BACKGROUND Simulated microgravity environment can lead to gastrointestinal motility disturbance.The pathogenesis of gastrointestinal motility disorders is closely related to the stem cell factor(SCF)/c-kit signaling p...BACKGROUND Simulated microgravity environment can lead to gastrointestinal motility disturbance.The pathogenesis of gastrointestinal motility disorders is closely related to the stem cell factor(SCF)/c-kit signaling pathway associated with intestinal flora and Cajal stromal cells.Moreover,intestinal flora can also affect the regulation of SCF/c-kit signaling pathway,thus affecting the expression of Cajal stromal cells.Cajal cells are the pacemakers of gastrointestinal motility.AIM To investigate the effects of Bifidobacterium lactis(B.lactis)BLa80 on the intestinal flora of rats in simulated microgravity and on the gastrointestinal motility-related SCF/c-kit pathway.METHODS The internationally recognized tail suspension animal model was used to simulate the microgravity environment,and 30 rats were randomly divided into control group,tail suspension group and drug administration tail suspension group with 10 rats in each group for a total of 28 days.The tail group was given B.lactis BLa80 by intragastric administration,and the other two groups were given water intragastric administration,the concentration of intragastric administration was 0.1 g/mL,and each rat was 1 mL/day.Hematoxylin&eosin staining was used to observe the histopathological changes in each segment of the intestine of each group,and the expression levels of SCF,c-kit,extracellular signal-regulated kinase(ERK)and p-ERK in the gastric antrum of each group were detected by Western blotting and PCR.The fecal flora and mucosal flora of rats in each group were detected by 16S rRNA.RESULTS Simulated microgravity resulted in severe exfoliation of villi of duodenum,jejunum and ileum in rats,marked damage,increased space between villi,loose arrangement,shortened columnar epithelium of colon,less folds,narrower mucosal thickness,reduced goblet cell number and crypts,and significant improvement after probiotic intervention.Simulated microgravity reduced the expressions of SCF and c-kit,and increased the expressions of ERK and P-ERK in the gastric antrum of rats.However,after probiotic intervention,the expressions of SCF and ckit were increased,while the expressions of ERK and P-ERK were decreased,with statistical significance(P<0.05).In addition,simulated microgravity can reduce the operational taxonomic unit(OTU)of the overall intestinal flora of rats,B.lactis BLa80 can increase the OTU of rats,simulated microgravity can reduce the overall richness and diversity of stool flora of rats,increase the abundance of firmicutes in stool flora of rats,and reduce the abundance of Bacteroides in stool flora of rats,most of which are mainly beneficial bacteria.Simulated microgravity can increase the overall richness and diversity of mucosal flora,increase the abundance of Bacteroides and Desulphurides in the rat mucosal flora,and decrease the abundance of firmicutes,most of which are proteobacteria.After probiotics intervention,the overall Bacteroidetes trend in simulated microgravity rats was increased.CONCLUSION B.lactis BLa80 can ameliorate intestinal mucosal injury,regulate intestinal flora,inhibit ERK expression,and activate the SCF/c-kit signaling pathway,which may have a facilitating effect on gastrointestinal motility in simulated microgravity rats.展开更多

In this paper, the central role of the stem cell factor (SCF)/c-kit signaling pathway in the pathogenesis of FD was systematically elucidated, and it was proposed that the Chai Gui Hewei Decoction repaired the ICC network through a three-fold mechanism: 1) Bupleurum saponin activated the c-kit receptor to promote the proliferation of ICC;2) Cinnamomum cassiae ingredient inhibited the tumor necrosis factor-α (TNF-α) to improve the inflammation microenvironment;3) Paeonia lactiflora glucoside regulates 5-hydroxytryptamine (5-HT) signaling to restore the balance of brain-gut axis. Based on the theory of “liver depression and spleen deficiency-ICC dysfunction”, a new hypothesis of multi-target repair of gastrointestinal pacing network by traditional Chinese herbal medicine compound was constructed, which provides a theoretical basis for the combination of Chinese and Western medicine.展开更多

