目的探讨心梗后心律失常钠通道泛素化的调节机制,深入研究lncRNA-CCRR在心肌梗死后调控心脏电生理重构的机制,对心梗后心律失常的防治具有重要意义。方法应用lncRNA-CCRR转基因鼠,C57BL/6小鼠在体注射lncRNA-CCRR过表达腺相关病毒,感染... 目的探讨心梗后心律失常钠通道泛素化的调节机制,深入研究lncRNA-CCRR在心肌梗死后调控心脏电生理重构的机制,对心梗后心律失常的防治具有重要意义。方法应用lncRNA-CCRR转基因鼠,C57BL/6小鼠在体注射lncRNA-CCRR过表达腺相关病毒,感染4周后结扎冠状动脉左前降支12 h构建小鼠急性心肌梗死模型,程序性电刺激检测心律失常发生率。将lncRNA-CCRR过表达/敲减腺相关病毒、阴性对照转染至原代乳鼠心肌细胞中,缺氧12 h。FISH检测lncRNA-CCRR表达,Western blot和实时荧光定量PCR检测Na_(v)1.5和UBA6蛋白以及Na_(v)1.5的mRNA表达,免疫荧光检测Na_(v)1.5和UBA6表达,RIP检测lncRNA-CCRR与UBA6作用关系,全细胞膜片钳检测细胞CCRR过表达及敲减后I_(Na)电流密度。结果心梗小鼠心肌梗死边缘区lncRNA-CCRR表达降低,心律失常发生率升高,CCRR和Na_(v)1.5 mRNA表达下调,Na_(v)1.5蛋白表达下调,UBA6蛋白表达上调;过表达CCRR可逆转上述变化。转染AAV-CCRR可逆转缺氧后下调的CCRR和Na_(v)1.5 mRNA水平,改善Na_(v)1.5和UBA6蛋白表达,且lncRNA-CCRR与UBA6蛋白具有相互结合作用;转染AAV-CCRR后I_(Na)密度增加;转染AAV-si-CCRR后I_(Na)密度降低。结论心梗后lncRNA-CCRR表达降低;lncRNA-CCRR可通过抑制UBA6,增加Na_(v)1.5蛋白表达和I_(Na)电流密度改善心梗后心律失常。 展开更多

Cardiac conduction regulatory RNA(CCRR)has been documented as an antiarrhythmic lncRNA in our earlier investigation.This study aimed to evaluate the effects of CCRR on SERCA2a and the associated Ca^(2+)homeostasis in myocardial infarction(MI).Overexpression of CCRR via AAV9-mediated delivery not only partially reversed ischemia-induced contractile dysfunction but also alleviated abnormal Ca^(2+)homeostasis and reduced the heightened methylation level of SERCA2a following MI.These effects were also observed in CCRR overexpressing transgenic mice.A conserved sequence domain of CCRR mimicked the protective function observed with the full length.Furthermore,silencing CCRR in healthy mice led to intracellular Ca^(2+)overloading of cardiomyocytes.CCRR increased SERCA2a protein stability by upregulating FTO expression.The direct interaction between CCRR and FTO protein was characterized by RNA-binding protein immunoprecipitation(RIP)analysis and RNA pulldown experiments.Activation of NFATc3 was identified as an upstream mechanism responsible for CCRR downregulation in MI.This study demonstrates that CCRR is a protective lncRNA that acts by maintaining the function of FTO,thereby reducing the m^(6)A RNA methylation level of SERCA2a,ultimately preserving calcium homeostasis for myocardial contractile function in MI.Therefore,CCRR may be considered a promising therapeutic strategy with a beneficial role in cardiac pathology. 展开更多

Model Modification and Application on Carrying Capacity of Relative Resources

Based on the retrospection of researches on carrying capacity, this article reviewed systematically the research progresses on carrying capacity of relative resources (CCRR). Then the viewpoint was put forward that CCRR is not an appropriate method of appraising the regional sustainability, but a sound way to obtain cognition for coordinating spatial location and flow of population and economy. However, as the most popular computing method of CCRR, the Weighting Linear Sum Model is defective in the random of weight choice and the neglect of matching among different resources. Therefore, this article established the Geometric Model on CCRR based on modifying Weighting Linear Sum Model, which can be used to appraise regions where resources are close matching. Employing the Geometric Model, the article empirically analyzed the population and economic CCRR in Hubei Province from 1978 to 2006. The result indicates that the population in Hubei Province is overloading while the economic carrying capacity is abundant compared to the whole country, and the economic insufficiency restricts the population carrying capacity. In the future, Hubei Province will become one of the core developing zones which are characterized by economic conglomeration. 展开更多
