fragments of OPS03<sub>620</sub> and OPS03<sub>580</sub> amplified with primer OPS03 co-dominantly segregated in the F<sub>2</sub> individuals. The genetic distance between OPS03<sub>620</sub> and the resistant gene is 8.7 cM. According to the co-dominant marker, the accuracy of predicting the homozygous and heterozygous resistant F<sub>2</sub> individuals were 100% and 97.6%, respectively. 展开更多

叶面喷施亚磷酸钾对甜菜褐斑病抗性及产质量的影响

【目的】研究亚磷酸钾处理能否通过增强感病甜菜的防御反应来降低病害胁迫的不利影响,为褐斑病的防治及病害条件下甜菜的高产、稳产提供理论依据。【方法】以甜菜品种‘KUHN1387’为试验材料,清水处理(CK)和杀菌剂处理(T1)为对照,设置... 【目的】研究亚磷酸钾处理能否通过增强感病甜菜的防御反应来降低病害胁迫的不利影响,为褐斑病的防治及病害条件下甜菜的高产、稳产提供理论依据。【方法】以甜菜品种‘KUHN1387’为试验材料,清水处理(CK)和杀菌剂处理(T1)为对照,设置杀菌剂与亚磷酸钾的混合处理(T2),亚磷酸钾设稀释400、800、1200倍3个浓度处理,以及喷施2次和4次处理。喷药后7~9 d调查田间病情指数,测定甜菜的株高、叶面积指数、过氧化氢含量、过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性,在收获期测定其产量和含糖率。【结果】亚磷酸钾显著提高了甜菜褐斑病的防治指数,在亚磷酸钾的不同处理中,杀菌剂和亚磷酸钾混合处理(T2)效果最优,防效达到了65.45%;亚磷酸钾处理促进了甜菜株高和叶面积的增加,降低了甜菜叶片中过氧化氢含量,提高了CAT和SOD活性,提高了甜菜对褐斑病的抗性,最终提高了甜菜的产量和含糖率,其中亚磷酸钾喷施4次、稀释800倍的处理效果最优,其产量及含糖率较CK分别提高了28.50%和2.19个百分点。【结论】亚磷酸钾处理可以缓解褐斑病对甜菜的危害,降低甜菜褐斑病的病情指数,促进甜菜的生长,提高甜菜的产量和含糖率。研究结果可为甜菜褐斑病防治和甜菜产量提高策略的制定提供参考。 展开更多

【目的】为明确内蒙古兴安盟地区甜菜褐斑病病原菌类型并筛选出防治效果较好的杀菌剂。【方法】将从内蒙古兴安盟扎赉特旗甜菜示范田采集的病样进行分离鉴定,通过离体叶片法和盆栽接种进行致病性测定,并利用菌丝生长速率法测定11种不同... 【目的】为明确内蒙古兴安盟地区甜菜褐斑病病原菌类型并筛选出防治效果较好的杀菌剂。【方法】将从内蒙古兴安盟扎赉特旗甜菜示范田采集的病样进行分离鉴定,通过离体叶片法和盆栽接种进行致病性测定,并利用菌丝生长速率法测定11种不同杀菌剂对该病原菌的毒力作用。【结果】从发病甜菜叶片上分离出的菌株ZHB-1,根据形态学和分子鉴定结果,将其鉴定为甜菜生尾孢菌(Cercospora beticola Sacc.)。供试11种杀菌剂对甜菜褐斑病病原菌菌丝生长均具有一定的抑制作用,其中430 g/L戊唑醇抑菌效果最强,EC_(50)值为0.0001 mg/L;其次是70%甲基硫菌灵、30%苯甲·丙环唑、70%三苯基氢氧化锡和8%氟硅唑,EC_(50)值分别为0.0007 mg/L、0.0008 mg/L、0.0525 mg/L和0.0666 mg/L;抑制效果最差的药剂是80%多菌灵,EC_(50)值为158.4890 mg/L。【结论】430 g/L戊唑醇对甜菜褐斑病有较强的抑制作用,试验结果可为杀菌剂的合理使用及甜菜褐斑病田间防治提供理论基础和数据依据。 展开更多