BACKGROUND Hypertension is a chronic cardiovascular disease characterized by persistently elevated arterial blood pressure.It is not only a significant risk factor for cardio-vascular and cerebrovascular diseases(such...BACKGROUND Hypertension is a chronic cardiovascular disease characterized by persistently elevated arterial blood pressure.It is not only a significant risk factor for cardio-vascular and cerebrovascular diseases(such as myocardial infarction and stroke)but also closely related to multiple organ damages(such as kidney disease and retinopathy),imposing a heavy health and economic burden on individuals and society.AIM To investigate the expression differences and relationships of endothelin-1(ET-1),interleukin-6(IL-6),stem cell factor(SCF),and its receptor(c-kit)in hypertensive patients with or without depression.METHODS A retrospective analysis was conducted on the clinical data of 163 hypertensive patients admitted to our hospital from March 2022 to January 2024.Based on the presence of depression,patients were divided into Group A(n=77,with depre-ssion)and Group B(n=86,without depression).Serum levels of ET-1 and IL-6 were measured using radioimmunoassay,while serum levels of SCF and c-kit were measured using ELISA.The differences in ET-1,IL-6,SCF,and c-kit levels between Groups A and B were compared.Additionally,the differences in these biomarkers among patients with varying degrees of depression in Group A were analyzed.Pearson correlation analysis was used to examine the relationship between ET-1,IL-6,SCF,c-kit levels,and Hamilton depression rating scale(HAMD)scores.Multivariate logistic regression analysis was performed to identify factors influencing hypertension with depression.The diagnostic efficacy of individual and combined biomarkers was analyzed using receiver operating characteristic(ROC)curves.Comparative statistical analysis of the area under the curve(AUC)values was performed using DeLong’s test to assess the superiority of combined biomarker detection.RESULTS The levels of ET-1 and IL-6 in Group A were significantly higher than those in Group B,while the levels of SCF and c-kit were significantly lower in Group A compared to Group B(P<0.05).In the severe depression subgroup,ET-1 and IL-6 levels were higher than those in the mild-to-moderate depression subgroup,while SCF and c-kit levels were lower(P<0.05).Pearson correlation analysis showed that ET-1 and IL-6 levels were positively correlated with HAMD scores(r=0.442,0.463,P<0.05),while SCF and c-kit levels were negatively correlated with HAMD scores(r=-0.429,-0.394,P<0.05).Multivariate logistic regression analysis revealed that high ET-1,high IL-6,low SCF,and low c-kit were independent influencing factors for hypertension with depression(P<0.05).ROC analysis revealed AUCs of 0.746(ET-1),0.801(IL-6),0.732(SCF),0.779(c-kit),and 0.884(combination).The combined diagnosis demonstrated significantly higher AUC than individual markers(DeLong's test,P<0.01),with superior sensitivity(90.24%)and specificity(85.37%).CONCLUSION Compared to patients with hypertension alone,patients with hypertension and depression exhibited higher serum levels of ET-1 and IL-6 and lower levels of SCF and c-kit.High ET-1,high IL-6,low SCF,and low c-kit were inde-pendent influencing factors for hypertension with depression.The combination of ET-1,IL-6,SCF,and c-kit demonstrated significant diagnostic value for hypertension with depression.展开更多
BACKGROUND For patients with advanced gastrointestinal stromal tumors(GISTs)carrying the ckit exon 11 mutation,imatinib(IM)at a standard dosage of 400 mg per day is the preferred first-line treatment.In cases where tr...BACKGROUND For patients with advanced gastrointestinal stromal tumors(GISTs)carrying the ckit exon 11 mutation,imatinib(IM)at a standard dosage of 400 mg per day is the preferred first-line treatment.In cases where treatment with IM fails,there is an urgent need for a more precise assessment method to determine whether to switch therapies or escalate the IM dosage.This approach will enhance clinical decision-making and optimize patient outcomes.AIM To investigate IM plasma concentration’s role in second-line treatment decisions for c-kit 11-mutated advanced GISTs post-IM failure.METHODS Patients with advanced GIST harboring c-kit 11 mutation who experienced failure with IM 400 mg per day as first-line treatment at our hospital were retrospectively analyzed.Patients were categorized into a low plasma(LP)concentration group(LP group,<1100 ng/mL)and high plasma(HP)concentration group(HP group,≥1100 ng/mL).Each group was further subdivided into Group A(dose-escalation group)and Group B(drug-switch group).Baseline characteristics were compared and Kaplan-Meier curves were used to analyze the survival of patients.RESULTS Seventy-five patients were included in the analysis.For the LP group(n=28),Group A(n=14)had longer overall survival(OS)than Group B(n=14)(P=0.02).No differences were observed between the two subgroups in disease control rate(DCR),objective response rate,and progression-free survival(PFS)(P>0.05).For the HP group(n=47),Group B(n=18)had a higher DCR and longer PFS than Group A(n=29)(P=0.008 and P=0.03,respectively).No difference in OS was observed between the two subgroups(P>0.05).CONCLUSION Increasing IM dosage for c-kit 11-mutated advanced GISTs post-IM failure may prolong OS if plasma concentration is<1100 ng/mL.Switching tyrosine kinase inhibitors may improve DCR and PFS if≥1100 ng/mL.展开更多

文摘BACKGROUND For patients with advanced gastrointestinal stromal tumors(GISTs)carrying the ckit exon 11 mutation,imatinib(IM)at a standard dosage of 400 mg per day is the preferred first-line treatment.In cases where treatment with IM fails,there is an urgent need for a more precise assessment method to determine whether to switch therapies or escalate the IM dosage.This approach will enhance clinical decision-making and optimize patient outcomes.AIM To investigate IM plasma concentration’s role in second-line treatment decisions for c-kit 11-mutated advanced GISTs post-IM failure.METHODS Patients with advanced GIST harboring c-kit 11 mutation who experienced failure with IM 400 mg per day as first-line treatment at our hospital were retrospectively analyzed.Patients were categorized into a low plasma(LP)concentration group(LP group,<1100 ng/mL)and high plasma(HP)concentration group(HP group,≥1100 ng/mL).Each group was further subdivided into Group A(dose-escalation group)and Group B(drug-switch group).Baseline characteristics were compared and Kaplan-Meier curves were used to analyze the survival of patients.RESULTS Seventy-five patients were included in the analysis.For the LP group(n=28),Group A(n=14)had longer overall survival(OS)than Group B(n=14)(P=0.02).No differences were observed between the two subgroups in disease control rate(DCR),objective response rate,and progression-free survival(PFS)(P>0.05).For the HP group(n=47),Group B(n=18)had a higher DCR and longer PFS than Group A(n=29)(P=0.008 and P=0.03,respectively).No difference in OS was observed between the two subgroups(P>0.05).CONCLUSION Increasing IM dosage for c-kit 11-mutated advanced GISTs post-IM failure may prolong OS if plasma concentration is<1100 ng/mL.Switching tyrosine kinase inhibitors may improve DCR and PFS if≥1100 ng/mL.
文摘目的通过分析肾透明细胞癌组织中c-kit的表达情况,并对差异表达的c-kit进行相关生物信息学分析,进一步探讨烟草与c-kit在肾透明细胞癌患者中的潜在作用机制。方法利用癌症基因组图谱(the Cancer Genome Atlas,TCGA)数据库,分析c-kit在肿瘤组织与正常组织中的表达差异,Meta分析验证上述结果。基于UALCAN数据库,分析影响基因表达的临床病理特征。通过Linkedomics进行c-kit共表达基因的基因富集通路及PPI(protein-protein interaction)网络图的获取。Timer分析c-kit表达与肿瘤免疫浸润的关系。免疫组化分析肾透明细胞癌患者中c-kit蛋白的表达情况以及与吸烟史的相关性。结果c-kit在肾透明细胞癌中呈现低表达,且其表达与患者肿瘤分期相关。KIT在肾细胞癌的免疫浸润中发挥着重要的作用。肾透明细胞癌患者中c-kit阳性占10%,但其表达与吸烟无相关性。结论吸烟作为肾癌的重要危险因素,可能通过激活肿瘤组织中c-kit以外的其他信号通路在肾透明细胞癌的发生发展过程中发挥重要作用。
